Biological Control of Rice Bakanae by an Endophytic Bacillus oryzicola YC7007

  • In our previous study, we reported that a novel endophytic bacterium Bacillus oryzicola YC7007 has suppressed bacterial diseases of rice via induced systemic resistance and antibiotic production. This endophytic strain, B. oryzicola YC7007 was used as a biological control agent against bakanae disease of rice caused by Fusarium fujikuroi, and its mechanism of interaction with the pathogen and the rice was further elucidated. Root drenching with B. oryzicola YC7007 suspension reduced the disease severity of bakanae significantly when compared with the untreated controls. The treatments of B. oryzicola YC7007 suspension ($2.0{\times}10^7cfu/ml$) to the rice rhizosphere reduced bakanae severity by 46-78% in pots and nursery box tests containing autoclaved and non-autoclaved soils. Moreover, in the detached rice leaves bioassay, the development of necrotic lesion and mycelial expansion of F. fujikuroi were inhibited significantly by spraying the culture filtrate of B. oryzicola YC7007. Drenching of ethyl acetate extracts of the culture filtrate to the rhizosphere of rice seedlings also reduced the bakanae disease severity in the plant culture dish tests. With the root drenching of B. oryzicola YC7007 suspension, the accumulation of hydrogen peroxide was observed at an early stage of rice seedlings, and a hormonal defense was elicited with and without pathogen inoculation. Our results showed that the strain B. oryzicola YC7007 had a good biocontrol activity against the bakanae disease of rice by direct inhibition, and was also capable of inducing systemic resistance against the pathogen via primed induction of the jasmonic acid pathway.

A Study on the Effect of Alleolopathy of Vitex negundo var. incisa Leaves Extracts (좀목형 엽 추출물의 Allelopathy에 관한 연구)

  • 마편초과의 좀목형 엽에 함유된 물질의 allelopathy 효과를 구명하기 위해 수용성 엽 추출액, 메탄올 엽 추출액, 엽 분말과 토양과의 혼화된 pot에 각각 상치, 이탈이란라이그라스, 개솔새, 억새 종자를 발아실험하여 발아억제물질을 분석한 결과는다음과 같다. 1. 좀목형 엽 분말을 증류수에 각각 24, 48, 72 시간 침지 후 추출물에 상치, 이탈리안라이그라스, 개솔개, 억새 종자를 파종한 후의 발아율은 각 처리구마다 대조구마다 높은 발아억제를 보였고, 48시간 침지시에는 각각 26.9%, 58.8%, 6.6%, 22.2%의 발아율을 보여 강한 발아억제현상르 보였다. 2. 좀목형 엽 분말을 메탄올 75%에 24, 48시간 침지 후 추출물에 이탈리아라이그사스, 개솔새, 억새 종자를 파종한 후의 발아율은 48시간 추출액때 각각 33.3%, 15.4%, 7.7%를 보여 48시간 수용성 추출액과 24시간 메탄올 추출액보다 강한 발아억제효과가 있었다. 3. 좀목형 엽 분말과 Vermiculite를 1: 99.5: 95, 10:90, 20:80(w/w)으로 구분한 pot에 공시수종을 파종한 결과 엽 분말의 비율이 높을수록 발아억제효과가 컸으며, 엽 분말이 20%인 경우 발아된 부분의 건중량의 비율은 실험구와 대조구간에 비교한 발아율의 비율보다 훨씬 낮아 생장억제효과가 있는 것으로 나타났다. 4. Column chromatography의 TLC를 이용하여 최종 7 fraction을 찾고 생물검정 결과, 5, 6번 fraction에 강한 활성을 나타냈다. 5. UV 와 IR분광분석 결과 UV 분광분석은 256 nm과 336nm에서 흡수 최대를 나타내었으며, 이들은 시르시트시약을 사용하여 검토한 결과 7위에 에테르 결합한 전형적인 플라본류로 추정되었고, IR 분광분석 결과 지방족에 에테르결합이 존재하는 것으로 생각되었다. 6. Py-GC-MS 분석으로부터 5, 6 fraction의 경우 플라본류에 1개의 당이 결합한 배당체 구조이고 B환의 경우 1개 또는 2개 이상의 페놀성 수산기의 존재가 예상되었다.

Inhibitory Activity of Lonicera caerulea Against Cell Proliferation in Human Colorectal Cancer Cells (댕댕이나무(Lonicera caerulea)의 대장암세포 생육억제 활성)

  • In this study, we evaluated the effect of the extracts from Lonicera caerulea leaves (LCLE), branches (LCBE) and fruits (LCFE) on the cell growth and migration in human colorectal cancer cells, HCT116 and SW480 cells. LCLE and LCBE dose- and time-dependently inhibited the proliferation of HCT116 and SW480 cells. However, LCFE did not affect the proliferation of HCT116 and SW480 cells. In addition, LCLE and LCBE dramatically cell migration and wound healing in HCT116 cells. LCLE and LCBE decreased β-catenin protein level but not mRNA level in HCT116 and SW480 cells. Furthermore, LCLE decreased TCF4 level in both protein and mRNA level in HCT116 and SW480 cells. However, LCBE decreased TCF4 protein level but not mRNA level in HCT116 and SW480 cells. Based on these findings, LCLE and LCBE may inhibit the cell proliferation and migration through blocking Wnt signaling activation in human colorectal cancer cells. Therefore, LCLE and LCBE may be a potential candidate for the development of chemopreventive or therapeutic agents for human colorectal cancer.

