Site-directed Mutagenesis of the Evolutionarily Conserved Tyr8 Residue in Rice Phi-class Glutathione S-transferase F3

  • To elucidate the role of the evolutionarily conserved Tyr8 residue in rice Phi-class GSTF3, this amino acid was replaced with alanine and phenylalanine by site-directed mutagenesis, respectively. The replacement of Tyr8 with Ala significantly affected the catalytic activity and the kinetic parameters, whereas the substitutions of Tyr8 with Phe had almost no effect. The Y8A mutant resulted in approximately 90-100% decrease of the specific activity. Moreover, the Y8A mutant resulted approximately in 2-fold increase of $K_m$, approximately 60-80% decrease of $k_{cat}$, and approximately 6.5-fold decrease in $k_{cat}/K_m$. From the pH/log $k_{cat}/K_m$ plot, $pK_a$ values of the GSH in the wild-type enzyme-GSH complex, Y8A-GSH complex and Y8F-GSH complex were estimated to be approximately 6.8, 8.5 and 6.9, respectively. From these results, we suggest that the evolutionarily conserved Tyr8 residue in OsGSTF3 seems to influence the structural stability of the active site of OsGSTF3 rather than directly its catalytic activity.

Immunomodulatory Activity of Ginsan, a Polysaccharide of Panax Ginseng, on Dendritic Cells

  • Ginsan, a Panax ginseng polysaccharide that contains glucopyranoside and fructofuranoside, has immunomodulatory effects. Although several biologic studies of ginsan have been performed, its effects on dendritic cells (DCs), which are antigen-presenting cells of the immune system, have not been studied. We investigated the immunomodulatory effects of ginsan on DCs. Ginsan had little effect on DC viability, even when used at high concentrations. Ginsan markedly increased the levels of production by DCs of IL-12 and TNF-${\alpha}$, as measured by ELISA. To examine the maturation-inducing activity of ginsan, we measured the surface expression levels of the maturation markers MHC class II and CD86 (B7.2) on DCs. It is interesting that ginsan profoundly enhanced the expression of CD86 on DC surfaces, whereas it increased that of MHC class II only marginally. In $^3H$-thymidine incorporation assays, ginsan-treated DCs stimulated significantly higher proliferation of allogeneic $CD4^+$ T lymphocytes than did medium-treated DCs. Taken together, our data demonstrate that ginsan stimulates DCs by inducing maturation. Because DCs are critical antigen-presenting cells in immune responses, this study provides valuable information on the activities of ginsan.

고등학교 물리의 기체 분자 운동론에서 웹 활용 모의실험이 학습에 미치는 효과 (The Effect of Web-Aided Laboratory on Molecular Dynamics of High School Physics Course)

  • 본 연구에서는 고등학교 물리교과의 '기체분자 운동론'단원에 대한 개별 웹 활용 모의실험을 개발 적용하여 과학 수업에 대한 태도, 학습동기, 학업성취도에서 미치는 효과를 조별 전통실험 집단과 비교하여 알아보았다. 농촌지역에 위치한 고등학생(조별 전통실험 집단 1학급, 개별 웹 활용 모의실험 집단 1학급)을 대상으로 기체 분자 운동론 단원에 대하여 실험 처치를 2차시동안 실시하였다. 모의 시험은 기체에 대한 실험에서 눈에 보이지 않는 기체를 시각화하여 눈에 보이는 입자로 표현되었고, 온도 및 압력을 자유롭게 변화시킬 수 있도록 개발되었다. 웹 활용 모의실험 집단은 개발된 모의실험을 이용하여 개별적으로 실험하였고, 전통실험 집단은 교과서에 나오는 실험을 4인 1조로 실시하였다. 사전 검사로 사전 과학 수업에 대한 태도 검사, 사전 학습동기(PALS) 검사, 논리적 사고력 검사를 실시하였다. 실험 처치 후 학생들의 사후 과학 수업에 대한 태도 검사, 사후 학습동기(IMMS) 검사, 학업 성취도 검사를 실시하였고, 자료를 수집하여 공변량 분석을 실시하였다. 분석 결과, 개별 웹 활용 모의실험과 조별 전통실험 집단간의 과학 수업에 대한 태도는 유의미한 차이가 없었다. 또한, 학습동기와 학업 성취도는 개별 웹 활용 모의실험 집단이 조별 전통실험 집단에 비해 유의미하게 높았다.

