D. kaki and D. lotus are used as rootstocks for astringent persimmons in Korea but characteristics of their seedlings have not been determined. In this experiment, their seeds were sown in 3-L pots on April 18 and the seedlings were grown until October 24. Growth and nutrient absorption were compared at the end of the season after destructively harvesting the seedlings. Seedling growth of D. lotus was much faster than that of D. kaki in terms of total stem length, stem diameter, and number of leaves. However, chlorophyll value and specific leaf weight were higher in D. kaki than in D. lotus. Dry weight of D. lotus was 3.6- and 3.7-fold higher than that of D. kaki in above-ground parts and the root, respectively. D. kaki seedlings were characterized by higher concentrations of N, P, K, Ca, and Mg in the leaves, stem, or the root. However, total contents of the elements were 1.8- to 3.7-fold higher in a D. lotus seedling due to its greater dry weight. Since D. lotus seedlings absorbed more inorganic elements on a tree basis and grew more vigorously than D. kaki seedlings, the level of fertilization for astringent persimmons should be adjusted depending on rootstocks to maintain the trees at the optimum vigor.
The leaves of Persimmon (Diospyros kaki L.) has long been used for tea in Korea since it was thought to be effective against hypertension. An anticoagulant fraction was purified through gel filtration G-100, hydrophobic, gel filtration G-150, and FPLC, Phenyl superpose column chromatographies. The purified fraction was homogenous and its Mr was estimated 10,000 Da by gel filtration and SDS-PAGE. The purified fraction was sensitive to treatment of subtilisin B, but not to heat and its activity was not changed after periodate oxidation, indicating that the activity was not due to carbohydrates. It delayed thrombin time (TT), activated partial thromboplastin time (APTT), and prothrombin time (PT) using human plasma. TT was more sensitive than APTT and PT, suggesting that the anticoagulant activity may be caused by a degradation or a defect of fibrin or thrombin. It did not cause the hydrolysis of fibrin after incubation. However, it inhibited thrombin-catalyzed fibrin formation with a competitive inhibition pattern. These results indicate that it may be an antithrombotic agent and that it is bound to fibrinogen binding sites of thrombin.
Phytochemical investigation of the methanolic extract of Diospyros kaki leaves led to the isolation of osmanthuside H (1) and a new phenol glycoside, named gamnamoside [4-(3-hydroxypropyl)-2-methoxyphenol $\beta$-D-apiofuranosyl( 1 $\rightarrow$ 6)$\beta$-D-glucopyranoside] (2) along with (-) catechin (3) through a series of reversed phase column chromatography and preparative C18 HPLC. The structures of the isolates were determined by spectroscopic methods including IR, UV, HRTOFMS, and 2D NMR. Compounds 1, 2, and 3, showed good inhibitory activities ($IC_{50}$) of 175.4, 94.4, and 126.6 ${\mu}g/mL$ respectively, whereas a reversible ADH inhibitor, 4-methylpyrazole, showed the $IC_{50}$ of 326.6 ${\mu}g/mL$ against alcohol dehydrogenase (ADH).
Changes in antioxidative substance levels in eleven different cultivars of persimmon leaves during growth were investigated. In general, the contents of soluble phenols, L-ascorbic acid and flavonoids in astringent persimmon leaves(APL) were higher than those of nonastringent persimmon leaves(NAPL). The soluble phenol contents in APL and NAPL showed a tendency to decrease throughout leaf growth. L-ascorbic acid content in APL decreased rapidly during growth, whereas its content in NAPL reached its highest value at the late of July, and then decreased rapidly. Major flavonoids in APL and NAPL were quercetin and Kaempferol which were present in conjugate forms. Before acid hydrolysis, the contents of kaempferol and quercetin in APL and NAPL remained at a relatively constant level until the late of July, and then decreased slightly. After acid hydrolysis, kaempferol contents in APL and NAPL varied significantly by cultivar and growth stage, while quercetin contents decreased slowly until the late of July, and then increased drastically, reached a maximum at the early of August, afterward continuously decreased. These results suggest that APL harvested at the early of June may be useful as potential sources of natural antioxidants.
