A method for sex determination of pigs was examined using polymerase chain reaction(PCR). Sex determining region Y(SRY) gene encoded on Y chromosome plays a key role for primary male development. Zinc finger X-Y(ZFX-ZFY) gene, one of the X-V homology gene group was found on the X and Y chromosomes, respectively, We tested for molecular sexing by amplification patterns of SRY and ZF genes. Genomic DNAs from various resources including porcine hairs and semen collected from domestic pig breeds and native pigs was used for PCR assay of each gene. The amplified products for porcine SRY gene were yielded only in males but not in females. On the other hand, two differential patterns were observed in amplification of ZF gene reflecting the chromosomal dimorphism by a length polymorphism between X and Y chromosomes. Of both, a common band was detected in all individuals tested so that this band might be amplified from ZFX gene as a PCR template, but another is specific for males indicated that from ZFY. The result of PCR assay provides identical information to that from investigation of phenotypic genders of the pigs tested. We suggest that this PCR strategy to determine porcine sexes using comparison of the amplification patterns of the SRY gene specific for Y chromosome and the dimorphic ZF gene between X and Y chromosomes may be a rapid and precise method for discrimination of two sexes and applied to DNA analysis of small samples such as embryonic blastomere, semen, and hairs.
The objective of this study is to investigate the effects of Cynanchum wilfordii (CW) on cell viability, anti-oxidant activity, volatile fatty acid (VFA) production and methane gas production. Collected rumen fluid incubated with CW powder (1% w/v) for 12 and 24 hours were analyzed for pH, VFAs and methane. Alamar blue assay showed no significant difference on the viability of 3T3-L1 and C2C12 cells treated with CW for 24 hours. TBARS data showed a dose dependent increase on the antioxidant activity of CW. VFAs increased in the CW-treated groups compared to the control group. In addition, propionate increased more than other VFAs by the treatment with CW. There was a significant decrease in methane gas production in batch culture treated with CW in 12hrs. In conclusion, it was suggested that Cynanchum wilfordii could manipulate rumen fermentation considered by increasing VFA production and inhibition of methanogenesis.

Jatropha curcas L. is an oil bearing species with many uses and considerable economic potential as a biofuel crop. Salt stress effect on growth, ion accumulation, contents of protein, proline and antioxidant enzymes activity was determined in callus and seedling to understand the salt tolerance of the species. Exposure of callus and seedling to salt stress reduced growth in a concentration dependent manner. Under salt stress Na content increased significantly in both callus and seedling whereas, differential accumulation in the contents of K, Ca, and Mg was observed in callus and seedling. Soluble protein content differed significantly in callus as compared to seedling, however proline accumulation remained more or less constant with treatments. The proline concentration was ~2 to 3 times more in callus than in seedling. Salt stress induced qualitative and quantitative differences in superoxide dismutase (SOD; E.C. 1.15.1.1) and peroxidase (POX; E.C. 1.11.1.7) in callus and seedling. Salt induced changes of the recorded parameters were discussed in relation to salinity tolerance.
A novel approach to synthesize silver nanoparticles (AgNPs) using leaf extract of Canna edulis Ker-Gawl. (CELE) under ambient conditions is reported here. The as-prepared AgNPs were analyzed by UV-visible spectroscopy, transmission emission microscopy, X-ray diffraction, Fourier transform-infrared spectroscopy, energy-dispersive analysis of X-ray spectroscopy, zeta potential, and dynamic light scattering. The AgNPs showed excellent antimicrobial activity against various pathogens, including bacteria and various fungi. The biocompatibility of the AgNPs was analyzed in the L929 cell line using NRU and MTT assays. Acridine orange/ethidium bromide staining was used to determine whether the AgNPs had necrotic or apoptotic effects on L929 cells. The concentration of AgNPs required for 50% inhibition of growth of mammalian cells is far more than that required for inhibition of pathogenic microorganisms. Thus, CELE is a candidate for the eco-friendly, clean, cost-effective, and nontoxic synthesis of AgNPs.
We designed and constructed a multichannel superconducting quantum interference device (SQUID) magnetometer system to measure magnetic fields from the human brain. We used a new type of SQUID, the double relaxation oscillation SQUID (DROS). With high flux-to-voltage transfers of the DROS, about 10 times larger than the dc SQUIDs, simple flux-locked loop circuits could be used for SQUID operation. Also the large modulation voltage of the DROS, typically being 100 $mutextrm{V}$, enabled stable flux-locked loop operation against the thermal offset voltage drift of the preamplifier. The magnetometers were fabricated using the Nb/AlOx/Nb junction technology. The SQUID system consists of 37 signal magnetometers, distributed on a semispherical surface, and 11 reference channels were installed to pickup background noises. External feedback was used to eliminate the magnetic coupling with the adjacent channels. The liquid helium dewar has a capacity of 29 L and boil-off rate of about 4 L/d with the total 48 channel insert. The magnetometer system has an average noise level of 3 fT/√Hz at 100 Hz, inside a shielded loon, and was applied to measure auditory-evoked fields.

