Effect of GdL Addition on Physico-chemical Properties of Fermented Sausages during Ripening

  • This study investigated the effects of glucono-δ-lactone (GdL) addition on physicochemical and microbiological characteristics of fermented sausages during ripening and drying. Five batches of sausages were produced under ripening conditions: without GdL and with 0, 0.1, 0.25, 0.5 and 0.75% of GdL addition. Samples from each treatment were taken for physicochemical and microbiological analyses on the 0, 1, 3, 5, 7, 10, 15, 20 and 25th day of ripening. Chemical analysis showed a significant decrease in moisture content of sausages with increasing amounts of GdL added (p<0.05). The moisture contents decreased, whereas the fat, protein and ash contents increased throughout ripening (p<0.05). Increasing levels of GdL caused a decrease in the pH values (p<0.05), which can have an inhibitory effect against microflora. Water holding capacity content of samples decreased with increasing GdL concentration (p<0.05). The shear force values of fermented sausages showed the highest in T4 (p<0.05). During ripening, the shear force values of sausages were increased on the 25th day compared to day 0 (p<0.05). The higher GdL level produced lighter and more yellow sausages. The addition of 0.75% GdL was effective in controlling bacteria counts. Addition of GdL in sausages resulted in the physicochemical and microbiological attributes equal to or better than no addition of GdL without any harmful effect.

Production of Mycelia and Water Soluble Polysaccharides from Submerged Culture of Lentinus lepideus in Bioreactor (생물반응기를 이용한 잣버섯(Lentinus lepideus)의 균사체 및 수용성 다당체 생산특성)

  • The mushroom Lentinus lepideus was used to produce mycelial as well as soluble polysaccharides in bioreactor cultures. To determine optimal submerged culture conditions, both growth characteristics and water soluble polysaccharides production were compared among four different types of bioreactor and culture conditions. For the production of mycelial biomass, the following bioreactors were proven to be effective in decreasing order: an external-loop type air-lift bioreactor (ETAB; 7g/l), a balloon type air bubble bioreactor (BTBB; 6.2g/l), a stirrer type bioreactor (STB; 6g/l), and a column type air bubble bioreactor (CTBB; 5g/l). Maxiaml production of water soluble exopolysaccharides (EPS; 0.62g/l) and endopolysaccharides (PPS; 7.7%) could also be obtained from BTBB. The mycelial biomass increased with increase in glucose concentration from 15g/l to 75g/l in the media. In contrast, PPS contents in the cells decreased with increase in glucose concentration in the media, showing the highest PPS content (7%) at 15g/l. Among different medium feeding types, fed-batch culture based on concentration control in media (10g/l) produced higher mycelia than fed-batch culture based on volume control of media (5.8g/l) or batch culture (3.4g/l). EPS production was also higher in fed-batch culture based on medium concentration control than that in other feeding types.

Optimization of Media Composition and Cultivation for the Mycelial Growth of Agrocybe Cylindracea (Agrocybe Cylindracea의 영양배지 조성 및 배양조건의 최적화)

  • Studies were made to optimize the meida composition and cultural conditions for the mycelial growth of Agrocybe cylindracea. Media composition for optimal growth was found to be starch 20.0g/l, bacto-soytone 4.0g/l. The media supplemented with $KH_2PO_4\;0.46g/l,\;K_2HPO_4\;1.0g/l,\;MgSO_47H_2O\;0.5g/l$ supported mycelial growth better than the media without mineral salts. Optimum temperature and pH for the growth was $28^{\circ}C$, and 6.0 respectively. Temperature range for the mycelial growth appeared to be $10-35^{\circ}C$ and the mycelium evidently lost the vitality at $40^{\circ}C$.

