Partial Purification of Bacteriocin Produced by Lactobacillus sp. GM7311 (Lactobacillus sp. GM7311이 생산하는 박테리오신의 부분 정제)

  • The bacteriocin produced by Lactobacillus sp. GM7311 was purified by sequential steps including n-propanol/acetone treatment, CM-cellulose chromatography, and gel filtration on Sephacryl HR-100. The relative activity of bacteriocin increased 493-fold after final purification step with a recovery of 8.3%. Two protein bands of ca. 8,200 and 2,500 were detected by SDSPAGE of bacteriocin purified through CM-cellulose and sephacryl HR-100 chromatography and both of them had bacteriocin activity.

  • The antimicrobial action of bacteriocin produced by lactobacillus sp. GM7311 against three Gram negative bacteria, Proteus mirabilis, Vibrio parahaemolyticus, and Escherichia coli, was investigated, When the bacteriocin was added to the culture at different stages, viable cells of all of the indicator strains tested were decreased, even though the most inhibited indicator cell growth stages were different. Transmission electron microscopic observation of indicator strains treated with bacteriocin revealed cell Iysis, indicating the cell membrane appears to be the primary site of action. The amino acids concentration of indicator strains treated with bacteriocin were diminished and fatty acids compositions were changed as compared with controls.

  • A lactic acid bacteria which showed antimicrobial activity was isolated from dairy products and identified as Lactobacillus sp. according to the morphological, physiological and biochemical properties, which was named Lactobacillus sp. SH 7311. The bacteriocin of Lactobacillus sp. GM 7311 showed a broad range of inhibitory spectrum against some gram positive and negative bacteria. Especially, Proteus mirabilis was highly sensitive to bacteriocin and used as indicator strain for further investigation. The optimal condition for the production of bacteriocin was showed on MRS broth at $37^{\circ}C$ and pM 6.0. Bacteriocin production of this strain cultured under optimal condition was increased late logarithmic phase to early stationary phase. This bacteriocin was fully active at the pH range $2.0\~5.0$, also was stable at $100^{\circ}C$ for 60 min. at pH 5.0, But about $40\%$ of bacteriocin activity was diminished by the treatment of acetone, ethanol, iso-butanol and ethyl ether during 2 hours at $4^{\circ}C$.

  • The bacteriocin produced by Lactobacillus sp. GM7311 showed strong inhibitory activity against the growth of three Gram positive bacteria, Listeria monocytogenes, Bacillus subtilis, and Staphylococcus aureus. When the bacteriocin was added to the culture at different phases, viable cells of all of the tested strains were decreased, although the most inhibited phase was different. Thereby, when the bacteriocin $(100\;Bu/m{\ell})$ was added to exponential and stationary phase of L. monocytogenes, the rapid reduction of viable cell counts occured. And, in the case of B. subtilis, the highest inhibitory effect occured at lag phase and mid-exponential phase by the addition of the bacteriocin under same condition as mentioned above. Also, we can observe the accelerated reduction of survivors counts for the all of the phase except stationary phase in the S. auresus. Transmission electron microscopic observation of L. monocrogenes and B. subtilis treated with bacteriocin revealed apparent Iysis of the cell wall and excretion of the cell contents, indicating bacteriolysis. Also, the amino acids and fatty acids compositions were different from controls. However, the Iysis of cell wall didn't occur in S. aureus, though the cytoplasmic materials were reduced. This result indicates that the bacteriocin inhibits the synthesis of nuclear materials such as DNA, RNA and proteins.

Production of the Bacteriocin from the Tofu-Residue (두부비지를 이용한 박테리오신 생산)

  • Growth and bacteriocin production by Lactobacillus sp. GM7311 in tofu residue treated with two commercial amylases were investigated. The optimal condition of amylase Ⅰ(liquefied enzyme for sauce) and Ⅱ(multienzyme 2,000) for the enzyme reaction was showed at pH 6.0 and 4.0, respectively. The optimal temperature was 40oC both. At the enzyme dosage 4% and 3% and reaction time 1hr, about 2% of reduced sugar needed bacteriocin production was obtained. The enzymatic treatment of tofu residue enhanced bacteriocin production by lactic acid bacteria, particularly in the tofu residues added 2.0% yeast extract. But, we couldn't see the increment of bacteriocin activity in the tofu residues added other nitrogen sources such as proteose peptone No. 3 and lab lemco powder. Also, in the comparision of amylase I and Ⅱ, bacteriocin activity in the tofu residue treated with amylase Ⅰ was better than that of amylase Ⅱ.

Anti-Helicobacter pylori Activity of Pediococcus acidilactici GMB7330 Isolated from Infant Feces (신생아 분변에서 분리한 Pediococcus acidilactici GMB7330의 Helicobacter pylori에 대한 항균활성)

  • In the present study, lactic acid bacterium that has antibacterial activity against Helicobacter pylori was isolated from feces of newborn baby. The selection was based on the ability to inhibit the growth of H. pylori and to withstand harsh environmental conditions such as acidic pH and high bile concentration. By biochemical test and 16S rDNA sequencing, selected strain was turned out to be an Pediococcus acidilactici, therefore designated to P. acidilactici GMB7330. In order to investigate the inhibitory effects of P. acidilactici GMB7330 on the growth of H. pylori, we have tested in vitro studies such as cell viability and urease test. These results showed that antibacterial activity of P. acidilactici GMB7330 significantly decreased the viable cell count and urease activity of H. pylori. Antibacterial activity of P. acidilactici GMB7330 against H. pylori remained after pH adjustment to neutral, and the concentration of lactate produced from P. acidilactici GMB7330 was not enough to inhibit H. pylori. On the basis of the analysis by transmission electron microscope, it demonstrated that addition of P. acidilactici GMB7330 destroyed the cell structure of H. pylori. These results strongly suggested that P. acidilactici GMB7330 produce antibacterial substances to be able to inhibit the growth of H. pylori other than lactic acid.
