Effect of Nepal Pseudo Ginseng Components on Lipolytic Action of Toxohormone-L from Cancerous Ascites Fluid (Nepal Pseudo Ginseng 성분이 Toxohormone-L에 의한 체지방 분해작용에 미치는 영향)

  • This study was divised to observe an inhibitory toward a lipolytic action of toxohormone-L from large root and small root Nepal pseudo ginseng(NPG ; Nepal products) components by water extract and ethanol precipitate in vitro. Toxohormone-L Is known to be a lipolytic factor that was partially purified from the ascites fluid of sarcoma 180-hearing mice and of patients with hepatoma. The inhibitory effect that inhibited the lipolytic action of toxohormone-L by ethanol precipitate component of large root NPG(mean 46.8%) was higher (mean 1.8 times) than that of water extract component in final reaction concentration ,5001g1m1, on the other side inhibitory effect of water extract component in small root NPG(mean 43.9%) was higher(mean 1. 2 times) than that of ethano1 precipitate component, respectively. In a way inhibitory effect of ethanol precipitate component in large root NPG(47.6%), when final reaction concentration of sample were 1,000 U g/ml, was about 4095 lower than that of Korean red ginseng, respectively.

Improved guggulsterone production from sugars, precursors, and morphactin in cell cultures of Commiphora wightii grown in shake flasks and a bioreactor

  • Cell cultures of Commiphora wightii (Arnott.) Bhandari were grown in shake flasks and a bioreactor and an increase in guggulsterone accumulation up to $18{\mu}g\;l^{-1}$ was recorded in cells grown in the production medium containing a combination of sucrose:glucose (4% total), precursors (phenylalanine, pyruvic acid, xylose, and sodium acetate), morphactin, and 2iP. A yield of $10g\;l^{-1}$ biomass and ${\sim}200{\mu}g\;l^{-1}$ guggulsterone was recorded in a 3-l flask and in a 2-l stirred tank bioreactor compared with 6.6 g biomass and $67{\mu}g\;l^{-1}$ guggulsterone in 250-ml flasks. Increased vessel size was correlated with increased biomass and guggulsterone accumulation. 2iP alone was not effective for biomass and guggulsterone accumulation in cell cultures of C. wightii.

  • Littorophiloscia visayanensis n. sp. is described from Visayan islands in the Philippines. In the shape of telson, L. visayaensis is close to L. riedli Strouhal, 1966), L. nipponensis Nunomura 1986, and L. formosana Kwon and Jeon, 1993, but it is easily distinguished by the peculiar shape of male pleopod 1 endopod.

Simulation of D-limonene Separation from Mandarine Extract in Simulated Moving Bed (SMB) (감귤 추출물로부터 D-리모넨 분리를 위한 유사 이동층 크로마토그래피(SMB) 전산모사)

  • Limonene is orange flavored natural material that is mainly contained in mandarine and lemon peels. D-limonene was extracted from cold-storaged mandarine peel by using Soxhlet extractor at $120^{\circ}C$ for 2 hours with ethanol as solvent. Henry constants of d-limonene and impurity were calculated as $H_{Lim}=8.55$ and $H_{imp}=0.223$ from the result of HPLC analysis. 4-bed SMB of limonene simulation with $0.46{\times}25cm$ columns was conducted by using Aspen chromatography program. Then effective condition for purity was found by changing $m_2$ and $m_3$ values in triangle diagram. The highest purity was 98.59% at $m_2=2.57$, $m_3=9.55$. For this case, feed, desorbent, extract, and raffinate flow rates were 1 mL/min, 1.19 mL/min, 0.857 mL/min and 1.34 mL/min, respectively. Scale-up simulation was also conducted by increasing column diameter from 0.46 cm to 1.6 cm for getting the same efficiency. The increased flow rates were 12 mL/min, 14 mL/min, 10 mL/min, and 16 mL/min for feed, desorbent, extract, and raffinate. It was possible to scale-up with maintaining same limonene purity because linear isotherms of limonene and impurity were assumed.

Effect of Plant Growth Regulators on Bulblet Formation and Plant Regeneration in Fritillaria thunbergii Miq. (패모조직배양에서 생장조절 물질이 자구형성 및 식물체 재생에 미치는 영향)

  • To improve the efficiency of mass propagation in vitro of Fritillaria thunbergii, bulb scale and nodes were cultured in LS medium supplemented with the combination of 2, 4-D and kinetin or NAA and BA. The number and size of bulb, the number of adventitious shoot, the ratio of callus formation, rooting, and the effects of light and dark on the culture, plant regeneration from calli, and the gelling substances were investigated. The combination of 2, 4-D and kinetin in media was more effective than the media of NAA and BA for the bulblet formation. The media supplemented with 2 mg/L 2, 4-D and 1 mg/L kinetin, $1{\sim}2\;mg/L$ 2, 4-D without kinetin and $1{\sim}3\;mg/L$ BAA only were effective in the adventitious shoot development. Callus formation and root formation, respectively were effective in the medium supplemented with 2mg/L 2, 4-D and 1mg/L kinetin. In bulb formation, the medium with 5 mg/L kinetin was effective, and the most of bulbs were formed from the axillary bud of node part. In bulb formation, shoot growth, callus and root formation, the light culture for 16 hours per day was better than that in the dark culture. Bulb was nicely formed in the medium with 0. 2 mg/L 2, 4-D, 1 mg/L kinetin. The medium without hormone was most effective for plant regeneration. The phytagel was more effective than agar in the medium as the gelling agent.

