Characterization of L-Galactono-1, 4-lactone Oxidase Purified from Saccharomyces cerevisiae (Saccharomyces cerevisiae에서 분리한 L-Galactono-1, 4-lactone Oxidase의 특성)

  • A partially purified preparation of L-galactonolactone oxidase which catalyzes the last step of L-ascorbic acid biosynthesis was obtained from Saccharomyces cerevisiae ATCc 26787. The purification procedures included Triton X-100 treatment, protamine sulfate precipitation, ammonium sulfate precipitation, DEAE-Sepharose CL-6B ion exchange chromatography, Sephadex G-150 gel filtration chromatography, and Phenyl-Sepharose CL-4B hydrophobic interaction chromatography. The optimum temperature for the enzyme activity was about $34^{\circ}C$ and the optimum pH was 6.8-7.0. The substrate specificity was confined to L-aldonolactones, L-galactono-1,4-lactone and L-gulono-1,4-lactone. An apparent Km value of 0.294mM with L-galactono-1,4-lactone as a substrate was found. By comparing the substrate specificities of this enzyme with those of isofunctional enzymes of higher plants and animals, it becomes evident that the enzyme of S. cerevisiae ATCC 26787 is rather similar to the L-gulonolactone oxidase of animals than the galactonolactone dehydrogenase of higher plants.

  • The medical use of Acer mono MAX. and Betula costata T. sap has had the long history in Korea. So the mineral and sugar contents of these sap gathered in Mt. Jiri area at spring were analyzed to investigate the scientific base of folk remedies by using ion liquid chromatography and high performance liquid chromatography. The values of chlorine and sulfate in Acer mono MAX. were 11.5 mg/l and 176.7 mg/l, and in Betula costata T. 26.5 mg/l and 162.4 mg/l, respectively. The values of potassium, sodium, calcium and magnesium in Acer mono MAX. were 67.9 mg/l, 5.6 mg/l, 73.8 mg/l and 4.5 mg/l, and in Betula costata T. were 152.1 mg/l, 9.7 mg/l, 39.2 mg/l and 5.7 mg/l, respectively. The values of copper, zinc and manganese in Acer mono MAX. were 0.057 mg/l, 0.483 mg/l and 5.071 mg/l, and copper, zinc, mangances and iron in Betula costata T. were 0.038 mg/l, 1.584 mg/l, 4.354 mg/l and 2.511 mg/l, respectively. The values of sucrose in Acer mono MAX. were 27.29 mg/l, glucose and fructose in Betula costata T. were 0.97 g/l and 3.05 g/l, respectively.

  • Six-coordinate molybdenum(Ⅴ)-oxo complexes, (R4N)[MoO(NCS)2L](R=CH3, C2H5, n-C4, H9) with S-methyl-3-(2-hydroxy-x-phenyl)methylenedithiocarbazate(L1: x=5-H) and its derivatives (L2:x=5-CH3, L3: x=3-CH3O, L4: x=5,6-C4H4 and L5: x=5-NO2) have been synthesized and the structural, spectral and electrochemical properties of the complexes have been characterized by elemental analysis, molar conductivity, UV-Vis, IR, 1H NMR, and CV (cyclic voltammetry).

  • This study was performed to compare the differences between total fat area, subcutaneous fat area and visceral fat area according to L1-S1 inter vertebral disc level in asymptomatic Korean patients with non-contrast abdominal CT scan. The location of the maximum fat ratio of the total fat area is the total fat area measured in male L2-3, L3-4, L4-5, female L4-5, and L5-S1. The location showing the maximum fat ratio of the subcutaneous fat area is the subcutaneous fat area measured in male L4-5, and the woman is the subcutaneous fat area measured in L4-5 and L5-S1. The location of the maximum fat ratio of visceral fat area was the visceral fat area measured in L1-2 and L2-3 males, and there was no difference in visceral fat area according to the measured position in females. Although abdominal fat measurement is common at the L4-5 inter vertebral disc level, fat distribution varies widely depending on nationality, race, and gender. Therefore, it is necessary to analyze through the research data which location to choose according to the research purpose, and to mention why the location is appropriate.

