Effects of Mugwort Powder Supplementation on Carcass and Meat Characteristics in the Finishing Period of Gilts (인진쑥 첨가가 비육기 암퇘지의 도체 및 육질 특성에 미치는 영향)

  • This study was conducted to determine the effect of mugwort powder on meat composition and sensory characteristics in finishing gilts $(LY{\times}D)$. Twenty pigs (75kg) were randomly allotted to four treatments. Mugwort powder was added to final concentrations of 0(C), 0.5(T1), 1.0(T2), and 1.5%(T3) in the basal diet. Pigs were fed with the specified diet for 50 days. Carcass characteristics and cooking loss were not significantly affected by mugwort powder supplementation. The WHC and cooking loss of T2 and T3 were not significantly different than the control. However, the pH values for meat from gilts fed the T1 and T3 diets were significantly lower than the control (p<0.05). Shear force values of fresh meat were highest with the T3 diet (p<0.05), while the shear force values of cooked meat were significantly lower than the control with the T2 and T3 diets. Texture values of hardness, brittleness, gumminess, chewiness, and adhesiveness were significantly higher with the T3 diet (p<0.05), however values for cohesiveness and springiness were not significantly affected by mugwort powder supplementation. These results indicate that the feeding of mugwort powder improves the shear force of cooked meat from gilts fed the T3 diet, however the carcass, WHC and cooking loss characteristics in gilts are not affected by mugwort powder supplementation.

Berchemia floribunda-mediated Proteasomal Degradation of CyclinD1 via GKS3β-dependent Threonine-286 Phosphorylation in Human Colorectal Cancer Cells (인간 대장암 세포에 대한 먹넌출 추출물의 GSK3β 의존성 threonine-286 인산화를 통한 Cyclin D1 분해)

  • In this study, we evaluated the anti-cancer activity and potential molecular mechanism of 70% ethanol extracts of the Berchemia floribunda (BF) which belongs to Rhamnaceae against human colorectal cancer cells. The treatment of BF decreased the cell proliferation in HCT116 cell and suppressed cellular accumulation of Cyclin D1 protein. Inhibition of proteasomal activity by MG132 attenuated BF-mediated Cyclin D1 downregulation and Cyclin D1 was decreased in the cell treated with BF. These findings indicates that BF-mediated Cyclin D1 downregulation may be result from Cyclin D1 proteasomal degradation. Additionally, BF-mediated Cyclin D1 degradation was blocked in the presence of LiCl, a GSK3β inhibitor, but not PD98059, SP600125, SB203580, Bay11-7082, LY294002 an ERK1/2 inhibitor, JNK inhibitor, p38 inhibitor, IκK inhibitor and PI3K inhibitor. Furthermore, BF phosphorylated Cyclin D1 at threonine-286 (Thr286), and LiCl-induced GSK3β inhibition reduced the BF mediated phosphorylation of Cyclin D1 at Thr286. These results suggested that BF may downregulate Cyclin D1 expression as a potential anti-cancer target through GSK3β dependent Cyclin D1 degradation. Therefore, this study provides that the extract of BF has anticancer activity against human colorectal cancer cells.

Purification and Properties of D-Xylose Isomerase from Lactococcus sp. JK-8 (Lactococcus sp. JK-8에서 생산된 D-Xylose isomerase의 정제와 특성에 관한 연구)

  • D-Xylose isomerase produced by Lactococcus sp. JK-8, isolated from kimchi, was purified 17-fold of homogeneity, and its physicochemical properties were determined. Although the N-terminal amino acid sequence of D-xylose isomerase was analysed to Ala-Tyr-Phe-Asn-Asp-Ile-Ala-Pro-Ile-Lys, it was not similar to that of Lactobacillus enzyme. The molecular weight of the purified enzyme was estimated to be 180 kDa by gel filtration, 45 kDa by SDS-PAGE and the enzyme was homotetramer. The optimum pH of the enzyme was around 7 and stable between pH 6 and 8. The optimum reaction temperature was 7$0^{\circ}C$ and stable up to 7$0^{\circ}C$ in the presence of 1 mM $Mn^{2+}$. Like other D-xylose isomerases, this enzyme required divalent cation, such as $Mg^{2+}$, $Co^{2+}$, or $Mn^{2+}$ for the activity and thermostability. $Mn^{2+}$was the best activator. Substrate specificity studies showed that this enzyme was highly active on D-xylose. The enzyme had an isoelectric point of 4.8, and fm values for D-xylose was 5.9 mM.

