병원성 미생물에 의한 감염을 일으키는 주요 독성인자 중 하나인 응집 물질은 자가응집 및 미생물막 형성으로 인체 건강에 부정적인 영향을 미칠 수 있다. 본 연구에서 Lacticaseibacillus rhamnosus LRH020 (DSM25568) 균주의 독성 유전자 분석을 통해 asa1 유전자를 확인하였고, 표현형으로의 발현 여부 확인을 위하여 미생물막 형성능과 자가응집능 활성실험을 진행하였다. 실험결과 LRH020은 양성대조군 Escherichia. faecalis ATCC 19433과 비교하였을 때 유의적으로 미생물막 형성능이 낮으며, 비교균주 Lacticaseibacillus rhamnosus GG (LGG)와 자가응집능에 차이가 없음을 확인하였다. 균주 LRH020은 asa1 유전자는 가지고 있으나, 표현형으로는 상업균주 LGG와 유사함으로 잠재적인 프로바이오틱스로서의 안전성을 확인하였다.

The use of dietary protein products has increased with interests in health promotion, and demand for sports supplements. Among various protein sources, milk protein is one of the most widely employed, given its economic and nutritional advantages. However, recent studies have revealed that milk protein undergoes fecal excretion without complete hydrolysis in the intestines. To increase protein digestibility, heating and drying were implemented; however, these methods reduce protein quality by causing denaturation, aggregation, and chemical modification of amino acids. In the present study, we observed that Lacticaseibacillus rhamnosus IDCC 3201 actively secretes proteases that hydrolyze milk proteins. Furthermore, we showed that co-administration of milk proteins and L. rhamnosus IDCC 3201 increased the digestibility and plasma concentrations of amino acids in a high-protein diet mouse model. Thus, food supplementation of L. rhamnosus IDCC 3201 can be an alternative strategy to increase the digestibility of proteins.

Atopic dermatitis (AD) is a chronic inflammatory disease caused by immune dysregulation. Meanwhile, the supernatant of lactic acid bacteria (SL) was recently reported to have anti-inflammatory effects. In addition, HaCaT keratinocytes stimulated by tumor necrosis factor alpha (TNF-α) and interferon gamma (IFN-γ) are widely used for studying AD-like responses. In this study, we evaluated the anti-inflammatory effects of SL from lactic acid bacteria (LAB) on TNF-α/IFN-γ-induced HaCaT keratinocytes, and then we investigated the strains' probiotic properties. SL was noncytotoxic and regulated chemokines (macrophage-derived chemokine (MDC) and thymus and activation-regulated chemokine (TARC)) and cytokines (interleukin (IL)-4, IL-5, IL-25, and IL-33) in TNF-α/IFN-γ-induced HaCaT keratinocytes. SL from Lacticaseibacillus rhamnosus MG4644, Lacticaseibacillus paracasei MG4693, and Lactococcus lactis MG5474 decreased the phosphorylation of nuclear factor-κB (NF-κB) and mitogen-activated protein kinase (MAPK). Furthermore, the safety of the three strains was demonstrated via hemolysis, bile salt hydrolase (BSH) activity, and toxicity tests, and the stability was confirmed under simulated gastrointestinal conditions. Therefore, L. rhamnosus MG4644, L. paracasei MG4693, and Lc. lactis MG5474 have potential applications in functional food as they are stable and safe for intestinal epithelial cells and could improve atopic inflammation.

Antibiotics are used to control infectious diseases. However, adverse effects of antibiotics, such as devastation of the gut microbiota and enhancement of the inflammatory response, have been reported. Health benefits of fermented milk are established and can be enhanced by the addition of probiotic strains. In this study, we evaluated effects of fermented milk containing Lacticaseibacillus rhamnosus (L. rhamnosus) SNUG50430 in a mouse model with antibiotic treatment. Fermented milk containing 2 × 105 colony-forming units of L. rhamnosus SNUG50430 was administered to six week-old female BALB/c mice for 1 week. Interleukin (IL)-10 levels in colon samples were significantly increased (P < 0.05) compared to water-treated mice, whereas interferon-gamma (IFN-γ) and tumor necrosis factor alpha (TNF-α) were decreased, of mice treated with fermented milk containing L. rhamnosus SNUG50430-antibiotics-treated (FM+LR+Abx-treated) mice. Phylum Firmicutes composition in the gut was restored and the relative abundances of several bacteria, including the genera Coprococcus and Lactobacillus, were increased in FM+LR+Abx-treated mice compared to PBS+Abx-treated mice. Interestingly, abundances of genus Coprococcus and Lactobacillus were positively correlated with IL-5 and IL-10 levels (P < 0.05) in colon samples and negative correlated with IFN-γ and TNF-α levels in serum samples (P < 0.001). Acetate and butyrate were increased in mice with fermented milk and fecal microbiota of FM+LR+Abx-treated mice were highly enriched with butyrate metabolism pathway compared to water-treated mice (P < 0.05). Thus, fermented milk containing L. rhamnosus SNUG50430 was shown to ameliorate adverse health effects caused by antibiotics through modulating immune responses and the gut microbiota.
