Nuclear transplantation technique is known as the most potential and efficient method for producing a large number of genetically identical animals from a single embryo. The technical development of nuclear transplantation in mammals and its application to the production of cloned animals are described. For the efficient and successful production of cloned embryos by nuclear transplantation, the right selection and micromanipulation of recipient eggs or embryos as capacious recipient cytoplasm, the adequate and benefitlal preparation of multiple totipotent embryonic cells as donor nuclei, and also the fusion technique are very critical. Recent studies approaching to these critical points are introduced and discussed. Up to date, the overall efficiency of production of cloned embryos and offspring in livestock is estimated to be low. Further technical development of nuclear transplantation will enable large-scale production of cloned livestock and in near future the commercial cloning of animals will become a reality.
Satsuma dwarf virus (SDV) seriously damages citrus production by reducing the quality and yield of fruit. To avoid contamination with SDV, mother trees are checked to be SDV-free in advance of nursery tree distribution. In this study, we compared an immunochromatographic assay (ICA) kit with double-antibody sandwich enzyme-linked immunosorbent assay (DAS-ELISA) for large-scale diagnosis of SDV in orchardgrown trees in Shizuoka Prefecture, Japan. The two methods gave conflicting results for 11 of 1,705 samples, all of which were negative by DAS-ELISA but positive by ICA. The samples scored as positive by either DAS-ELISA or ICA were analyzed by reverse transcription polymerase chain reaction and all were confirmed to be positive. These results validate the use of ICA as a screening method for large-scale diagnosis. Strain discrimination revealed that 16 of 22 isolates belonged to SDV, while citrus mosaic virus (CiMV) infection only and co-infection (SDV and CiMV) were in a minority.

The aim of this study was to assess the associations of some potential risk factors and occurrence of calf mortality in large-scale dairy farms. Njoro area of the Rift valley, Kenya was selected due to its potential of large-scale dairy farms, since the time of the Europeans settlers. The study was retrospective and focused on the calves dying from January 1996 through October 1998. Sample of studied population consisted of 105 calves extracted from the farm records. Data was collected using a questionnaire and were grouped into farm-level and animal-level factors. Calf mortality was 15.6% and important risk factors for calf mortality were sex of calf, season of birth, pneumonia disease, age of dam when calf was born and house type for calves. Female calve born during colder wet seasons and born to dams of 2-4.5 years of age were equally at higher risk. Calves raised in movable pens compared to those raised in permanent pens were at higher risk of mortality from pneumonia. Animal level factors were major causes of calf mortality in commercial farms used in this study and therefore details study is needed in these factors in controlling the calf mortality rates.
Hydrogen as an environmentally friendly energy carrier has received special attention to solving uncertainty about the presence of renewable energy and its dependence on time and weather conditions. This material can be prepared from different sources and in various ways. In previous studies, fossil fuels have been used in hydrogen production, but due to several limitations, especially the limitation of the access to this material in the not-too-distant future and the great problem of greenhouse gas emissions during hydrogen production methods. New methods based on renewable and green energy sources as energy drivers of hydrogen production have been considered. In these methods, water or biomass materials are used as the raw material for hydrogen production. In this article, after a brief review of different hydrogen production methods concerning the required raw material, these methods are examined and ranked from different aspects of economic, social, environmental, and energy and exergy analysis sustainability. In the following, the current position of hydrogen production is discussed. Finally, according to the introduced methods, their advantages, and disadvantages, solar electrolysis as a method of hydrogen production on a small scale and hydrogen production by thermochemical method on a large scale are introduced as the preferred methods.
This paper is aims at studying the economics of scale of sweet potato cultivation. Sweet potato is one of the decreasing crop. Its cultivated areas has been decreasing more rapidly than soybean or corn. In this paper 23 farmers were surveyed, including 8 families with less than 1 ha, 7 families with 1-5 ha, and 8 families with more than 5 ha. The results are as follows; First, sweet potato cultivation is good for specialization, considering the high income per acre. Second, there is no good evidence explaining for the economy of scale of sweet potato cultivation, but there is an evidence for the profit of specialization. Large scale farmers have more incomes per acre, lather than more production or less management costs, by means of good sales methods. Third, mechanization of sweet potato cultivation is one of the key factor for successful large scale sweet potato cultivations.
