Common plasma protein marker LCAT in aggressive human breast cancer and canine mammary tumor

  • Breast cancer is one of the most frequently diagnosed cancers. Although biomarkers are continuously being discovered, few specific markers, rather than classification markers, representing the aggressiveness and invasiveness of breast cancer are known. In this study, we used samples from canine mammary tumors in a comparative approach. We subjected 36 fractions of both canine normal and mammary tumor plasmas to high-performance quantitative proteomics analysis. Among the identified proteins, LCAT was selectively expressed in mixed tumor samples. With further MRM and Western blot validation, we discovered that the LCAT protein is an indicator of aggressive mammary tumors, an advanced stage of cancer, possibly highly metastatic. Interestingly, we also found that LCAT is overexpressed in high-grade and lymph-node-positive breast cancer in silico data. We also demonstrated that LCAT is highly expressed in the sera of advanced-stage human breast cancers within the same classification. In conclusion, we identified a possible common plasma protein biomarker, LCAT, that is highly expressed in aggressive human breast cancer and canine mammary tumor.

고콜레스테롤혈증 흰쥐의 유리지방산, Creatine Phosphokinase 및 LCAT 활성에 돌복숭아(Prunus persica Batsch var. davidiana Max.)추출액이 미치는 영향 (Effect of Prunus persica Batsch var. davidiana Max. Extract on the Free Fatty Acid, Creatine Phosphokinase and LCAT Activities in Hypercholesterolemic Rats)

  • 식이성 고콜레스테롤혈증 유발 Sprague Dawley계 숫 흰쥐를 사용하여 돌복숭아(Prunus persica Batsch var. davidiana Max.) 생리활성물질 추출액을 급여하므로서, 혈청 유리지방산, creatine phosphokinase 및 lecithin cholesterol acyltrasferase(LCAT) 활성 등 생체 내 지질 대사 이상 및 혈청 지질 개선 효과를 생리생화학적 측면에서 검토하기 위하여 본 실험을 수행하였다. 기본 식이에 물 급여군인 대조군(BW군)과 돼지기름, 콜레스테롤 등에 의한 고콜레스테롤 유발 식이에 물 급여군(BCW군), 고콜레스테롤혈증 유발 식이에 돌복숭아 $5.0 g\%$ 추출액을 급여한 군(BCP군)을 5주간 실험 사육하였다. 혈청 중 전해질 농도(Na, K, Cl)는 고콜레스테롤혈증 군에 돌복숭아 추출액을 급여하므로서 농도가 다소 감소되는 경향을 보였지만 전해질 농도의 저하효과는 유의성 있게 관찰되지 않았다. 혈청 중 유리지방산, 과산화지질 농도 및 creatine phosphokinase 활성 등은 돌복숭아 추출액을 급여한 군에서 농도와 활성이 감소되는 것으로 나타났다. 반면, 혈청 LCAT 활성은 고콜레스테롤 유발군에 돌복숭아 추출액 급여에 의해 증가되는 것 등으로 미루어 보아, 돌복숭아 중의 생리활성물질이 식이성 고콜레스테롤혈증 흰쥐에 대한 혈청 지질 개선 효과가 있는 것으로 사료된다.

  • The effect of milk in low and high cholesterol diets were investigated on serum cholesterol esterification. Weanling male Sprague-Dawley rats were divided into low(0.01% w/w) and high (1.01% w/w) cholesterol-diets groups. Bothlow and high cholesterol groups were consisted of three groups : control , LM(low heat milk), and HM (high heat milk) groups. After feeding these experimental diets for six weeks, serum cholesterol (free cholesterol and cholestryl ester) concentration and activity of lecithin.: cholesterol acyltransferase (LCAT) were measured, and serum lipoprotein profile was examined using gel column chromatography. According to the result, activity of LCAT was elevated independently by intakes of high cholesterol and milk , which resulted in the increase of daily turnover of serum cholesteryl ester. However, the turnover of HDL-cholesteryl ester increased only by milk. LCAT activity was moderately correlated with levels of total-and HDL-free choelsterol. It is concluded from the present study that milk had the cholesterol-lowering effect which partly appears to be mediated through facilitated reverse cholesterol transport.

고 불포화 지방산 식이가 인체 혈청 Cholesterol의 Esterification과 Transfer에 미치는 영향 (Effects of High Polyunsaturated Fat Diet on Human Plasma Cholesterol Esterification and Transfer)

