A New Single Cross Maize Hybrid for Grain and Silage, 'Pyeongangok' (내도복 다수성 종실 및 사일리지 옥수수 신품종 '평강옥')

  • Pyeongangok, a new single cross variety, is an yellow dent maize hybrid (Zea mays L.) developed by the maize breeding team at the National Institute of Crop Science (NICS), RDA in 2011. This hybrid, which has a high yield of grain and dry matter, was produced by crossing two inbred lines, KS160 and KS155. KS160 is the seed parent and KS155 is the pollen parent of Pyeongangok. Silking date of Pyeongangok is 2 days earlier than that of check hybrid, Jangdaok, and equal to that of another check hybrid, Kwangpyeongok. Plant height of Pyeongangok is longer than that of Jangdaok and similar to that of Kwangpyeongok. Ear numbers per 100 plants of Pyeongangok is more than that of Jangdaok. Ear length of Pyeongangok is shorter than that of Jangdaok. 100 seeds weight of Pyeongangok is lighter than that of Jangdaok. Ear rate of Pyeongangok is lower than that of Kwangpyeongok. Stay-green of Pyeonganok is not greatly different with that of Kwangpyeongok. It has moderately resistance to southern corn leaf blight (Bipolaris maydis), black streaked dwarf virus (BSDV) and corn borer. It has strong resistance to northern corn leaf blight (Exserohilum turcicum). It has resistance to lodging. Pyeongangok was evaluated for the yields of grain and dry matter at four locations from 2009 to 2011. The yield of Pyeongangok in grain was 7.66 ton/ha. The yield of Pyeongangok in dry matter was 19.80 ton/ha. The yield of Pyeongangok in total digestible nutrient (TDN) was 13.32 ton/ha. Seed production of Pyeongangok has gone well due to a good match during crossing between the seed parent, KS160, and the pollen parent, KS155, in Yeongwol.

Effects of a Soil-Born Paenibacillus spp. Strain KPB3 on Suppression of Bacterial Wilt Disease Caused by Ralstonia solanacearum (토양에서 분리한 Paenibacillus spp. KPB3의 Ralstonia solanacearum에 의한 세균성 풋마름병 억제 효과)

  • Two hundred bacterial strains were isolated from the soil around healthy tomato plants in a polyvinyl house, where most of the other plants showed bacterial wilt symptoms. The strains were screened in vitro for their antibacterial activity. Among them, a strain, KPB3 showed strong bactericidal activity against bacterial wilt pathogen, Ralstonia solanacearum. The strain KPB3 was identified using physiological and biochemical tests, and 16S rRNA analyses. Based on these tests, the strain was found to be closer to genus Paenibacillus. To control the bacterial wilt caused by R. solanacearum, greenhouse experiments were conducted to determine the effectiveness of the Paenibacillus strain KPB3. Drench application of this strain ($4{\times}10^8$ CFU $mL^{-1}$) into the pots containing tomato plants, post-inoculated with the pathogen, R. solanacearum could drastically reduce the disease severity, compared to the non-treated plants. To evaluate effectiveness of this strain under field conditions, experiments were carried out in polyvinyl houses infested with R. solanacearum, during spring and autumn of the year 2006. It was observed that, during spring, bacterial wilt was more prevalent compared to the autumn. During spring, 50.9% disease incidences occurred in non-treated controls, while, Paenibacillus strain KPB3 treated plants showed 24.6% disease incidences. Similarly, during autumn, around 17.2% plants were infected with bacterial wilt in non- treated polyvinyl houses, compared to the Paenibacillus strain KPB3 treated plants, which showed 7.0% disease incidences. These results demonstrated that, Paenibacillus strain KPB3 is a potential biological control agent against bacterial wilt caused by R. solanacearum, effective under greenhouse as well as field conditions. This is the first report showing biocontrol of R. solanacearum using a Paenibacillus spp. under field conditions.

