Studies on the Interspecific and Intergeneric Hybridization in Herbage Grasses II. Morphological physiological charateristics of Italian ryegrass X tall fescue hybrid (화본과목초의 종.속간 잡종에 관한 연구 II. Italian ryegrass X tall fescue F1 잡종의 형태 및 생리적 특성)

  • Tall fescue(Festuca amndinacea Schreb.) is a productive pasture grass that is widely adapted to the soil and climate of Korea. It is regarded, however, as relatively low in palatability and nutritive value. On the other hand, Italian ryegrass(lo1ium rnultz$om Lam.) is highly nutritious and palatable, but is essentially weak in cold tolerance. Numerous investigators have succeeded in producing hybrids between Lolium spp. and Festuca spp. but no improved varieties have resulted. Therefore, intergeneric hybridization of Italian ryegrass with tall fescue is employed in 1984 to improve the adaptive and cold resistance quality of Italian ryegrass. In the mean time, F1-hybrid plants obtained by crossing Italian ryegrass x tall fescue were observed in open field. The results are summarized as follows: 1. Leaf-size and silica cell hair were intermediate between the types of parents but the shape of auricle tended to show Italian ryegrass and auricle-cilia to show tall fescue. 2. Roots of $F_1$-hybrid plants were mainly distributed at top soil and did not produced florescence material. 3. Head type of hybrids was intermediate between the types of parents but ray length was somewhat less than the length of tall fescue ray. Typical Italian ryegrass or tall fescue-like phenotypes were not observed. 4. Anthers of $F_1$-hybrid plants were formed but pollen were not formed. Even though pollen were formed, they were all empty. 5. Hybrid plants were more winterhardy and summer persistency than Italian ryegrass. They showed a equivalent growth to Italian ryegrass in spring and to tall fescue in summer. 6. Chromosome of $F_1$-hybrids was allopolyploid(2n=4~=28) and all $F_1$-hybrid plants were sterile.

Development of Culture System for Masspropagation and Acclimatization of Tissue Cultured Plantlets (유식물체 증식.순화용 배양시스템 개발)

  • In mass production of seed-potato plantlets, the processes for in vitro propagation and ex vitro acclimatization with a high cost should be improved by a culture system with environmental control using scaled-up culture vessels. The experiment was conducted to design a hydroponic culture system for enhancement of growth and development of seed-potato (Solanum tuberosum) plantlets cultured under photoautotrophic (without sugar in culture medium) conditions with controlled light intensity and ventilation rate. The culture system was consisted of scaled-up culture vessels, ventilation pipes, a multi-cell tray and an environmental control system (ECS) for optimum controlling in temperature, light intensity, ventilation rate, and culture-medium supply. Growth and development of the plantlets was significantly increased under the ECS compared with a conventional culture system (CCS) of photomixotrophic culture (with sugar in culture medium) using small scale vessels. For 21 days, leaf area of the plantlets was expanded more than 2 times, and number of internodes also approximately 4 times greate. under the ECS. In addition, the photoautotrophic growth in sweetpotato (Ipomoea batatas) and chrysanthemum (Chrysanthemum morifolium) plantlets was greater more than 2 times compared with the CCS.

Inhibitory Effects of Campsis grandiflora on HIV-1 reverse Transcriptase, HIV-1 Protease and α-glucosidase (능소화 잎 및 줄기 추출물의 Human Immunodeficiency Virus Type I 억제활성)

  • For the elucidation of action mechanism on anti-HIV of natural resources, the extracts of $Campsis$ $grandiflora$ were tested for their inhibitory effects on HIV-1 replication and its essential enzymes as the reverse transcriptase (RT), protease and ${\alpha}$-glucosidase. In the assay of HIV-1-infected human T-cell line, water extracts of stem inhibited the HIV-1-induced cytopathic effects with IC (inhibitory concentration) of 100 ${\mu}g$/ml. Moreover water extracts (100 ${\mu}g$/ml) of stem showed strong activity of 37.9% on anti-HIV-1 RT using Enzyme Linked Oligonucleotide Sorbent Assay (ELOSA) method. In the HIV-1 protease inhibition assay, methanol extracts of stem and leaf extract showed 33.6% and 31.5% inhibition of the enzyme activity to cleave an oligopeptide resembling one of the cleavage sites in the viral polyprotein which can only be processed by HIV-1 protease, but did not exhibited glucosidase inhibitory activities. From these results, it is suggested that the inhibition of the viral replication $in$ $vitro$ is due to the inhibition of reverse transcriptase by water extracts of stem of $Campsis$ $grandiflora$.