Polyphenol Analysis and Peroxynitrite Scavenging Effect of the Extracts from Eight Korean Mountainous Vegetable (한국 산채류 8종 추출물의 폴리페놀 함량분석 및 Peroxynitrite 소거효과)

  • Mountainous vegetables are usually used in the early stage of plant growth. We attempted to identify the quantity of polyphenols (caffoeoylquinic acids (CQs) and flavonoids) and peroxynitrite scavenging effect ($ONOO^-$) of eight Korean mountainous vegetables. The prominent characteristics were as follows: Only the roots of two plants Taraxacum platycarpum and Ixeris dentata contained chicoric acid. Five CQs were identified in the leaves of Cacalia firma whereas only 3-p-coumaroylquinic acid in the petioles of the plant was shown. The quantities of polyphenols such as quinic acid or tartaric acid derivatives in the mountainous vegetables were generally high, though those of flavonoids were very low. The $IC_{50}$ of chlorogenic acid, chicoric acid and kaempferol were 0.31, 0.12 and $0.25\;{\mu}g/ml$, respectively.

Cholera Toxin B Subunit-Porphyromonas gingivalis Fimbrial Antigen Fusion Protein Production in Transgenic Potato

  • Porphyromonas gingivalis, the gram-negative anaerobic oral bacterium, initiates periodontal disease by binding to saliva-coated oral surface. The cholera toxin B subunit (CTB) genetically linked to FimA1 (1-200 aa) or FimA2 (201-337 aa) of the P. gingivalis fimbrial antigen were introduced into Solanum tuberosum cells by Agrobacterium tumefaciens-mediated transformation method. The integration of CTB-FimA1 or CTB-FimA2 fusion genes were confirmed in the chromosome of transformed leaves by genomic DNA PCR amplification method. Synthesis and assembly of the CTB-FimA fusion proteins into oligomeric structures with pentamer size was detected in transformed tuber extracts by immunoblot analysis. The binding activities of CTB-FimA fusion proteins to intestinal epithelial cell membrane receptors were confirmed by GM1-ganglioside enzyme-linked immunosorbent assay (GM1-ELISA). The ELISA showed that the expression levels of the CTB-FimA1 or CTB-FimA2 fusion proteins were 0.0019, 0.002% of the total soluble protein in transgenic tuber tissues, respectively The synthesis of CTB-FimA monomers and their assembly into biologically active oligomers in transformed potato tuber tissues demonstrates the feasibility of using edible plants for the production of enterocyte targeted fimbrial antigens that could elicit mucosal immune responses.

Antiproliferative Evaluation and Apoptosis Induction in MCF-7 Cells by Ziziphus spina christi Leaf Extracts

  • Background: Herbal medicine has becoming a potential source of treatment for different types of cancer including breast cancer. It has been shown that plants from the family Rhamnaceae possess anticancer activity. Objective: In this study, we determined the antiproliferative influence of Ziziphus spina christi- a species from this family- on the MCF-7 (human breast adenocarcinoma) cell line. Materials and Methods: The cytotoxicity of the total extract, ethanol, ethanol-aqueous (1:1) as well as aqueous fractions of Ziziphus spina christi leaves was evaluated through MTT assay against MCF-7 cell line. Cell cycle inhibition and apoptosis induction were assessed by flowcytometry cycle RNase/PI analysis and Annexin V-FLUOS, respectively. Apoptosis was also analyzed by immunoblotting assay. Results: Our results indicated that the ethanolic fraction had the lowest $IC_{50}$ value (0.02 mg/ml), induced cell cycle arrest at the G1/S phase as well as apoptosis after a 48h of treatment. Conclusions: This is the first report on anticancer effect of Ziziphus spina christi ethanolic fraction on breast cancer cells, providing a scientific basis for its utility in traditional medicine. However, further in-depth studies are needed to confirm the precise mechanisms.