Conservation and Reproduction of Cheongpung Buwongun Kim Wumyeong's Funeral Bier (청풍부원군 김우명 상여의 보존 및 복제)

  • Cheongpung Buwongun Kim Wu-myeong's Funeral Bier, an important folklore cultural property No.120, possessed by Chuncheon National Museum was donated in 2002 (by Kim Seonggu). It consists of a bier, yoyeo(腰輿), myeongjeongdae(銘旌臺), and manjangdae(輓章臺). It has a high value as the oldest royal bier. The bier which had a resting time in the storage for special exhibition of "The great cultural treasure of Gangwon province" was inspected in September 2012 and colored pigment layer of the wooden part had the risk of peeling off and surface damage of the textile was serious. Therefore, conservation treatment was conducted. In addition, knots and susiks(垂飾) were severely damaged and their exhibition was impossible. Therefore, a reproduction to replace them through a close investigation was made. All parts of the funeral bier were in separation except for the basic furniture. Conservation was made by dividing the parts into wooden parts and textile parts. Yoyeo was reinforced after disassembling bujae from it and then was reassembled. Paraloid B-72 2 wt% (in ethyle acetate), acrylic resin, was applied to the wooden part of the bier in order to reinforce the colored pigment layer with the addition of sodium alginate 2 wt%(in stilled water) and glue 4 wt%(in stilled water). The pollutants on the surface of the textile part were removed (vacuuming) and its creases were smoothed out (steaming). Fat-soluble pollutants were removed using an nonionic surfactant(Saponin, concentration at 0.25 to 0.5 g/𝑙, in de-ionized water). After the disassembly of the yoyeo from the broken wooden, it was bonded with glue (3 wt% for the first gluing, 35 wt% for gluing), and pine wood was used to restore missing parts. In the process of connecting Wongak(雲角), the original metal hinge and nails were reused to complete the assembly.

Effects of Pinacidil, a Potassium-Channel Opener, on Biodistribution of Thallium-201 in Tumor-Bearing Mice ($K^+$ 통로개방제 Pinacidil이 종양이식 생쥐에서 Tl-201의 체내분포에 미치는 영향)

  • Purpose: Thallium behaves similarly to potassium in vivo. Potassium channel opener (K-opener) opens ATP-sensitive $K^+$-channel located at cell membrane, resulting in potassium efflux from cytosol. We have previously reported that K-opener can alter biokinetics of Tl-201 in cultured cells and in vivo. Malignant tumor cells have high Na-K ATPase activity due to increased metabolic activities and dedifferentiation, and differential delineation of malignant tumor can be possible with Tl-201 imaging. K-opener may affect tumoral uptake of Tl-201 in vivo. To investigate the effects of pinacidil (one of the potent K-openers) on the localization of the tumor with Tl-201 chloride, we evaluated the changes in biodistribution of Tl-201 with pinacidil treatment in tumor-bearing mice. Materials and Methods: Baltic mice received subcutaneous implantation of murine breast cancer cells in the thigh and were used for biodistribution study 3 weeks later. $100{\mu}g$ of pinacidil dissolved in $200{\mu}l$ DMSO/PBS solution was injected intravenously via tail vein at 10 min after 185 KBq ($5{\mu}Ci$) Tl-201 injection. Percentage organ uptake and whole body retention ratio of Tl-201 were measured at various periods after injection, and values were compared between control and pinacidil-treated mice. Results: Pinacidil treatment resulted in mild decrease in blood levels of Tl-201, but renal uptakes were markedly decreased at 30-min, 1- and 2-hour, compared to control group. Hepatic, intestinal and muscular uptake were not different. Absolute percentage uptake and tumor to blood ratios of Tl-201 were lower in pinacidil treated mice than in the control group at all time points measured. Whole body retention ratio of Tl-201 was lower in pinacidil treated mice ($58{\pm}4%$ ), than in the control group ($67{\pm}3%$) at 24 hours after with injection of $100{\mu}g$ pinacidil. Conclusion: K-opener did not enhance, but rather decreased absolute tumoral uptake and tumor-to-blood ratios of Tl-201. Decreased whole body retention ratio and renal uptake were observed with pinacidil treatment in tumor-bearing mice.