Effect of nitrogen compounds on the chemical composition and biological activity of mainstream smoke (질소 화합물이 담배연기성분 및 안전성에 미치는 영향)

  • The objective of this study was to investigate the effect of nitrogen compounds such as protein on the chemical composition and toxicity of cigarette mainstream smoke. BSA protein was treated into the tobacco leaf of original 2R4F cigarette at 1~4 % level. The studies were performed which included a bacterial mutagenicity assay and a mammalian cell cytotoxicity assay for total particulate matter(TPM), and glutathione(GSH) consumption assay for gas/vapor phase(GVP) and determination of smoke chemical constitute. Cigarettes treated with protein were observed dose-dependent increase in yield of volatiles, semi-volatiles and aromatic amines compared with control cigarette. However, carbonyl compounds such as acrolein was lower than that of control cigarette when calculated on an equal TPM basis. The cytotoxicity of TPM obtained from the protein-added cigarettes was not different from that of control cigarette. However, the mutagenicity of the TPM from protein-treated cigarettes(1~4 %) was up to 10-27 % higher than that of control. On the other hand, toxicity of GVP from protein-treated cigarette(4 %) was significantly decreased compared with control cigarette. An overall assessment of our data suggests that nitrogen compounds such as protein should be important for the chemical composition and biological activity of cigarette mainstream smoke.

Antioxidant Activities and Whitening Effect from Lindera obtusiloba BL. Extract (생강나무 추출물의 항산화 활성과 미백효과)

  • In this study we investigated antioxidant activity of against several free radicals and skin whitening effect of 70% ethanol extract (leaf extracts and branch/stem mixed) of Lindera obtusiloba BL. Antioxidant activity was assessed by DPPH, superoxide radical and hydroxyl radical assays. The Lindera obtusiloba BL. extract had antioxidant activity dose dependently with an ${IC}_{50}$ value of 243.14 and 181.10 ${\mu}g$/ml for DPPH, 165.77 and >1500 ${\mu}g$/ml for non-enzymatic system of superoxide radical assay, 35.47 and >100 ${\mu}g$/ml for enzymatic system of superoxide radical assay, 1.21 mg/ml for hydroxyl radical assay. In addition we tested tyrosinase inhibition activity and melanin contents on B16 melanoma F10. B16 melanoma cell was treated by such sample as 1, 5, 10 and 50 ${\mu}g$/ml for 72 hr and tyrosinase inhibition was tested. Melanogenesis was inhibited to 22% at the dose of 50 ${\mu}g$/ml and tyrosinase was inhibited to 45.2% at the same dose. In conclusion Lindera obtusiloba BL had potent antioxidant activity and inhibitory activity of tyrosinase and melanin formation. It could be developed as the health functional food and functional cosmetic resources.

Chemical Composition and Antimicrobial Activity of Essential Oil Extracted from Eucalyptus citriodora Leaf

  • Eucalyptus oil is a rich source of bioactive compounds with a variety of biological activities and is widely used in traditional medicine. Eucalyptus citriodora is cultivated for the production of essential oils. However, the mode of antibacterial action of essential oils from E. citriodora is not well-known. This study aimed to determine the chemical components, microbial inhibitory effect, and mechanism of action of the essential oil from E. citriodora. The oil was extracted from E. citriodora leaves by hydro-distillation and the chemical components were analyzed using gas chromatography-mass spectrometry. The antibacterial activities of eucalyptus oil against gram-positive bacteria (Bacillus subtilis, Staphylococcus aureus, and Staphylococcus intermedius) and gram-negative bacteria (Escherichia coli and Pseudomonas aeruginosa) were screened by disc diffusion method and quantitative analysis was conducted by the microdilution method. The mechanism of action of the extracted essential oil was observed using SEM and analyzed by SDS-PAGE. The major components of E. citriodora oil were citronellal (60.55 ± 0.07%), followed by dl-isopulegol (10.57 ± 0.02%) and citronellol (9.04 ± 0.03%). The antibacterial screening indicated that E. citriodora oil exhibited prominent activity against all tested strains. The minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) against B. subtilis were 0.5% and 1.0%, respectively. The MIC and MBC concentrations against S. aureus, S. intermedius, E. coli, and P. aeruginosa were 1% and 2%, respectively. As observed by SEM, the antibacterial mechanism of E. citriodora oil involved cell wall damage; SDS-PAGE revealed decrease in protein bands compared to untreated bacteria. Thus, E. citriodora oil showed significant antimicrobial properties and caused cellular damage.