Radical Intermediate Generation and Cell Cycle Arrest by an Aqueous Extract of Thunbergia Laurifolia Linn. in Human Breast Cancer Cells

  • Thunbergia Laurifolia Linn. (TL) is one of the most familiar plants in Thai traditional medicine that is used to treat various conditions, including cancer. However, the antitumor activity of TL or its constituents has never been reported at the molecular level to support the folklore claim. The present study was designed to investigate the antitumor effect of an aqueous extract of TL in human breast cancer cells and the possible mechanism(s) of action. An aqueous crude extract was prepared from dried leaves of TL. Folin-Ciocalteu colorimetric assays were used to determine the total phenolic content. Antiproliferative and cell cycle effects were evaluated in human breast adenocarcinoma MCF-7 cells by MTT reduction assay, cell growth inhibition, clonogenic cell survival, and flow cytometric analysis. Free radical generation by the extracts was detected using electron paramagnetic resonance spectroscopy. The exposure of human breast adenocarcinoma MCF-7 cells to a TL aqueous extract resulted in decreases in cell growth, clonogenic cell survival, and cell viability in a concentration-dependent manner with an $IC_{50}$ value of $843{\mu}g/ml$. Treatments with extract for 24h at $250{\mu}g/ml$ or higher induced cell cycle arrest as indicated by a significant increase of cell population in the G1 phase and a significant decrease in the S phase of the cell cycle. The capability of the aqueous extract to generate radical intermediates was observed at both high pH and near-neutral pH conditions. The findings suggest the antitumor bioactivities of TL against selected breast cancer cells may be due to induction of a G1 cell cycle arrest. Cytotoxicity and cell cycle perturbation that are associated with a high concentration of the extract could be in part explained by the total phenolic contents in the extract and the capacity to generate radical intermediates to modulate cellular proliferative signals.

Antimicrobial Effects of Natural Plant and Mushroom, Dicyophora indusiata Extracts on Fish Pathogenic Bacteria (어류질병세균에 대한 천연식물 및 망태버섯 (Dicyophora indusiata) 추출물의 항균활성)

  • Ethanol extract obtained from bark, roots, stem, leaves and seeds of 30 species of plants and mushroom, Dicyophora indusiata were examined antimicrobial effect on fish pathogenic bacteria, Listoneria anguillamm, Lactococcus garvieae, Streptococcus iniae by disk method. Ethanol extract of D. indusiata was found to be the most active against all fish pathogenic bacteria in test system. Ethyl acetate fraction from ethanol extract of D. indusiata showed the strongest compared with those from other solvent fractions such as dichloromethane, n-buthanol and water. The antimicrobial effect of ethyl acetate fraction was particularly evident against S. iniae.

Animal and Clinical Study of the Efficacy of Mulberry Extract Complex on Degenerative Arthritis (상심자추출복합물의 퇴행성관절염 개선 효능에 대한 동물실험 및 임상연구)

  • Objectives : This study was performed to investigate the efficacy and safety of the Mulberry Extract Complex in a placebo-controlled randomized clinical trial and animal study on degenerative arthritis. Methods : Animal study: Mulberry Extract Complex is composed of extracts of mulberry (Morus alba L.) fruit, mulberry leaves and black beans (Glycine max (L.) Merr.). To evaluate the serum level of interleukin-2, interferon-$\gamma$, and prostaglandin E2, an animal model of degenerative arthritis induced by monosodium iodoacetate was employed. Clinical study: The efficacy index (VAS, K-WOMAC) was compared among patients with symptoms of degenerative arthritis before and after Mulberry Extract Complex ingestion as well as the one in groups. Evaluations of the improvement by the subjects and by doctor assessment were also performed. Results : Animal study: Mulberry Extract Complex reduced the serum level of interferon-$\gamma$ and prostaglandin E2 in an animal model with degenerative arthritis. Clinical study: The VAS change showed statistical significance in the experimental groups after 4 weeks (PP set) and 8 weeks (ITT set) of ingestion. When the K-WOMAC was analyzed using a modified ITT set to determine the effectiveness, statistically significant results were obtained in the fields of pain & symptom within the Mulberry Extract Complex group as well as between the Mulberry Extract Complex and placebo groups after 8 weeks of ingestion. Results from the improvement evaluation by subjects and the assessment of improvement by doctors showed statistical significance in the experimental groups (PP set) after 8 weeks. Conclusions : Mulberry Extract Complex could be useful for the improvement of various symptoms of degenerative arthritis based on its anti-inflammatory activity and its reduction of VAS and K-WOMAC pain scores.

Extraction and Identification of Volatile Isothiocyanates from Wasabi using Supercritical Carbon Dioxide (초임계 이산화탄소를 이용한 고추냉이로부터 휘발성 Isothiocyanates류 추출 및 동정)

  • The aim of this study was to identifyisothiocyanates (ITCs) from wasabi (Wasabi japonica Matsum) using supercritical carbon dioxide ($SCO_2$) and to compare the composition in the extracts between $SCO_2$ and organic solvents extraction. A semi-continuous high pressure apparatus was used to extract wasabi (roots, stems and leaves) at following conditions pressure 80$\sim$120 bar, temperature $40\sim50^{\circ}C$. Ether, ethanol, chloroform and dichloromethane were used as organic solvents. The ITCs extracted by means of both separation technologies were analyzed by a gas chromatography system. As the results of study, AITC and ITCs were highly extracted at 40$^{\circ}C$ and 80 bar. To extract AITC from wasabi, $SCO_2$ extraction is more effective than organic solvents extraction, resulted in thermal degeneration and remaining of organic solvents.
