Development of a Highly Active Fluorescence-Based Detector for Yeast G Protein-Coupled Receptor Ste2p

  • Twenty analogs of $[Orn^6,D-Ala^9]{\alpha}-factor$ were synthesized and assayed for their biological activities: seven analogs of $[Orn^6,X^9]{\alpha}-factor$, seven analogs of $[X^6,D-Ala^9]{\alpha}-factor$, five analogs of $[X^5,X^6,D-Ala^9]{\alpha}-factor$, and native ${\alpha}-factor$ (X = amino acids). Their biological activities (halo, gene induction, and affinity) were measured using S. cerevisiae Y7925 and LM102 and compared with those of native ${\alpha}-factor$ (100%). G protein-coupled receptor was expressed in strain LM102 containing pESC-LEU-STE2 vector. $[Dap^6,D-Ala^9]{\alpha}-factor$ with weak halo activity (10%) showed the highest receptor affinity (> 230%) and the highest gene induction activity (167%). $[Arg^6,D-Ala^9]{\alpha}-factor$ showed the highest halo activity (2,000%). The number of active binding sites per cell (about 20,000 for strain LM102) was determined using a newly-designed fluorescence-based detector, $[Arg^6,D-Ala^9]{\alpha}-factor-Edan$, with high sensitivity (12,500-fold higher than the absorption-based detector $[Orn^6]{\alpha}-factor-[Cys]_3$).

  • These studies were conducted to investigate the effect of dietary supplemental lincomycin at different food waste supplements on performance of broiler chicks. Diets were based corn, soybean meal and contained 3,100kcal/kg ME, 21% CP for the rest two weeks. Three levels of dietary lincomycin(LM; 0,2.2, 4.4mg/kg) and food waste(FW; 0, 7.5, 15%) were respectively inclusion in both experiments. In experiment 1, weight gain of birds fed 4.4mg/kg LM was significantly higher than no LM treatments(P<0.05). It tended to increase in 7.5% food waste treatment compared to that of control, but was not significantly different. FCR of chicks fed 2.2mg/kg LM supplement was significantly improved compared to that of other treatments Chicks fed food waste showed inferior FCR to control as dietary FW level increased(P<0.05). There was no significance in blood components among treatments. In experiment 2, weight gain of chicks fed 15% FW exhibited higher weight gain than that of other treatments. As FW supplement increased, the feed intake increased significantly(P<0.05) There was no significance in blood components except serum calcium. The results of these experiments indicated that hicks fed 4.4mg/kg LM supplements maximized growth rate of all treatments, and improved FCR in 2.2mg/kg LM.

양만장 사육조에서 분리한 Edwardsiella tarda의 약제 내성과 R Plasmid (Antibiotic Resistance and R Plasmids in Edwardsiella tarda)

  • 서해안 일원의 가온 양만장 사육조로부터 분리한 E. tarda균에 대한 약제내성검사 및 내성균의 내성인자 전달 시험을 실시하였다. 공시한 항생제에 대한 내성률은 LM, PM, SD, SF 등에는 100%, AP, CH, SM, OT 및 CP 등에는 89% 이상, OA 및 KM에는 70% 정도 그리고 AK, GM 및 EF 등에는 30% 이하의 내성률을 보였으며, 이와같은 내성률은 각 양만장에서 분리한 균주간에 차이를 보이지 않았다. 내성유형은 총 34유형으로 모든 분리균주가 8~14제의 다제내성을 보였으며, 그 중 LM PM SD SF CP CH SM OT AP OA KM(22주, 21.4%) 및 LM PM SD SF CP CH SM OT AP OA(12주, 11.7%) 유형등이 고빈도로 출현하였다. 내성균의 내성인자 전달유형은 총 55형으로 단제에서 13제까지의 다양한 내성인자가 전달되었는데, 그 중 LM PM SD SF CP CH SM OT AP OA(10주. 10%), LM PM SD SF CP CH SM OT AP OA KM(7주, 7%) 및 LM PM SD SF CP CH AP OA(7주, 7%) 등의 다제내성유형이 고빈도의 전달률을 보였으며, 약재별로는 GM, EF 및 AK 등에는 저빈도(0~20%), OT, SM, KM 및 OA 등에는 중등도(49~62%), AP, PM, LM, SF, CP, CH 및 SD 등에는 고빈도(71~94%)로 내성인자를 전달하였다.