Conformational Transition of Form II to Form Ⅰ PoLy(L-proline) and the Aggregation of Form Ⅰ in the Transition: Water-Propanol Solvent System

  • The conformational transition of poly(L-proline) (PLP), Form Ⅱ → Form Ⅰ and the intermolecular aggregation of the product, Form Ⅰ, during and after the transition in water-propanol (1:7, 1:9, 1:15.7, and 1:29 v/v) were studied. For the study, the viscosity change and excess light scattering intensity were measured in the course of the transition which was determined by the Form Ⅰ fraction, fI of the sample solution. For the PLP sample of molecular weight Mv=31,000 the experimental results show that the reaction course is roughly divided into three regions: in the first region [fI=0.27 to 0.40 (- [α]D=400 to 330)], the conformational change of Form Ⅱ → Form Ⅰ occurs with decrease of viscosity, in the second region [fI=0.40 to 0.80 (- [α]D=330 to 120)], a partial side-by-side (p-S-S) type aggregation in which Form Ⅰ blocks interact with each other, which induces the increase of viscosity, starts to occur, and in the third region [fI=0.80 to 1.00 (- [α]D=120 to 15)], a side-by-side type (raft like) aggregation of Form Ⅰ or an end-to-end (E-E) type aggregation occurs according to the solvent situation, i.e., in a water-rich medium [water-propanol (1:9 or 1:7 v/v)], the (S-S) type aggregation with a gross decrease in viscosity occurs while in a water-poor medium [water-propanol (1:29 or 1:15.7 v/v), the (E-E) type aggregation with a large increase in viscosity occurs. The (S-S) type aggregation was promoted at high temperatures. Based on the structure of PLP, a reasonable mechanism for the (p-S-S) and (S-S) aggregation which occurs with the transition of Form Ⅱ → Form Ⅰ is considered. The suggested mechanism was also supported by the result of chain length effect of PLP for the aggregation.

  • This study was carried out to investigate effects of diazinon, an organophosphate, and nicotine, a plant~originated toxic chemical, on larval mortality and activity of cytochrome P-45D monooxygenases, a major detoxifwation enzyme system, in Helicoverpa assulta and Spodoptera exigua. Diazinon treatment gave a higher mortaliLy to H. assulta larvae than S. exigua larvae. In contrast to the case of diazinon, nicotine caused a higher mortahty to S. exigua than to H. assulla larvae. It was partly due to the fact that nicotine induced the actiVIty of midgut cytochrome P-450 monooxygenases (MFO) more than diazinon did in If assulta larvae. When If. aSSlllta larvae were reared on their host p\am. NicotwlIa tuoocum leaves, other componentS were mostly metabolized with the exception of dietary nicotine.

  • An optimal Stabilization technique for a bilinear in duction model is introduced. This technique includes to o parts; the one is an stabilization control using Lyap unov Function which has the form of a sum of linear and quadratic function of the state variables, and the other is an optimal control using the performance index which depends on the choice of the elements of the Ly apunov matrices concerning both the state variables and the input variables. Therefore, induction motor is drived with the shorter transient time of the state variables and with the smaller overshoot of the ones, simulation results are obtained from a digital computer. Experimental ones are obtained from implementation of the optimizing controller using 8086 microprocessor kits and analog circuits are compared.

