무(Raphanus sativus L.) 첨가 속성발효 멸치(Engraulis japonicus) 액젓의 제조 및 품질 (Processing and Quality Characteristics of Rapidly Fermented Anchovy Engraulis japonicus Sauce with Radish Raphanus sativus L.)

  • To develop a value-added anchovy Engraulis japonicus sauce, the processing conditions and quality characteristics of a rapidly fermented and high-purity anchovy sauce (RRAS) were examined by adding 5-10% (w/w) radish Raphanus sativus L. RRAS exhibited higher yield, total nitrogen content, and amino nitrogen and calcium contents as well as lower salinity than those of conventional anchovy sauce (control). The salinity, yield, and total amino acid contents of RRAS and control were 17.2-17.7% and 19.6%, 81.2-88.7% and 61.0%, and 13,117.8-14,174.9 mg/100 g and 10,041.1 mg/100 g, respectively. The major amino acids recorded were aspartic acid, glutamic acid, alanine, valine, isoleucine, leucine, phenylalanine, ornithine, lysine, and histidine. The histamine contents of RRAS and control were 8.3-8.6 mg/100 g and 19.2 mg/100 g, respectively, while the protease activity levels were 0.901-0.958 unit/mg and 0.695 unit/mg, respectively. Overall, this study establishes that RRAS can not only significantly shorten the salt fermentation period, but can also serve as an anchovy sauce with superior nutritional quality and higher levels of amino acid and calcium.

Protective Effects of Lacticaseibacillus rhamnosus IDCC3201 on Motor Functions and Anxiety Levels in a Chronic Stress Mouse Model

  • Growing evidence indicates a crucial role of the gut microbiota in physiological functions. Gut-brain axis imbalance has also been associated with neuropsychiatric and neurodegenerative disorders. Studies have suggested that probiotics regulate the stress response and alleviate mood-related symptoms. In this study, we investigated the effects of the probiotic Lacticaseibacillus rhamnosus IDCC3201 (L3201) on the behavioral response and fecal metabolite content in an unpredictable chronic mild stress (UCMS) mouse model. Our study shows that chronic stress in mice for three weeks resulted in significant changes in behavior, including lower locomotor activity, higher levels of anxiety, and depressive-like symptoms, compared to the control group. Metabolomic analysis demonstrated that disrupted fecal metabolites associated with aminoacyl-tRNA biosynthesis and valine, leucine, and isoleucine biosynthesis by UCMS were restored with the administration of L3201. Oral administration of the L3201 ameliorated the observed changes and improved the behavioral alterations along with fecal metabolites, suggesting that probiotics play a neuroprotective role.

Synthesis and Properties of Novel Flame-Retardant and Thermally Stable Poly(amideimide)s from N,N'-(bicyclo[2,2,2]oct-7-ene-tetracarboxylic)-bis-L-amino Acids and Phosphine Oxide Moiety by Two Different Methods

  • N,N'-(bicyclo[2,2,2]oct-7-ene-tetracarboxylic)-bis-L-amino acids 3a-g were synthesized by the condensation reaction of bicyclo[2,2,2]oct-7-ene-2,3,5,6-tetracarboxylic dianhydride 1 with two equimolars of Lalanine 2a, L-valine 2b, L-leucine 2c, L-isoleucine 2d, L-phenyl alanine 2e, L-2-aminobutyric acid 2f and L-histidine 2g in an acetic acid solution. Seven new poly(amide-imide)s PAIs 5a-g were synthesized through the direct polycondensation reaction of seven chiral N,N'-(bicyclo[2,2,2]oct-7-ene-tetracarboxylic)-bis-L-amino acids 3a-g with bis(3-amino phenyl) phenyl phosphine oxide 4 by two different methods: direct polycondensation in a medium consisting of N-methyl-2-pyrrolidone (NMP)/triphenyl phosphite (TPP)/calcium chloride ($CaCl_2$/pyridine (py), and direct polycondensation in a tosyl chloride (TsCl)/pyridine (py)/N,N-dimethylformamide (DMF) system. The polymerization reaction produced a series of flame-retardant and thermally stable poly(amide-imide)s 5a-g with high yield. The resulted polymers were fully characterized by FTIR, $^1H$ NMR spectroscopy, elemental analyses, inherent viscosity, specific rotation and solubility tests. Data obtained by thermal analysis (TGA and DTG) revealed that the good thermal stability of these polymers. These polymers can be potentially utilized in flame retardant thermoplastic materials.