  • Lecithin Cholesterol Acyltransferase(LCAT)와 cholesteryl ester transfer protein(CETP)은 간과 간외조직간의 혈청 cholcsterol 항상성을 유지하는데 중요한 기전인 reverse cholestrol transport system(RCT)에 매우 중요한 인자들이다. 본 연구의 목적은 RCT 기전의 식이 지질 효과를 추정하여 항 고지혈증 치료식이 연구의 중요한 자료를 제공하는데 있다. 4주의 인체대사 실험에서, 새당사 12명의 여성들은 실험 1주 전에 고 포화지질로써 적응시기를 거친 후, 각각 6명씩 high PUFA(corn oil)군과 high SFA(butter)군에 무작위로 배정되었다. Butter군은 total-34(%), esterified-(96%), $HDL_3$-(23%), LDL-(20%) 및 VLDL+LDL-(35%) cholestetol을 감소시켰다. Corn oil군은 esterified(2.5%) 및 LDL-(15%) cholesterol과 triglycerde(27%)를 감소시켰다. Corn oil과 butter fat군간의 식이 효과 차이는 total-(p=0.0001), esterified-(p=0.0001), total HDL-(p=0.005), $HDL_2$-(p=0.01) 및 LDL-(p=0.0001) cholesterol에서 유의적이다. LCAT activity는 두 군에서 변화가 없으나, 4주 후 CETP activity는 butter군에서 2.5배 증가하였다. 이는 VLDL+LDL cholesterol 농도가 butter군에서 증가한 결과와 일치한다. LCAT activity는 corn oil군에서 증가된 HDL의 apo A-I 농도와 free cholesterol과 정의 상관관계가 높은 반면, CETP activity는 total cholesterol과 LDL 및 VLDL+LDL cholesterol과 정의 상관관계가 높았다.

동충하초(Cordyceps militaris) 추출물이 당뇨성 질환 모델의 NEFA 및 LCAT 활성 조절작용에 관한 연구 (Regulatory Actions of the Cordyceps Militaris Extract on the NEFA and LCAT Activity in Diabetic Disease Model System)

  • This study was conducted to investigate the effects of the Cordyceps militaris extract on the improvement of the glucide metabolism in serum of streptozotocin (STZ, 50 mg/kg BW, IP injection)-induced diabetic rats (SD strain, male) fed the experimental diets for 5 weeks. Concentrations of creatinine, lipid peroxide and NEFA in sera were significantly higher in the STZ-induced diabetic group (group DMG) and STZ+Cordyceps militaris extract administration group (group DMS) than those in the control group (group BD). However, the concentrations of creatinine, lipid peroxide and NEFA in sera were reduced in the group DMS than those in the group DMG. The activity of creatine phosphokinase (CPK) in serum was lower in the group DMS than in the group DMG. The activity of LCAT in serum was increased in the group DMS (Cordyceps militaris extract administration) than in the group DMG. The results indicate that Cordyceps militaris extract were effective in the improvement of the glucide metabolism in the sera of STZ-induced diabetic rats.

Hypocholesterolemic Effects of Soybean Lecithin in Cholesterol-Fed Rats

  • The present study was performed to elucidate the hypocholesterolemic action of lecithin on the diet-induced hypercholesterolemia in rats. Male Sprague-Dawley rats (n=24) were fed lecithin-free (control) diet or diets containing $2\%\;or\;5\%$ lecithin for 4 weeks. Hypercholesterolemia was induced by adding $1\%$ cholesterol and $0.5\%$ cholic acid to all diets. No difference was found in food intake and body weight gain among groups. The lecithin treated groups showed significant improvement in the plasma levels of total cholesterol and LDL-cholesterol (p<0.05) compared to the control group, while the plasma triacylglyceride was not significantly affected 1he atherogenic index and HDL-cholesterol level were decreased in the lecithin groups. The diets with $2\%\;or\;5\%$ lecithin significantly decreased the activity of cholestetyl ester transfer protein (CETP) by $14\%\;or\;17\%$, respectively. Also, lecithin diets increased the activity of lecithin: cholesterol acyltransferase (LCAT). These results suggest that lecithin accounts for the hypocholesterolemic effect due to the decreased CETP activity and increased LCAT activity.

식이지방의 종류와 수준이 흰쥐의 Lecithin : Cholesterol Acyltransferase 활성 및 콜레스테롤대사에 미치는 영향 (Effects of Dietary Lipid Sources and Levels on Lecithin : Cholesterol Acyltansferase Activity and Cholesterol Metabolism in Rats)

