Kinetics of Kojic Acid Fermentation by Aspergillus flavus Link S44-1 Using Sucrose as a Carbon Source under Different pH Conditions

  • Kojic acid production by Aspergillus flavus strain S44-1 using sucrose as a carbon source was carried out in a 250-mL shake flask and a 2-L stirred tank fermenter. For comparison, production of kojic acid using glucose, fructose and its mixture was also carried out. Kojic acid production in shake flask fermentation was 25.8 g/L using glucose as the sole carbon source, 23.6 g/L with sucrose, and 6.4 g/L from fructose. Reduced kojic acid production (13.5 g/L) was observed when a combination of glucose and fructose was used as a carbon source. The highest production of kojic acid (40.2 g/L) was obtained from 150 g/L sucrose in a 2 L fermenter, while the lowest kojic acid production (10.3 g/L) was seen in fermentation using fructose as the sole carbon source. The experimental data from batch fermentation and resuspended cell system was analysed in order to form the basis for a kinetic model of the process. An unstructured model based on logistic and Luedeking-Piret equations was found suitable to describe the growth, substrate consumption, and efficiency of kojic acid production by A. flavus in batch fermentation using sucrose. From this model, it was found that kojic acid production by A. flavus was not a growth-associated process. Fermentation without pH control (from an initial culture pH of 3.0) showed higher kojic acid production than single-phase pH-controlled fermentation (pH 2.5, 2.75, and 3.0).

Influence of the pH and Enantiomer on the Antioxidant Activity of Maillard Reaction Mixture Solution in the Model Systems

  • This study was designed to investigate the influence of the pH and enantiomer on the antioxidant activity of Maillard reaction mixture solution in model systems. The loss of glucose in MRPs did not show different characteristics for the different amino acid enantiomers; however, the concentration of glucose decreased as the pH levels increased. The enolization of sugars was observed in all MRP samples according to increase of pH levels. In addition, D-amino acids were detected in L-amino acid systems and L-amino acids could also be observed in D-amino acid systems. Formation of the isomer was the highest in the Glc/L-Lys system. The browning development increased as pH levels increased; however, browning development did not show different characteristics based on the use of L- versus D-isomers of the same amino acid. The L- and D-isomers show different absorption values in the UV-Vis spectra, but the absorption patterns display a similar shape. The antioxidant activities of MRPs derived from the Glc/Gly, Glc/L-Asn and Glc/D-Asn systems at pH 7.0 were greater compared to those of pH 4.0 and pH 10.0. The antioxidant activities of MRPs derived from the Glc/L-Lys and Glc/D-Lys systems decreased as the pH increased. In addition, the results show that the MRPs derived from the D-isomers have similar antioxidant activities as those from L-isomer. Therefore, the MRPs have the different antioxidant activities on the basis of the pH level, but not on the basis of different amino acid enantiomers.

  • For obtaining the m.l.e. of $\Sigma$ from p-variate Non-singular Normal parent, $N_p(\mu, \Sigma)$, Andersous' Procedure based on the invariance property of the m.l.e. seems to be generally Preferred in the view of its simplicity. This paper shows that his approach with respect to $\Sigma^{-1}$ rather than $\Sigma$ itself, he burther applicable to deriving the m.l.e. of parametersinvolved in the common factor model an dsimplex model as well.

Optimization of Pasta with the Addition of Letinus edodes Powder (표고버섯분말 첨가 파스타의 제조조건 최적화)

  • The study aimed to determine the optimal mixing ratio of two different amounts of Letinus edodes powder and egg for preparation of pasta. The complete analysis was conducted using the Design Expert 7 program (State-Easy, Minneapolis, MN). Response surface methodology revealed 10 experimental points, including two replicates for L. edodes powder and egg. L. edodes pasta formulation was optimized using rheology. Lightness and redness displayed a lineal model pattern, whereas yellowness was represented by a quadratic model. The sensory evaluation parameters of texture, color, flavor, appearance and overall quality showed significant differences for color (p<0.01), flavor (p<0.05), texture (p<0.05) and overall quality (p<0.05). All sensory parameters showed significant values in a quadratic model. The optimum formulations processed by numerical and graphical optimization were determined as 15 g of L. edodes powder and 37.5 g of egg.