This study was conducted to develop a preservation technology that can induce changes in physicochemical properties to effectively utilize of persimmon leaves. The application effects of steaming or freezing technique were investigated. Astringent persimmon leaves were steam-blanched (100℃, 30 sec) or frozen (-20℃, 15 d), followed by hot-air drying (50℃). The physicochemical properties of the extract obtained by hot-water leaching from the dried leaves were compared. The extract of leaves dried without pretreatment was used as a control. L* value was higher in steamed than in control and frozen. a* value was highest in the control. The browning index was higher in the frozen and lower in the steamed than in the control. Soluble solids were the highest in the steamed and the lowest in the frozen. Sucrose content was relatively high in the steamed, and the glucose and fructose contents were relatively high in the frozen. Total polyphenol content and DPPH radical scavenging activity were higher in steamed and lower in frozen than in control. Thus, it was confirmed that steam or freeze pretreatment after harvesting persimmon leaves affects the extraction yield, color, antioxidant capacity and component changes of dried persimmon leaves. Unlike steaming, freezing pretreatment showed the effect of promoting decomposition and browning reactions, and it is considered useful when such an effect is needed.

In order to inspect safety and function regarding the reduction of cadmium poisoning by polyphenol compounds prepared from persimmon leaves (Diospyros kaki folium), animal test was done. Rats, group P-1 and P-2, were treated with cadmium and polyphenols of persimmon leaves. A control was just treated with cadmium. GOT activities in P-1 and in P-2 were lower than that of the control without considerable difference, whereas GPT activities decreased in P-1 and P-2. The SOD activity of liver tissue increased in P-l and P-2 compared with that of the control. GST activity was higher than that of the control without considerable difference in P-1; however, it increased in P-2. Lipid peroxide value considerably increased in both P-1 and P-2.
Salinity stress limits plant cultivation in many areas worldwide; however, persimmon (Diospyros spp.) has high tolerance to salt. Five accessions of Diospyros [three of Diospyros lotus (accession numbers 824, 846, and 847); one of Diospyros kaki var. sylvestris (869); and one of Diospyros virginiana (844)] were chosen for analysis of salinity stress. We compared the effects of salt stress on plant growth, relative water content (RWC), malondialdehyde (MDA), electrolyte leakage (EL), hydrogen peroxide content ($H_2O_2$), and antioxidative enzyme activities (superoxide dismutase, SOD; catalase, CAT; peroxidase, POD; and ascorbate peroxidase, APX) in leaves of healthy potted seedlings from each of the five accessions after salt treatment for 25 days. Salt stress affected the growth of plants in all five accessions, with all three D. lotus accessions showing the most severe effect. Salt stress increased membrane lipid peroxidation in all accessions, but a stronger increase was observed in the three D. lotus accessions. Moreover, accumulation of $H_2O_2$ was faster in salt-sensitive D. lotus compared to salt-tolerant D. virginiana 844. The activities of all antioxidant enzymes increased in D. virginiana 844 and in D. kaki var. sylvestris 869; the activities of SOD, CAT, and APX were at similar levels in D. virginiana 844 and D. kaki var. sylvestris 869, but POD activity was stimulated to a greater extent in D. virginiana 844. The activities of all antioxidant enzymes (except POD) decreased in D. lotus 824 and increased (except for SOD) in D.lotus 846. The activities of SOD and APX decreased in D. lotus 847, whereas POD and CAT activities both increased. Relative water content decreased significantly in D. lotus. No significant changes in lipid peroxidation or relevant antioxidant parameters were detected in any of the accessions in controls treated with 0.0% NaCl. D. virginiana 844 had higher antioxidant capacity in response to salinity compared to other persimmon rootstocks. These results indicate that changes of these key physiological variables are related to salinity resistance in different accessions of persimmon.
