Cloning and Biochemical Characterization of Aspartate Aminotransferase from Xanthomonas oryzae pv. oryzae (Xanthomonas oryzae pv. oryzae로 부터 aspartate aminotransferase 유전자의 분리 및 생화학 특성)

  • The gene encoding a putative aspartate aminotransferase in Xanthomonas oryzae pv. oryzae (Xoo) was cloned using PCR technique. The gene was ligated with pET-21(a) vector containing His6 tag and expressed in E. coli BL21(DE3). Affinity purification of the recombinant aspartate aminotransferase with Ni-NTA resin resulted in one band by SDS-PAGE analysis. The purified enzyme showed a molecular weight of 43 kDa, as expected. The enzyme was the most active toward L-aspartate as an amino donor, indicating that the purified enzyme is one of aspartate aminotrans-ferases exist in Xoo. Optimal activity of the enzyme was observed at around pH 7.5 and stability was much higher at alkaline pH rather than acidic pH values. The enzyme was considerably activated by the presence of manganese ion, showing about 157% of control activity at 1.0 mM.

Overexpression and Activity Analysis of Cystathionine γ-Lyase Responsible for the Biogenesis of H2S Neurotransmitter (새로운 신경전달물질 H2S 발생 효소, cystathionine γ-lyase의 대량발현 조건과 활성측정)

  • There is a growing recognition of the significance of $H_2S$ as a biological signaling molecule involved in vascular and nervous system functions. In mammals, two enzymes in the transsulfuration pathway, cystathionine ${\beta}$-synthase (CBS) and cystathionine ${\gamma}$-lyase (CGL), are believed to be chiefly responsible for $H_2S$ biogenesis. Genetic inborn error of CGL leads to human genetic disease, cystathioninuria, by accumulating cystathionine in the body. This disease is secondarily associated with a wide range of diseases including diabetes insipidus and Down's syndrome. Although the human CGL (hCGL) overexpression is essential for the investigation of its function, structure, reaction specificity, substrate specificity, and protein-protein interactions, there is no clear report concerning optimum overexpression conditions. In this study, we report a detailed analysis of the overexpression conditions of the hCGL using a bacterial system. Maximum overexpression was obtained in conditions of low culture temperature after inducer addition, performing low aeration during overexpression, and using a low concentration inducer (0.1 mM, IPTG) for induction. Expressed hCGL was purified by His-tag affinity column chromatography and confirmed by Western blot using hCGL antibody and enzyme activity analysis. We also report that the His tag with TEV site attached protein exhibits 76% activity for ${\alpha}-{\gamma}$ elimination reaction with L-cystathionine and 88% for ${\alpha}-{\beta}$ elimination reaction with L-cysteine compared to those of wild type hCGL, respectively. His tag with TEV site attached protein also exhibits a 420 nm absorption maximum, which is attributed to the binding cofactor, pyridoxal 5'-phosphate (PLP).

Amino Acid, Amino Acid Metabolite, and GABA Content of Three Domestic Tomato Varieties

  • To determine the nutritional value of domestic tomatoes, the levels of amino acids, amino acid metabolites, and the bioactive compound ${\gamma}-aminobutyric-acid$ (GABA) were analyzed in three domestic tomato varieties (Rafito, Momotaro, and Medison). Eighteen free amino acids were found, and total free amino acid content was 3,810.21~4,594.56 mg/100 g (dry weight). L-glutamic acid (L-Glu) was the most abundant amino acid, ranging from 1,866.60 mg/100 g for Momotaro to 2,417.45 mg/100 g for Medison. The next most abundant amino acids were L-glutamine (L-Gln) and L-aspartic acid (L-Asp). The three tomato varieties had a good balance of all the essential amino acids except tryptophan. Total essential amino acid content was 274.26~472.71 mg/100 g (dry weight). The following amino acid metabolites were found: L-carnitine (L-Car), hydroxylysine (Hyl), o-phosphoethanolamine (o-Pea), phosphoserine (p-Ser), ${\beta}-alanine$ (${\beta}-Ala$), N-methyl-histidine (Me-His), ethanolamine (EtNH2),and L-citrulline(L-Cit). Large quantities of GABA were found in all three varieties: 666.95-868.48 mg/100g (dry weight). These results support the use of these tomato varieties as nutritious food materials.

