Expression of Chinese Cabbage Glutathione Reductase Gene in Lettuce (Lactuca sativa L.) (형질전환 상추에서 배추 Glutathione Reductase 유전자의 발현)

  • Cotyledon explants of lettuce were cocultured with Agrobacterium tumefaciens LBA4404::pBKS-GR1 harboring glutathione reductase(GR) gene in MS medium supplemented with 0.1 mg/L NAA and 1.0 mg/L 2ip for 48 hr. These explants were transferred to MS medium supplemented with 0.1 mg/L NAA and 1.0 mg/L 2ip, 50 mg/L kanamycin, and 500 mg/L carbenicillin. After 4 weeks of subculture, kanamycin-resistant shoots were obtained on selection medium. Leaves of putative transformants survived on selection medium containing 100 mg/L kanamycin. Incoporation of the GR gene into lettuce was confirmed by PCR analysis of genomic DNA. Southern blot analysis showed that ECL-labeled GR gene was hybridized to the expected amplified genomic DNA fragment of about 1.8 kb from transgenic lettuce. The presence of mRNA in transgenic lettuce was confirmed by RT-PCR with total RNA of transgenic lettuce. In progeny test of transformants, R$_1$ seeds were resistant to kanamycin (200mg/L) on MS medium.

Transformation of Rice (Oryza sativa L.) with Sucrose Transporter cDNA from Potato (Solanum tuberosum L.) (감자 Sucrose Transporter 유전자의 벼 Genome 내로의 도입)

  • The transport and allocation of photosynthetic assimilate is an important regulatory factor in plant productivity, In order to modify assimilate partitioning in rice, transgenic plants containing a potato sucrose transporter (SuT) gene were developed. Calli derived from rice seeds (Oryza sativa L. cv Dongjin) were cocultured with A. tumefaciens LBA 4404 harboring the SuT gene. Calli were transferred to MS medium supplemented with 50 mg/L hygromycin, 500 mg/L carbenicillin, 2 mg/L kinetin, 0.1 mg/L NAA. After 2 weeks, hygromycin resistant shoots were obtained from the calli on the selection medium. Roots were induced from the putative transgenic shoots on rooting MS medium supplemented with 250 mg/L cabenicillin. Plant regeneration rate from the calli was about 150%. Stable incorporation of the potato SuT gene into rice genomic DNA was confirmed by PCR and Southern blot analysis.

The increased GUS gene inactivation over generation in Arabidopsis transgenic lines (애기장대 형질전환 식물체의 세대경과에 따른 GUS유전자의 비활성화에 관한 연구)

  • The effect of transgene inactivation in T2, T3 and F2 generations was analyzed in progeny seedlings which had been generated by Agrobacterium (LBA4404/pBI121)-mediated transformation in Arabidopsis thaliana. In a system which investigated in the expression of $\beta$-glucuronidase(GUS)gene in kanamycin-resistant (ke $n^{R}$)seedlings, GUS inactivated seedlings were observed in 5 of 12 tested lines of T2 generation and the frequency of GUS inactivation was approximately 2.3%. Lines with multi-copies of T-DNA exhibited severe GUS gene inactivation with the frequency of 5.8% in T2 generation. In T3 generation lines exhibited GUS gene inactivation with the frequency of 1.3%. In contrast, inactivation increased dramatically up to 12.6% in multi-copy T-DNA line. A similar phenomenon was also found in F2 progeny from a transgenic line which had been crossed with wild-type Arabidopsis plant, WS-O (GUS gene inactivation frequency 9.9%). These results indicate that the foreign gene introduced into the plant was inactivated progressively in its transmission during subsequent generations and the transgenic line with multi-copies of T-DNA tended to show more increased inactivation.