두유박 분말 첨가가 식빵 반죽에 미치는 영향 (Effect of Soybean Milk Residues Powder on the Quality of Dough)

  • The rheological properties of dough made the substitution of wheat flour(composite flour) at the levels of 0%, 5%, 10% and 15% soymilk residue flour, with addition of vital wheat gluten at the levels of 3, 6 and 9% were investigated. And nutrition contents of soymilk residue flour were analyzed. The results were as follows; Principal components of soymilk residue flour were 22.0% crude protein, 13.2% crude lipid, 54.3% carbohydrate, 27.2% dietary fiber and $220{\mu}g/g$ isoflavones. Free amino acid component of soymilk residue were L-glutamic acid, L-leucine, L-lysine, L-valine, L-phenylalanine, L-isoleucine, L-threonine, L-methionine and L-cystine. Total dietary fiber content of bread with soymilk residue and wheat flour were 5% soymilk residue; 3.50%, 10% soymilk residues; 4.65%, 15% soymilk residues; 5.96%, and wheat flour bread: 2.1% respectively Mixing water absorption capacity was increased by increasing amounts of added soymilk residue and vital wheat gluten. Dough development time was increased by increasing amounts of added soymilk residues, while decreased by increasing amounts of vital wheat gluten. The dough volume of composite flour with 5%, 10% and 15% soymilk residue flour were the smaller than wheat flour dough. But the dough volume was increased by added vital wheat gluten, and the composite flour with 5% soymilk residue flour and 9% vital wheat gluten was better than the others. This study proved that the dough volume of composite flour with 5% soymilk residue flour and 9% vital wheat gluten was better than the others. On the other hand, the soymilk residue flour contains dietary fiber, isoflavone, protein, lipid and carbohydrate. Therefore the soymilk residue flour will be very useful as food material.

간질(Fasciola hepatica)의 Branched Chain 아미노산 Aminotransferase의 정제 및 성상 (Purification and Properties of Branched Chain Amino Acid Arminotransferase from Fasciola hepatica)

  • The distribution and Properties of branched chain amino acid aminotransferase (EC 2.6. 1.42) was investigated in adult Fasciola hepatica. Fascicla hepatica was fractionated by differential centrifugation into nuclear, mitochondrial and cytosolic fractions. The activity of branched chain amino acid aminotransferase was measured by the method of Ichihara and Koyama (1966) . Isozyme patterns of this enzyule was also examined by DEAE-cellulose column chromatography. The results obtained were as follows; 1. The activity in homogenate was found to be 12.69 units/g wet tissue. The activity of this enzyme was relatively high compared with those in rat tissues. 2. The distribution of branched chain amino acid aminotransferase in the subcellular organelles showed that 87.8% of the activity was in cytosolic, 10.9% in mitochondrial and 1.3% was in nuclear fraction. 3. Cytosolic fraction of Fasciola hepatica contained Enzyme I, but not Enzyme II and III, of branched chain amino acid aminotransferase. Ensyme I was eluted by 50mM phosphate buffier from DEAE-cellulose column and catalyzed the transamination of all three branched chain amino acids. 4. The Enzyme I was purified about 22-folds increase in specific activity after chromatography on DEAE-cellulose. 5. The best substrate among three amino acids (leucine, isoleucine and valise) was L-isoleucine. 6. The optimal temperature of Enzyme I was $45^{\circ}C$ and the optimal pH was 8.2. 7. The Km value for leucine of Enzyme I was 4.17 mM. 8. The Km values for a-ketoglutarate and pyridoxal phosphate of Enzyme I were 0.41mM and $4.76{\times}10^{-3}{\;}mM$, respectively.

Isoleucine at position 150 of Cyt2Aa toxin from Bacillus thuringiensis plays an important role during membrane binding and oligomerization

  • Cyt2Aa2 is a mosquito larvicidal and cytolytic toxin produced by Bacillus thuringiensis subsp. darmstadiensis. The toxin becomes inactive when isoleucine at position 150 was replaced by alanine. To investigate the functional role of this position, Ile150 was substituted with Leu, Phe, Glu and Lys. All mutant proteins were produced at high level, solubilized in carbonate buffer and yielded protease activated product similar to those of the wild type. Intrinsic fluorescence spectra analysis suggested that these mutants retain similar folding to the wild type. However, mosquito larvicidal and hemolytic activities dramatically decreased for the I150K and were completely abolished for I150A and I150F mutants. Membrane binding and oligomerization assays demonstrated that only I150E and I150L could bind and form oligomers on lipid membrane similar to that of the wild type. Our results suggest that amino acid at position 150 plays an important role during membrane binding and oligomerization of Cyt2Aa2 toxin.

