Role of the Vestibular and Medullary Reticular Neuclei for the Motor Evoked Potentials in Rats (흰쥐의 운동유발전위에 대한 전정신경핵과 연수망상핵의 역할)

  • The motor evoked potentials (MEPs) have been advocated as a method of monitoring the integrity of spinal efferent pathways in various injury models of the central nervous system. However, there were many disputes about origin sites of MEPs generated by transcranial electrical stimulation. The purpose of present study was to investigate the effect of major extrapyramidal motor nuclei such as lateral vestibular nucleus (VN) and medullary reticular nucleus (mRTN) on any components of the MEPs in adult Sprague-Dalwey rats. MEPs were evoked by electrical stimulation of the right sensorimotor cortex through a stainless steel screw with 0.5mm in diameter, and recorded epidurally at T9 - T10 spinal cord levels by using a pair of teflon-coated stainless steel wire electrodes with 1mm exposed tip. In order to inject lidocaine and make a lesion, insulated long dental needle with noninsulated tips were placed stareotoxically in VN and mRTN. Lidocaine of $2{\sim}3\;{\mu}l$ was injected into either VN or mRTN. The normal MEPs were composed of typical four reproducible waves; P1, P2, P3, P4. The first wave (P1) was shown at a mean latency of 1.2 ms, corresponding to a conduction velocity of 67.5 m/sec. The latencies of MEPs were shortened and the amplitudes were increased as stimulus intensity was increased. The amplitudes of P1 and P2 were more decreased among 4 waves of MEPs after lidocaine microinjection into mRTN. Especially, the amplitude of P1 was decreased by 50% after lidocaine microinjection into bilateral mRTN. On the other hand, lidocaine microinjection into VN reduced the amplitudes of P3 and P4 than other MEP waves. However, the latencies of MEPs were not changed by lidocaine microinjection into either VN or mRTN. These results suggest that the vestibular and reticular nuclei contribute to partially different role in generation of MEPs elicited by transcranial electrical stimulation.

Enhancement of Growth and Survival of Populus alba × P. glandulosa Cuttings Inoculated with Ectomycorrhizal Fungus, Pisolithus tinctorius under Fumigated Nursery Condition (모래밭버섯 균근균(菌根菌)의 인공접종(人工接種)에 의(依)한 포플러 삽목묘(揷木苗)의 생장촉진(生長促進) 및 활착률(活着率) 증진(増進))

  • Populus alba ${\times}$ P. glandulosa cuttings in nursery bed were inoculated with mycelium of ectomycorrhizal fungus, Pisolithus tinctorius (Pt) to evaluate effectiveness of the fungus in growth stimulation of poplar. Pt was cultured in 1l glass bottles with vermiculite-peatmoss mixture moistened with modified Melin-Norkrans' solution. The nursery bed was arranged for microplots of $1{\times}2m$ in size and fumigated with methyl bromide before inoculation and cutting placement. Fifty cuttings were placed in each microplot and two treatments (fumigation only and fumigation plus Pt inoculation) were replicated three times. At the end of the first growing season, inoculated plants grew 19% faster in height and produced 49% more dry weight (above-ground portion) than uninoculated plants. Survival rate of inoculated cuttings was also improved by 20% over that of uninoculated cuttings. Inoculated cuttings developed abundant fine root system with golden brown zigzag tips. In the middle of September a sporocarp of Pt was produced from an inoculated plot, suggesting successful establishment of mycorrhiza between poplar and Pt fungus.

