Effect of Paclobutrazol on Growth, and High Temperature and Drought Stress in Perennial Ryegrass (Paclobutrazol 처리가 Perennial Ryegrass의 생육 및 고온과 건조 Stress에 미치는 영향)

  • This study was conducted to investigate the effect of paclobutrazol [(2 RS , 3 RS )1-(4- chlor-ophenyl )-4, 4- dimethyl -2- (1, 2, 4- triazol -1- yl )- pentan -3-01] on the tolerance of hi-gh temperature and drought stress as related to growth retardation , iranspiration rate , soil water content , nitrogen level and photosynthetic rate in perennial ryegrass ( Loliurn perenne L . ' Omega H , ). Plants were given a 30 ml soil drench of paclohutrazol at the concentrations of 0, 0.01, 0.1, 1.0, 10.. 0, mg / 6 .5cm- diameter pot . The rcsults were as follows : 1. Increasing concentrations of paclohutrazul reduced plant height , leaf area , fresh weight and dry weight , hut increased chlorophyll content per unit area . The number of tillers and leaf width were not affected hy the paclobutrazol concentrations . 2. The proper concentration of paclohatrazol on growth retardation in perennial ryegrass was about I mq /pot , hut leaf deformity and severe growth retardation were shown at high concentration of 10 mq / pot . 3. Perennial ryegrasses grown at 30˚C were shown significantly short plant height and low leaf nitrogen level compared with those grown at 20˚C. Increasing concentrations of paclohutrazol at 20˚C increased nitrogen level hut it could not increase nitrogen level at 30˚C . 4. During the drought stress , increasing temperatures significantly promoted transpiration rate and wilting time . It took about 5 days at 20˚C and 3 days at 30˚C to reach wilting time of leaves from water stress treatment . Soil water contents at wilting time of non-treated controls were averaged 6. 871% at 20˚C and 6. 17% at 30˚C 5. Paclohutrazol reduced transpiration rate at high temperature and drought stress . Wilting appeared at the lower water content of soil according to increasing concentrations of paclobutrazol at 30˚C hut there were no differences among concentrations of at 20˚C. 6.Paclohutrazol treatment at 1 rag /pot reduced injury rate of leaves from 67.1 % and 100 % in control plants to 15.7% and 80% at 20˚C and 3010, respectively. 7. Photosynthetic rate per unit area was significantly reduced at high temperature . Paclohutrazol stimulated photosynthetic rate with increase of concentrations at 20˚C but there was no increasing effect at 30˚C.

Factors affecting the vertical distribution of Betula platyphylla var. japonica and Betula ermanii on Mt. Neko in Nagano Prefecture, Japan

  • Betula platyphylla var. japonica and Betula ermanii segregate vertically at an elevation of approximately 1,850 m on Mt. Neko in Nagano Prefecture, Japan. B. platyphylla var. japonica and B. ermanii were the dominant species below and above this altitude, at which the mean-annual and growing-season air temperatures were $4^{\circ}C$ and $14.1^{\circ}C$, respectively. Based on a modification of Kira's warmth index which employs cumulative temperature represented as $^{\circ}C$ day, leaf unfolding in both species was observed to be initiated at $58^{\circ}C$ day and $169^{\circ}C$ day, respectively. In 1996, leaf unfolding was initiated on 18 May in B. platyphylla var. japonica (+/-6 days) and on 5 June in B. ermanii (+/-8 days), shortly after the last frost which occurred on 5 May 1995 above 1,850 m; below this elevation there was no risk of frost at the time. At elevations above 1,850 m, the unfolded leaves of B. platyphylla were damaged by late frost, while B. ermanii escaped injury because the leaves were still protected by winter buds. The optimum temperature for seed germination in both B. platyphylla and B. ermanii was $30^{\circ}C$. Temperature alternation from 10 to $30^{\circ}C$ and moist storage of seeds at $4^{\circ}C$ (stratification) prior to incubation increased germination rates in both species. The seedlings of B. ermanii had a greater survival rates than those of B. platyphylla var. japonica when planted above 1,850 m. Comparisons of the timing of leaf unfolding and the latest frost at a site appeared to be the main factors affecting the vertical distribution of these species.

Growth, Physiological Responses and Ozone Uptake of Five Betula Species Exposed to Ozone