소나무 잔나비버섯의 아미노산, 비타민 및 무기물 분석 (Analysis of Amino Acids, Vitamins and Minerals of Fruiting Body of Fomitopsis pinicola)

  • 소나무 잔나비버섯은 약용으로 이용하고자 구성아미노산, 유리아미노산, 비타민, 무기물의 성을 분석 검토한 결과 건조버섯 100 g당 총아미노산은 2901.5 mg, 유리아미노산 68.67 mg, 비타민 276.91 mg, 무기물은 347.37 mg이었다. 총 아미노산의 구성아미노산은 18종의 아미노산으로 구성되었으며, glutamic acid가 457 mg로 가장 많이 함유되었고, threonine, glycine, aspartic acid, valine등의 순으로 각각 279 mg, 241 mg, 233 mg, 233 mg의 함량이었다. 유리아미노산 중에서는 glutamine이 17.61 mg으로 가장 많이 함유되어 있고, arginine, tryptophan, threonine, glutamic acid, isoleucine, phenylalanine, tyrosine, valine이 각각 6.51 mg, 5.65 mg, 4.38 mg, 4.02 mg, 3.89 mg, 2.69 mg, 1.51 mg, 1.87 mg등의 순으로 구성되었다. 소나무잔나비 버섯의 무기물 구성 중, 성분 P가 77.57 mg로 가장 많이 함유되었고 다음으로 Mg, Fe, Ca, Na, Zn,등의 순이었다. 풍부한 아미노산 및 무기물 함량과 더불어 소나무잔나비 버섯의 비타민 성분 중, vitamin E는 275.76 mg으로 노화방지에 효과가 높을 것으로 사료되고, 추후 기능성 식품으로 이용적 가치가 높을 것으로 판단된다.

캐놀라 식물체내에서 클로르설푸론의 약해 유발 요인 (Chlorsulfuron-induced Phytotoxicity in Canola(Brassica napus L.) Seedlings)

  • Acetolactate synthase를 저해하는 sulfonylurea계 제초제 chlorsulfuron은 감수성 식물 체내에서 많은 생리, 생화학적 변화를 유도한다. 이 연구의 목적은 chlorsulfuron이 유발하는 독성이 감수성 식물체내에서 branched-chain 아미노산 생합성경로의 최종산물인 leucine, valine과 isoleucine의 결핍에 의한 것인지 혹은 branchedchain 아미노산 생합성 경로중 독성 대사산물의 축적에 의한 것인지를 결정하는 것이다. Chlorsulfuron에 처리된 캐놀라의 성장은 저해되었으며, 처리된 식물은 백화 현상, 잎말이 현상 그리고 안토시아닌 축적과 같은 약해증상을 보였다. Branched-chain 아미노산이 첨가된 영양배양액에서 생장하는 캐놀라에 chlorsulfuron을 처리하였을 경우 생장저해와 약해는 단지 부분적으로 완화되었다. 이와 같은 사실은 chlorsulfuron에 처리된 캐놀라의 약해는 branched-chain 아미노산의 결핍 이외에 또 다른 요인이 있음을 시사하는 것이다. 독성 대사산물로 알려진 2-ketobutyrate에 처리된 캐놀라 식물체내에서의 대사산물 변화와 chlorsulfuron에 처리된 캐놀라 식물체내에서의 대사산물 변화는 서로 다른 양상을 보였다. 본 연구에서 얻어진 결과들은 chlorsulfuron에 처리된 감수성 식물이 나타내는 독성은 부분적으로 branched-chain 아미노산의 결핍에 의한 것이라는 점을 시사한다.

체외생산된 소 배반포강 내의 아미노산 농도 (Amino Acid Concentrations in the Blastocoelic Fluid of In Vitro-Produced Bovine Blastocysts)

  • 체외생산된 소 배반포 및 탈출 배반포강 내의 유리 아미노산 농도를 측정하였다. 체외 배양은 소 혈청알부민이 함유된 합성난관배양액(synthetic oviduct fluid; SOF)에서 실시하였으며 수정 후 180시간의 배반포 및 216시간 후의 탈출 배반포를 실험에 공여하였다 아미노산 측정은 알부민 대신 Polyvinyl alcohol이 함유된 SOF 미소적에 수정란을 분주하고 미세조작기를 이용하여 배반포강 내의 액을 추출하여 20종류의 아미노산을 측정하였다. 탈출 배반포는 isoleucine, leucine 및 methionine 농도가 배반포보다 유의적(p〈0.05)으로 높게 나타났으며, glutamate, aspartate는 두 군간에 차이를 나타내지 알았다. 반면 alanine 및 threamine (p〈0.01)과 cystine을 제외한 나머지 12종류의 아미노산(p〈0.001)은 배반포가 탈출 배반포에 비해서 유의적으로 높은 측정치를 나타내었다. 비록 glutamine의 경우 배양액 내에 첨가되지 않았으나 두 군, 특히 배반포 내에서 높은 측정치를 보였다. 본 연구결과로 보아 체외생산된 배반포 및 탈출 배반포는 내강에 필수 및 비필수 아미노산을 각기 다른 농도로 함유하고 있는 것으로 사료된다.