  • A 50 kW class rotating body type wave energy converter consisted of two floating bodies and a PTO (Power Takeoff) unit is studied. As an wave energy extractor, the body is designed to have a VLCO (Variable Liquid-Column Oscillator) having a liquid filled U-tube with air chambers. Owing to the oscillation of the liquid in the U-tube caused by the air spring effect of the air chambers, the amplitude of response of the VLCO becomes significantly amplified for a target wave period. The PTO converts the rotational moment introduced from the relative motion of the hinged bodies to an hydraulic power by means of a cylinder. A high pressure accumulator, hydraulic motor and a generator are equipped in the PTO to convert the hydraulic power to electric power. A control law for adjusting the oscillation period of the VLCO is proposed for the efficient operation of the VLCO with various wave conditions. It is found that the effect of the air spring has an important role to play in making the oscillation of the VLCO match with the ocean wave. In this way, the wave energy converter equipped with the VLCO provides the most effective mode for extracting energy from the ocean wave.

Human Cytomegalovirus Inhibition of Interferon Signal Transduction

  • Cytomegalovirus (CMV), a beta-herpesvirus with worldwide distribution, exhibits host persistence, a distinguishing characteristic of all herpesviruses. This persistence is dependent upon restricted gene expression in infected cells as well as the ability of productively infected cells to escape from normal cell-mediated anti-viral immunosurveillance. Type I (IFN-α/β) and type II (IFN-γ) interferons are major components of the innate defense system against viral infection. They are potent inducers of MHC class I and II antigens and of antigen processing proteins. Additionally, IFNS mediate direct antiviral effects through induction effector molecules that block viral infection and replications such as 2′, 5-oligoadenylate synthetase (2, 5-OAS). IFNS function through activation of well-defined signal transduction pathways that involve phosphorylation of constituent proteins and ultimate formation of active transcription factors. Recent studies have shown that a number of diverse viruses, including CMV, EBV, HPV mumps and Ebola, are capable of inhibiting IFN-mediated signal transduction through a variety of mechanisms. As an example, CMV infection inhibits the ability of infected cells Is transcribe HLA class I and II antigens as well as the antiviral effector molecules 2, 5-OAS and MxA I. EMSA studies have shown that IFN-α and IFN-γ are unable to induce complete signal transduction in the presence of CMV infection, phenomena that are associated with specific decreases in JAKl and p48. Viral inhibition of IFN signal transduction represents a new mechanistic paradigm for increased viral survival, a paradigm predicting widespread consequences in the case of signal transduction factors common to multiple cytokine pathways.

  • 항공기용 전기식 구동장치는 유압식 구동장치에 비해 소형/경량화에 유리하고, 힌지라인 형태의 구동장치는 공기저항이 작고 스텔스 등의 특수 목적에 적합한 특징을 가지고 있다. 본 논문에서는 10,000 lbf-in급 힌지라인 이중화 전기식 구동장치의 동작성능시험을 위한 시스템 설계 내용을 기술하였다. 영구자석전동기의 자속 기준 제어와 이중화 구동기의 torque fighting을 고려한 제어기를 설계하고, 구동장치 시스템 모델을 수립하여 성능 시뮬레이션을 수행하였다. 성능시험을 수행하여 시뮬레이션 결과와 비교하였으며 목표성능 만족여부를 확인하였다.

  • 본 논문에서는 자심 테슬라 변압기를 이용한 400kV급 소형 고전압 펄스 발생기를 개발하였다. 테슬라 변압기의 효율 및 출력전압 향상을 위해 4분할 적층 원통형 자심 구조를 새롭게 제안 하였다. 제안 된 자심 구조를 적용하여 테슬라 변압기를 제작 한 결과, 최대 출력전압 425kV, 에너지 변환 효율 50%를 확인하였다.

  • The purpose of the present paper is to introduce a new class of almost para-contact metric manifolds namely, Golden para-contact metric manifolds. Then, we are particularly interested in a more special type called Golden para-Sasakian manifolds, where we will study their fundamental properties and we present many examples which justify their study.

  • The aim of this paper is two-fold. First, we study the Chinea-Gonzalez class C12 of almost contact metric manifolds and we discuss some fundamental properties. We show there is a one-to-one correspondence between C12 and Kählerian structures. Secondly, we give some basic results for Riemannian curvature tensor of C12-manifolds and then establish equivalent relations among 𝜑-sectional curvature. Concrete examples are given.

  • In this study, we establish some new characterizations for a class of anisotropic Herz spaces in which all exponents are considered as variables. We also provide a description of these spaces based on bloc decomposition. As an application, we investigate the boundedness of certain sublinear operators within these function spaces.