Appropriate Block Level in Neurolytic Lumbar Sympathetic Ganglion Block (신경파괴적 요부교감신경절 차단의 적정 부위)

  • Background: The lumbar sympathetic ganglia are variable in both position and numbers. The aim of this study was to detect the appropriate lumbar vertebral level where the lumbar sympathetic ganglia primarily aggregate. Methods: Forty patients comprising of hyperhidrosis (26 cases), complex regional pain syndrome (10 cases), peripheral artery occlusion disease (3 cases) and postherpetic neuralgia (1 case) underwent lumbar sympathetic block. We randomly selected one of two (L2 or L3) levels and the L4 level. The position of the needle tip and distribution of dye was verified by injection of a mixture of radio-opaque dye (1.5 ml) and 4% lidocaine (1.5 ml) and subsequently confirmed by L-spine anteroposterior and a lateral view X-ray. We considered the response positive when the skin temperature increased more than $1^{\circ}C$ in 5 min. Results: In general, the positive response ratio was greater when the needle tip located at the L2 or L4 level vice L3 and when the drug was distributed on the lower half of the L4 body and in the L4/5 intervertebral disc space. In a right side block, the positive response ratio was greater when the drug was distributed on the lower half of the L4 body and in the L4/5 intervertebral disc space, although in a left side block there was no significant difference seen between the levels. The complications of the neurolytic block were alcohol neuritis (7.5%) and hypoesthesia (5%) on the L1 or L2 dermatome. Conclusions: The best effect with least chance of complication may be induced by spreading the drug on the lower half of the L4 body and/or into the L4/5 intervertebral disc space by placing the needle tip on the L4 body.

Comparison of Synthetic Lysine Sources on Growth Performance, Nutrient Digestibility and Nitrogen Retention in Weaning Pigs

  • We compared the effects of supplementing $L-lysine{\cdot}SO_4$ to L-lysine HCl on growth performance, nutrient digestibility and nitrogen retention in weaning pigs. A total of 96 crossbred pigs, weaned at $21{\pm}3$ days of age and with an average initial body weight (BW) $6.23{\pm}0.01kg$, were given one of 4 treatments, which translated into 6 replicates of 4 pigs in each pen. The animals were randomly assigned to four dietary treatments according to a randomized completely block design (RCBD) as follows: 1) control-no synthetic lysine, lysine deficient (0.80% total lysine); 2) L-C (= 0.2% L-lysine HCl); 3) K-L-S (= 0.332% $L-lysine{\cdot}SO_4$, A company); 4) C-L-S (= 0.332% $L-lysine{\cdot}SO_4$, B company). Diets were formulated with corn, soy bean meal, and corn gluten meal as the major ingredients, and all nutrients except the lysine met or exceeded NRC requirements (1998). The lysine content of supplemented synthetic lysine was the same in all treatment groups except the control. No clinical health problems associated with the dietary treatments were observed. During the entire experimental period, body weight, average daily gain (ADG) and feed efficiency (G:F ratio) increased (p<0.01) in pigs fed the experimental diets supplemented with L-lysine??HCl or $L-lysine{\cdot}SO_4$ produced by A company, irrespective of the two synthetic lysine sources. Although the supplementation of $L-lysine{\cdot}SO_4$ produced by B company tended to improve the ADG and G:F ratio, significant differences were not seen among all treatments and tended to be lower than the L-C (L-lysine HCl) and K-L-S ($L-lysine{\cdot}SO_4$ groups using the product from A company). The digestibility of crude protein (CP) was increased by the supplementation of synthetic lysine (p<0.05), irrespective of the L-lysine source (L-C, K-L-S, C-L-S). The results of this study showed that ADG, G:F ratio, and CP digestibility improved when $L-lysine{\cdot}SO_4$ or L-lysine HCl was supplemented into the weaning pigs' diet. There was a clear difference in efficacy between the two $lysine{\cdot}SO_4$ products based upon the growth performance of weaning pigs. Consequently, the bioavailability of $lysine{\cdot}SO_4$ products should be evaluated before supplementation of synthetic lysine in swine diets.

Morphometric Analysis of the Ureter with Respect to Lateral Lumbar Interbody Fusion Using Contrast-Enhanced Computed Tomography

  • Objective : To analyze the anatomical location of the ureter in relation to lateral lumbar interbody fusion and evaluate the potential risk of ureteral injury. Methods : One hundred eight patients who performed contrast-enhanced computed tomographic scans were enrolled in this study. The location of the ureter from L2-L3 to L4-L5 was evaluated. The distances between the ureter and psoas muscle, intervertebral disc, and retroperitoneal vessels were also recorded bilaterally. Results : Over 30% of the ureters were close to the working corridor of extreme lumbar interbody fusion at L2-L3. Most of the ureters were close to working corridor of oblique lumbar interbody fusion, especially at L4-L5. The distance from the ureter to the great vessels on the left side was significantly narrowing from L2-L3 to L4-L5 (28.8±9.5 mm, 22.0±8.0 mm, 15.5±8.4 mm), and it was significantly larger than that on the right side (12.3±6.1 mm, 7.4±5.7 mm, 5.4±4.4 mm). Conclusion : Our findings indicate that the location of the ureter varies widely among individuals. To avoid unexpected damage to the ureter, it is imperative to directly visualize it and verify the ureter is not in the surgical pathway during lateral lumbar interbody fusion.