  • After modernism, artistic movements such as Cubism, Futurism, De stijl and Bauhaus demonstrated a connection to architecture and other plastic arts and developed a tendency towards a synthetic art. Above all, the unification of painting and architecture is the most noticeable point; a blueprint and a multi-coloring space that are applied on 3-dimensions painting brought new dimensions into relief. Following this artistic trend, it is easy to point out that Le Corbusier's works; which were created under the name of Charles Edouard Jeanneret, show the pictorial peculiarity in the Space Design. Even if white space that he has designed under the slogan such as standardization and mechanization, the variety and metamorphism that he has tried to express on his work have widely been recognized. Especially, his late architectural works were produced as the figurative arts expressing the ambiguity of painting and hence led a step towards a synthetic art. The study analyses Le Corbusier's 2-dimensional Painting as well as his 3-dimensional Space Design. Furthermore, it aims at embarking on the unification of painting and architecture in terms of Le Corbusier's synthetic design philosophy.

Acute Viral Hepatitis A with Cholestatic Hepatitis Treated with Traditional Korean Medicine : A Case Report

  • More and more patients suffering from hepatitis A, the number of patients representing symptoms that are uncharacteristic to hepatitis A is also increasing. There are occasions of serious complications among adult hepatitis A patients that can cause fulminant hepatitis or cirrhosis which can be fatal. A 43-year-old male was diagnosed as acute hepatitis A and was treated at a Western hospital over the course of three months, which included two months of hospitalization. However, his symptoms did not improve, so he was admitted to Oriental hospital. We prescribed Chunggan extract (CGX) four times a day. Acupuncture was performed twice a day. From the twenty-fourth day of admission, the total bilirubin fell under 10 mg/dL for the first time and next day he was discharged. Following his discharge, he consistently took CGX three times a day. 21 days after discharge all of his symptoms disappeared. This study demonstrated that traditional Korean medicine (TKM) for acute viral hepatitis with cholestatic hepatitis might be effective and safe with no adverse effects.

  • Vincenzo Scamozzi as the successor of Palladio stands as a major accomplishment of the task of cleanup to the classicism in 16th century. In addition, unlike the trend seen over Palladian, he shows succession and change of Renaissance villa. "L'Idea dell' Archittectura Universale; The Idea of a Universal Architecture"(1615) is a book of Scamozz's representative. This book is represented his idea for a residential building. His concepts for a residential building were analyzed through the analysis of his book and work. Scamozzi thought that domestic architecture should be designed according to he owner's social status and reputation. These concepts as decorum and this is divided into three categories. This is a threefold order, the first category and highest encompassing reigning princess and their families, who were more or less regarded as God's representatives on earth. The second category comprised noblemen and high office holders whose houses were to be, in all respects, a degree less grand, costly and dignified than the prince's residence. The third category was made up of prominent citizens and wealthy merchants whose houses were to have only moderate commoner did not come into this classification at all. - the magnifiche, honorevoli, and commune style of speech.

The Reality of Child Abuse in Korea (한국 아동학대의 현실)

  • A trauma surgeon is always concerned about child abuse when he or she meets injured children. Abused children will be neglected if trauma surgeons only concentrate on the injured site or physical dynamics. Lately, violence on children has increased in Korea. Therefore, in this study, we considered child abuse through a review of the literatures. An eleven-year-old boy visited the emergency room vomiting with abdominal distension. He had been kicked in the abdomen by his step-mother 10 days earlier. The computed tomography revealed a transected pancreas tail and neck with a large pesudocyst (Fig. 1) and laboratory findings showed an elevated amylase level of more than 6,500 IU/L. Because he complained of severe pain with rebound tenderness on the whole abdomen, he underwent an emergent laparotomy, a distal pancreatectomy of the tail portion with an anti-leakage procedure on the cut surface of the pancreas. However, he underwent a distal pancreatectomy again on the neck portion of the pancreas because of a continuing pseudocyst with severe pain that could not be controlled with conservative managements. After that, his symptoms were improved and he returned to his daily life.

