Genetic Parameter Estimation of Carcass Traits of Duroc Predicted Using Ultrasound Scanning Modes

  • A total of 6,804 records for Duroc breed were collected from three farms registered at the Korean Animal Improvement Association (KAIA) from 1998 to 2004 of which both records from two ultrasound modes (A and B) were analyzed to estimate the variance components of carcass traits. Three carcass traits backfat thickness (bf), loin eye muscle area (lma) and lean meat percentage (lmp) were measured. These traits were analyzed separately as bf1, lma1 and lmp1 for ultrasound mode A and bf2, lma2 and lmp2 for ultrasound mode B with multiple trait animal model by using MTDFREML (Boldman et al., 1993). All the traits revealed medium heritability values. Estimated heritabilities for bf1, bf2, lma1, lma2, lmp1 and lmp2 were 0.45, 0.39, 0.32, 0.25, 0.28 and 0.39, respectively. Estimated genetic correlations for traits bf1 and bf2, lma1 and lma2, lmp1 and lmp2 were positive but low. Specifically, genetic correlations between bf1 and bf2 was 0.30 while the estimates for lean traits between lma1 and lma2 and between lmp1 and lmp2 were 0.15 and 0.18, respectively. Conversely, high negative genetic correlations existed between bf1 and the lean traits lma2, lmp2. Likewise, the estimated genetic correlations between lma1 and lma2 and lmp1 and lmp2 were low.

  • Hematological malignancies (HM) are a group of neoplasms derived from the cells of the bone marrow and lymphatic system. Genetic factors leading to susceptibility to HM have been investigated for years but little is known yet. Low molecular weight polypeptide (LMP) 2 and LMP7 genes are important subunits of the immunoproteasome and play significant role in antigen presentation. The polymorphisms of LMP genes have been reported to be risk factors for various types of diseases. The aim of this study was to investigate the association of LMP2 and LMP7 polymorphisms with the occurrence of particular types of HM. A total of 132 patients with HM and 130 control subjects were investigated. No significant difference was obtained in the distribution of genotype and allele frequencies of LMP7 gene in HM patients and the control group. On the other hand, the prevalence of LMP2-AA genotype was found to be higher in acute myeolid leukemia (AML) patients while it was significantly lower in multiple myeloma (MM) cases than in the control subjects. Our results suggested that LMP7 could not be a risk factor for susceptibility to HM, whereas LMP2 polymorphisms could play a role in the development of AML and MM.

Isolation and Characterization of Listeria phages for Control of Growth of Listeria monocytogenes in Milk

  • In this study, two Listeria bacteriophages, LMP1 and LMP7, were isolated from chicken feces as a means of biocontrol of L. monocytogenes. Both bacteriophages had a lytic effect on L. monocytogenes ATCC 7644, 15313, 19114, and 19115. Phages LMP1 and LMP7 were able to inhibit the growth of L. monocytogenes ATCC 7644 and 19114 in tryptic soy broth at $10^{\circ}C$ and $30^{\circ}C$. Nevertheless, LMP1 was more effective than LMP7 at inhibiting L. monocytogenes ATCC 19114. On the contrary, LMP7 was more effective than LMP1 at inhibiting L. monocytogenes ATCC 7644. The morphology of LMP1 and LMP7 resembled that of members of the Siphoviridae family. The growth of L. monocytogenes ATCC 7644 was inhibited by both LMP1 and LMP7 in milk; however, the growth of L. monocytogenes ATCC 19114 was only inhibited by LMP1 at $30^{\circ}C$. The lytic activity of bacteriophages was also evaluated at $4^{\circ}C$ in milk in order to investigate the potential use of these phages in refrigerated products. In conclusion, these two bacteriophages exhibit different host specificities and characteristics, suggesting that they can be used as a component of a phage cocktail to control L. monocytogenes in the food industry.

  • The combustion test of LMP-103S, a propellant based on ADN(Ammonium Dinitramide), was performed with a 50 mN scale micro-thruster. The micro-thruster was made with photosensitive glass using MEMS manufacturing process. $Pt/{\gamma}-Al_2O_3$ was used as a catalyst to decompose LMP-103S. After injecting 90 wt.% hydrogen peroxide into combustion chamber to preheat the catalyst, LMP-103S was injected for the combustion test. As a result, the ignition and combustion of LMP-103S was confirmed in platinum catalyst environment with the combustion chamber temperature going up to $650^{\circ}C$.

