This study was conducted to analyze penicillin G (PEG), streptomycin (STR) and neomycin (NEO) residues in milk of healthy lactating cows. Milk samples were collected from all four quarters of 12 dairy cows 2−7 days after intramammary infusions of an ointment containing PEG, STR and NEO once (n = 4; group I) or twice (n = 4, group II) daily. Ultra-performance liquid chromatography coupled with electrospray tandem mass spectrometry was used to determine the antibiotic residues in the samples. The correlation coefficient ($r^2$) of the calibration curves for all antibiotics was > 0.999 and the limits of detection and quantification were $0.002-0.005{\mu}g/mL$ and $0.007-0.02{\mu}g/mL$, respectively. Recovery rates were ranged from 75.5 to 92.3%. In group I, PEG, STR and NEO residues were detected in milk at 2, 3 and 2 days post-treatment, respectively, which were below the maximum residue limit (MRL). In group II, PEG, STR and NEO residues were detected in milk at 2, 3 and 3 days post-treatment, respectively, which were bellow the MRL. These results suggest that a 3-day for milk withdrawal period after the ointment treatment might be sufficient for reduction of the antibiotic residues below the MRL.
No. Sperm can be sexed with 90% accuracy by flow cytometry/cell sorting. No. The current speed of sexing is about 5,000 live sperm of each sex per second, remarkably fast considering that each sperm is individually sexed. No. Although fast, sperm sexing is not fast enough to use standard numbers of sperm per AI dose. No. With well managed heifers, pregnancy rates with low doses of sexed, frozen sperm are 70-80% of those with unsexed sperm with normal sperm numbers. Pregnancy rates are lower in lactating dairy cows. No. Calves from sexed sperm appear to be normal. No. Sexed, frozen semen from a few bulls currently is available commercially in the United Kingdom, and likely will be available in several other countries in 2002, probably at a premium of US $30-50 per straw. Objective: The study aimed to evaluate sugarcane bagasse as roughage in lactating cow on feed intake, digestibility, ingestive behavior, milk production and composition, and microbial protein synthesis. Methods: Ten Girolando cows at initial body weight of $450{\pm}25.6kg$ and at $143.7{\pm}30.7days$ in milk were assigned in two $5{\times}5$ Latin square designs. Five 21-day experimental periods were adopted ($1^{\circ}$ to 14-day: diets adaptation period; $15^{\circ}$ to 21-day: data collection and sampling period). The diets consisted of four different levels of sugarcane bagasse (45%, 50%, 55%, and 60%) and a control diet, commonly adopted in the region, based on spineless cactus (25% sugarcane bagasse), formulated to meet 12 kg/d milk yield. Results: The dry matter (DM), organic matter (OM), and total digestible nutrients intakes and DM and OM digestibilities observed for 45% and 50% bagasse inclusion were similar to control diet, while that 55% and 60% bagasse inclusion were lower. Cows fed control diet, and bagasse diets of 45%, and 50% levels had the nutritional requirements attended, that guaranteed 12 kg/d of milk yield. The crude protein intake and digestibility of cows fed 45%, 50%, and 55% of bagasse inclusion were similar to control diet. The neutral detergent fiber (NDF) intake and digestibility differ for all bagasse diets related to control diet, while the non-fiber carbohydrates intake and digestibility for cows fed 45% of bagasse were similar for control diet. The intakes and digestibilities of nutrients decreased linearly in function of bagasse inclusion; NDF and indigestible NDF intakes did not vary. The ruminating time, feeding and rumination efficiency, microbial protein synthesis and milk yield decreased linearly with sugarcane bagasse inclusion. Conclusion: Sugarcane bagasse decreases milk production; however, its inclusion level in between 45% to 50% associated to concentrate could replace diets based on spineless cactus for crossbred dairy cow's producing 12 kg/d of milk.

Limiting amino acids (AAs) for milk production in dairy cows fed on a concentrate diet of corn grain and soybean meal was evaluated in this study. Four lactating and multiparous Holstein cows (in third or fourth parities, with an average body weight of 633 ± 49.2 kg), 8 to 9 weeks into their lactation period, were used in a 4 × 4 Latin square design. The experiment comprised four dietary treatments: (1) no intravenous infusion (control); (2) control plus intravenous infusion of an AA mixture of 6 g/d methionine, 19.1 g/d lysine, 13.8 g/d isoleucine, and 15.4 g/d valine (4AA); (3) control plus intravenous infusion of the AA mixture without methionine (no-Met); and (4) control plus intravenous infusion of the AA mixture without lysine (no-Lys). All animals were fed on a controlled diet (1 kg/d alfalfa hay, 10 kg/d silage, 14 kg/d concentrate mixture, ad libitum timothy hay). The AA composition of the diet and blood were determined using an automatic AA analyzer. Milk composition (protein, fat, lactose, urea nitrogen, and somatic cell counts) was determined using a MilkoScan. The results showed that feed intake for milk production did not differ from that of intravenous infusion using a limiting AA mixture. The 4AA treatment numerically had the highest milk yield (32.4 kg/d), although there was no difference when compared with the control (31.2 kg/d), no-Met (31.3 kg/d), and no-Lys (31.7 kg/d) treatments. The concentration of AAs in blood plasma of cows in all treatments, mainly isoleucine and valine, increased significantly compared with that of control. The no-Met treatment increased (p < 0.05) the concentration of lysine in the blood relative to the control and no-Lys treatments, whereas the no-Lys treatment increased (p < 0.05) the concentration of methionine relative to the control and no-Met treatments. In conclusion, milk production increased when feeding 10 g/d methionine to the cows, together with their concentrate diet of corn grain and soybean meal.