Construction of an Agroinfectious Clone of a Korean Isolate of Sweet Potato Symptomless Virus 1 and Comparison of Its Infectivity According to Agrobacterium tumefaciens Strains in Nicotiana benthamiana

  • Sweet potato symptomless virus 1 (SPSMV-1) is a single-stranded circular DNA virus, belonging to the genus Mastrevirus (family Geminiviridae) that was first identified on sweet potato plants in South Korea in 2012. Although SPSMV-1 does not induce distinct symptoms in sweet potato plants, its co-infection with different sweet potato viruses is highly prevalent, and thus threatens sweet potato production in South Korea. In this study, the complete genome sequence of a Korean isolate of SPSMV-1 was obtained by Sanger sequencing of polymerase chain reaction (PCR) amplicons from sweet potato plants collected in the field (Suwon). An infectious clone of SPSMV-1 (1.1-mer) was constructed, cloned into the plant expression vector pCAMBIA1303, and agro-inoculated into Nicotiana benthamiana using three Agrobacterium tumefaciens strains (GV3101, LBA4404, and EHA105). Although no visual differences were observed between the mock and infected groups, SPSMV-1 accumulation was detected in the roots, stems, and newly produced leaves through PCR. The A. tumefaciens strain LBA4404 was the most effective at transferring the SPSMV-1 genome to N. benthamiana. We confirmed the viral replication in N. benthamiana samples through strand-specific amplification using virion-sense- and complementary-sense-specific primer sets.

Interrelationships and Differences of Brachial-Ankle Pulse Wave Velocity (baPWV), Body Composition and Cardiovascular Variables between Genders Who Have Been Exercised or Not in Elderly (운동참여 여부와 성별에 따른 노인의 맥박파전파속도, 신체구성과 심혈관계 변인의 상관성 및 재변인의 차이)

  • This study sought to investigate interrelationships and differences of brachial-ankle pulse wave velocity (baPWV), body composition and cardiovascular variables between genders who have been exercised or not in elderly. One hundred fifty participants were classified four groups. Especially out of variables, in male elderly group who had participated in exercise program the PWV range were significantly correlated with muscle mass (r=0.357, p=0.026), SBP (r=0.468, p=0.003), right baPWV (r=0.406, p=0.010), and left baPWV (r=0.333, p=0.038). In male elderly group who had not participated in exercise program, the PWV range were significantly correlated with heart rate (r=0.395, p=0.014), right baPWV (r=0.598, p=0.000), and left baPWV (r=0.602, p=0.000). In female elderly group who had participated in exercise program, the PWV range were significantly correlated with name of diagnosis (r=0.321, p=0.044), SBP (r=0.399, p=0.011), DBP (r=0.545, p=0.000), right baPWV (r=0.648, p=0.000), and left baPWV (r=0.676, p=0.000). In female elderly group who had not participated in exercise program, the PWV range were significantly correlated with age (r=0.471, p=0.003), right baPWV (r=0.836, p=0.000), and left baPWV (r=0.801, p=0.000). The PWV among four groups were not significant different after experiment. However, there were significant differences in the waist/hip ratio (F=9.197, p=0.000), muscle mass (F=74.295, p=0.000), and %fat (F=35.045, p=0.000) from body composition. And there were significant differences in the SBP (F=3.525, p=0.017) after experiment. In conclusion, these data show that regular exercise is associated with arterial compliance (PWV range, right or left PWV) and differed from genders. In other words, this paper may support the concept that regular exercise program may exert a protective effect on arterial compliance, body composition and cardiovascular systems.

The Expression of Egg Plant Flavonoid 3',5'-Hydroxylase Gene in Tobacco Plants (Nicotiana tabacum cv. Xanthi)

  • The anthocyanin gene encoding flavonoid 3',5'-hydroxylase(F3,5H) was normally expressed in Nicotiana tobacco (Xanthi) plants cocultivated with Agrobacterium tumefaciens LBA4404 carrying egg plant flavonoid 3',5'-hydroxylase cDNA. Northern blot analysis showed the normal expression of F3', 5'H gene from transgenic plants. Here we found the phenotypic differences between transgenic plants and wild-type plants. The petal shape of transgenic plants showed more round shape and around petal tube area was compared to that of wild-type tobacco plants. And the petal color of transgenic plants was much lighter than that of wild-type tobacco plants.