Growing evidence indicates a crucial role of the gut microbiota in physiological functions. Gut-brain axis imbalance has also been associated with neuropsychiatric and neurodegenerative disorders. Studies have suggested that probiotics regulate the stress response and alleviate mood-related symptoms. In this study, we investigated the effects of the probiotic Lacticaseibacillus rhamnosus IDCC3201 (L3201) on the behavioral response and fecal metabolite content in an unpredictable chronic mild stress (UCMS) mouse model. Our study shows that chronic stress in mice for three weeks resulted in significant changes in behavior, including lower locomotor activity, higher levels of anxiety, and depressive-like symptoms, compared to the control group. Metabolomic analysis demonstrated that disrupted fecal metabolites associated with aminoacyl-tRNA biosynthesis and valine, leucine, and isoleucine biosynthesis by UCMS were restored with the administration of L3201. Oral administration of the L3201 ameliorated the observed changes and improved the behavioral alterations along with fecal metabolites, suggesting that probiotics play a neuroprotective role.
본 연구는 6개월 미만의 4명의 영아 분변에서 생육이 용이한 우점균을 분리하였고, 이들 중 Lactobacillus속의 유용한 균주를 발굴, 이를 이용하여 발효시켜 제조한 미나리 배양물에 대한 기능적 특성을 살펴보았다. 8주의 분리균주들 중 항생제 내성, 용혈성 등의 안전성을 확인한 후, L. rhamnosus L28과 L. reuteri 11-2 등 2주를 항염증 효능 평가를 위해 선발하였다. 선발한 시험균주 단독과 이들로 미나리를 발효시켜 제조한 미나리발효물 등의 시료들을 각각 처리하여 병원성 세균 4종에 대한 생육 억제능을 살펴본 결과, L. rhamnosus L28 균주 단독처리만으로도 양성대조구인 L. rhamnosus LGG균에 비해 B. cereus와 Sta. aureus 등에 대한 높은 항균활성을 나타내었고, 이 균주를 이용하여 발효시킨 미나리배양물(Oj+L28) 역시도 동일한 병원균들에 대해 25.03±0.00-36.14±0.00 mm의 범위로 억제환을 나타내어 높은 유해균 생육 억제능을 보였다. 또한, LPS로 자극시켜 대식세포 RAW cell에서 분비된 NF-kB/AP-1 전사인자, 활성산소(NO) 및 염증성 매개물질(TNF-α와 IL-6)의 활성 또는 생성량 억제 정도를 살펴본 결과, 양성대조구(LGG) 대비 시험균주 L. rhamnosus L28 단독 처리와 이 균으로 제조한 미나리 발효물(Oj+L28) 처리구는 항염증 효능을 나타내는 모든 지표에서 통계적 유의수준(p<0.05)으로 높은 억제 활성을 나타내었다. 특히, 사이토카인 IL-6 항목에서 L. rhamnosus L28 단독 처리의 억제율이 처리량별로 83, 96 및 99%로 L. reuteri 11-2균주보다 높은 억제 효과를 보였으며, Oj+L28 발효물 역시도 59-72%로 Oj 원액 및 Oj+LGG 배양물보다 낮은 생성량을 보였다. 또한 염증성 매개물질인 TNF-α와 IL-6의 생성량 역시도 L. rhamnosus L28 단독 처리 시 통계적 유의수준(p<0.05)으로 가장 낮은 값을 보여, LPS와 같은 항원 제시에 의해 자극된 면역세포의 기능조절에 도움을 줄 것으로 사료된다. 따라서, 본 연구에서 처음으로 발굴한 L. rhamnosus L28 균주와 이 균주로 발효시켜 제조한 미나리발효물의 높은 항염증 효과를 확인하여, 향후 동물시험 수준의 효능검증 및 기전 구명 등 심화연구를 통해 건강기능성 소재로서의 활용성을 높이고자 한다.

