Characterization of Heavy Metals Bioleaching from Fly Ash by a Sulfur-Oxidizing Bacterium Thiobacillus thiooxidans: Effect of Solid Concentrations (황산화세균 Thiobacillus thiooxidans에 의한 fly ash의 중금속 제거 특성:고형물 농도의 영향)

  • The bioleaching of heavy metals from fly ash was performed by Thiobacillus thiooxidans MET isolated from the enrichment culture of an anaerobically digested sludge. The effect of solid concentrations on the efficiency of metal leaching was studied in shaken flasks. In the range of solid concentrations 20 g.L­$^1$to 100 g.L­$^1$T. thiooxidans MET oxidized S$^{0}$ to sulfate without any lag period. The final pH of slurry solution was decreased to below pH 1, and the final oxide-redox potential (ORP) was increased to over 420 mV in the solid concentrations below 100 g.L­$^1$. However, the initial lag period of 4 to 8 days was required to obtain the pH reduction and ORP increase of the slurry solutions in the range of solid concentrations 150 g.L­$^1$to 300 g.L­$^1$. The sulfur oxidation rate of T. thiooxidans MET in 20~100 g.L­$^1$solid concentrations was 0.70~0.75 g-S.L­$^1$ㆍ d­$^1$, but its sulfur oxidation activity was remarkably inhibited with increasing solid concentration over 150 g.L­$^1$. Increasing fly ash solids concentration in the range of solids concentration 20 g.L­$^1$ to 200 g.L­$^1$decreased the removal efficiency of Zn, Cu, Mn, Cr and Pb. The solubilization of heavy metals from fly ash was strongly correlated with the pH value of slurry solution. When the pH of slurry solution was reduced to 3, the solubilization process of Zn, Cu and Mn started, and their solubilization efficiency of Zn, Cu and Mn was progressively increased below pH 2. However, the solubilization process of Cr and Pb started at pH 2.5 and 2.0, respectively.

  • S-adenosyl-L-homocysteine(AdoHcy) is the product of all biological methylation in which S-adenosyl-L-methionine (AdoMet) is utilized as a methyl donor and is reversibly hydrolyzed to L-homocysteine and adenosine by AdoHcy hydrolase physiologically. Inhibition of this enzyme results in intracelluar accumulation of AdoHcy leading to a feedback inhibition of AdoMet-dependent methylation reactions which are essential for viral replication.   • The effect of sludge digestion on the leaching efficiency of heavy metals from sludge by bioleaching with Thiobacillus thiooxidans MET was investigated. The used sludges were a non- and anaerobically digested. The leaching efficiency of heavy metals was strongly dependent on the pH of the sludge solution rather than the sludge condition and stolid concentration. The lower the pH the more heavy metal was leached from 3.0 of pH. The sequent orders of leaching heavy metals were Zn, Cu, and Cr. Although the buffering capacity of non-digested sludge was similar to anaerobically digested sludge, the pH decrease rate of the anaerobically digested sludge solution was faster than that of the non-digested sludge solution due to the higher sulfur oxidation rate of T. thiooxidans MET in the anaerobically digested sludge. The amount of leached heavy metals from the anaerobically digested sludge showed higher than that of non-digested sludge at the same pH value. This result might be caused by the difference of the insoluble metal types, which were contained in the sludge. An increase in sludge solids concentration decreased the leaching efficiency of heavy metals in the range of solids concentration 10 g/L to 70 g/L. The optimum ratio of S° to sludge stolid was 0.1 in both the sludge. The bioleaching process of heavy metals with T. thiooxidans MET showed the disinfecting effect over 90% as well as the reduction effect in sludge weight of 20%.

The Wine Yeast Strain-Dependent Expression of Genes Implicated in Sulfide Production in Response to Nitrogen Availability

  • Sulfur metabolism in S. cerevisiae is well established, but the mechanisms underlying the formation of sulfide remain obscure. Here, we investigated by real-time RT-PCR the dependence of expression levels of MET3, MET5/ECM17, MET10, MET16, and MET17 along with SSU1 on nitrogen availability in two wine yeast strains that produce divergent sulfide profiles. MET3 was the most highly expressed of the genes studied in strain PYCC4072, and SSU1 in strain UCD522. The strains behaved differently according to the sampling times, with UCD522 and PYCC4072 showing the highest expression levels at 120 h and 72 h, respectively. In the presence of 267 mg assimilable N/l, the genes were more highly expressed in strain UCD522 than in PYCC4072. MET5/ECM17 and MET17 were only weakly expressed in both strains under any condition tested. MET10 and SSU1 in both strains, but MET16 only in PYCC4072, were consistently upregulated when sulfide production was inhibited. This study illustrates that strain genotype could be important in determining enzyme activities and therefore the rate of sulfide liberation. This linkage, for some yeast strains, of sulfide production to expression levels of genes associated with sulfate assimilation and sulfur amino acid biosynthesis could be relevant for defining new strategies for the genetic improvement of wine yeasts.