Rabbit Model for in vivo Study of Intervertebral Disc Degeneration and Regeneration

  • Objective: The purpose of this study is to verify the usefulness of the rabbit model for disc degeneration study. Materials: The L1-L2, L2-L3, L3-L4. or L4-L5 lumbar intervertebral disc (IVD) of 9 mature male New Zealand White rabbits were injured by inserting a 16-gauge needle to a depth of 5 mm in the left anterolateral annulus fibrosus while leaving L5-L6 IVD uninjured. Three other rabbits also received intradiscal injections of rabbit disc cells transfected with adenovirus and bone morphogenetic protein-2 (ad-BMP-2) at L4-L5 in addition to injury by 16-gauge needle at the L1-L2 level. Using digitized radiographs, measurements of IVD height were made and analyzed by using the disc height index (DHI). Magnetic resonance imaging (MRI) scans of the injured discs, injected discs, and uninjured L5-L6 discs were performed at 15 weeks post surgery and compared with preoperative MRI scans. Results: All twelve rabbits showed consistent results of disc degeneration within 15 weeks following annular puncture. DHIs of injured discs were significantly lower than that of the uninjured L5-L6 discs (p<0.05). The mean value of disc degeneration grade of injured discs was significantly higher than that of uninjured discs (p<0.05). The injection of disc cell transfected with ad-BMP-2 did not induce disc regeneration at 15 weeks after injection. Conclusion: This study showed that the injured disc had a significant change in DHI on simple lateral radiograph and disc degeneration grade on MRI scans within 15 weeks in all rabbits. Rabbit annular puncture model can be useful as a disc degeneration model in vivo.

A Simple Analytical Model for the Study of Optical Bistability Using Multiple Quantum Well p-i-n Diode Structure

  • A simple analytical model has been presented for the study of the optical bistability using a $GaAs-Al_{0.32}Ga_{0.68}As$ multiple quantum well (MQW) p-i-n diode structure. The calculation of the optical absorption is based on a semi-emperical model which is accurately valid for a range of wells between 5 and 20 nm and the electric field F< 200kV/cm . The electric field dependent analytical expression for the responsivity is presented. An attempt has been made to derive the analytical relationship between the incident optical power ( $(P_{in})$ ) and the voltage V across the device when the diode is reverse biased by a power supply in series with a load resistor. The relationship between $P_{in}$ and $P_{out}$ (i.e. transmitted optical power) is also presented. Numerical results are presented for a typical case of well size $L_Z=10.5nm,\;barrier\;size\;L_B=9.5nm$ optical wave length l = 851.7nm and electric field F? 100kV/cm. It has been shown that for the values of $P_{in}$ within certain range, the device changes its state in such a way that corresponding to every value of $P_{in}$ , two stable states and one unstable state of V as well as of $P_{out}$ are obtained which shows the optically controlled bistable nature of the device.

Effects of Ultra-high Pressure Homogenization on the Emulsifying Properties of Whey Protein Isolates under Various pH

  • The effect of ultra-high pressure homogenization on the emulsifying properties of whey protein was investigated in a model emulsion made with whey protein isolate and soya oil under various pH. The emulsifying properties, the average diameter of the oil droplets ($d_{vs}$), and the protein load, were measured for each emulsion produced at different homogenization pressures (50 to 200 MPa) and pH values (4.6 to 8.0). According to the results of variance analysis and response surface, the pH had more influence on oil droplet size and protein load than homogenization pressure. The model equations, which were obtained by response surface analysis, show that pH and homogenization pressure had the major effect on oil droplet size and protein load. Higher homogenization pressure decreased the average droplet size and the protein load. Homogenization at high pressure, as opposed to low pressure, causes no overprocessing, but the effect was pH-dependent. The average diameter of the oil droplets increased slightly by decreasing the pH from 8.0 to 6.5 and then increased dramatically toward the isoelectric point of whey protein (i.e., at pH 4.6). Moreover associated droplets were found at acidic pH and their size was increased at high temperature.

Evaluation of Removal Properties of Cu(II) from Aqueous Solutions by Inflated Vermiculites (팽창질석에 의한 수용액내의 구리 제거능 평가)

  • The main objective of this study was to examine the removal properties of Cu from water by inflated vermiculites. The component of vermiculites was analyzed by XRF and the concentration of Copper ion was measured by UV-VIS. Serial batch Kinetic tests and batch sorption tests were conducted to determine the removal characteristics for Cu in aqueous solutions. The result shows that removal rate, $K_{obs}$, of Cu are 0.73, 1.52, and 1.71 for initial pH 3, pH 4, pH 5, respectively, and are 3.19, 1.90, and 0.73 for the initial concentration of $1mg\;L^{-1}$, $5mg\;L^{-1}$, $10mg\;L^{-1}$, respectively. It leads to the conclusion that the removal rates are inversely proportional to the initial Cu concentration and are increased proportionally to the initial pHs. Finally, Sorption data were correlated with both Langmuir and Freundlich isotherms. As a result, Langmuir and Freundlich models were well fitted to batch isotherm data with good values of the determination coefficient. but the determination coefficient value for the Freundlich model fit was slightly higher than that of Langmuir model (0.965 for the Freundlich model and 0.936 for the Langmuir model). Using the Langmuir model, the maximum sorption capacity ($Q_{max}$), Freundlich partition coefficient, and the numerical value of n wrer estimated as $1,250mg\;kg^{-1}$, $635.1L\;kg^{-1}$ and 1.69, respectively. These results show that the inflated vermiculites could be used as an excellent adsorbent for copper contained in various types of aqueous solutions.