Two experiments were conducted to examine the production and quality of cassava hay and its utilization in diets for dairy cows. In experiment I, a $2{\times}2$ Factorial arrangement in a randomized complete block design with 4 replications was carried out to determine the effects of different initial (IC) and subsequent cutting (SC) on yield and composition of cassava plant. The results revealed that cassava could produce from 4 to 7 tonne of DM and 1.2 to 1.6 tonne of CP for the first six months after planting. CP content in cassava plant ranged from 20.8 to 28.5% and was affected by different SC regimes. Condensed tannin in cassava foliage ranged from 4.9 to 5.5%. Initial cutting at 2 months with subsequent cutting at 2 month intervals was the optimal to obtain high dry matter and protein yield. In the second experiment, five crossbred Holstein-Friesian cows in mid lactation with an initial live-weight of 505${\pm}6.1kg$ and average milk yield of 10.78${\pm}1.2kg/d$ were randomly assigned in a $5{\times}5$ Latin square design to study the effects of 2 levels of CH (1 and 2 kg/hd/d) and concentrate (1 to 2 kg of milk and 1 to 3 kg of milk) on milk yield and milk composition. The results showed that cassava hay increased rumen $NH_3-N$ and milk urea nitrogen (MUN) (p<0.05). Cassava hay tended to increase milk production and 4% FCM. Milk protein increased in cows fed cassava hay (p<0.05). Moreover, cassava hay could reduce concentrate levels in dairy rations thus resulting in increased economic returns. Cassava hay can be a good source of forage to reduce concentrate supplementation and improve milk quality.

This study was conducted to evaluate the effects of the particle size (PS) of alfalfa hay (AH) and soybean hull (SH) on milk production of dairy cows and the population of major cellulolytic bacteria in the rumen. Eight lactating Holstein cows, averaging $590{\pm}33\;kg$ BW and $47{\pm}13$ days in milk (DIM), were assigned in a $4{\times}4$ Latin square design to a $2{\times}2$ factorial arrangement of treatments: alfalfa hay particle size (fine vs. coarse) combined with soy hull (zero or substituted as 50% of AH). The cows were fed diets formulated according to NRC (2001). Physically effective factor (pef) and physically effective fiber (peNDF) contents of diets increased by increasing AH particle size and inclusion of SH in the diets (p<0.01). Dry matter intake was not significantly affected by treatments but intake of peNDF was increased marginally by increasing the PS of AH (p = 0.08) and by SH inclusion (p<0.01) in the diets. Milk production was increased by feeding diets containing SH (p = 0.04), but it was not affected by the dietary PS. Milk fat content was increased by increasing AH particle size (p = 0.03) and decreased by SH substitution for a portion of AH (p<0.01). The numbers of total bacteria and cellulolytic species were not affected by PS of AH or by SH. F. succinogenes was the most abundant species in the rumen followed by R. albus and R. flavefaciens (p<0.01). This study showed that SH cannot replace the physically effective fiber in AH having either coarse or fine particle size. In diets containing SH, increasing of diet PS using coarse AH can maintain milk fat content similar to diets without SH. Particle size and peNDF content of diets did not affect the number of total or fibrolytic bacteria in the rumen.
Objective: The objectives of the study were to assess the effect of cacao bean husk as bedding material in free-stall barn on the behavior, productivity, and udder health of dairy cattle, and on the ammonia concentrations in the barn. Methods: Four different stall surfaces (no bedding, cacao bean husk, sawdust, and chopped wheat straw) were each continuously tested for a period of 1 week to determine their effects on nine lactating Holstein cows housed in the free-stall barn with rubber matting. The lying time and the milk yield were measured between d 4 and d 7. Blood samples for plasma cortisol concentration and teat swabs for bacterial counts were obtained prior to morning milking on d 7. The time-averaged gas-phase ammonia concentrations in the barn were measured between d 2 and d 7. Results: The cows spent approximately 2 h more per day lying in the stalls when bedding was available than without bedding. The milk yield increased in the experimental periods when cows had access to bedding materials as compared to the period without bedding. The lying time was positively correlated with the milk yield. Bacterial counts on the teat ends recorded for cows housed on cacao bean husk were significantly lower than those recorded for cows housed without bedding. Ammonia concentration under cacao bean husk bedding decreased by 6%, 15%, and 21% as compared to no bedding, sawdust, and chopped wheat straw, respectively. The cortisol concentration was lowest in the period when cacao bean husk bedding was used. We observed a positive correlation between the ammonia concentrations in the barn and the plasma cortisol concentrations. Conclusion: Cacao bean husk is a potential alternative of conventional bedding material, such as sawdust or chopped wheat straw, with beneficial effects on udder health and ammonia concentrations in the barns.