Effects of Application Rates of Liquid Pig Manure on Rice Growth, Quality and Soil Properties (돈분액비의 시용수준이 벼 생육과 품질 및 토양에 미치는 영향)

  • The effects of liquid pig manure (LM) on the yield and quality of rice as well as soil chemical properties were determined in the field of sandy loam soil under the different fertilizer management. Treatments consisted of 100%, 130% and 160% N application rates of liquid manure as calculated on the basis of the recommended rate of nitrogen (9 kg N/10a) for rice cultivation. Chemical fertilizer (CF) was used as control. Concentrations of T-N and T-P in paddy water were measured by 5-day intervals up to 20days after application. LM treatments significantly increased T-N concentrations in paddy water proportionally with increasing rates of LM (13.2 to 25.7 mg/L). Similarly Total-P content in paddy water was increased right after LM applications but was well below the quality standard of wastewater and manure. Plant height and tillers in 100% and 130% N LM treatments were lower than those in CF control. In the 160% LM treatment, however, plant height and numbers of tillers were higher than those in the CF control. Yields in 100% LM and 160% LM plots were decreased by 3 and 5%, respectively, as compared with 422 kg per 10a in the CF plot. Rice protein contents were similar between 100% LM and CF control (about 6.8%) but it was increased to 7.2% and 7.7% in 130% LM and 160% LM treatments, respectively. Toyo-taste value in the 100% LM treatment was higher than in CF control plot. The proportions of perfect grain of the brown rice were lower in 130% LM and 160% LM treatments than that in CF control. Soil organic matter content, heavy metal and exchangeable cations were highest in the 160% LM plot. Thus considering yield and quality of rice and heavy metals contents in soil, 130% N basal application of liquid manure can be recommended for rice cultivation in this experiment.

Occurrence and Survival Rate of the Larvae of Ark Shell Anadara broughtonii in Chinhae Bay (진해만(鎭海灣)에서의 피조개 Anadara broughtonii 부유유생(浮游幼生)의 출현(出現)과 생존율(生存率))

  • In order to set up a predictive model for an effective spat collection of ark shells, Anadara broughtonii, the survival rate and the time required for each developmental stage of planktonic larvae were investigated during the period from July 1 to October 30 in 1974, in one of the main ark shell seed collection areas, Chinhae Bay, in the southern part of Korea. The advent of D-shaped larvae ca. $94.3{\times}72.7{\mu}m$ long had three peaks during the surveyed period: August 25, August 31 and September 9, umbo-shaped larvae ca. $141.6{\times}108.4{\mu}m$ and full grown larvae ca. $269.3{\times}221.7{\mu}m$ long also showed three peaks: September 6, September 12 and September 20 for the former, and September 20, September 25 and October 5 for the latter, respectively. About 11 to 12 days was required for D-shaped larvae to develop to umbo-shaped stage. At this intermorphological stage, the daily survival rate was 0.93 with a total survival rate of 45% for the stage. The time required for umbo-shaped larvae to develop to full grown larvae varied from 13 to 15 days with a daily survival rate of 0.93 and with a total survival rate of 36% in the period. Twenty-five to twenty-six days were required for each peak group of the D-shaped larvae to reach a full grown stage, and their total survival rate was 16% during this developmental stage.

Dispersal of sugar beet cyst nematode (Heterodera schachtii) by water and soil in highland Chinese cabbage fields (고랭지 배추 재배지에서 사탕무씨스트선충의 물과 토양을 통한 포장 간 분산)

  • Sugar beet cyst nematode, Heterodera schachtii, causes serious economic losses worldwide in Brassicaceae crops. In 2011, this nematode was first found in highland vegetable cultivation areas in Korea, and thereafter spread to the surrounding healthy Chinese cabbage fields. However, little has been documented on the biological and ecological characteristics of the sugar beet nematode in highland vegetable cultivation areas. In this study the dispersal of the sugar beet cyst nematode was examined, focusing on spreading through soil and/or water infested with the nematode. When farmers with work boots trampled on Chinese cabbage fields for 10 minutes, the number of cysts recovered from the soil attached to the working differed depending on the research sites. Under irrigation conditions of 2, 4, and 8 liters of water per $m^2$, the amounts of soils attached on the bottom of the work boots and the number of healthy cysts in the soils increased significantly with the increase in soil moisture, although the total number of cysts, eggs, and juveniles did not increase significantly. After driving on the farm road adjacent to cabbage fields infested with the sugar beet cyst nematode, cysts were also recovered from the soil attached to the vehicle's tires, suggesting that the sugar beet cyst nematode can spread to new fields through soil carried by vehicles and by farmers. An excavator and a motor truck could deliver 41 kg and 224 g, respectively, of soil on the shovel shoes and the wheels to other locations during the Chinese cabbage harvest, suggesting that farming equipment and transportation vehicles may be vital means by which the cyst nematode spreads to non-infested fields in the highland area of Korea. Runoff water also contained cysts, whose amounts increased after water irrigation onto the ridges to simulate rainy conditions, with no significant difference in the number of cysts with increasing amounts of irrigation water. All of these results confirmed that the sugar beet cyst nematode spreads through soil attached to work boots, harvesting tools, and transportation vehicles, especially during the harvest time, and through runoff water on rainy days. These observations suggest that personal sanitization and cleaning of working tools and vehicles are one of the most important means to prevent the dispersal of the sugar beet cyst nematode in fields.