Vesicular stomatitis virus G glycoprotein (VSV-G) has been widely used for pseudotyping retroviral, lentiviral, and artificial viral vectors. The objective of this study was to establish a potential approach for large-scale production of VSV-G. To this end, VSV-G was cloned with an N-terminal His-tag into Pichia pastoris expression vector pPIC3.5K. Three clones ($Mut^s$) containing the VSV-G expression cassette were identified by PCR. All clones proliferated normally in expansion medium, whereas the proliferation was reduced significantly under induction conditions. VSV-G protein was detected in cell lysates by western blot analysis, and the highest expression level was observed at 96 h post induction. VSV-G could also be obtained from the condition medium of yeast protoplasts. Furthermore, VSV-G could be incorporated into Ad293 cells and was able to induce cell fusion, leading to the transfer of cytoplasmic protein. Finally, VSV-G-mediated DNA transfection was assayed by flow cytometry and luciferase measurement. Incubation of VSV-G lysate with the pGL3-control DNA complex increased the luciferase activity in Ad293 and HeLa cells by about 3-fold. Likewise, incubation of VSV-G lysate with the pCMV-DsRed DNA complex improved the transfection efficiency into Ad293 by 10% and into HeLa cells by about 1-fold. In conclusion, these results demonstrate that VSV-G could be produced from P. pastoris with biofunctionalities, demonstrating that large-scale production of the viral glycoprotein is feasible.
For mass production of poly(3-hydroxybutyrate) (PHB), high cell density cultures of Ralstonia eutropha were carried out in 2.5-1 and 60-1 fermentors by two fed-batch culture techniques of nitrogen and phosphate limitation. When the nitrogen limitation technique was employed using both an on-line glucose monitoring and control system, a high concentration level of PHB (121g/l) was obtained in the small-scale fermentor of 2.5 1. However, the PHB concentration obtained in a large-scale fermentor of 60 1 only turned out to be 60g/l. In contrast, when another fed-batch culture technique of the phosphate-limitation employing dissolved oxygen (DO) stat glucose feeding was used, a large amount of PHB was successfully produced in both 60-1 and 2.5-1 fermentors. In a 2.5-1 fermentor, concentrations of PHB and cells obtained in 58 h were 175 and 210 g/l, respectively, which corresponded to the PHB productivity level of 3.02 g/l/h. In a 60-1 fermentor, a final cell concentration of 221 g/l and a PHB concentration of 180 g/l with PHB productivity level of 3.75 g/l/h were obtained in 48h. PHB content and yield from glucose were 81% and 0.38g PHB/g glucose, respectively. These data suggest that the phosphate limitation technique is more effective compared to nitrogen limitation in the mass production of PHB by R. eutropha of a large scale.
Solid oxide fuel cells (SOFCs) have been under development for a variety of power generation applications. Power system sizes considered range from small watt-size units (e.g., 50-W portable devices) to very large multi-megawatt systems (e.g., 500-MW base load power plants). Because of the reversibility of its operation, the SOFC has also been developed to operate under reverse or electrolysis mode for hydrogen production from steam (In this case, the cell is referred to as solid oxide electrolysis cell or SOEC.). Potential applications for the SOEC include on-site and large-scale hydrogen production. One critical requirement for practical uses of these systems is long-term performance stability under specified operating conditions. Intrinsic material properties and operating environments can have significant effects on cell performance stability, thus performance degradation rate. This paper discusses potential applications of the SOFC/SOEC, technological status and current research and development (R&D) direction, and certain aspects of long-term performance degradation in the operation of SOFCs/SOECs for power generation/hydrogen production.

Environment, price, regulation, and other factors influence the price of agricultural products, which is a social signal of product supply and demand. The price of many agricultural products fluctuates greatly due to the asymmetry between production and marketing details. Horticultural goods are particularly price sensitive because they cannot be stored for long periods of time. It is very important and helpful to forecast the price of horticultural products which is crucial in designing a cropping plan. The proposed method guides the farmers in agricultural product production and harvesting plans. Farmers can benefit from long-term forecasting since it helps them plan their planting and harvesting schedules. Customers can also profit from daily average price estimates for the short term. This paper study the time series models such as ARIMA, SARIMA, and neural network models such as BPN, LSTM and are used for wheat cost prediction in India. A large scale available data set is collected and tested. The results shows that since ARIMA and SARIMA models are well suited for small-scale, continuous, and periodic data, the BPN and LSTM provide more accurate and faster results for predicting well weekly and monthly trends of price fluctuation.
Anaerobic digestion (AD) is the most promising method of treating and recycling of different organic wastes, such as OFMSW, household wastes, animal manure, agro-industrial wastes, industrial organic wastes and sewage sludge. During AD, i.e. degradation in the absence of oxygen, organic material is decomposed by anaerobes forming degestates such as an excellent fertilizer and biogas, a mixture of carbon dioxide and methane. AD has been one of the leading technologies that can make a large contribution to producing renewable energy and to reducing $CO_2$ and other GHG emission, it is becoming a key method for both waste treatment and recovery of a renewable fuel and other valuable co-products. A classification of the basic AD technologies for the production of biogas can be made according to the dry matter of biowaste and digestion temperature, which divide the AD process in wet and dry, mesophilic and thermophilic. The biogas produced from AD plant can be utilized as an alternative energy source, for lighting and cooking in case of small-scale, for CHP and vehicle fuel or fuel in industrials in case of large-scale. This paper provides an overview of the status of biogas production and utilization technologies.
