Relationships between Ovulation and Fertilization Rate in Different Species of Pigs

  • The aim of the present study was to investigate the ovulation rate and its relationship to fertilization ability in Landrace, Durock and Crossbred pigs. Gilts were natural mated at a body weight of at least 120 kg under the same hormone treatment. Embryos were surgically collected 1 day after natural mating (Day 0). Embryos derived from in vivo-fertilized oocytes were cultured in medium PZM-3. The ovaries were examined and the pathological findings were recorded. The number of corpus hemorrhagicum was counted, and was assumed to equal the ovulation rate. There was no difference in the number of corpus hemorrhagicum (20.4, 28.8 and 23.2) and ovulation (13.5, 26.8 and 17.2) in the Landrace, Durock and Crossbred pigs. The two pronucleus formation was 76.0, 80.0 and 86.9%. The Day-7 embryos had blastocyst rates of 68.0, 75.0 and 73.9%. There was no difference in the number of total cells and apoptotic cells. In the future, more studies require determining relationships between ovulation and fertilization rate in different species of pigs.

Circular RNA expression profiles in the porcine liver of two distinct phenotype pig breeds

  • Objective: An experiment was conducted to identify and characterize the circular RNA expression and metabolic characteristics in the liver of Jinhua pigs and Landrace pigs. Methods: Three Jinhua pigs and three Landrace pigs respectively at 70-day were slaughtered to collect the liver tissue samples. Immediately after slaughter, blood samples were taken to detect serum biochemical indicators. Total RNA extracted from liver tissue samples were used to prepare the library and then sequence on HiSeq 2500. Bioinformatic methods were employed to analyze sequence data to identify the circRNAs and predict the potential roles of differentially expressed circRNAs between the two breeds. Results: Significant differences in physiological and biochemical traits were observed between growing Jinhua and Landrace pigs. We identified 84,864 circRNA candidates in two breeds and 366 circRNAs were detected as significantly differentially expressed. Their host genes are involved in lipid biosynthetic and metabolic processes according to the gene ontology analysis and associated with metabolic pathways. Conclusion: Our research represents the first description of circRNA profiles in the porcine liver from two divergent phenotype pigs. The predicted miRNA-circRNA interaction provides important basis for miRNA-circRNA relationships in the porcine liver. These data expand the repertories of porcine circRNA and are conducive to understanding the possible molecular mechanisms involved in miRNA and circRNA. Our study provides basic data for further research of the biological functions of circRNAs in the porcine liver.

Detection of Porcine Stress Syndrome from Genomic DNA of Hair Follicle by PCR-RFLP in Breeding Pig (종돈의 모근 Genomic DNA를 이용한 스트레스 증후군 검색)

  • This study was carried out to investigate PSS (Porcine Stress Syndrome) with the PSE (Pale, Soft, Exudative) in 319 different pigs(Yorkshire 150; Landrace 89 and Duroc 80). The PCR-RFLP method was adapted to detect the ryanodine receptor (RYR 1) gene mutation and to estimate the genotype frequency of the RYR1 gene in breeding pig population. The DNA samples were collected from hair follicles of pigs of Yorkshire, Landrace and Duroc. After DNA amplification by PCR, the PCR products were digested by restriction enzyme, Cfo I. Primary PCR products of ryanodine receptor gene were length of 659 bp in hair follicle and their second PCR products were length of 522 bp in hair follicle. The exon region (522 bp) including point mutation ($C \arrow T; Arg \arrow Cys$) in the porcine ryanodine receptor gene, which is a causal mutation for PSS, was digested with Cfo I restriction enzyme. The RYR1 gene was classifed into three genotypes by agarose gel electrophoresis. The normal homozygous (NN) individuals showed two DNA fragments consisted of 439 and 83 bp. The mutant homozygous (nn) individuals showed only one DNA fragment 522 bp. In addition, all three fragments (522, 439 and 83 bp) were showed in heterozygous (Nn) carrier animals. The normal homozygous (NN), heterozygous (Nn) and mutant homozygous (nn) were 98.00, 2.00 and 0.00% in Yorkshire pigs, 87.64, 11.24 and 1.12% in Landrace, 100.00, 0.00 and 0.00% in Duroc, respectively. The gene frequencies of N and n were 0.990 and 0.010 in Yorkshire pigs, 0.933 and 0.067 in Landrace, 1.000 and 0.000 in Duroc, respectively.