  • The objectives of this study were to examine the physiological responses to ozone and to measure ozone uptake rates of Betula species exposed to relatively high concentration of pollutants. At the end of the growing season, photosynthesis, pigments contents, antioxidants (SOD and GR) and ozone uptake rates were measured or estimated at the leaves of five Betula species (Betula costata, B. davurica, B. platyphylla var. japonica, B. schmidtii and B. ermanii) exposed to 100ppb ozone concentration. On the termination of the experiment, growth effects were determined by measuring leaf area and dry weights of leaf, stem and root. Ozone treatment showed the significant reduction the leaf area and dry weight of four Betula species, except for B. ermanii. Shoot / root (SR) ratio of five species represented two different types. SR ratio of B. costata and B. davurica were lower than control, in contrast, SR ratio of B. platyphylla var. japonica, B. schimidtii and B. emani, were higher than that of control. The photosynthetic responses of five species were different in responses to ozone exposure. Four species, except for B. emanii, maintained or increased the stomatal conductance, but B. emanii decreased both stomatal conductance and photosynthesis. SOD activities of five species decreased by the ozone exposure, especially B. ermanii showed the largest reduction, GR activities of B. platyphylla var. japonica and B. schmidtii increased, B. costata and B. emanii decreased. Instantaneous ozone uptake rate was the highest at the leaves of B. ermanii and B. costata, ozone uptake per seedling was the highest at the leaf of B. schmidtii and B. emanii. It was concluded that B. costata, B. davurica and B. platyphylla var. japonica, appeared the growth reduction and visible ozone injury, were sensitive species to ozone, and B. schmidtii with the increased antioxidant activity and B. ermanii without the growth reduction were relatively resistant species to high ozone concentration at the early growing stage.

Effects of simulated acid rain on garlic in spraying copper hydroxide solution (쿠퍼 수화제 살포시 마늘에 미치는 인공산성비의 영향)

  • This experiment was conducted to investigate the effects of acid rain in spraying copper hydroxide 77% wettable power(WP) on leaf surface morphology and crop injury of the garlic(Allium sativum L.). Leaf chlorosis, hooked leaves, wrinkled epidermal cells and increased stomata open of garlic were induced by foliar application of simulated acid rain(SAR). It was become to be severe in application of copper hydroxide solution after application of SAR, but those yield of garlic was not significantly different from the untreated control. Leaf chlorosis was sharply increased above 100 ppm of copper ions. Amount of solved copper ions in copper hydroxide solution was remarkably increased below pH 5.5 and the residue in garlic leaves was significantly increased by application of copper hydroxide solution after spraying SAR compared with application of copper hydroxide solution only. When calcium carbonate was applied together with copper hydroxide leaf chlorosis was less than copper hydroxide only.

Effects of Salt Treatment on Yield and Physiological Characteristics of Flag Leaf at Heading Stage in Winter Barley (보리 출수기 염처리가 수량 및 지잎의 생리적 특성에 미치는 영향)

  • This research was conducted to obtain the fundamental data on salt injury and different responses among cultivars in winter barley (Hordeum vulgare L.). Salts did not affect yield components including number of panicles, stem length, grain number per ear and grain yield while reduced stem dry weight and thousand seed weight significantly with increasing concentrations of salt from 60 to 180 mM. NaCl had less injury effect on barley straw dry weight and thousand seed weight than did $MgSO_4$. Chlorophyll content and relative turgidity in flag leaf were reduced when treated with both salts, while free proline in the salt-treated leaf was increased. Content of proline in salt-treated barley was about 10 folds compared to the control. Based on yield components and physiological traits of flag leaf, the tolerance to salt injury was the greatest in Baegdong, followed by Dongbori#1, Mogpo#55, and Gangbori. The results suggested that salt- stressed barley at reproductive stage had higher free proline content, and that special management in this stage must be considered because salt stress at heading stage affect flag leaf growth as well as yield components Harmfully.

Physiological and Biochemical Responses of Fifteen Rice Cultivars to UV-B Radiation

  • This study was conducted to examine the physiological and biochemical responses against UV-B radiation in the seedling of 15 different rice cultivars, having the different physiological sensitivities. Out of 15 rice cultivars tested, moderate and susceptible groups showed significant decreases in biomass and RGR (relative growth rate). Contents of total chlorophyll were reduced remarkedly by irradiation of UV-B. In all rice cultivars tested, the content of chlorophyll a was strongly decreased, while the contents of chlorophyll b were slightly reduced without showing clear different among three groups and 15 cultivars. Carotenoid content was largely reduced by UV-B radiation, whereas polyamine content was moderately increased. The contents of MDA (malondialdehyde) that reflect the level of lipid peroxidation of cell membranes were clearly increased by UV-B stress, showing higher content in susceptible cultivars than moderate and torelant cultivars. The physiological important parameters highly related to visible injury were leaf color, chlorophyll, carotenoid, and lipid peroxidation, whereas biomass and polyamines were not closely correlated. Based on this results, it was concluded that changes of visible injury and the contents of chlorophyll and MDA could be adequately applied and utilized as physiological indicators to UV-B radiation.

Agronomic Characteristics as Affected by Polyethlene Film Mulching and Sowing Date in Vegetable Perilla (잎들깨 멀칭재배 및 파종기에 따른 생육특성)

  • In case of a semi-forcing culture for vegetable perilla, the following research results on the effect of black polyethylene mulching, the optimum sowing season, and the seedling stand improvement method were drawn. Soil temperature was higher and the emergence was faster in a black polyethylene mulching culture than in a non-mulching culture. However, the mature was late, the main stem were larger, and the seed yield, as well as the leaf yield, was greater in a mulching culture than in a non-mulching culture. Considering growth and chilling injury, the stable sowing season of vegetable perilla was judged to be the early in January. The covering materials for improvement of the seedling stand can be a non-woven fabric and hyaline polyethylene. However, the non-woven fabric seemed to be quite suitable in view of the stability and convenience of control after the emergence of perilla.