As a foundational study for notifing excellence of persimmon leaves tea, the chemical component and antioxidant activity were investigated in persimmon leaves from Dungsi, Gabjubaekmok, Weulhasi and Cheongdobansi and green tea leaves. Total sugar contents in all persimmon leaves more higher than that of green tea leaves, and the highest free sugar contained in persimmon and green tea leaves was sucrose. Free sugars present in persimmon and green tea leaves were composed of sucrose, glucose, fructose, maltose and xylose. Sucrose and fructose took more than 70% of total sugar contents. 31∼32 kinds of amino acid were detected in persimmon leaves and 35 kinds in green tea leaves. And total amino acids contained in persimmon leaves were Dungsi, Gabjubaekmok, Weulhasi and Cheongdobansi, respectively 60.40 nmol/${\mu}$L, 53.21 nmol/${\mu}$L, 52.29 nmol/${\mu}$L and 47.58 nmol/${\mu}$L. Total amino acid contents in green tea leaves was the most abundant of all as 114.72 nmol/${\mu}$L. The contents of vitamin C in persimmon and green tea leaves were in the range of 0.015∼0.089% and 0.01%, respectively. Vitamin C was significant higher content in the persimmon leaves than in green tea leaves. Caffeine was not detected in all persimmon leaves, but the caffeine content of green tea leaves was 6.63 mg/l00 g. The content of catechin was showed in the orders of Cheongdobansi, Gabjubaekmok, Weulhasi, Dungsi and green tea leaves; 0.35%, 0.34%, 0.24%, 0.18% and 0.07%, respectively. The contents of gallic acid in Dungsi and Gabjubaekmok were 0.32% and 0.20%. That of green tea was 1.41%, it was the highest content in all samples. The content of calcium in Chengdobansi was most abundant in all samples as 3516.14 ppm, it was 4∼5 times as that of green tea leaves. Flavor component pattern among persimmon leaves was similar, but that of green tea leaves was different. The IC50(${\mu}$g) value of Dungsi, Weulhasi, Gabjubaekmok, Cheongdobansi and green tea were 64.5, 42.0, 47.0, 64.0 and 19.0 respectively.
The efforts were made to develop persimmon leaf beverages on the basis of the results of sensory evaluation. The result of sensory evaluation for the development of persimmon leaf beverages showed that when the concentration of extracts was 10%, the aroma and flavor of persimmon leaves was best. The optimum concentration of sugar was 4.5% and apple juice was 15%. The change of pH had the tendency to keep stable range(3.82-3.71) during the period of 30 days and free-sugar had little change according to the temperature and time of preservation. The preservation of the change of ascorbic acid revealed that the range of decrease in the case of preservation at $4^{\circ}C$ was a bit narrower than in the case of preservation at $25^{\circ}C$. Nonvolatile organic acids were four kinds of acids, oxalic acid, levulinic acid, malic acid, and citric acid, among which levulinic acid was on the increase for the first 10 days during the preservation period and then on the decrease regardless of the temperature of preservation. The browning of beverages was on the slight increase.
As a series of study on the removal of astringency and production of tannin substances, this experiment was conducted to investigate changes of tannin contents and microscopic observation on leaves of persimmon(Diospyros kaki L.). Changes of tannin contents of leaves and fruits in Daegu Bansi(astringent variety) were of similar tendency in content of total and soluble tannin during growth. It seemed therefore that tannin contents of leaves in astringent variety were closely concerned with tannin of fruits. But Fuyu(sweet variety) was different from each other during its growth. Histological form of leaves in microscopic observation had similar tendency between astringent and sweet varieties. It was observed that tannin substances were contained in epidermis cells of levels, that tannin substances were transfered in phloems of nerves system, and that tannin cell existed too in the vicinity of nerves system.