Effect of Drinking Fermented Milk on the Improvement of Defecation in Constipated Female Students (농후 발효유에 의한 여대생의 변비개선 효과)

  • The aim of this study was to investigate the effect d concentrated fermented milk on the improvement of defecation in female students with constipation. This yogurt was fermented with Bifidobacterium, L. acidophilus, and S. thermophilus and supplemented with oligosaccharides, dietary fiber, and yeast and mushroom extracts. 44 female students with constipation were sampled as subjects and fed 150ml fermented milk per day for 2 weeks period. After each defecation, the subjects were examined for the status of stool evacuation by questionnaires. The frequency of stool evacuation, fecal amount, time spending for evacuation, fecal color and shape, and refreshing feeling after evacuation were remarkably improved after ingestion of fermented milk for 2 weeks. These effects were greater in the heavy constipation group than in the mild group. The fecal odor did rot decrease significantly after 2 weeks of intake of fermented milk, but the odor thended to decrease after the intake of fermented milk These results indicated that the ingestion of concentrated fermented milk is effective to improve the constipation.

Phosphorylation of Eukaryotic Elongation Factor 2 Can Be Regulated by Phosphoinositide 3-Kinase in the Early Stages of Myoblast Differentiation

  • We have previously reported that phosphorylation of eukaryotic elongation factor 2 (eEF2) is related to the differentiation of chick embryonic muscle cells in culture. In the present study, we found that eEF2 phosphorylation declined shortly after induction of differentiation of L6 myoblasts, when the cells prepare for terminal differentiation by withdrawing from the cell cycle. This decrease in phosphorylation was prevented by inhibitors of phosphoinositide 3-kinase (PI3-kinase) that strongly inhibit myoblast differentiation. We hypothesized that PI3-kinase plays an important role in myoblast differentiation by regulating eEF2 phosphorylation in the early stages of differentiation. To test this hypothesis, myoblasts were synchronized at in $G_2/M$ and cultured in fresh differentiation medium (DM) or growth medium (GM). In DM the released cells accumulated in $G_0$/$G_1$ while in GM they progressed to S phase. In addition, cyclin D1 was more rapidly degraded in DM than in GM, and eEF2 phosphorylation decreased more. Inhibitors of PI3-kinase increased eEF2 phosphorylation, but PI3-kinase became more activated when eEF2 phosphorylation declined. These results suggest that the regulation of L6 myoblast differentiation by PI3-kinase is related to eEF2 phosphorylation.

Real-time Detection of Trace Copper in Brain and Kidney of Fish for Medical Diagnosis

  • For the detection of trace copper to be used in medical diagnosis, a sensitive handmade carbon nanotube paste electrode (PE) was developed using voltammetry. Analytical optimized conditions were found at 0.05 V anodic peak current. In the same conditions, various common electrodes were compared using stripping voltammetry, and the PE was found to be more sharply sensitive than other common electrodes. At optimum conditions, the working ranges of $3{\sim}19{\mu}gL^{-1}$ were obtained. The relative standard deviation of $70.0{\mu}gL^{-1}$ was determined to be 0.117% (n = 15), and the detection limit (S/N) was found to be $0.6{\mu}gL^{-1}$ ($9.4{\times}10^{-9}M$). The results were applied in detecting copper traces in the kidney and the brain cells of fish.

  • Mekong River is the world's $10^{th}$ longest river and runs through China's Yunnan province, Burma, Thailand, Laos, Cambodia and Vietnam. And Tonle Sap Lake, the largest fresh water body in Southeast Asia and the heart of Mekong River system, covers an area $2,500-3,000Km^2$ in dry season and $10,000-16,000Km^2$ in wet season. As previously noted, the water within Sap river flows from the Mekong River to Tonle Sap Lake in flood season (between June and October) and backward to Mekong River in dry season. Recently the flow regime of Sap River might be significantly affected by the development of large dams in upstream region of Mekong River. This paper aims at basic study about the large scale flood inundation of Cambodia using by CAESAR-Lisflood. CAESAR-Lisflood is a geomorphologic / Landscape evolution model that combines the Lisflood-FP 2d hydrodynamic flow model (Bates et al, 2010) with the CAESAR geomorphic model to simulate flow hydrograph and erosion/deposition in river catchments and reaches over time scales from hours to 1000's of years. This model is based on the simplified full Saint-Venant Equation so that it can simulate the interacted flow of between Mekong River and Tonle Sap Lake especially focusing on the flow direction change of Sap River by season.