Reproductive Performance in Post-weaning Sows Using PG600 and Inseminated with Frozen Semen (동결정액과 PG600의 이용이 이유종빈돈의 번식능력에 미치는 영향)

  • This study was carried out to investigate the fertility and farrowing date in post-weaning sows using PG600 and inseminated with frozen semen. A total of 48 sows of Landrace, Large White and Duroc after 7-week lactation were used at the Chungnam Provincial Animal Breeding Station. The results obtained were summarized as follows: 1. Motility had no significant differences between the breeds, but NAR acrosome was highest in Landrace, followed by Duroc and Large White(p<.01). 2. Interval from weaning to estrus and length of estrus were, respectively, 3.7 days and 52.6hours of sows treated with PG600, and 6.5 days and 53.8 hours for control sows. The average interval from weaning to onset of estrus was significantly(p<.01) shorter by 2.8 days in PG600 treated sows compared to control sows. 3. In Landrace, Duroc and Large White, farrowing rate and number of pigs born alive per litter were 55.0%, 10.0; 43.8%, 8.1; and 16.7%, 3.5, respectively. Average pig weight at birth and survival rate at 56 days had no significant differences between the breeds. 4. Farrowing rate, number of pigs born alive per litter, average pig weight at birth and survival rate at 56 days were, respectivey, 45.8%, 101, 1.56kg and 94.5% for sows treated with PG600, and 37.5%, 7.0, 1.66kg and 93.8% for control sows. Sows treated with PG 600 had an average of 3.1 more pigs at farrowing compared to control sows.

Effects of gender and gonadectomy on growth and plasma cholesterol levels in pigs

  • We conducted two studies to determine the effect of gender, gonadectomy (GDX) on growth and plasma cholesterol levels in pigs. In experiment 1, five sham-operated and five GDX female Landrace pigs (26kg) were allowed to have free access to water and feed up to market weight (approximately 100kg). Body weight and feed consumption were recorded biweekly, and daily body weight gain, daily feed intake and feed efficiency (gain/feed) were calculated during the feeding period. In experiment 2, 10 male (26kg) and 10 female (26kg) Landrace pigs were used; five male and five female pigs were assigned to sham-operated or GDX. Pigs were allowed to have free access to water and a diet without added cholesterol (Table 1) until they were 6 months old (male 104 and female 98kg) and thereafter they were fed a hypercholesterolemic diet (Table 1) containing 0.5% cholesterol and 0.1% cholate for 10 days. GDX of female pigs increased average daily gain (P<0.05), compared with their sham-operated counterparts during the growing-finishing period, but had no effect (P>0.05) on feed efficiency. Plasma cholesterol levels in pigs fed a hypercholesterolemic diet for 10 days were much higher (P<0.05) in females than in males (161 vs 104mg/100mL plasma), and were increased by GDX only in male pigs. HDL-cholesterol/LDL+VLDL-cholesterol ratio appeared to be higher in males than in females, and was not influenced by GDX in either sex. Results suggested that the lower growth rate of female pigs than their male counterparts is attributable to the ovarian activity, and the lower plasma cholesterol level in male than in female pigs fed a hypercholesterolemic diet is due to the testicular activity.