Flexible Organic Light-Emitting Diodes Using Modified Graphene Anodes

  • Graphene films have a strong potential to replace indium tin oxide anodes in organic light-emitting diodes (OLEDs), to date. However, the luminous efficiency of OLEDs with graphene anodes has been limited by a lack of efficient methods to improve the low work function and reduce the sheet resistance of graphene films to the levels required for electrodes. Here, we fabricate flexible OLEDs by modifying the graphene anode to have a high work function and low sheet resistance, and thus achieve extremely high luminous power efficiencies (37.2 lm/W in fluorescent OLEDs, 102.7 lm/W in phosphorescent OLEDs), which are significantly higher than those of optimized devices with an indium tin oxide anode (24.1 lm/W in fluorescent OLEDs, 85.6 lm/W in phosphorescent OLEDs). We also fabricate flexible white OLED lighting devices using the graphene anode. These results demonstrate the great potential of graphene anodes for use in a wide variety of high-performance flexible organic optoelectronics.

Effects of Plane of Nutrition on Growth Performance and Meat Quality Traits in Finishing Pigs

  • The present study was performed to investigate the effects of the plane of nutrition on growth performance and meat quality traits, especially marbling and PSE (pale, soft, and exudative), in finishing pigs. One hundred and four barrows and 102 gilts born to Yorkshire ${\times}$ Landrace dams and Duroc sires were randomly allocated to 16 pens at approximately 80-kg body weight, with 13 animals housed per pen except for two pens. Each eight pens received a diet containing either 3.4 Mcal ME/kg and 0.98% total lysine (high plane) or 3.1 Mcal ME/kg and 0.81% total lysine (medium plane). All animals were slaughtered at approximately 112 kg BW on an average, after which the Longissimus muscle (LM) was analyzed physicochemically. The ADG, ADFI, gain:feed as well as marbling score did not differ between the two dietary groups, whereas backfat thickness was greater in the high-plane group than in the medium-plane group (24.1 vs. 22.5 mm P<0.05). No LM exhibited PSE, i.e. $L^*$ (lightness) > 50, pH < 6.0 and drip loss > 5.0%, nor were these PSE-related variables influenced by the dietary treatment. Contents of crude protein and fat in LM did not differ between the two dietary groups. In conclusion, the present results suggest that growth performance and the occurrence of the PSE or 'watery' carcass are not influenced significantly by increasing the plane of nutrition in finishing pigs when the finisher is adequate nutritionally for supporting maximal lean growth. However, increasing the plane of nutrition is likely to result in an increase in fat deposition.

Effects of Stabilization Exercise Applying Sling Neurac Method on Pain and Cervical Alignment in Young Adults with Forward Head Posture

  • 이 연구의 목적은 전방머리자세를 가진 젊은 성인을 대상으로 슬링뉴렉안정화운동이 중재방법이 통증 및 목뼈 앞굽음각, 중력중심선에 어떠한 영향을 미치는지 알아보고자 실시되었다. 연구의 대상자는 슬링뉴렉안정화운동그룹(SNSEG) 10명, 통제그룹(CC) 10명으로 선정하였으며, 8주간 주3회 70분간 실시하였다. 이 연구 결과 슬링뉴렉안정화운동은 통증은 감소(p<.001), 목뼈 앞굽음각은 증가(p<.001)와 중력중심선은 감소에(p<.01) 유의한 효과가 나타났다. 이상의 결과 종합해 보면 슬링뉴렉안정화운동은 속근육(LM)군의 고유수용성감각 개선 및 근활성화에 효과적이며, 대근육(GM)군과 공동활성화하여 앞굽은각과 중력중심선에 개선에 효과적인 것으로 나타났다. 따라서 깊은목굽힘근과 뒤통수밑근의 재활성화는 통증 조절과 자세 정렬에 중요한 요소로 전방머리자세 개선시킬 수 있는 효과적인 중재방법으로 제안한다.

  • This study addresses evaluation of performance of hardening model for a titanium alloy (Ti6Al4V) based on the artificial neural network (ANN) regarding the strain rate and the temperature. Uniaxial compression tests were carried out at different strain rates from 0.001 /s to 10 /s and temperatures from 575 ℃ To 975 ℃. Using the experimental data, ANN models were trained and tested with different hyperparameters, such as size of hidden layer and optimizer. The input features were determined with the equivalent plastic strain, strain rate, and temperature while the output value was set to the equivalent stress. When the number of data is sufficient with a smooth tendency, both the Bayesian regulation (BR) and the Levenberg-Marquardt (LM) show good performance to predict the flow behavior. However, only BR algorithm shows a predictability when the number of data is insufficient. Furthermore, a proper size of the hidden layer must be confirmed to describe the behavior with the limited number of the data.