Development of Predictive Growth Model of Vibrio parahaemolyticus Using Mathematical Quantitative Model (수학적 정량평가모델을 이용한 Vibrio parahaemolyticus의 성장 예측모델의 개발)

  • Predictive growth model of Vibrio parahaemolyticus in modified surimi-based imitation crab broth was investigated. Growth curves of V. parahaemolyticus were obtained by measuring cell concentration in culture broth under different conditions ($Initial\;cell\;level,\;1{\times}10^{2},\;1{\times}10^{3},\;and\;1{\times}10^{4}\;colony\;forming\;unit\;(CFU)/mL$; temperature, 15, 25 37, and $40^{\circ}C$; pH 6, 7, and 8) and applying them to Gompertz model. Microbial growth indicators, maximum specific growth rate (k), lag time (LT), and generation time (GT), were calculated from Gompertz model. Maximum specific growth rate (k) of V. parahaemolyticus increased with increasing temperature, reaching maximum rate at $37^{\circ}C$. LT and GT were also the shortest at $37^{\circ}C$. pH and initial cell number did not influence k, LT, and GT values significantly (p>0.05). Polynomial model, $k=a{\cdot}\exp(-0.5{\cdot}((T-T_{max}/b)^{2}+((pH-pH_{max)/c^{2}))$, and square root model, ${\sqrt{k}\;0.06(T-9.55)[1-\exp(0.07(T-49.98))]$, were developed to express combination effects of temperature and pH under each initial cell number using Gauss-Newton Algorism of Sigma plot 7.0 (SPSS Inc.). Relative coefficients between experimental k and k Predicted by polynomial model were 0.966, 0.979, and 0.965, respectively, at initial cell numbers of $1{\times}10^{2},\;1{\times}10^{3},\;and\;1{\times}10^{4}CFU/mL$, while that between experimental k and k Predicted by square root model was 0.977. Results revealed growth of V. parahaemolyticus was mainly affected by temperature, and square root model showing effect of temperature was more credible than polynomial model for prediction of V. parahaemolyticus growth.

Effect of Rebamipide on nepato-Renal Dysfunction Caused by E.coli Lipopolysaccharide in Rat (E. coli Lipopolysaccharide 유발 간-신 기능장애에 있어서 Rebamipide의 효과)

  • The present study was aimed to investigate the preventive effects of rebamipide on the multiple organ dysfunction in a rat model of circulatory shock induced by bacterial endotoxin (E. coli lipopolysaccharide; LPS) in comparison with that of methotrexate. Endotoxemia for 6 hours resulted in little change in the levels of hemoglobin and neutrophils. However, treatment with methotrexate decreased significantly the numbers of circulating neutrophils. Significant increases in serum alanine aminotransferase (ALT,958 $\pm$ 250 lU/L, p<0.001) and aspartate aminotransferase (AST, 1350 $\pm$ 295 lU/L, p<0.001) levels induced by endotoxemia were significantly decreased by rebamipide and methotrexate. The increased level of lactic acid dehydrogenase (LDH) by LPS (2850 $\pm$ 467 lU/L, p<0.05) was significantly inhibited by rebamipide, but not by methot.elate. The elevated serum creatinine (1.2$\pm$0.1, p.0.05) and urea levels (55.3$\pm$6.5 mg/dL, p.0.01) by LPS were also decreased by rebamipide, but not by methotrexate. In line with these results, the plasma concentration of tumor necrosis factor-$\alpha$ (TNF-7,167 $\pm$ 20 pg/mL) was significantly increased upon injection of endotoxin at 1 hour by 1570$\pm$100 pg/mL, and declined to 312$\pm$35 pg/mL at 6 hours. The TNF-$\alpha$ level at 6 hours was significantly decreased by rebamipide to 207$\pm$8 pg/mL (P<0.05). Taken together, it is summarized that rebamipide inhibits the development of multiple ogran dysfunction by inhibition of neutrophil activation in association with inhibition of TNF-$\alpha$ formation in a murine model of endotoxemia.