Probiotics, specifically Lacticaseibacillus rhamnosus, have garnered attention for their potential health benefits. This study focuses on evaluating the probiotic properties of candidate probiotics L. rhamnosus IDCC 3201 (3201) using the Caenorhabditis elegans surrogate animal model, a well-established in vivo system for studying host-bacteria interactions. The adhesive ability to the host's gastrointestinal tract is a crucial criterion for selecting potential probiotic bacteria. Our findings demonstrated that 3201 exhibits significantly higher adhesive capabilities compared with Escherichia coli OP50 (OP50), a standard laboratory food source for C. elegans and is comparable with the widely recognized probiotic L. rhamnosus GG (LGG). In lifespan assay, 3201 significantly increased the longevity of C. elegans compared with OP50. In addition, preconditioning with 3201 enhanced C. elegans immune response against four different foodborne pathogenic bacteria. To uncover the molecular basis of these effects, transcriptome analysis elucidated that 3201 modulates specific gene expression related to the innate immune response in C. elegans. C-type lectin-related genes and lysozyme-related genes, crucial components of the immune system, showed significant upregulation after feeding 3201 compared with OP50. These results suggested that preconditioning with 3201 may enhance the immune response against pathogens. Metabolome analysis revealed increased levels of fumaric acid and succinic acid, metabolites of the citric acid cycle, in C. elegans fed with 3201 compared with OP50. Furthermore, there was an increase in the levels of lactic acid, a well-known antimicrobial compound. This rise in lactic acid levels may have contributed to the robust defense mechanisms against pathogens. In conclusion, this study demonstrated the probiotic properties of the candidate probiotic L. rhamnosus IDCC 3201 by using multi-omics analysis.
The microbial starter used to produce kefir beverages, kefir grain, contains a microbial exopolysaccharide called kefiran. Kefir grain consisting of water-insoluble polysaccharides, proteins, and fats, which can be applied as a multi-functional biopolymer. The mass of kefir grain can increase in the fermentation process of Kefir, but it is considered very slow. The purpose of this research is to study the impact of ammonium sulfate supplementation and yeast extract on reconstituted skim milk to increase the mass kefir grain and physical properties of kefiran. Results showed that the ammonium sulfate-supplemented substrate increased the mass of kefir grain by 547% in 14 days, with the condition that the substrate must be renewed every 2 days. Refreshing the substrate is considered one of the important factors. Supplementation on substrate did not appear to affect the viability of bacterial and yeast cells. Kefir grain produced from supplemented substrate also yields better thermal stability properties and has more functional groups than without supplementation. Two Lacticaseibacillus rhamnosus (RAL27 and RAL43) and one Limosilactobacillus fermentum (RAL29) were found to produce EPS. The three isolates also showed good skim milk fermentation ability after purification from kefir grain. The kefir grain produced in this study has the potential for wider application. This study also showed that kefir grain can be adjusted in quantity and quality through fermentation substrate engineering.
프로바이오틱스 제품에 대한 수요가 지속적으로 증가하고 있으며, Lactobacillus 균주가 가장 대중적인 프로바이오틱스로 널리 사용되고 있다. 프로바이오틱스는 기준에 적합한 균수의 확보가 중요하며 제조원가나 시간 등을 낮추기 위해 배양법의 개발이 필요하므로 Lactobacillus 생산을 위한 배양 조건이 최적화되었다. 반응표면방법론에 의한 통계적 최적화에서 반응 변수에 영향을 미치는 독립 변수의 최적 조건은 Lactobacillus acidophilus의 경우 22.55 시간(배양시간), 25℃(배양온도), 3.41%(프리바이오틱스 농도); Lactiplantibacillus plantarum의 경우 24시간, 30.86℃, 2.00%; Lacticaseibacillus rhamnosus의 경우 66.67시간, 35℃, 3.41%이었다. Lactobacillus의 최적 배양조건은 예측한 결과와 실제 결과가 밀접하게 일치하는 것을 확인하였다. 이러한 데이터는 수율 높은 Lactobacillus를 생산하는데 중요한 포인트를 제공할 것이다.