Effect of Sodium Hydrosulfite Solution on Agrobacterium-Mediated Chinese Cabbage Transformation and Transient Expression

  • We investigated chemical-wounding effect on Agrobacterium-mediated Chinese cabbage transformation via vacuum infiltration. Pre-germinated or germinating Chinese cabbage seeds were infiltrated with Agrobacterium tumefaciens LBA4404 cells carrying either GUS gene (pBI121) or hepatitis B virus surface antigen DNA (pBIHBsAg). Prior to agroinfiltration process, the seeds were soaked in sodium hydrosulfite (SHS) solution or just in sterile water as a control. Comparative transformation efficiency was determined by both of histochemistry and ELISA. We could demonstrate that SHS solution treatment especially to 1-day or 2-days old germinating seeds efficiently improved transformation process, and therefore, transient expression level. This strongly indicated that Agrobacterium infection could be facilitated indeed by SHS-causing wounds on Chinese cabbage seeds.

Double-Stranded RNA-Dependent Protein Kinase Gene Expression in Tobacco Plant (연초식물체에서의 dsRNA 의존성 인산화 효소 유전자 발현)

  • 동물계에서 항바이러스와관련된 dsRNA 의존성 인산화 효소(PKR)의 유전자를 식물체에서 발현시킬 경우 PKR에 의한 단백질합성 및 식물바이러스의 증식조절 가능성에 대한 기초자료를 확보하기 위하여 사람에서 분리된 PKR cDNA를 Agrobacterium 방법에 의하여 연초식물체(Nicotiana tabacum cv. Xanthi-nc)로 형질전환시켰다. HindIII/PstI처리에 의해 얻어지는 약 1.8kb의 phPKR cDNA절편을 일련의 유전자 조작 방법을 통하여 식물발현벡터인 pBI121에 도입하여, p12168을 재조합하였다. 이를 A. tumefaciens LBA 4404에 형질전환시켜 연초식물체형질 전환에 이용하였다. 2mg/l BA와 0.5mg/l NAA가 포함되고 100$\mu\textrm{g}$/ml의 kanamycin이 첨가된 MS배지에서 shooting시킨 후 phytohormone이 첨가되지 않은 MS배지상에서 rooting을 시켜 형질전환 연초식물체를 얻었으며, 형질전환식물체는 정상식물체와 유사한 생육양상을 나타내었다. 형질전환식물체의 유전자도입은 hPKR cDNA의 전사부여는 RT-PCR 방법에 의하여 확인되었다.

Honey Bee Based Load Balancing in Cloud Computing

  • The technology of cloud computing is growing very quickly, thus it is required to manage the process of resource allocation. In this paper, load balancing algorithm based on honey bee behavior (LBA_HB) is proposed. Its main goal is distribute workload of multiple network links in the way that avoid underutilization and over utilization of the resources. This can be achieved by allocating the incoming task to a virtual machine (VM) which meets two conditions; number of tasks currently processing by this VM is less than number of tasks currently processing by other VMs and the deviation of this VM processing time from average processing time of all VMs is less than a threshold value. The proposed algorithm is compared with different scheduling algorithms; honey bee, ant colony, modified throttled and round robin algorithms. The results of experiments show the efficiency of the proposed algorithm in terms of execution time, response time, makespan, standard deviation of load, and degree of imbalance.

The Effect of Jujube Extract on the Growth of Lactic Acid Bacteria (대추 추출물이 유산균의 생육에 미치는 영향)

  • This study was carried out to survey the effect of Jujube extract on the growth of 3 strains of lactic acid bacteria(LBA) starter cultures in the MRS broth by the addition of 0, 0.1, 0.5, 1 and 2% extract The pH, titratable acidity and OD of LAB were investigated in order to get fundamental knowledge for the development a new product. The effects of Jujube extract on the growth of LAB were variable depending upon the LAB species and concentrations of Jujube extract significantly (p<0.05). In the results, Jujube extract enhanced the acid production and propagation by the 3 LAB strains with increasing concentration of Jujube extracts until 2.0% added was to the MRS broth medium (p<0.05). Addition of Jujube extract markedly stimulated the acid production and propagation of Lactobacillus acidophilus, Streptococcus thermophilus and Bifidobacterium longum.