Comparison of pork belly characteristics and weights of primal cuts between gilt and barrow of Landrace × Yorkshire × Duroc pigs measured by AutoFomIII

  • Currently, pigs breed in Korea are LYD (Landrace × Yorkshire × Duroc) crossbred pigs. Pigs used as fresh meat are gilts and barrows. However, the current supply of pork is not satisfying Korean consumers. In addition, the comparison of carcasses between gilts and barrows only studies carcass weight, backfat thickness, or meat quality, and there are very few studies comparing carcass characteristics. The purpose of this study was to compare characteristics of 7 primal cuts of gilts and barrows as measured by AutoFom III. A total of 350,179 pigs were used, including 176,461 gilts and 173,718 barrows. Characteristics of seven primal cuts were measured using AutoFom III. In the case of carcass weight, there was no significant difference in grade 1+. For all other survey items except for grade 2, gilts showed significantly (p < 0.05) higher values. For all grades except for pork belly, amounts of the remaining six primal cuts were higher in gilts (all p < 0.05,). In addition, the ratio of intermuscular fat in the pork belly of barrows showed a higher value than that in the pork belly of gilts (p < 0.05). The amount of pork belly, which is the most popular among consumers in Korea, not only produced more production than gilts in barrows, but also showed a higher value than gilts in barrows for the ratio of intermuscular fat affecting taste. In summary, gilts produced higher yields than barrows in all parts except pork belly. For the production of only pork belly, barrows showed higher production than gilts.

Association between Numerical Variations of Vertebrae and Carcass Traits in Jeju Native Black Pigs, Landrace Pigs, and Crossbred F2 Population (제주재래흑돼지와 Landrace, 교배집단 F2에서 척추의 수적 변이와 도체형질의 상관관계)

  • The number of thoracic and lumbar vertebrae is known to be an unfixed trait among mammals. This study focused on the relationship between numerical variations of cervical (CER), thoracic (THO), and lumbar (LUM) vertebrae and the total number of vertebrae (TNV) and carcass traits in Jejunative black pigs (JBPs), Landrace pigs, and their intercrossed $F_2$ population. There were no numerical variations in CER vertebrae. On the other hand, the numbers of THO and LUM vertebrae and the TNV varied in all three populations. Of the traits investigated in the three populations, only the means${\pm}$SE of the LUM vertebrae did not show statistical significance (p>0.05). The carcass weights (CW), meat color (MC), marbling score (MS), backfat thickness (BFT), carcass length (CLE), THO vertebrae, and TNV all showed statistical significance (p<0.05). The JBP had 14-16 THO vertebrae, 5-6 LUM vertebrae, and 27-29 TNV. The Landrace pigs had 15-16 THO vertebrae, 5-7 LUM vertebrae, and 28-29 TNV. The $F_2$ population had 14-17 THO vertebrae, 5-7 LUM vertebrae, and 27-30 TNV. In the F2 population, increased numbers of THO vertebrae and TNV were associated with a significant increase in the CW, CLE, and BFT (p<0.05). In particular, the increase in the TNV was caused by an increase in the number of THO rather than LUM vertebrae. Although the animals with a greater number of THO and TNV had thicker backfat, they had a longer CLE and a heavier CW. Both these traits are economically more important than the level of backfat when determining the productivity level. These results suggest that genetic selection to increase the number of vertebrae, especially in Landrace pigs, JBPs, and their related populations, may be an excellent strategy for improving productivity.

Effect of Fibre Level and Fibre Source on Gut Morphology and Micro-environment in Local (Mong Cai) and Exotic (Landrace×Yorkshire) Pigs

  • The effect of genotype, fibre level and fibre source on gut morphology, environment and microflora was studied using 18 Mong Cai (MC) and 18 Landrace${\times}$Yorkshire (LY) pigs, aged around 60 d. The diets were based on maize, rice bran, soybean meal, fish meal and soybean oil, and cassava residue (CR) or brewer's grain (BG) as fibrous ingredient sources in the high-fibre diets (HF). A low-fibre diet (LF), containing around 200 g NDF/kg dry matter (DM), was formulated without CR and BG as feed ingredients. The HF diets (HF-CR and HF-BG) were formulated to contain around 270 g NDF/kg DM. The experiment was arranged according to a $2{\times}3$ factorial completely randomized design with six replications, and lasted 30 d. Crypt density in ileum was lowest (p<0.05) and villus height in jejunum and ileum were the greatest (p<0.05) in pigs fed diet HF-BG. Villus width in ileum was greatest in pigs fed diets HF-CR and HF-BG (p<0.05). Lactic acid bacteria (LAB) counts in stomach were greatest (p<0.05) and E. coli counts in ileum and colon were lowest (p<0.05) in pigs fed diet HF-CR. The concentration of total organic acids in ileum, caecum and colon were greatest (p<0.05), and pH in ileum and colon were lowest (p<0.05) in pigs fed diet HF-CR. Crypt density in ileum was lowest, and villus height in ileum and villus width in jejunum and ileum was greatest in LY pigs (p<0.05). LAB counts in stomach and ileum were greatest, and E. coli counts in ileum were lowest in MC pigs (p<0.05). The concentration of total organic acids in ileum, caecum and colon were greatest (p<0.05) and pH lowest (p<0.05) in MC pigs.