Gadolinium Complex of 1,4,7,10-Tetraazacyclododecane-1,4,7-trisacetic acid (DO3A) Conjugate of [(p-aniline benzothiazole)methyl]pyridine as a Tumor-Targeting MRI Contrast Agent

  • The synthesis of a DO3A conjugate of [(p-aniline benzothiazole)methyl]pyridine ($L^2H_3$) and its gadolinium complex of the type [$Gd(L^2)(H_2O)$] ($GdL^2$) is described. The $R_1$ relaxivity ($=4.50mM^{-1}sec^{-1}$) and kinetic inertness of $GdL^2$ compares well with those of structurally analogous Dotarem$^{(R)}$ ($R_1=3.70mM^{-1}sec^{-1}$), a typical extracellular (ECF) MRI contrast agent (CA). Yet, by comparison with Dotarem$^{(R)}$, $GdL^2$ exhibits non-covalent interactions with human serum albumin (HSA) as evidenced by the ${\varepsilon}^*$ titration curve along with in vivo MR signal enhancement in both aorta and heart. Liver-specific nature of $GdL^2$ is also observed as excretion is made through gallbladder. Most notably, $GdL^2$ further demonstrates specificity toward the MDA-MB-231 breast cancer.

Preparation and Properties of New Di-N-alkylated 14-Membered Tetraaza Macrocycles and Their Nickel(II) and Copper(II) Complexes

  • New 14-membered tetraaza macrocycles 1,8-diallyl-3,5,7,7,10,12,14,14-octamethyl-1,4,8,11-tetraazacyclotetradecane $(L^2)$ and 1,8-bis(n-propyl)-3,5,7,7,10,12,14,14-octamethyl-1,4,8,11-tetraazacyclotetradecane $(L^3)$ have been prepared by direct react ion of 2,5,5,7,9,12,12,14-octamethyl-1,4,8,11-tetraazacyclotetradecane $(L^1)with$ allyl bromide or n-propyl bromide. The nickel(II) and copper(II) complexes of $L^2andL^3have$ been prepared. The macrocycles show high copper(II) selectivity against nickel(II) ion in methanol solutions containing water. The wavelengths (ca. 505 nm) of the d-d bands for the nickel(II) complexes are extraordinarily longer than those for the complexes of $L^1and$ other related di-N-alkylated 14-membered tetraaza macrocycles. Crystal structure of $[NiL^2](ClO4)_2$ shows that the average Ni-N bond distance $(1.992\AA)$ of the complex is distinctly longer than those of other related nickel(II) complexes. Effects of the N- and C-substituents on the properties of the macrocyclic compounds are discussed.

Plant regeneration through the callus culture induced from bulb scales of an endangered species Lilium cernum Komarvo. (멸종위기종인 솔나리(Lilium cernum Komarvo.)의 인편 유래 캘러스 유도 및 기내 식물체 재분화)

  • Lilium cernum Komarvo. is an important endangered plant belonging to the family Liliaceae. A method was developed for the rapid micropropagation of L. cernum through plant regeneration from bulb scales explant-derived calli. The bulb scales segments were cultured on Murashige and Skoog (MS) medium supplemented with 0, 0.5, 1.0, $3.0mg{\cdot}L^{-1}$ kinetin and 0, 0.1, 0.5, $1.0mg{\cdot}L^{-1}$ NAA or 2,4-D for callus induction. In media with $0.5{\sim}3.0mg{\cdot}L^{-1}$ kinetin and $0.1{\sim}1.0mg{\cdot}L^{-1}$ NAA and 2,4-D, 95~100% of explants produced callus. They were then transferred to MS medium supplemented with various concentrations of NAA (0, 0.01, 0.05 and $1.0mg{\cdot}L^{-1}$) in combination with BA (0, 1.0 and $2.0mg{\cdot}L^{-1}$) for bulbet formation. Bulbet induction (78%), weight (468 mg) and size (15.5 mm) were obtained the highest on MS medium containing $2.0mg{\cdot}L^{-1}$ BA and $1.0mg{\cdot}L^{-1}$ NAA. In vitro frequency of plant regeneration was not significantly treated in strength of MS and sucrose concentration. Chlorophyll contents in 1/2MS with $50g{\cdot}L^{-1}$ sucrose treatments were higher than those in control and another treatment. This in vitro propagation protocol will be useful for conservation and mass propagation of this endangered plant.

Optimum Dissolved Oxygen Level for the Growth of the Israeli Strain of Common Carp, Cyprinus carpio in the Recirculating Water System (순환여과식 사육장치내에서 이스라엘계 잉어(Cyprinus carpio)의 성장을 위한 최적용존산소량)

  • A growth experiment of the Israeli strain of common carp (Cyprinus carpio) under varying dissolved oxygen levels in the recirculating water system was conducted at the Fish Culture Experiment Station of the National Fisheries University of Pusan from August 28, 1985 to September 17, 1985. Five tanks with a capacity of $5m^3$ of water each were used under the same condition of water parameters except for dissolved oxygen levels which were designed to maintain at 2.0, 2.5, 3.0, 3.5 and 4.0 mg/l ranges. The weight of fish in the beginning was about 300g and each tank was stocked with 200kg of fish. DO level of 3.5mg/l was found to be the best level with a feed coefficient of 1.57 and a daily growth rate of $1.411\%$ whereas 4.mg/l showed a slightly decreased performance of 1.63 and $1.365\%$ respectively. The amounts of feed consumed in 3.5 and 4.0mg/l DO levels were almost the same. Below 3.0mg/l DO levels the growth rate markedly decreased. Furthermore, in 2.0 and 2.5 mg/l groups, the fish did not accept feed vigorously and after feeding the fish usually concentrated around the inflow point showing oxygen deficiency response, The experiment indicates that the DO range of 3.5 to 4.0mg/l is the optimum level for the best growth at $27.5^{\circ}C$. DO concentration above these levels is considered a waste of energy resulting in uneconomical performance, and on the other hand, below these levels, the carp certainly shows a poor growth performance.