Effect of Defoliation Interval on Regrowth and Morphological Characters in White Clover (예취주기가 White Clover의 재생 및 형태적 특성에 미치는 영향)

  • Weak persistence of white clover (Trifolium repens L.) under continuous grazing management has been limited its availability in the mixture with grasses. The experiment was carried out to determine the effect of defoliation interval on the regrowth and morphological characters of the clover cultivars. Individual plants of Regal (large leaf), Grasslands Huia (medium-small leaf) and Aberystwyth S184 (small leaf) were grown in 15cm plastic pot containing a 1: 2: 1 soil:sand:Promix mixture for 55 days, and then clipped to remove all fully expanded leaves every 7, 14 or 28 days. For the analysis of the cultivar response, plants were sampled on the final harvest date(0), and 1, 3, 7, 14 and 28 days after the final harvest date. Harvested dry weight of all cultivars declined as defoliation interval was increased, and that of Regal was the highest compared to the other cultivars. During the regrowing period, increase of total plant dry weight was due to that of leaf and petiole dry weight, and that of Osceola was greater than the others. Although total leaf area and mean single leaf area were increased during the regrowing period, they were reduced with increased defoliation interval and those of Osceola were the greatest until 14-days regrowth. S184 possessed the most number of leaves and Osceola did the longest petiole since 14-days regrowth although the more defoliation, the less nunber of leaves per plant and the shorter petiole. Stolon length and growing tips of all cultivars increased steeply during regrowth, while they were decreased with increased defoliation interval and those of S184 were the highest. More frequent defoliation had detrimental effects on regrowth of white clover, although larger leaf type was productive but less persistent in a mixture with grasses than smaller leaf type.

Application of Slow-Freezing Cryopreservation Method for the Conservation of Diverse Potato (Solanum tuberosum L.) Genotypes

  • Cryopreservation has been recognized as a practical and efficient tool for the long-term storage of vegetatively propagated plants. This study was conducted to investigate the effects of slow-freezing techniques on the cryopreservation of potato. In vitro plantlets of the potato genotypes of 'Atlantic', 'Superior’, 'Namseo', 'J138', and 'CTO5-5' were cold acclimated, and the excised axillary buds were precultured, osmoprotected, exposed to plant vitrification solution, frozen slowly to $-40^{\circ}C$ and then rapidly plunged into liquid nitrogen, thawed and finally plated on the regeneration medium. It was found that the higher the sucrose concentrations in the subculture medium of donor plantlets, the higher the survival rates of shoot tips after cryopreservation, and the highest survival (20%) was observed in the medium added with 0.25 M sucrose. As for the effect of cooling, $0.3^{\circ}C/min$ cooling speed showed the highest survival (25%). Different varieties showed different responses over different cryopreservation treatments. Survival rate was increased by slow-freezing technique method as compared with that of the basic cryopreservation method of vitrification alone in the diverse potato genotypes. Leaf and tuber morphologies of potatoes regenerated after cryopreservation using slow freezing technique were similar to those derived from the in vitro stock plantlets.

Banana Blast Caused by Pyricularia angulata Hashioka (Pyricularia anguiata Hashioka에 의한 바나나 도열병 (가칭))

  • Banana blast was frequently encountered in greenhouses of Jeju province during a local survey of plan diseases from 1985 to 1986. The disease incidence on banana was very severe up to $100\%$ in a greenhouse. The causal fungus was identified as Pyricularia angulata Hashioka based on mycological characteristics and Path genicity of the fungus. The symptom of blast produced on leaves of banana was circular to oval, dark brow spots with yellow margins which measured 1-10mm in diameter, and the symptom on fruits was circular to oval reddish brown or dark brown, depressed spots which measured up to 20mm in diameter. The symptoms were scattered not only on leaves and fruits of banana but. also on petioles, leaf sheaths, bunch stalks and crowns The pathogen was characterized that conidia were hyaline, 2 septated, ovate to pyriform with a small hilum solitary at ends of den tides of conidiophores, and measured $16.0-34.0\times7.0-12.0{\mu}m$ (average $22.5\times9.0{\mu}m$) and conidiophores were mostly 2 septated, occasionally 2 or 3 branched, denticulate at the tips, and measured $7.5-100.0\times3.0-5.0{\mu}m$(average $90.0\times4.0{\mu}m$). The optimum temperature range for mycelial growth of the fungus on PDA was $26-28^{\circ}C$. The result of pathogenicity tests revealed that P. angulata had pathogenicity only on banana. On the other hand Pyricularia grisea (Cke.) Sacc. isolated from Digitaria sanguinalis(L.) Scopo. was not pathogenic to banana. It was revealed that the leaves were the most susceptible to P. angulata among several parts of banana in inoculation tests. Thirty-day-old fruits of banana were more susceptible to the fungus than 60-day-old fruits, and bunch stalks were more susceptible than the fruits.

