Upregulation of HIF-1α by Hypoxia Protect Neuroblastoma Cells from Apoptosis by Promoting Survivin Expression

  • Apoptosis is one of main types of neural cell death and is reversible and is a major target of therapeutic interventions. However, detailed apoptotic cascades still need to be recognized. In present study, we determined the promotion of HIF-$1{\alpha}$ and survivin in brain samples of a mouse model of hypoxic-ischemia and in neuroblastoma SH-SY5Y cells post hypoxia treatment. Then gain-of-function and loss-of-function strategies were adopted to manipulate the HIF-$1{\alpha}$ in SH-SY5Y cells, and hypoxia-induced survivin upregulation and cell apoptosis were determined. Results demonstrated that the HIF-$1{\alpha}$ and survivin were significantly promoted in a mouse model of hypoxic-ischemia or in SH-SY5Y cells post hypoxia in vitro. Manually upregulated HIF-$1{\alpha}$ could promote the hypoxia-induced survivin upregulation and improve the hypoxia-induced SH-SY5Y cell apoptosis. On the other hand, the HIF-$1{\alpha}$ knockdown by RNAi reduced the hypoxia-induced survivin upregulation and cell apoptosis. Therefore, the present study confirmed the protective role of HIF-$1{\alpha}$ and survivin in the hypoxia-induced SH-SY5Y cell apoptosis, and the survivin upregulation by hypoxia is HIF-$1{\alpha}$-dependent. Promotion of HIF-$1{\alpha}$ and survivin might be a valuable stragegy for therapeutic intervention for hypoxic-ischemic encephalopathy.

BA침종기간이 숙주나물의 형태와 생장에 미치는 영향 (Effect of Benzyladenopurine Soaking Period on Growth of Mungbean Sprouts)

  • 숙주나물의 세근 발생을 방지하기 위한 BA 처리는 주로 침종기간에 행하여진다. 본 연구는 숙주나물 생산에서 BA 흡수량과 관련된 침종기간의 설정에 필요한 정보를 제공하고자 3개 공시품종(금성녹두, 어울녹두, 중록1호)을 이용하여 침종기간(3, 5, 7시간)에 따른 숙주나물의 생장과 형태, 이들 품종의 흡수량으로 BA 소모량을 조사한 결과를 요약하면 다음과 같다. 1. 종자의 수분흡수에서 어울녹두는 침종기간 내내 가장 낮았던 반면, 금성녹두와 중록1호는 침종초기에서는 중록1호가 많았으나 침종 후기에는 거의 비슷한 것으로 나타났다. 2. 상품성이 가장 높은 하배축 길이가 7cm이상의 비율은 중록1호에서 가장 높았고, 금성녹두, 어울녹두 순으로 낮아졌으나, 상품화가 불가능한 4cm 이하의 비율은 7cm 이상과는 반대의 결과를 보였다. 침종기간이 7시간으로 길어질수록 하배축 길이가 7cm이상의 비율이 감소되는 중록1호를 제외하고는 침종기간간에 뚜렷한 차이가 없었다. 3. 세근은 침종기간이 길수록 적게 형성되었으나 3시간에서 5시간으로 연장할 경우 현저히 줄어드는 경향을 보였다. 하배축과 뿌리 길이는 침종기간이 길수록 짧아졌으나 하배축 직경은 5시간 침종시 대체로 굵은 것으로 나타났다. 4. 개체당 전체 생체중과 건물중은 생장이 상대적으로 빠른 중록1호에서 가장 많았으며, 침종기간의 장단간에는 차이가 없었다. 5. 흡수량으로 평가된 BA 소모량은 침종기간이 5시간보다 짧을 경우 중록1호에서 가장 많았던 반면, 7시간 침종시킬 경우 금성녹두와 중록1호가 거의 비슷하게 많은 것으로 분석되었다.

An analytical approach for nonlinear response of elastic cable under complex loads

  • In this paper a general analytical approach is proposed to analyse the nonlinear response of elastic cable under complex loads. The effect of temperature change on the cable is also considered. From the vertical equilibrium equations of cable, the general analytical formula of vertical displacement is derived. Based on the vertical displacement formula and on the compatibility condition of the cable, the dimensionless equation with respect to cable tension is established. By means of such analytical procedures, the exact solutions of various cable problems can be obtained quickly. The example given in this paper shows that the new procedure is efficient for practical analysis and can be easily implemented by a general computer program without the superposition problem which there has always been in traditional analytical methods.