Effect of Medium Compositions on the Mycelial Growth of Inonotus obliquus (차가버섯의 균사체 성장에 대한 배지성분의 영향)

  • Effect of temperature($22-32^{\circ}C$), pH(5-7) and medium composition on the mycelial growth for the submerged culture of Inonotus obliquus. The concentrations of glucose, starch, peptone, yeast extract, $K_2HPO_4$, $MgSO_4{\cdot}7H_2O$ and $CaCl_2$ were examined in the ranges of 30-120 g/L, 0-10 g/L, 0-20 g/L, 0-15 g/L, 0-2 g/L, 0-1.5 g/L and 0-0.5g/L, respectively. The maximum mycelial growth of Inonotus obliquus was obtained for $26-27^{\circ}C$ and pH 6. The concentrations of glucose, yeast extract and $CaCl_2$, which gave the maximum mycelial growth of Inonotus obliquus, were 70 g/L, 5 g/L and 0.1 g/L, respectively. In the cases of starch, peptone and $K_2HPO_4$, the mycelial growth of Inonotus obliquus increased with increasing the concentrations. However, as the concentration of $MgSO_4{\cdot}7H_2O$ increased, the mycelial growth of Inonotus obliquus decreased. The medium for maximum mycelial growth of Inonotus obliquus consisted of (per 1 L): glucose, 70 g; peptone, 5-20 g; starch, 10 g; yeast extract, 5 g; $K_2HPO_4$, 2 g and $CaCl_2$, 0.1 g.

  • The spinel L $i_{1-x}$ M $n_2$ $O_4$has been synthesized by the solid-state reaction. L $i_{l-x}$M $n_2$ $O_4$which includes a mixture of LiOH . $H_2O$ and Mn $O_2$prepared by preliminary heating at 35$0^{\circ}C$ for 12hr. L $i_{l-x}$M $n_2$ $O_4$fired at temperature range from 75$0^{\circ}C$ for 48hr. The structure and the electrochemical characteristics of spinel to L $i_{1-x}$ M $n_2$ $O_4$which is fabricated by changing sintering condition from starting materials are investigated. The cyclic voltammetric measurement was performed using 3 electrode cells. Electrode specific capacity and cycle life behavior were tested in a 3.0~4.2V range at a constant current density of 0.45mA/c $m^2$. To improve the cycle performance of spinel L $i_{l-x}$M $n_2$ $O_4$as the cathode of 4V class lithium secondary batteries, spinel phases L $i_{1-x}$ M $n_2$ $O_4$were Prepared at various lithium. The results showed that discharge capacity of L $i_{l-x}$M $n_2$ $O_4$varied at lithium quantity decrease with increasing lithium add quantity. The discharge capacities of L $i_{0.925}$M $n_2$ $O_4$and LiM $n_2$ $O_4$revealed 108 and 117mAh/g, respectively.spectively.y.

Synthesis and In-vitro Evaluation of N4-Amino Acid Derivatives of Cytarabine for Improving the Oral Delivery of Cytarabine

  • The present study aimed to investigate the in-vitro characteristics of N4-amino acid derivatives of cytarabine for the oral delivery of cytarabine. After the synthesis of L-Ile-cytarabine, L-Leu-cytarabine and L-Arg-cytarabine, the gastrointestinal stability of each prodrug was examined using artificial gastric juice and intestinal fluids. The cellular uptake characteristics of prodrugs were also examined in Caco-2 cells. While L-Ile-cytarabine and L-Leu-cytarabine appeared to be stable in all the tested biological media during 4-hr incubation, L-Arg-cytarabine was rapidly disappeared within 5 min. Accordingly, the cellular uptake of L-Ile-cytarabine and L-Leu-cytarabine was significantly higher than that of its parent drug, cytarabine in Caco-2 cells but the cellular uptake of L-Arg-cytarabine was similar to that from its parent drug. The cellular uptake of L-Ile-cytarabine and L-Leu-cytarabine appeared to be saturable as drug concentration increased from 0.4 to 4 mM. Collectively, L-Ile-cytarabine and L-Leu-cytarabine could be promising candidates to improve the oral absorption of cytarabine via a saturable transport pathway.