  • A water culture experiment was conducted to investigate the effect of As content in a culture solution on the water status and growth of rice plants. Rice (Oryza sativa L. Line. Iri 316) seeds were germinated in bentonite and cultivated there for 30 days. Rice seedlings were transplanted into 3.5l pots containing the culture solution on May 1, 1985 and allowed to grow without As treatment for one month. Afterwards, they were grown in a culture solution maintaining the final concentration of As, 0, 1, 5, 10 and 15mg/1 renewing in the solution dissolved sodium arsenate at intervals of 3 to 7 days. Plants were cultivated in the green house during the growing period and harvested 60 days after As treatment. The results obtained were as follows: Transpiration of rice plants was decreased with the increase of the As level in the culture solution. Stomatal diffusive resistance and leaf temperature increased with increase of As levels though the humidity and the air flow rate in leaf decreased. Air flow rate, transpiration and stomatal diffusive resistance showed a highly significant correlation with As contents in shoots and roots of rice plants: Espally The air flow rate and transpiration revealed a significantly higher correlation with As contents in the root than that in the shoot, but diffusive resistance showed adverse tendency. High levels of As in the culture solution depressed plant height, no. of tillers, leaf width and dry weight of plant remarkably. Typical symptoms of As toxicity were root discoloration, and necrosis of leaf tips and margins, and leaf rolling during the sunny daytime were also another symptoms.

Effects of Initial Defoliation Stage and Defoliation Interval on the Growth of White Clover Cultivars Differing in Leaf Size (최초예취시간 및 예취간격이 엽의 크기가 다른 White Clover 품종들의 생장에 미치는 영향)

  • Availability of white clover (Trifolium repens L.) has been limited due to its poor introduction to swards and lack of persistence under improper grazing management. This experiment was conducted to determine the effects of initial defoliation stage and defoliation interval on the growth of white clover varieties. Individual plants of Regal (large leaf), Louisiana S.1 (medium-large leaf), Grasslands Huia (medium-small leaf) and S184 (small leaf) were grown in containers (4.5 $\times$ 13.5 cm) containing a 1:2:1 soil:sand:Promix until reaching to the stage of 1, 4, or 8 trifoliolates, and then clipped to remove all fully expanded leaves every 7 or 28 days (d). For analysis of morphological parameters, plants were sampled on the final harvest date (0 d), and 7, 14, 21, and 28 d after the final harvest date. Harvested dry weight (dw) of all varieties declined as defoliation interval declined or initial defoliation was made earlier. That of Regal was the highest as initial defoliation was delayed. On the 7 d regrowth shoot and root dw were increased as initial defoliation was delayed or interval lengthened, whereas on the 28 d regrowth the trend was alleviated. Root dw and biomass of Regal were higher than the other varieties during the whole regrowing period, when the increase of biomass resulted from that of shoot dw. Leaf areas and petiole lengths of all varieties declined under 7 d defoliation interval. The area and the length declined with earlier initial defoliation on the 7 d regrowth but not on the 28 d regrowth. Stolon length and growing tips of S184 were the highest and increased more steeply during regrowth, while those of Regal were the lowest and did slightly. It is concluded that the continuous defoliation and the first defoliation at earlier growth stage have detrimental effects on growth of white clover, although larger leaf types are more productive but less persistent in a sward than smaller leaf types.