Higher concentrations of folic acid reduced the dietary requirements of supplemental methionine for commercial broilers

  • Objective: An experiment was conducted to study the effect of supplementing DL methionine (DL Met) at graded concentrations on performance, carcass variables, immune responses and antioxidant variables in broiler chicken fed folic acid (FA) fortified (4 mg/kg) low-methionine diet. Methods: A basal diet (BD) without supplemental DL Met, but with higher level (4 mg/kg) of FA and a control diet (CD) with the recommended concentration of methionine (Met) were prepared. The BD was supplemented with DL Met at graded concentrations (0%, 10%, 20%, 30%, 40%, and 50% supplemental DL Met of CD). Each diet was fed ad libitum to 10 replicates of 5 broiler male chicks in each from 1 to 42 d of age. Results: Body weight gain (BWG) reduced, and feed conversion ratio (FCR) increased in broilers fed low-Met BD. At 30% and 20% inclusion of DL met, the BWG and FCR, respectively were similar to those fed the CD. Similarly, supplementation of 10% DL Met to the BD significantly increased ready to cook meat yield and breast meat weight, which were similar to those of the CD fed broilers. Lipid peroxidation reduced, the activity of antioxidant enzymes (GSHPx and GSHRx) in serum increased and lymphocyte proliferation increased with increased supplemental DL Met level in the BD. The concentrations of total protein and albumin in serum increased with DL Met supplementation to the BD. Conclusion: Based on the data, it can be concluded that supplemental Met can be reduced to less than 50% in broiler chicken diets (4.40, 3.94, and 3.39 g/kg, respectively in pre-starter, starter and finisher phases) containing 4 mg/kg FA.

Usefulness of $^{11}C-Methyl-L-and$ D-Methionine PET in Gliomas : with Special Attention to Recurrence

  • Objective : This study concernes the usefulness of $^{11}C-methyl-L-and$ D-methionine[Met]-positron emission tomography[PET] for glioma grading and detection of recurrence in gliomas, compared with fluorine-18, 2-fluoro-deoxyglucose[FDG]-PET. Methods : Eighty patients underwent Met-PET study for evaluation of glioma : 37 astrocytomas [WHO grade II, 3; III, 8; IV, 26]. 27 oligodendrogliomas [WHO grade II, 16; III, 11]. and 12 suspicious recurrent gliomas. All images were taken within 2 weeks before operation. For suspicious recurrent cases on magnetic resonance images, both FDG-PET and Met-PET were performed. Results : In astrocytoma, Mean maximum standard uptake value[SUV] of region of interest[ROI] was not different between WHO grades [p=0.108]. but ROI/normal contralateral tissue SUV [T/N] ratio was statistically different between WHO grades [p=0.002]. T/N ratio was more closely related to visual scale than maximum SUV of ROI [p<0.001 and p=0.107 respectively]. In oligodendroglioma, there was no statistical difference between WHO grades in view of maximum SUV and T/N ratio. For recurrent gliomas, sensitivity of FDG-PET and Met-PET was 25% and 100%, while specificity of FDG-PET and Met-PET were 100% and 80%, respectively. Conclusion : Met-PET might be an appropriate tool for tumor grading in astrocytoma and be more sensitive for detection of recurrence in gliomas than FDG-PET.