The Induction of ROS-dependent Autophagy by Particulate Matter 2.5 and Hydrogen Peroxide in Human Lung Epithelial A549 Cells (미세먼지와 산화적 스트레스에 의한 인간 폐 상피 A549 세포에의 ROS 의존적 자가포식 유도)

  • Recently, interest in the harmful factors of particulate matter (PM), a major component of air pollution, has been increasing. In particular, PM2.5 with a diameter of less than 2.5 ㎛ is well known to induce oxidative stress accompanied by autophagy in human lung epithelial cells. However, studies on whether PM2.5 increases autophagy under oxidative stress and whether this process is reactive oxygen species (ROS)-dependent are insufficient. Therefore, in this study, we investigated whether PM2.5 promotes autophagy through the generation of ROS in human alveolar epithelial A594 cells. According to our results, cells co-treated with PM2.5 and hydrogen peroxide (H2O2) showed a lower cell viability than cells treated with each alone, which was associated with increased total and mitochondrial ROS production. The co-treatment of PM2.5 and H2O2 also increased autophagy induction, which was confirmed through Cyto-ID staining, and the expression of autophagy biomarker proteins increased. However, when ROS generation was artificially blocked by N-acetyl-L-cysteine pretreatment, the reduction in cell viability and induction of autophagy by PM2.5 and H2O2 co-treatment were markedly attenuated. Therefore, the present results suggest that PM2.5-induced ROS generation may play a critical role in autophagy induction in A549 cells.

Selection of Forage Corn Varieties Adapted to High Latitude (The South of Mt. Suyang) (고위도 기후대 재배 적합 국산 사료용 옥수수 품종 선발)

  • Since maize (Zea mays L.) originated in central and south America, it requires warm climate conditions throughout its growing season. Growth halts when night-time temperatures drop below 10℃, and the plant may die if temperature reach -1.7℃. Thus, temperature should be maintained between 10 and 30℃ from seeding to maturity. The germination temperature for maize should be at least 8-11℃, whit an optimal range 32-34℃. Since temperature significantly affects the germination rate and period, it plays a crucial role in maize growth. In this study, we evaluated the quantity and feed value of 11 major varieties to determine those best suited for maize cultivation as feed in higher latitude, specifically in Democratic People's of Republic of Korea, below 38 degrees north. A cultivation test was also conducted in Suwon in Republic of Korea, to assess adaptability in areas south of Mt. Suyang. Among the varieties tested, Shinhwangok2 reached silking the fastest, in 65 days, while Gwangpyeongok took the longest at 75 days. The stem length of all varieties exceeded 230 cm. Gwangpyeongok had the tallest stems, while Daanok and Shinhwangok2ho displayed the highest ear ratios. Dacheongok presented the highest values in both dry matter and TDN quantity, with 31,420 kg/ha and 21,66 kg/ha respectively. Pyeonggangok had the highest crude protein content at 8.0%. TDN (%) ranged from 57-68%, with Hwangdaok reaching up to 68%. Based on these findings, Dacheongok and Pyeonggangok appear to be the most suitable varieties for cultivation in terms of both quantity and feed value.