Video-assisted Thoracoscopic Surgery for Treatment of Earlystage Non-small Cell Lung Cancer

  • Objectives: To evaluate the safety, efficacy, and invasiveness of lobectomy by video-assisted thoracoscopic surgery (VATS) in the treatment of stage I/II non-small cell lung cancer (NSCLC). Methods: A total of 148 patients presenting with Stage I or II NSCLC were enrolled into our study, comprising 71 who underwent VATS and 77 patients undergoing conventional thoracotomic lobectomy, in combination with systematic lymph node resection. Results: It was found that VATS was superior to conventional thoracotomy in terms of the duration of surgery, intraoperative blood loss, frequency of the need to administer postoperative analgesia, thoracic intubation indwelling time, post-operative hospital stay, and survival rate (P<0.05). We saw no obvious difference in the number of resected lymph nodes with either approach. Conclusions: VATS lobectomy is a safe and reliable surgical approach for the treatment of Stage I/II NSCLC, characterized by significantly minimal invasiveness, rapid post-operative recovery, and markedly lower loss of blood.

  • Using OpenSees as a framework, constitutive models of reinforced, prestressed and prestressed steel fiber concrete found by the panel tests have been implemented into a finite element program called Simulation of Concrete Structures (SCS) to predict the seismic behavior of shear-critical reinforced and prestressed concrete structures. The developed finite element program was validated by tests on prestressed steel fiber concrete beams under monotonic loading, post tensioned precast concrete column under reversed cyclic loading, framed shear walls under reversed cyclic loading or shaking table excitations, and a seven-story wall building under shake table excitations. The comparison of analytical results with test outcomes indicates good agreement.

Study on the PET image quality according to various scintillation detectors based on the Monte Carlo simulation

  • Purpose: Positron emisson tomography (PET) is a crucial medical imaging scanner for the detection of cancer lesions. In order to maintain the improved image quality, it is crucial to apply detectors of superior performance. Therefore, the purpose of this study was to compare PET image quality using Monte Carlo simulation based on the detector materials of BGO, LSO, and LuAP. Materials and Methods: The Geant4 Application for Tomographic Emission (GATE) was used to design the PET detector. Scintillations with BGO, LSO and LuAP were modelled, with a size of 3.95 × 5.3 mm2 (width × height) and 25.0 mm (thickness). The PET detector consisted of 34 blocks per ring and a total of 4 rings. A line source of 1 MBq was modelled and acquired with a radius of 1 mm and length of 20 mm for 20 seconds. The acquired image was reconstructed maximum likelihood expectation maximization with 2 iteration and 10 subsets. The count comparison was carried out. Results and Discussion: The highest true, random, and scatter counts were obtained from the BGO scintillation detector compared to LSO and LuAP. Conclusion: The BGO scintillation detector material indicated excellent performance in terms of detection of gamma rays from emitted PET phantom.

  • $Ce^{3+}$ 이온을 도핑한 $(Ce_xLu_{1-x})_3MgAl_3SiO_{12}$ 형광체 샘플을 고상합성법으로 합성하였다. 열처리 온도를 1250부터 $1550^{\circ}C$ 온도영역에서 조절하면서 5 h 동안 소성하였으며, 소성된 샘플들의 XRD와 PL 특성을 조사하여 가장 최적의 열처리 온도를 구하고자 하였다. 또한 $Ce^{3+}$ 이온의 도핑농도를 2.0에서 10.0 mol%로 변화시키면서 $Ce^{3+}$ 이온의 도핑농도와 광학적 특성과의 관계를 고찰하였다. $Ce^{3+}$ 이온의 도핑농도에 따른 형광체들의 PL 강도, 피크위치, 반치폭을 계산하여 실제적인 LED 패키징에 가장 적절한 Ce 도핑 농도를 구하고자 하였다. 또한 최적의 소성온도와 Ce 도핑농도 조건에서 합성한 형광체 샘플의 입도와 입형을 분석하였다.