  • $\beta$-Glucuronidase (GUS) gene of Escherichia coli was introduced into sweet potato (Ipomoea batatas (L.) Lam.) cells by particle bombardment and expressed in the regenerated plants. Microprojectiles coated with DNA of a binary vector pBI121 carrying CaMV35S promoter-GUS gene fusion and a neomycin phosphotransferase gene as selection marker were bombarded on embryogenic calli which originated from shoot apical meristem-derived callus and transferred to Murashige and Skoog (MS) medium supplemented with 1 mg/L 2,4-dichlorophenoxyacetic acid and 100 mg/L kanamycin. Bombarded calli were subcultured at 4 week intervals for six months. Kanamycin-resistant calli transferred to MS medium supplemented with 0.03 mg/L 2iP, 0.03 mg/L ABA, and 50 mg/L kanamycin gave rise to somatic embryos. Upon transfer to MS basal medium without kanamycin, they developed into plantlets. PCR and northern analyses of six regenerants transplanted to potting soil confirmed that the GUS gene was inserted into the genome of the six regenerated plants. A histochemical assay revealed that the GUS gene was preferentially expressed in the vascular bundle and the epidermal layer of leaf, petiole, and tuberous root.

  • Nonionic surfactants, diglycerol N-acyl glutamates were prepared through the esterification of N-acryl glutamic acids and glycidol in the presence of quarternary ammonium salts. Their cmc evaluated by the surface tension method was $0.15{\sim}0.75mol/{\ell}$, and the surface tension of aqueous solution was decreased to 28~35dyne/cm. L-DGLG with acyl group of carbon number of 12 showed good foaming power and emulsifying power, and the emulsifying power in toluene was stronger than in soybean oil. Dispersion effect of L-DGPG and L-DGSG with acyl group of carbon number of 16 and 18 in carbon black and ferric oxide were higher than L-DGLG. All of them showed good biodegradability at $20^{\circ}C$.

  • To evaluate the anti-oxidant and anti-tumor potential of the green pepper (Capsicum annuum L. cv. DangZo), total polyphenol content, radical scavenging activities and anti-tumor properties were measured. The total polyphenol content of the 70% ethanol extracts from green pepper (Capsicum annuum L. cv. DangZo) was 30.29 mg gallic acid equivalent/g extract. The DPPH radical and hydroxyl radical scavenging activities of 70% ethanol extracts of green pepper (Capsicum annuum L. cv. DangZo) were documented at 2.87 and 10.55, respectively. For ${\alpha}$-glucosidase and ${\alpha}$-amylase inhibitory activity, 70% ethanol extracts of green pepper (Capsicum annuum L. cv. DangZo) were documented at 35.67% and 58.41% respectively. The green pepper (Capsicum annuum L. cv. DangZo) demonstrated greater capability in terms of anti-neoplastic activity vis-a-vis colon cancer cell lines when compared to other cancer cell lines.s. er (Capsicum annuum L. cv. DangZo) higher activities of anticancer activities on colon cancer cell lines compared to other cancer cell lines.

Effects of Dietary Supplementation of Lysine Cell Mass (LCM) in Juvenile Israeli Carp, Cyprinus carpio (치어기 잉어에 있어 사료내 Lysine 부산물의 첨가효과)

  • This experiment was conducted to evaluate the lysine cell mass (LCM) as a dietary fish meal (EM) protein replacer in juvenile Israeli carp, Cyprinus carpio. Fishmeal, a major animal protein source in the control diet, was replaced by tCM on the protein equivalent base, Fish averaging 1,7 $\pm$ 0.1 g (Mean $\pm$ SD) fed one of nine diets containing isonitrogenous and isocaloric basis of $38\%$ crude protein and 15.2 kJ available energy/g diet: control, $100\%$ $FM; LCM_20$, $80\%$ $FM+20\%$ $LCM; LCM_40$, $60\%$ $FM+40\%$ $LCM; LCM_60$, $40\%$ $FM+60\%$ $LCM; LCM_100$, $100\%$ $LCM; LCM_20$l, $80\%$ $FM+20\%$ $LCM+0.07\%$ $Lysine; LCM_40$l, $60\%$ $FM+40\%$ $LCM+0.14\%$ $Lysine; LCM_60$l $40\%$ $FM+60\%$ $LCM+0.22\%$ Lysine; LCM_100l, $100\%$ LCM+$0.35\% Lysine. After 6 weeks of feeding trial there was no significant difference in weight gain (WG), feed efficiency (FE), protein efficiency ratio (PER) and specific growth rate (SGR) among fish fed control and $LCM_20$ (P>0.05), while fish fed $LCM_40,\;LCM_60,\;LCM_100,\;LCM_40l,\;LCM_60l\;and\;LCM_100l$ diets had a significantly lower WG, FE, PER and SGR than did fish fed control diet (P<0.05). There was no significant difference in WG, PER and SGR among fish fed control and $LCM_20$l diets (P>0.05), while fish fed $LCM_20$l S had a significantly lower FE than did fish fed control diet (P<0.05). No significant difference was observed in hematocrit and condition facto, among fish fed nine diets (P>0.05). Therefore, these results indicated that LCM could replace FM up to $20\%$ and dietary synthetic lysine supplementation did not show any positive growth effects in juvenile Israeli carp.