Plant Regeneration from Zygotic Embryo-Derived Callus in Citrus junos SIEB. (유자(Citrus junos SIEB.)의 접합배로부터 캘러스 유도 및 식물체 재분화)

  • Calli were successfully induced from immature embryos of Citrus junos SIEB. cultured on 1/2 MS medium supplemented with 40.4 BA. Plant were regenerated from immature embryo derived callus on MS medium with 5 $\mu$M BA. The calli were morphologically characterized by two types: one was whitish and the other was yellowish. After 16 weeks of culture, shoots and root were formed on calli. Plantlets were transplanted to soil and successfully grown to a whole plant Also, the arrangement of the cells showed many differences according to developmental stages of callus and organogenesis. The small cells were compact in callus cultured for 6 weeks and the extended cells which divided actively appeared in it after 8 weeks of culture. The globular protrusion of compacted cells occurred in callus after 10 weeks of culture, and the neighboring cells were liquefied. Oil sac surrounded by the liquefied cell was observed in the leaf and was formed by rupture of liquefied cells.

Phosphate Fertilizer Influences Growth and Photosynthesis of Pepper Seedlings (인산시비가 고추 플러그묘의 생장과 광합성에 미치는 영향)

  • Seedlings of ‘Nokgwang’ and ‘Kwari’ green pepper were cultured in 32-cell plug trays in TK$_2$ medium to find out optimal concentrations of phosphate. Seedling growth of both ‘Nokgwang’ and ‘Kwari’ cultivars was affected by phosphate strength. Applying P fertilizer to the plug system caused a marked increase in plant height and leaf area at 1.0 phosphate strength. On the other hand, total dry weight increased with increasing phosphate strength. Higher chlorophyll content was observed in ‘Nokgwang’ than ‘Kwari’ in all treatments. Phosphate strength greater than 0.5 gave similar chlorophyll content. Photosynthetic rate was higher for plants fertilized with 1.0 strength for ‘Nokgwang’ and 0.5 strength for ‘Kwari’than the other treatments. Higher concentrations of phosphate reduced photosynthesis in both cultivars. With ‘Nokgwang’increasing concentration of phosphate up to 1.0 strength resulted in increase in stomatal conductance and transpiration rate.

Response of Growth and Photosynthesis of Pepper Seedlings to Potassium Fertilizer (칼리처리가 고추 플러그묘의 생육과 광합성에 미치는 영향)

  • Experiments were conducted to estimate optimal potassium strength in the nutrient solution for ‘Nokgwang’and‘Kwari’green pepper seedlings cultured in small plug-cell trays. Plant height, stem diameter, and leaf area increased with increasing K concentration, with greater effect on ‘Kwari’ than ‘Nokgwang’. Total dry weight increased by increasing potassium strength.‘Kwari’had heavier dry weight than ‘Nokgwang’Chlorophyll content differed by cultivars and potassium strength. The highest chlorophyll content was obtained at 2.0 strength potassium in‘Nokgwang’, but not in‘Kwari’Net photosynthesis was greater in plants fertilized with 1.5 strength potassium for‘Nokgwang’and 2.0 strength potassium for ‘Kwari’than the other treatments.

Two unrecorded species of Najas L. (Hydrocharitaceae) from Korean flora: N. orientalis and N. oguraensis (나자스말속(자라풀과)의 미기록 식물 2종: 동아나자스말, 큰톱니나자스말)

  • We report Najas orientalis Triest & P. Uotila and N. oguraensis Miki as unrecorded species from the Korean flora. N. orientalis is distinct from other Najas species in having rounded leaf sheaths and raised testa in the cell walls, and in that this species occurs in the southern region of Korea (Gimhae, Gangjin). N. oguraensis is close to N. minor, but is distinct from this species by having larger male flowers and 4-celled anthers. This species is distributed in the southern region of Korea (Changwon, Jangheung). We also give a new Korean common name for N. gracillima (A. Braun ex Engelm.) Magnus, which is widely distributed in South Korea. A taxonomic key to the six species of Najas is given.