Dosimetric Analysis of a Phase I Study of PSMA-Targeting Radiopharmaceutical Therapy With [177Lu]Ludotadipep in Patients With Metastatic Castration-Resistant Prostate Cancer

  • Objective: 177Lutetium [Lu] Ludotadipep is a novel prostate-specific membrane antigen targeting therapeutic agent with an albumin motif added to increase uptake in the tumors. We assessed the biodistribution and dosimetry of [177Lu]Ludotadipep in patients with metastatic castration-resistant prostate cancer (mCRPC). Materials and Methods: Data from 25 patients (median age, 73 years; range, 60-90) with mCRPC from a phase I study with activity escalation design of single administration of [177Lu]Ludotadipep (1.85, 2.78, 3.70, 4.63, and 5.55 GBq) were assessed. Activity in the salivary glands, lungs, liver, kidneys, and spleen was estimated from whole-body scan and abdominal SPECT/CT images acquired at 2, 24, 48, 72, and 168 h after administration of [177Lu]Ludotadipep. Red marrow activity was calculated from blood samples obtained at 3, 10, 30, 60, and 180 min, and at 24, 48, and 72 h after administration. Organand tumor-based absorbed dose calculations were performed using IDAC-Dose 2.1. Results: Absorbed dose coefficient (mean ± standard deviation) of normal organs was 1.17 ± 0.81 Gy/GBq for salivary glands, 0.05 ± 0.02 Gy/GBq for lungs, 0.14 ± 0.06 Gy/GBq for liver, 0.77 ± 0.28 Gy/GBq for kidneys, 0.12 ± 0.06 Gy/GBq for spleen, and 0.07 ± 0.02 Gy/GBq for red marrow. The absorbed dose coefficient of the tumors was 10.43 ± 7.77 Gy/GBq. Conclusion: [177Lu]Ludotadipep is expected to be safe at the dose of 3.7 GBq times 6 cycles planned for a phase II clinical trial with kidneys and bone marrow being the critical organs, and shows a high tumor absorbed dose.

  • Gas pressure sintering (GPS) of reaction bonded silicon nitride (RBSN) was performed using $Lu_2O_3-SiO_2$ additive and the properties were compared with those of specimens prepared using high purity $Si_3N_4$ powder. The relative density of RBSN and compacted $Si_3N_4$ powder were 68.9 and 47.1%, and total linear shrinkage after sintering at $1900^{\circ}C$ were 14.8 and 42.9%, respectively. High nitrogen partial pressure (5MPa) was required during sintering at $1900^{\circ}C$ in order to prevent the decomposition of the nitride and to promote the formation of SiC. The relative density and 4-point bending strength of RBSN and $Si_3N_4$ powder compact were 97.7%, 954MPa and 98.2%, 792MPa, respectively, after sintering at $1900^{\circ}C$. The sintered RBSN also showed high fracture toughness of 9.2MPam$^{1/2}$.

An HPLC-UV-based quantitative analytical method for Chrysanthemum morifolium: development, validation, and application

  • A simple and reliable analytical method based on high-performance liquid chromatography-ultraviolet detection was established for the analysis of the flowers of Chrysanthemum morifolium (CM). Luteolin-7-O-glucoside (LU7G) was chosen as a target analyte considering its content, availability, and ease of analysis. Chromatographic separation of LU7G was achieved using a Phenomenex Gemini $C_{18}$ column ($250{\times}4.6mm$, $5{\mu}m$) run with a mobile phase consisting of 0.5 % acetic acid in water and 0.5 % acetic acid in acetonitrile at a flow rate of $1.0mL\;min^{-1}$. The detection wavelength and column temperature were set at 350 nm and $40^{\circ}C$, respectively. Method validation was performed according to the AOAC guidelines and the method was specific, linear ($R^2=0.9991$ for $50-300{\mu}g\;mL^{-1}$), precise (${\leq}3.91%$RSD), and accurate (100.1-105.7 %). The limits of detection and quantification were 3.62 and $10.96{\mu}g\;mL^{-1}$, respectively. The established method was successfully applied to determine the contents of LU7G in various batches of bulk CM extracts and labscale CM extract. The developed method is a readily applicable method for the quality assessment of CM and its related products.