A Study on the Spatial Configuration and Design Method Represented in the Hotel Project by Philippe Starck (필립 스탁의 호텔 프로젝트에서 나타나는 공간구성과 디자인 방법에 관한 연구)

  • One of the most famous designers of our age, Philippe Starck designed eight hotels among fan Schrager's of which hotels are not so big but have huge popularity all around the world located in major cities such as New York, London, Miami, L.A., and San Francisco. These hotels have brought significant influence particularly to the aspects of their recent fashion and strategies in Boutique hotels. Therefore, this paper intends to examine the space composition and design methods by analyzing the eight hotel projects by him. The research started with reviewing his design background first and analyzed the characteristic of his designs of above hotels. The analysis results were arranged from the aspects of narrative spatial configuration and dramatic presentation. From the aspect of narrative spatial configuration, the hotels by Philippe Starck mainly consisted of 1) boundaries and gateways, 2) spaces of passage, 3) center spaces, and 4) the spaces surrounding the center spaces. For the latter's aspects, it was found that various design methods such as 1) objectification, 2) intended ambiguity, 3) depaysement 4) eclectic hybrid, 5) distortions of scale, and 6) sensuous clues were employed. A study on the hotels by tan Schrager, which owe their big success to the emotional design of Philippe Starck's, not only widens the understanding of his design world, but also serves to the appreciation of today's hotel marketing strategies and design.

An Experience of Judicial Autopsy for a Death by Muscular Dystrophy: An Autopsy Case (근이영양증으로 인한 사망의 사법부검 사례 경험: 증례 보고)

  • Progressive muscular dystrophy (PMD) is a primary muscle disease characterized by progressive muscle weakness and wasting, which is inherited by an X-linked recessive pattern and occurs mainly in males. There are several types of muscular dystrophies classified according to the distribution of predominant muscle weakness including Duchenne and Becker, Emery-Dreifuss, facioscapulohumeral, oculopharyngeal, and limb-girdle type. Clinical manifestations of PMD are clumsy, unsteady gait, pneumonia, heart failure, pulmonary edema, hydropericardium, hydrothorax, aspiration, syncopal attacks, and sudden cardiac death. The deceased was a 34-year-old man, and the onset of the first clinical symptom, gait disturbance, was in his late teens. His elder brother had the same disease and experienced brain death after a head trauma and died after mechanical ventilation was discontinued. After an autopsy, we found contracture of the joints, pseudohypertrophy of the calf, wasting and fat replacement of the thigh muscle, pericardial effusion (80 mL), fibrosis and fat replacement of the cardiac ventricular wall, pulmonary edema, and froth in the bronchus. The cause of death was heart failure and dyspnea due to muscular dystrophy. There was no sign or suspicion of foul play in his death.

Probiotics and Prolongation of Life (유산균 Probiotics와 생명의 연장에 대한 고찰)

  • One hundred one years have passed since Metchnikoff made his first scientific contribution to probiotics study. Intestinal lactic acid bacteria (LAB) for humans are closely associated with the host's health because LAB is an important biodefense factor in preventing colonization and subsequent proliferation of pathogenic bacteria in the intestine. A probiotic is recently defined as "live microorganisms that when administered in adequate amount confer a health benefit on the host". Some species of LAB have been claimed as probiotics, such as Lactobacillus acidophilus, L. casei, L. fermentum, L. plantarum, L. reuteri, and Lactococcus lactis. For understanding the general mechanism of probiotics, this paper would explore the early studies relating to probiotics and intestinal microbiota, and briefly introduce the Prolongation of Life written by Elie Metctmikoff.