Studies on the Breeding of the Response to short photoperiod, Fiber weight, and Qualitative characters and of the Associations Among these characters in Kenaf (섬유용양마의 육종에 관한 연구 -단일반응성과 섬유종의 유전 및 연소)

  • It was shown that the most desirable characters for kenaf are high-fiber weight and moderately early maturity. Therefore, the objectives of this research on this crop is to find varieties possessing these characteristics. The experiments covered in this report provided new information relative to segregation, mode of inheritance, estimate of the number of genes involved in fiber weight and their response to short day length of 10 hours and the qualitative characters, such as, color of stem, capsule, petiole and shape of leaves. The associations which exist among these characters are also indicated. Fiber weight per plant, days to flowering, Stem color, Petiole color, Capsule color, and shape of leaves were studied in parental, $F_1$.$F_2$and backcross populations of a cross between Dashkent, a low-fiber weight but early maturing kenaf variety, and G 38 F-1, a high-fiber weight but late maturing kenaf variety. Crosses were made using the varieties, Dashkent and G 38 F-1 as parents. The Dashkent parent had the following characteristics: green stems, capsules and petioles and lobed shaped leaves; 105.8234 mean-days to flowering in the field, and 106.9222 mean-days under 10 hours short day treatment. The other parent, G 38 F-1 had red stems yellow capsules and red petioles and unlobed shaped leaves; 149.8921 mean-days to flowering in the field, and 62.3684 mean-days under 10 hours short day treatment. Both of the parents, $F_1$, $F_2$, $BC_1$ ($F_1$ X Dashkent, ) and $BC_2$($F_1$ ${\times}$ G38F-1) of the kenaf cross were grown at the Crops Experiment Station, Suwon, Korea in 1965. Color of stems, petioles and capsules, and shape of leaves were noted to be simply inherited as a single factor. Red stem color was dominant over green stem color, red petiole color was dominant over green petiole, lobed shaped leaves were dominant over unlobed shaped leaves and yellow capsules were dominant over green capsule. It was, also, noted that the factor for color of petiole was linked with the factor for shape of leaf with a 11.9587 percent recombination value, however no interaction or linkage were found among the color of stem and capsule color. Using Powers partitioning method, theoretical means and frequency distributions for each population, the days to flowering were calculated with the assumption that two gene pairs were involved. The values obtained fitted the theoretical values. In general this would indicate that Dashkent and G 38 F -1 were differentiated by two gene pairs. Heritability values were calculated as the percent of additive genetic variance. Heritability value of days to flowering, 89.5% in the broad sense and 79.91% in the narrow sense, indicated that the selection for this character would be effective in relatively early generations. Particularly, high positive correlations were found between days to flowering and the color of petioles and shape of leaves. However, there was no relation between days to flowering and capsule color nor between these and stem color. On the basis of the results of this experiment there is evidence that the hereditary factor for shape of leaves and the color of petioles is linked with an effective factor or factors for the characters of days to flowering. The association was sufficiently close to offer a possible simple and efficient means of selection for moderately early mat. uring plants by leaf shape and petiole color selection. Again using Powers partitioning method the frequency distribution for each population to the fiber weight were calculated with the assumption that two gene pairs, AaBb, were involved. Both phenotypic and genotypic dominance were complete. The obtained value did not agree with the theoretical value for $F_2$ and $BC_1$ ($F_1$ ${\times}$ Dashkent.) It seems that Dashkent and G 38 F-1 were differentiated by two major gene pairs but some the other minor genes are necessary. It is certain that the hereditary factor for shape of leaves and color of petioles is linked with an effective factor or factors for fiber weight. Also, high. yielding plants with moderately early maturity were found in the $F_2$ population. Thus, simultaneous selection for high-fiber yield and moderately early maturing plants should be possible in these populations. Phenotypic and genotypic correlation coefficients between fiber weight per plant and days to flowering, stem height and stem diameter were calculated. In general, genotypic correlations are higher than the phenotypic correlation. The highest correlation is found between stem height and fiber weight per plant (0.7852 in genotypic and 0.4103 in phenotypic) and between days to flowering and fiber weight per plant (0.7398 in genotypic and 0.3983 in phenotypic.) It was also expected that the selection of high stem height and moderately early maturing plants were given the efficient means of selection for high fiber weight.