Studies on Mechanism and Damage Occurrence for Foliar Spray of NaCl Solutions in Fruit Trees (NaCl 엽면살포에 따른 과수의 피해양상과 기작에 관한 연구)

  • We investigated the mechanism on occurring of briny injury in four fruit species (apple, pear, grape, peach). Briny injury of apple, pear, peach, and grape was induced by the foliar splay of saline solutions. Browning symptom was initiated one hour after the spray of 3% NaCl in apple, pearl and peach trees. while two hour were needed for grape, but the latter developed the symptom faster than the formers. Foliar spray of 3% NaCl resulted in 4-fold increase in ethylene production 24 h after the spray, and the production was returned to early stage rate after 72 h. Ethylene production of control was nearly constant with slight decrease after 4 h. Browning and defoliation response to the spray of 3% NaCl on lower side were not different from the spray on both sides regardless of fruit species, while no injury symptoms occurred by the spiny on upper side. In saline damage for treated date, lateral bud was only germinated in apple at 30 July. A shoot apex was 100% grew in all fruit tree. In result for hormonal content of leaf and shoot apex, control was high t-zeatin content in leaf. However, IAA and ABA content was increasing in high saline concentration.

Low temperature and Salt Tolerances of Native Zoysiagrass (Zoysia spp.) Collected in South Korea (국내 자생 한국잔디류의 내한성 및 내염성 조사)

  • This study was carried out to select salt tolerant zoysiagrass breeding lines. Eighty two native zoysiagrasses collected from S. Korea were used in this study. Saline water were prepared by mixing sea water and tap water. ECw levels of saline water treated ranged from 2 to $3dS{\cdot}m^{-1}$. Zoysiagrass planted in pot by sprigging were soaked into the plastic box containing saline water. Winter injury was investigated under the pot condition. Most of Z. japonica types did not show winter injury. But Z. tenuifolia type, Z. matrella type, and Z. sinica type showed winter injury under the pot condition at Cheonan area. NaCl level in soil was increased from 0% to 0.51% by treatment of saline water. Soil ECe measurement showed upto $170dS{\cdot}m^{-1}$. Z. tenuifolia type (Z5034), Z. matrella type ('Konhee', Z4109, 'Semill'), Z. japonica type (Z1055, Z1040, Z1008, 'Zenith', 'Millock') and medium leaf type zoysiagrass (Z6096, Z6118, Z6021, Z6074) resulted in below 30% leaf firing under the saline condition. This approach might be useful for selecting salt tolerant breeding lines.

Protective effects of four types of Taraxaci Herba water extract on carbon tetrachloride-induced acute liver injuries in mice (사염화탄소 유도성 급성 간 손상 모델에서 포공영(蒲公英) 열수 추출물 4종의 간 보호 효능 연구)

  • Objectives : Present study investigated hepatoprotective effects of four types of Taraxaci Herba water extract (TL-F, Taraxaci Herba leaf originated from foreign country; TR-F, Taraxaci Herba root originated from foreign country; TL-K, Taraxaci Herba leaf originated from Korea; TR-K, Taraxaci Herba root originated from Korea) on carbon tetrachloride (CCl4)-induced acute liver injury. Methods : Mice were administered orally with 200 mg/kg of TL-F, TR-F, TL-K, or TR-K for seven days, and intraperitoneally injected with 0.5 mL/kg of CCl4 1 h after last Taraxaci Herba treatment. Silymarin (100 mg/kg) was used as a positive drug. Body weight gain, relative liver weight, serum biochemistry, histopathological and immunohistochemical analyses, and hepatic antioxidant capacity were determined to investigate the hepatoprotective effect of four extracts. Results : Administration of four types of Taraxaci Herba extract increased body weight gain, and decreased relative liver weight in CCl4-injected mice. As compared to CCl4 group, TL-F and TR-F significantly decreased serum aspartate aminotransferase activity, while four extracts reduced CCl4-induced alanine aminotransferase activity. In addition, TL-F and TR-F significantly decreased the numbers of degenerated hepatocytes, infiltrated inflammatory cells, cleaved caspase-3 positive cells, and cleaved PARP positive cells in hepatic tissues. Moreover, TL-F, TR-F, and TR-K administration reduced the lipid peroxidation and nitrotyrosine, and increased glutathione, superoxide dismutase, and catalase activities in hepatic tissues. There were no statistical differences between TL-F- and silymarin-treated group. Conclusion : Of four extracts tested, present results suggest that TL-F is the most favorable candidate against CCl4-induced acute liver injury through enhancing antioxidant activity.