The breeding work was initiated by utilizing two Japanese hybrids namely C135${\times}$N134 and N137${\times}$C146 along with two evolved Indian breeds, J14 and A24. The breed CSR16 which is characterized with marked larvae and white dumbbell cocoons was evolved from the Japanese hybrid C135${\times}$N134 by crossing with J14, while the breed CSR17 which is characterized with plain larvae and white oval cocoons was evolved from the Japanese hybrid N137${\times}$C146 by crossing with A24. The hybrid of these evolved breeds i.e., CSR16${\times}$CSR17 has shown superior over control hybrid $KA{\times}{NB_4}{D_2}$ and on par with the ruling hybrid of CSR2${\times}$CSR4. The hybrid CSR16${\times}$CSR17 was authorized during 1999 by Central Silk Board, Bangalore, Government of India for commercial exploitation during favourable months based on national level race authorization test.
The mechanism of interaction between low energy ions and biological organisms has been paid much attention recently. In order to clarify the microstructural response to low energy ion irradiation embryonic cells of Paeonia rockii L. implanted by $Fe^{1+}$ ions with the energy of 80KeV were investigated by Optical Microscopy (OM), Scanning electron Microscopy(SEM) and Transmission Electron Microscopy(TEM). At the dose of 1$\times$1015 ions/$\textrm{cm}^2$, apparent cellular damage was observed in the outer several layers of the radicle. The shape of the cells was obviously deformed from regular polygon to irregular. The cell walls became obscure. SEM micrographs showed that the surface of the radicle was etched severely. It was observed by TEM that nucleus of the implanted cell was elongated and tended to fracture. Nuclear envelope lost its integrity. The implanted $Fe^{1+}$ ions were detected by Energy Dispersive Spectroscopy (EDS). These observations showed that low energy ions could damage to the plant organisms with the thickness of about 30~50$\mu\textrm{m}$. The possible reasons for radiation damage in the biological organisms were discussed.

Higenamine [1-(4-hydroxy-6, 7-dihydroxy-l, 2, 3, 4-tetrahydroisoquinoline) is a cardiotonic constituent of Aconiti tuber, one of the most widely prescribed oriental medicines. S-(-)higenamine was reported to have a stronger cardiotonic activity than R-(+)-higenamine and known as a central intermediate in the biosynthesis of various benzyl isoquionoline alkaloids in plants. The separation of higenamine enantiomers has been accomplished with capillary electrophoresis using cyclodextrins (CDs) as chiral selectors. Good resolution of this enantiomers was obtained using a 50 mM sodium phosphate buffer containing hydroxypropyl $\beta$-CDs using 27 cm fused silica capillary (50${\mu}{\textrm}{m}$ i.d., 20 cm to detector) at 25 $^{\circ}C$. With the electric field of 340 V/cm, the separation time of higenamine enantiomers was less than 6 min. Under this optimum conditions, the relative standard deviations of migration time and peak area were less than 1.6% and 3.2%. A 512-channel diode array detector was confirmed for the higenamine. The detection limits (S/N = 3) of these enantiomers are $1.5mutextrm{m}$/mL. We confirmed the chiral form of higenamine in medicinal plants.

Parthenogenetic development, hybrid vigour and cocoon shape variability were studied in bivoltine F1 hybrids of the silkworm, Bombyx mori L. Considerable breed differences were observed in parthenogenetic ability. Among the parental breeds, CSR18 exhibited maximum parthenogenetic development (79.65%) followed by CSR4 (67.90%). Among bivoltine F1 hybrids, CSR18 ${\times}$ CSR4 showed highest parthenogenetic development (73.32%) followed by CSR2 ${\times}$ CSR4 (55.43%). Study on hybrid vigour demonstrated that CSR18 ${\times}$ CSR19 expressed maximum significant hybrid vigour for all the seven economic characters over Mid Parent Value (MPV) followed by CSR2 ${\times}$ CSR4 and CSR18 ${\times}$ CSR4 exhibiting significant heterosis for six characters. Cocoon shape measurement study revealed that among ten bivoltine F1 hybrids, cocoons of three hybrids were found comparatively uniform. Importance of this study to know the level of paythenogenetic development, hybrid vigour and cocoon shape variability in different bivoltine F1 hybrids have been discussed.
