Light Quality and Photoperiod Affect Growth of Sowthistle (Ixeris dentata Nakai) in a Closed-type Plant Production System (밀폐형 식물생산시스템에서 광질과 광주기에 따른 씀바귀의 생육)

  • This study was conducted to examine the optimal environmental condition for promoting the growth of sowthistle as affected by light quality and photoperiod in a closed-type plant production system. Seeds were sown in 240-cell plug trays and then germinated for 3 days at a 24-hour photoperiod in a closed-type plant production system with LED lights (R:B:W = 8:1:1). Seedlings were transplanted and grown under 3 types of LED (R:B:W = 8:1:1, R:W = 3:7, or R:B = 8:2) and 4 photoperiods (24/0, 16/8, 8/16, or 4/20 hours) with $230{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ light intensity at a density of $20cm{\times}20 cm$. The experimental design was a randomized complete block design. Plants were cultured for 40 days un der the condition of $21{\pm}2^{\circ}C$ and $70{\pm}10%$ relative humidity after transplanting. Plants were fed with a recycling nutrient solution (pH 7.0 and EC $2.0dS{\cdot}m^{-1}$) contained in a deep floating tank. Fresh weight and dry weight of shoot or root, leaf length, and leaf area were the greatest in the photoperiod of 24/0 (light/dark) with RW LED. The highest number of leaves occurred in the photoperiod of 16/8 (light/dark) with RB LED, while the incidence of tip burn was higher in the photoperiod of 24/0 (light/dark) compared to the other treatments. Chlorophyll value was the highest in the 16/8 (light/dark) photoperiod and there was no significant difference by light quality. Chlorophyll fluorescence was the lowest in the photoperiod of 24/0 (light/dark) compared with other treatments. Therefore, in terms of economic feasibility and productivity for Ixeris dentata Nakai cultivation in a closed-type plant production system, the results obtained suggest that plants grew the best when kept in a photoperiod of 16/8 (light/dark) and light quality of combined LED RW (3:7).

  • We characterized the one-electron reduction of oligodeoxynucleotides with a 2-oxopropyl group on a thymine base ($d^{oxo}T$) and applied the reaction to the radiolytic activation of DNAzyme function. We designed a system in which the DNAzyme function of cleaving mRNA was suppressed by introduction of $d^{oxo}T$ into the strand of DNAzyme. Hypoxic X-irradiation led to recovery of the cleavage ability because the 2-oxopropyl group was removed to form unmodified DNAzyme. We characterized the DNAzyme function by monitoring the fluorescence change of fluorophore- and quencher-labeled target strands. We confirmed that the DNAzyme function could be regulated by hypoxic X-irradiation and the reaction of $d^{oxo}T$.

  • A FRET-based high-throughput screening system was developed for the discovery of competitive smallmolecule Hsp90 inhibitors. The biarsenical fluorescein derivative FlAsH and dabcyl-conjugated Hsp90 inhibitor GM were employed as the FRET donor and quencher, respectively. The spatial proximity perturbation between FlAsH-labeled Hsp90N and GM-dabcyl upon treatment of a small molecule led to changes in the FRET-induced fluorescence, monitored in a high-throughput fashion.

Toxicity and Biomedical Imaging of Fluorescence-Conjugated Nanoparticles in Hematopoietic Progenitor Cells

  • Cellular uptake of nanoparticles for stem cell labeling and tracking is a critical technique for biomedical therapeutic applications. However, current techniques suffer from low intracellular labeling efficiency and cytotoxic effects, which has led to great interest in the development of a new labeling strategy. Using silica-coated nanoparticles conjugated with rhodamine B isothiocyanate (RITC) (SR), we tested the cellular uptake efficiency, biocompatibility, proliferation or differentiation ability with murine bone marrow derived hematopoietic stem/progenitor cells. The bone marrow hematopoietic cells showed efficient uptake with SR with dose or time dependent manner and also provided a higher uptake on hematopoietic stem/progenitor cells. Biocompatibility tests revealed that the SR had no deleterious effects on cell cytotoxicity, proliferation, or multi-differentiation capacities in vitro and in vivo. SR nanoparticles are advantageous over traditional labeling techniques as they possess a high level of cellular internalization without limiting the biofunctionality of the cells. Therefore, SR provides a useful alternative for gene or drug delivery into hematopoietic stem/progenitor cells for basic research and clinical applications.