Serum vitamin D status and metabolic syndrome: a systematic review and dose-response meta-analysis

  • BACKGROUD/OBJECTIVES: Evidence has suggested an association between serum vitamin D and metabolic syndrome (MetS), but prospective studies are very limited. The objective was to assess the dose-response association between serum vitamin D concentration and MetS risk using a systematic review and meta-analysis of updated observational studies. MATERIALS/METHODS: Using MEDLINE, PubMed, and Embase, a systematic literature search was conducted through February 2020 and the references of relevant articles were reviewed. A random-effects model was used to estimate the summary odds ratio/relative risk and 95% confidence interval (CI). Heterogeneity among studies was evaluated with I2 statistic. In total, 23 observational studies (19 cross-sectional studies, and four cohort studies) were included in the meta-analysis. RESULTS: The pooled estimates (95% CI) for MetS per 25-nmol/L increment in serum vitamin D concentration were 0.80 (95% CI, 0.76-0.84; I2 = 53.5) in cross-sectional studies, and 0.85 (95% CI, 0.72-0.98; I2 = 85.8) in cohort studies. Similar results were observed, irrespectively of age of study population, study location, MetS criteria, and adjustment factors. There was no publication bias for the dose-response meta-analysis of serum vitamin D concentrations and MetS. CONCLUSIONS: Dose-response meta-analysis demonstrated that a 25-nmol/L increment in the serum vitamin D concentration was associated with 20% and 15% lower risks of MetS in cross-sectional studies and cohort studies, respectively.

Evaluation of Clinical Usefulness of Gamma Glutamyl Transferase as a Surrogate Marker for Metabolic Syndrome in Non Obese Adult Men (비만하지 않은 성인 남성에서 대사증후군의 대리 표지자로서 감마 글루타밀 전이효소의 임상적 유용성 평가)

  • This study was to evaluate the usefulness of gamma glutamyl transferase (GGT) as a surrogate marker predicting metabolic syndrome. 7,155 non obese men over the age of 20 were studied as subjects. The criteria for diagnosing MetS were the National Cholesterol Education Program - Third Adult Treatment Panel (NCEP-ATP III). The risk of developing MetS according to GGT was conducted logistic regression analysis, and the ROC (receiver operating characteristic) curve was obtained to confirm GGT ability to predict the risk of MetS. Regardless of age and body mass index, MetS had a 7.09 times higher risk of onset in the fourth quartile than in the first quartile of GGT (p<0.001). The AUC (area under the curve) of GGT for the diagnosis of MetS was 0.715, and the cutoff value of GGT was 40.0 U/L, the sensitivity was 65.0%, and the specificity was 70.2%. Therefore, GGT is considered to be a useful diagnostic index for diagnosing MetS.

Gamakamide C and D as Two New Analogues of Bitter-Tasting Cyclic Peptide with Hydantoin Structure from Oyster Crassostrea gigas

  • Two new bitter-tasting cyclic peptides comprising six amino acids, namely gamakamide C and D, were isolated from cultured oysters Crassostrea gigas. Dimethylaminoazobenzene sulfonyl-amino acid analysis detected Val and Leu in gamakamide C and Ile and Leu in gamakamide D. The molecular formula of gamakamide C was determined as $C_{43}H_{60}N_{7}O_8S$ by high-resolution fast atom bombardment mass spectroscopy (HR FAB-MS) ($[M+H]^+m/z822.4200{\Delta}-2.4mmu$), and that of gamakamide D was determined as $C_{43}H_{62}N_7O_8S$ by HR FAB-MS ($[M+H]^+m/z836.4379{\Delta}-2.0mmu$). Comparison of amino acid analyses and fragment ions by MS/MS among gamakamide C, D, and E (known), the structures of gamakamide C and D were confirmed $as-{\small{L}}-Val-{\small{L}}-Met(SO)-{\small{L}}-NMe-Phe-{\small{L}}-Leu-{\small{D}}-Lys-{\small{L}}-Phe-$ and $-{\small{L}}-Ile-{\small{L}}-Met(SO)-{\small{L}}-NMe-Phe-{\small{L}}-Leu-{\small{D}}-Lys-{\small{L}}-Phe-$, respectively.

A New Process for Liquid Phase Sintering of W-Cu Composite; Fluidized Beds Reductio Method (W-Cu 합금의 액상소결을 위한 새로운 공정의 개발:유동층 환원법)

  • A new process for uniform coating of copper to submicron tungsten powder has been developed. W-Cu alloy where copper can be uniformly distibuted has been made by the liquid phase sintering of thus prepared tungsten powder. It has been found that copper content can be lowered less than IOwt. % in our new process, maintaining the uniform distribution of copper in W-Cu alloy. Relative density above 96% was obtained after the liquid phase sintering when small amount of cobalt was added. It was revealed that the rapid increase of densification rate was due to the enhancement of wettability between tungsten particle and liquid copper.