Effects of the plane of nutrition during the latter grower and entire finisher phases on grow-finish pig performance in summer

  • The present study was undertaken to investigate the effects of the plane of nutrition (PN) for growing-finishing pigs on growth performance and meat quality in summer. One hundred and two barrows and 102 gilts weighing approximately 44 kg were placed on a high-, medium-, or low-plane grower diet (HPG, MPG, or LPG) with ME and lysine concentrations ranging from 3.33 to 3.40 Mcal/kg and 0.93% to 1.15%, respectively, for 29 days in 6 replicates (pens) in total. Pigs from each grower pen were divided into two finisher pens and provided with a high-plane finisher diet (HPF) containing 3.40 Mcal ME and 9.5 g lysine/kg and a low-plane finisher diet (LPF; 3.25 Mcal ME and 8 g lysine/kg), respectively, up to approximately 110 kg, and slaughtered. Growth performance of the pigs, including average daily gain (ADG), average daily feed intake (ADFI), and gain:feed ratio, was not influenced by the grower-phase PN during any of the grower phase, a 31-d finisher phase I, and ensuing phase II. However, both the ADG and gain:feed ratio were greater (p < 0.05) for the HPF group than for the LPF group during the finisher phase I (748 vs. 653 g with SEM = 13 g and 0.333 vs. 0.299 with SEM = 0.008, respectively). The ADG, but not gain:feed ratio, was greater for the HPF group vs. LPF during the finisher phase II (673 vs. 623 g with SEM = 15 g for ADG and 0.322 vs. 0.323 with SEM = 0.005 for the gain:feed ratio). The carcass backfat thickness (BFT) was greater for the LPF group vs. HPF within the pigs which had been placed on LPG during the grower phase, but not within the pigs from the HPG or MPG group. Physicochemical characteristics of the longissimus dorsi muscle (LM) and sensory quality attributes of fresh and cooked LM were not influenced by either the grower-phase or finisher-phase PN. In conclusion, high PN is necessary for finishing pigs during the hot season to minimize the reduced rate of weight gain and also to prevent the increase of BFT that could result from low PN.

Effects of the plane of nutrition for grower pigs on their grow-finish performance and meat quality in winter

  • Little is known about the effects of the plane of nutrition on growth performance and meat quality of grow-finish pigs under commercial production conditions. The present study was thus addressed to this virtually unanswered question. One hundred and two barrows and 102 gilts weighing approximately 24 kg were fed phase I and II grower diets with a high, medium, or low plane of nutrition (HP, MP, or LP) to approximately 43 and 70 kg, respectively, in 6 replicates (pens). Subsequently, the HP and MP groups were fed the HP and MP1 finisher diets, respectively, the LP group being fed a second MP (MP2) finisher diet (LP1 group). Moreover, 68 LP-grower-fed barrows and gilts were added to the feeding trial and fed the MP1 and LP finisher diets to approximately 95 kg and thereafter, respectively (LP2 group). All MP diets had the lysine:calorie ratios comparable to the RNC recommendations, with < 18% differences between those of the HP and LP diets. The finisher pigs were reared in 16 pens and slaughtered at approximately 115 kg. The gain:feed ratio, but not average daily gain (ADG), was greater for the HP group than for the MP and LP during the grower phase I whereas during the grower phase II, ADG was greater (p < 0.05) for the HP and LP groups vs. MP. During the finisher phase I, ADG was less for the LP (LP1 + LP2) group vs. HP and MP, with no difference between the HP and MP groups; the gain:feed ratio was less for the LP vs. MP group. Backfat thickness was greater for the LP vs. HP group. The water holding capacity of fresh longissimus dorsi muscle (LM) and the sensory juiciness score for cooked LM were greatest for the LP group, the sensory flavor and tenderness scores being greater for the LP group vs. MP. In conclusion, results suggest that compensatory growth occurred for the LP and MP groups during the grower phase II and finisher phase I, respectively, with fat deposition increased for the LP group and that meat quality could be improved by the use of LP.