  • Transoral microsurgery has come a long way as a go-to surgical intervention technique for head and neck cancers. This minimally invasive procedure had gained acceptance through comparative clinical studies against radical neck surgical procedures, radiotherapy, and chemotherapy. Laser technology has vastly improved the oncological outcomes of this procedure and brought about an appreciation of transoral laser surgery (TLM) as a mainstay for re-sectioning malignant tumors along the throat. As an established procedure, TLM has undergone several upgrades regarding the different energy devices used for cutting, ablation, and hemostasis. Continued advances in automation have eventually led to surgical robotics, resulting in the emergence of transoral robotic surgery (TORS) as a viable advanced alternative for TLM. Similarly, expansions and enhancements (image-based guidance, fluorescence spectroscopy, and advanced robotic system) have also been investigated as potential upgrades for TORS. This paper reviews a selection of publications on the significant technological advancements to TLM and TORS over the past five years.

Detecting Incipient Caries Using Front-illuminated Infrared Light Scattering Imaging

  • A new method for early caries diagnosis was proposed and tested through a home-made optical examination system that used quantitative light fluorescence (QLF) and digital imaging fiber optic transillumination (FOTI) (DIFOTI), with light sources across a wide spectral range, from 350 nm to 1,000 nm. The front-illuminated infrared light scattering image (FIR) showed similar diagnostic abilities to that of DIFOTI. The FIR method was invented based on the observation that caries lesions lose the high transmittance and low scattering properties of benign enamel tissue. There are various methods for the early diagnosis of caries, such as visual examination, exploration, X-ray radiography, QLF, FOTI, and infrared fluorescence (diagnodent). Among them, methods based on optical properties are regarded as having the most potential. A comparative study was performed between the FOTI, QLF, diagnodent, optical coherence tomography, and FIR scattering image methods, using 20 extracted teeth samples with early caries. A scale of lesion measurement based on optical image contrast was proposed. The statistical analysis showed a significant correlation between the DIFOTI and FIR methods (r = 0.35, p < 0.05). However, the QLF and diagnodent methods showed little association with FIR images, as they have different detection principles as compared with FIR. Tomographic images obtained by OCT, using 1,330 nm super luminescent LED as a gold standard of tooth structure, verified that the FOTI and FIR results correctly represented the lack of homogeneity in dental tissue. The newly proposed FIR method attained similar diagnostic results to those of FOTI, but with an easier approach.

Changes in Spectroscopic and Molecular Weight Characteristics of Dissolved Organic Matter in an Agriculture Reservoir during a Summer Monsoon (장마시기에 따른 농업용 저수지 내 용존 유기물 분광특성과 분자량 변화)

  • In this study, we investigated the variations of dissolved organic matter (DOM) in an agricultural reservoir during the monsoon period (June to October, 2020) with respect to the organic carbon concentration (DOC), molecular weight distribution, and optical properties. The monsoon period was divided into three phases - beginning storm (BS), during storm (DS), and after storm (AS). Our results showed significant differences in the concentrations and characteristics of DOM during the summer monsoon. The DOC concentrations were decreased after the monsoon, probably due to a dilution effect. In contrast, increasing trends were observed in the specific UV absorbance (SUVA), and relative abundances of humic-like fluorescence and larger-sized compounds. These observations implied that the large-sized and humic-like organic components with terrestrial origins strongly affected the reservoir DOM after the summer monsoon. Meanwhile, biopolymer size fraction, which is associated with algal activity, became more abundant after the monsoon. These results suggest that DOM with autochthonous sources became dominant as a result of the inflow of nutrients into the reservoir after the storm. Spatial changes in DOM within the reservoir were not pronounced as much as the temporal variations. All taken, it can be concluded that the summer monsoon simply led to the decrease of DOM concentrations while the sources and the quality of DOM underwent substantial changes, which may enrich refractory organic matter in the reservoir. This study reveals the importance of in-depth DOM quality monitoring before and after summer monsoon for effective water quality management in agricultural reservoirs.