  • 본 실험은 식이내 지방의 종류와 수준이 흰쥐의 LCAT활성 및 콜레스테롤 대사에 어떻게 영향을 미치는가를 알아보기 위하여 실시되었다. 식이지바의 종류는 정어리유($\omega$3 linolenic acid) 및 옥수수기름 ($\omega$6 linolic acid)의 네 처리로 나누고 각 처리마다 지방의 수준을 총 에너지의 10%, 20%, 40%등 세수준으로 모두 12군을 두었으며, 식이지바의 무첨가군을 대조구로 두었다. 총 78마리의 Sprague-Dawley 계통 흰쥐 수컷에게 시험식이를 6주간 자유급여하였다. 사양시험 종료 후 간조직에서의 콜레스테롤 합성능력을 알아보기 위해 흰쥐의 간조직을 채취하여 in vitro 배양시험을 실시하였다. 본 시험에서 얻어진 결과를 요약하면 다음과 같다. 1) 혈장 총 콜레스테롤, HDL-콜레스테롤. LDL-콜레스테롤농도 및 HDL-C/T-C비율은 식이지방의 종류에 의해 유의하게(p<0.001) 영향을 받았다. ($\omega$3지방산을 많이 함유한 군(정어리유와 들기름)이 ($\omega$6지방산과 포화지방산을 많이 함유한 군(옥수수기름과 우지)에 비해 혈장내 HDL-콜레스테롤농도가 유의성있게(p<0.001)증가하였고, LDL-콜레스테롤농도는 유의성있게(p<0.05) 감소하였다. 혈장내 콜레스테롤농도는 우지군이 다른지방군에 비해(p<0.001),무지방식이군이 식이지방첨가군에 비해 유의하게(p<0.05)영향을 받았다. 2) 혈장 lecithiin : cholesterol acyltransferase(LCAT) 활성은 식이지방첨가군이 무지방식이군보다(p<0.001), 식물성유지군이 동물성지방군보다(p<0.001), 정어리유군이 우지군보다(p<0.001), 들기름군이 옥수수기름군보다(p<0.05), 정어리유군이 들기름군보다 유의성있게(p<0.05) 증가하였다. 저지방식이군이 중지방식이군과 고지방식이군에 비해 혈장내 LCAT 활성이 유의하게9p<0.001) 증가하였다. 3) 간의 콜레스테롤합성 능력은 정어리유군이 다른지방군보다(p<0.001), 무지방식이군이 식이지방첨가군보다(p<0.001), 동물성지방군의 식물성유지군보다 유의하게(p<0.001) 증가하였으나, 식이 지방의 수준에 의한 차이는 나타나지 않았다.

동충하초 엑스의 고콜레스테롤혈증 대사효소 활성 변동에 관한 효과 (Studies of the Cordyceps militaris Extract Administration on the Metabolic Enzyme Activities in Hypercholesterolemia)

  • The objective of this study was to investigate the effects of the feeding Cordyceps militaris extract on the improvement of the free fatty acid, lipid peroxide, creatinine and enzyme (creatine phosphokinase; CPK, lactate dehydrogenase; LDH, alkaline phosphatase; ALP, lecithin cholesterol acyltransferase; LCAT) activities in the sera of dietary hypercholesterolemic rats (SD strain, male) fed the experimental diets for 5 weeks. Concentrations of free fatty acid, lipid peroxide and CPK, LDH, ALP activities in sera were fairly reduced in the Cordyceps militaris extract administration group (CHE) than in the hypercholesterolemic diet group (CHD). However, no significance was found in the effect of an creatinine concentration among the groups. The LCAT activity in serum was increased in the Cordyceps militaris extract administration (CHE) than in the hypercholesterolemic diet group (CHD). From these results, Cordyceps militaris extracts were effective on the improvement of the lipid components and metabolic enzyme activities in sera of dietary hypercholesterolemic rats.

Studies on the Distribution of Plasma Lipid Profiles and Body Fatness According to Apo E Polymorphism in Normolipidemic Korean Women

  • Apo E polymorphism(e2, e3, e4) was among the first reported genetic polymorphism that explained part of the normal vairation in plasma cholesterol concentrations. Both alleles E2 and E4 are significantly more frequent in patients with mixed forms of hyperlipidemia and contribute on the observed differences in CHD risk among different populations. Effects of apo E polymorphism on the distribution of plasma lipid profiles were studied in 89 normolipidemic healthy females, aged 19 up to 22 years. The relative frequencies of E3/3 was 0.787, E3/2 was 0.101, E3/4 allele was 0.112 and no E2/2, E2/4 and E4/4 were found. Weight, height and %LBM were elevated in E2 than those in E3&E4. No differences in the blood pressure among apo E isomers were found, otherwise the pulsation was higher in E4 than that in the others. There were no differences in plasma total-, total DL-, HDL$_3$-, HDL$_2$ cholesterol, apo B-100 and apo A-I, However, phenotype means rank E3/2>E3/3>E3/4 in average TG levels(p<0.0001) significantly, and rank E3/4>E3/3>E3/2 in LDL cholesterol levels. These results were related to the correlation between atherogenic indiced (AI) such as LDL/HDL, (TC-HDL)/HDL, HDL$_3$/HDL$_2$. The ratio of HDL$_3$& HDL$_2$was significantly increased in E2 & E4 than that in E3(P=0.043). LCAT activity was not different between E2 and E3 but was highly increased in E4 (p<0.0001 among apo E isomers), but CETP was not different. Since the negative correlation between LCAT and CETP in apo E2(r=-0.491) was stronger than that in apo E3, E2 allele impacts the clearance of plasma apo E mediated lipoproteins. In conclusion firstly, E4 mediated alteration through LDL or E receptors results in lower TG or higher $\beta$-lipoprotein levels and E2 shows reciprocal effects of E4, respectively. Second, E4 allele was more atherogenic than E2 allele because the higher levels of AI such as HDL$_3$/HDL$_2$ were criticized.