Effect of dietary supplementation of tapioca on growth performance and meat quality in pigs

  • This study evaluated whether dietary supplementation of tapioca could alleviate the negative effects of palm kernel meal (PKM) on growth performance and meat quality in growing and finishing pigs. In experiment 1, 120 73-dold crossbred growing pigs [$(Yorkshire{\times}Landrace){\times}Duroc$], with an average body weight of $31.7{\pm}4.5kg$, were used in a 3-week trial. In experiment 2, 120 108-d-old crossbred finishing pigs [$(Yorkshire{\times}Landrace){\times}Duroc$], with an average BW of $52.6{\pm}4.2kg$ were used in a 10-week trial. Treatments were: CON, a corn-soybean meal-based diet; PKM, 8% PKM, and TPKM, 8% PKM and 10% tapioca. No difference was observed in growth performance or meat quality among treatments in growing pigs. In finishing pigs, no difference was observed in growth performance or meat quality among CON and TPKM dietary treatments. Finishing pigs fed PKM decreases in final BW and ADG compared with those fed CON. Meat quality was not affected by dietary treatments. In conclusion, dietary supplementation of tapioca alleviated anti-nutritional effect of PKM on growth performance in growing and finishing pigs. Thus, the PKM with tapioca could be an available alternative energy source to reduce the cost of pig diets.

Nutritional Value Evaluation of Spray-Dried Egg Protein Containing Egg Yolk Antibody (난황항체를 함유한 분무건조전난의 영양적 가치 평가)

  • For the Exp. 1, twelve Duroc $\times$ Yorkshire $\times$ Landrace pigs (6.12 $\pm$ 0.24 kg average initial body weight and 21 d average age) were used in an 8-d metabolic assay to determine the effects of replacing spray-dried plasma protein (SDPP) with spray-dried egg Protein containing egg yolk antibody (SDCEP) on nutrient and amino acids digestibility in weaning Pigs. Experimental animals were fed diets containing SDPP, SDCEP (spray-dried commercial egg protein) and SDAEP. Protein content in the SDPP, SDCEP and SDAEP were 58.20, 45.83 and 41.85%, respectively. Pigs fed the SDPP diet tended to increase the apparent digestibility of dry matter and nitrogen compared to pigs fed the SDAEP diets without significant differences. The ileal digestibility of lysine and methionine for the SDAEP were greater than those for the SDPP, however, there are no significant differences between groups. For Exp.2, 36 Duroc $\times$ Yorkshire $\times$ Landrace pigs (4.11 $\pm$ 0.05 kg average initial body weight and 14 d average age) were used in a 10-day growth assay to determine the effects on growth performance and nutrient digestibility of replacing SDPP with SDAEP in early-weaning pigs. Experimental animals were fed diets containing CON (corn-dried whey-SBM based diet), SDAEP3 (corn-dried whey-SBM based diet + 3% SDAEP), SDAEP6 (corn-dried whey-SBM based diet + 6% SDAEP). The average daily weight gain of the pigs fed the SDAEP3 diet was higher than that for the pigs fed the CON and SDAEP6 diets (p < 0.05). SDAEP3 significantly increased the digestibility of dry matter and nitrogen compared to the CON and SDAEP6 diets (p < 0.05).