Changes in Root Water Uptake and Chlorophyll Fluorescence of Rice (Oryza sativa L. cv. Dongjin) Seedling under NaCl Stress (NaCl 스트레스에 따른 벼 유식물의 뿌리 수분흡수와 엽록소형광의 변화)

  • The physiological and photochemical responses of rice seedling to NaCl stress were investigated through measuring leaf relative water content (RWC), root water uptake and chlorophyll fluorescence. When plants were exposed to increased salinity stress, the visual symptoms of injury were significant at ${\geq}$500 mM NaCl concentration for 4 and 5 day stress periods. The differences in Fv/Fm between control treatment and plants treated with 500 mM and 1,000 mM NaCl were evident after 5 day and 4 day, respectively, whereas in root water uptake its effect was observed at 500 mM and 1,000 mM NaCl at 2 day of salt-stressed periods. Leaf RWC in salt-stressed plants decreased gradually with increasing salinity in exogenous solution and duration of salt stress, and these decrease showed leaf RWC of 58-68% atduration over 2 day stress of 1,000 mM NaCl treatment and 88% at 1 day stress. NaCl stress led to a significant inhibition of the light-induced greening in etiolated rice plants, especially in 4 and 5 day salt-stressed plants, which linearly decreased with NaCl concentration ($R^2$=0.812 and 0.918, respectively). The effects of NaCl stress in rice seedlings indicate that water uptake in root is more sensitive to increasing NaCl concentration and stress duration than Fv /Fm in leaves compared with the same NaCl concentration.

Steroidal Saponins from Paris polyphylla Induce Apoptotic Cell Death and Autophagy in A549 Human Lung Cancer Cells

  • Background: Paris polyphylla (Chinese name: Chonglou) had been traditionally used for a long time and shown anti-cancer action. Based on the previous study that paris polyphylla steroidal saponins (PPSS) induced cytotoxic effect in human lung cancer A549 cells, this study was designed to further illustrate the mechanisms underlying. Materials and Methods: The mechanisms involved in PPSS-induced A549 cell death were investigated by phase contrast microscopy and fluorescence microscopy, flow cytometry and western blot analysis, respectively. Results: PPSS decreased the proportion of viable A549 cells, and exposure of A549 cells to PPSS led to both apoptosis and autophagy. Apoptosis was due to activations of caspase-8, caspase-3, as well as cleavage of PARP, and autophagy was confirmed by up-regulation of Beclin 1 and the conversion from LC3 I to LC3 II. Conclusions: PPSS was able to induce lung cancer A549 cell apoptosis and autophagy in vitro, the results underlining the possibility that PPSS would be a potential candidate for intervention against lung cancer.

Peroxynitrite Scavenging Activity and its Mechanism of Cheonga-hwan (청아환의 Peroxynitrite 제거 활성 및 기전)

  • Objectives: Peroxynitrite ($ONOO^{-}$), formed from the reaction of superoxide <${\cdot}O_2^{-}$) and nitric oxide (NO), is a cytotoxic species that can oxidize several cellular components such as proteins, lipids and DNA. It has been implicated in diseases such as aging process, Alzheimer's disease, rheumatoid arthritis, cancer and arteriosclerosis. Due to the lack of endogenous enzymes responsible for $ONOO^{-}$ inactivation, developing a specific $ONOO^{-}$ scavenger is of considerable importance. The aim of this study was to evaluate $ONOO^{-}$ scavenging activity and its mechanism in Cheonga-hwan (CAH). Methods: The $ONOO^{-}$ scavenging activity in CAH was assayed by measuring oxidized dihydrorhodamine 123 (DHR 123) by fluorescence. The scavenging efficacy was expressed as $IC_{50}$, showing the concentration of each sample required to cause 50% inhibition of DHR 123 oxidation. In a separate study, the protective effect of CAR on $ONOO^{-}$-induced nitration of bovine serum albumin (BSA) was investigated using immunoassay with a monoclonal anti-nitrotyrosine antibody, and a horseradish peroxidase-conjugated anti-mouse secondary antibody from sheep. Results: CAH showed potent scavenging activities of $ONOO^{-}$, NO and ${\cdot}O_2^{-}$. The data demonstrated that CAH led to decreased $ONOO^{-}$-mediated nitration of tyrosine through electron donation. CAH showed significant inhibition on nitration of bovine serum albumin by $ONOO^{-}$ in a dose-dependent manner. Conclusions: CAH can be developed as an effective peroxynitrite scavenger for the prevention of the $ONOO^{-}$ involved diseases.