  • The phytopathogenic bacterium Pectobacterium chrysanthemi secretes multiple isozymes of plant cell wall disrupting enzyme such as pectate lyase and cellulase. The cel gene, existing in tandem with the pel gene, was isolated previously [10]. The role of Cel5Z and PelL1 in P. chrysanthemi PY35 pathogenicity on potato tissues was assessed by mutagenizing cloned cel gene and pel gene in tandem and recombining them with the chromosomal alleles. Strains with the Km cassette interposon in pelL1 or a double mutant showed a delay in the appearance of symptoms, suggesting that P. chrysanthemi PY35 pectate lyase PelL1 may playa minor role in soft-rot pathogenesis.

Hepatoprotective Effects of Allium monanthum MAX. Extract on Ethanol-Induced Liver Damage in Rat

  • This study investigated the effects of an ethanol extract of Allium monanthum MAX. (AME) on ethanol-induced hepatotoxicity in rat liver. Sprague-Dawley rats weighing 100~150 g, were divided into 5 groups; normal group (NOR), AME 200 mg/kg treated group (S1), ethanol (35%, 10 mL/kg) treated group (S2), AME 200 mg/kg and ethanol (35%, 10 mL/kg) treated group (S3) and AME 400 mg/kg and alcohol (35%, 10 mL/kg) treated group (S4). AME was fractionated by the following solvents: n-hexane, chloroform, EtOAC and n-BuOH. Antioxidant index of the n-BuOH fraction was 600 ppm, highest among fractions. The growth rate and feed efficiency ratio were decreased by ethanol, but gradually increased to the corresponding level of the normal group by administering AME. The serum ALT activities that were elevated by ethanol were significantly decreased by AME administration. It was also observed that the hepatic activities of SOD, catalase, xanthine oxidase and GSH-Px that were increased by ethanol were also markedly decreased in the AME treated group with compared to ETB. These results suggest that ethanol extracts of Allium monanthum MAX. may have a protective effect on ethanol-induced hepatotoxicity in rat liver.

Expression of resveratrol synthase gene and accumulation of resveratrol in transgenic potatoes (Solanum tuberosum L.)

  • A resveratrol synthase (RS) gene was isolated from peanut (Arachis hypogaea, L. cv. Jinpoong) plants. This gene was placed under the control of the cauliflower mosaic virus 35S promoter (CaMV35S) and introduced into two Korean varieties of potato (Solanum tuberosum L. cvs. Jasim and Jowon) plants by Agrobacterium-mediated gene transfer. Putative transformants were screened by PCR with primers designed from CaMV 35S promoter, NOS terminator and RS gene. Most of selected transgenic potato plants showed the amplification of expected fragments by PCR of genomic DNA with gene-specific primers, while they were absent in untransformed control plants. Expression of the resveratrol synthase gene was also examined by northern blot analysis. The transformants showed a band which was lacking in the control plant, confirming that the introduced gene is transcribed into mRNA in the transformants. The strength of the band, which reflected the level of mRNA expression, differed among the individual transformants. Among the transformants obtained, the highest trans-resveratrol content in the transgenic young leaves of purple-fleshed "Jashim" was $2.11{\mu}gg^{-1}$ fresh weight and that in the microtubers in vitro of purple fleshed "Jashim" was $8.31{\mu}gg^{-1}$ fresh weight. This amount of resveratrol may have a positive biological effect on human health.