Anti-oxidation and Anti-wrinkling Effects of Jeju Horse Leg Bone Hydrolysates

  • This study focused on the anti-oxidative and collagenase- and elastase inhibition effects of low molecular weight peptides (LMP) from commercial Jeju horse leg bone hydrolysates (JHLB) on pancreatin, via enzymatic hydrolysis. Cell viability of dermal fibroblasts exposed to UVB radiation upon treatment with LMP from JHLB was evaluated. Determination of the antioxidant activity of various concentrations of LMP from JHLB were carried out by assessing 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2-azino-bis-3-ethybenzothiazoline-6-sulphonic acid (ABTS) radical scavenging activity, ferric reducing antioxidant power (FRAP), and oxygen radical absorbance capacity (ORAC). The DPPH radical scavenging activity of LMP from JHLB (20 mg/mL) was 92.21% and ABTS radical scavenging activity (15 mg/mL) was 99.50%. FRAP activity (30 mg/mL) was $364.72{\mu}M/TE$ and ORAC activity (1 mg/mL) was $101.85{\mu}M/TE$. The anti-wrinkle potential was assessed by evaluating the elastase- and collagenase inhibition potential of these LMP. We found that 200 mg/mL of LMP from JHLB inhibited elastase activity by 41.32%, and 100 mg/mL of LMP from JHLB inhibited collagenase activity by 91.32%. The cell viability of untreated HS68 human dermal fibroblasts was 45% when exposed to a UVB radiation dose of $100mJ/cm^2$. After 24 h of incubation with $500{\mu}g/mL$ LMP from JHLB, the cell viability increased to 60%. These results indicate that LMP from JHLB has potential utility as an anti-oxidant and anti-wrinkle agent in the food and cosmetic industry. Additional in vivo tests should be carried out to further characterize these potential benefits.

  • In this report, the creep properties and creep life estimation by Larson-Miller Parameter(LMP) Method for 2.25Cr1Mo steel to be used as power plant tubes or other components were presented at the high temperatures of 500, 550, and $600^{\circ}C$. It was confirmed experimentally and quantitatively that a creep life estimation equation at such various high temperatures was well derived by LMP and could be used very effectively within the creep life of 10$^3$ hours, but very unreliable and even dangerous for design in a long term of creep life such as 10$^4$ or $10^5$ hours.

Application of the New Ballard Scale with Extended Scoring System in Full-term Newborns (New Ballard Scale(NBS)의 확장 적용)

  • Purpose: An exploratory study was done to examine the validity of the new Ballard scale with extended scoring system(eNBS) in estimating gestational age(AG) in full-term newborns. Method: The eNBS scoring system was extended to include all numbers of total score of NBS and GA to allow a 3-days variation in GA estimatio compared to the original scale which has a 2-week variation due to the application of a 5-score interval for the total NBS score and only even numbers for GA. GA by eNBS(GA-eNBS) was compared with GA by LMP(GA-LMP) and GA by standard NBS(GA-sNBS) in 133 full-term newborns. Difference between GA-LMP and GA-eNBS was analyzed for each GA. Results: Positive correlations were observed in GA-sNBS and GA-eNBS with GA-LMP. There was no difference between GA-LMP and GA-eNBS at 39GA and 40GA. At 37GA and 38GA, GA-eNBS overestimated GA-LMP up to 1 week, while underestimating up to 1 week at 41GA. Conclusions: The accuracy of eNBS was validated within 3 days of variation in GA estimation at 39-40GA. Overestimation by eNBS suggests the possible acceleration of fetal maturity in premature newborns, while underestimation, of the deceleration of fetal maturity in postterm newborns.

Deletion Analysis of the Major NF-${\kappa}B$ Activation Domain in Latent Membrane Protein 1 of Epstein-Barr Virus

  • Latent membrane protein 1 (LMP1) of the Epstein-Barr virus (EBV) is an integral membrane protein with six transmembrane domains, which is essential for EBV-induced B cell transformation. LMP1 functions as a constitutively active tumor necrosis factor receptor (TNFR) like membrane receptor, whose signaling requires recruitment of TNFR-associated factors (TRAFs) and leads to NF-${\kappa}B$ activation. NF-${\kappa}B$ activation by LMP1 is critical for B cell transformation and has been linked to many phenotypic changes associated with EBV-induced B cell transformation. Deletion analysis has identified two NF-${\kappa}B$ activation regions in the carboxy terminal cytoplasmic domains of LMP1, termed CTAR1 (residues 194-232) and CTAR2 (351-386). The membrane proximal C-terminal domain was precisely mapped to a PXQXT motif (residues 204-208) involved in TRAF binding as well as NF-${\kappa}B$ activation. In this study, we dissected the CTAR2 region, which is the major NF-${\kappa}B$ signaling effector of LMP1, to determine a minimal functional sequence. A series of LMP1 mutant constructs systematically deleted for the CTAR2 region were prepared, and NF-${\kappa}B$ activation activity of these mutants were assessed by transiently expressing them in 293 cells and Jurkat T cells. The NF-${\kappa}B$ activation domain of CTAR2 appears to reside in a stretch of 6 amino acids (residues 379-384) at the end of the carboxy terminus.

Determination of Frequency of Epstein-Barr Virus in Non-Hodgkin Lymphomas Using EBV Latent Membrane Protein 1 (EBV-LMP1) Immunohistochemical Staining

  • Background: The presence of Epstein-Barr virus (EBV) in Non-Hodgkin's lymphoma can be identified by immunohistochemistry for detection of EBV latent membrane protein (LMP). The role of EBV as an etiologic agent in the development of non-Hodgkin lymphoma has been supported by detection of high levels of latent membrane protein 1 (LMP-1) expression in tumors. However, no study has been conducted in a Pakistani population up till now to determine the frequency of Epstein-Barr virus positivity. The objective of our study was to determine a value for non-Hodgkin lymphoma patients using EBV LMP-1 immunostaining in our institution. Materials and Methods: This study was carried out at the Department of Histopathology, Armed Forces Institute of Pathology (AFIP), Pakistan from December 2011 to December 2012. It was a cross sectional study. A total of 71 patients who were diagnosed with various subtypes of NHL after histological and EBV LMP-1 immunohistochemical evaluation were studied. Sampling technique was non-probability purposive. Statistical analysis was achieved using SPSS version 17.0. Mean and SD were calculated for quantitative variables like patient age. Frequencies and percentages were calculated for qualitative variables like subgroup of NHL, results outcome of IHC for EBV and gender distribution. Results: Mean age of the patients was $53.6{\pm}16$ years (Mean${\pm}$SD). A total of 50 (70.4%) were male and 21 (29.6%) were female. Some 9 (12.7%) out of 71 cases were positive for EBV-LMP-1 immunostaining, 2 (22.2%) follicular lymphoma cases, 1 (11.1%) case of T-cell lymphoblastic lymphoma, 4 (44.4%) cases of diffuse large B cell lymphomas, 1 (11.1%) mantle cell lymphoma and 1 (11.1%) angioimmunoblastic T cell lymphoma case. Conclusion: In our study, frequency of EBV in NHL is 12.7% and is mostly seen in diffuse large B cell lymphoma. This requires further evaluation to find out whether this positivity is due to co-infection or has a role in pathogenesis.

Residual Life Assessment of mod HP Reformer Tubes Using Surface Replication Methods (표면복제를 사용한 mod HP강 개질로 튜브의 잔여수명 평가에 대한 연구)

  • Residual life assessment of reformer tubes of mod HP steel used in the refinery and petrochemical industry were investigated by quantitative metallographic evaluation method. Area fractions of precipitated carbides were measured by using surface replication and its relationship with the remaining life of the tube was investigated. The microstructures of the virgin and other tubes were examined using optical microscopy (OM) for service periods of 1.8, 6.0, 7.2, 8.5, 9.7, and 16.2 years at 950 ℃ and the area fractions of the precipitates were measured by image analysis using ImageJ software. As the area fraction of the precipitates increased, the residual life was decreased accordingly. The results showed that the LMP was the highest in the virgin and the LMP decreased as the service time increased and the residual life decreased. A regressional equation showing the relation between the area fraction of carbides and LMP was experimentally derived. The obtained relationship can be used with the surface replication method for on-site residual life assessment.
