Ethanol extract of Plantago asiatica L. controls intracellular fat accumulation and lipid metabolism in 3T3-L1 Adipocytes (차전초의 에탄올추출물이 3T3-L1 지방세포의 지방축적 및 지질대사에 미치는 영향)

  • Objectives : The effects of ethanol extract of Plantago asiatica L. were investgated on adipocyte differentiation, lipopogenesis, lipolysis and apoptosis using differnentiated 3T3-L1 adipocytes. Methods : Plantago asiatica L. was extracted with ethanol (CCE). We carried on MTT assay for cell proliferation, Oil Red O staining for determination of cell differentiation and intracelluar adipogenesis. TUNEL staining assay for cell apoptosis, and Western blot analysis for measurement of pAMPK and pACC, $C/EBP{\alpha}$, $PPAR{\gamma}$ protein expressions were performed. Results : The addition of CCE up to 0.2 mg/ml into cell culture media showed no cytotoxicity. Treatment of 0.2 mg/ml CCE significantly inhibited differentiation in 3T3-L1 preadipocytes. Lipid accumulation of the CCE treated cells was decreased compared with that of control. Induction of cell apoptosis was increased in CCE treated cells compared with that of control. AMPK and ACC levels of the cells with 0.2 mg/ml CCE were led to phosphorylation and also expressions of $C/EBP{\alpha}$ and $PPAR{\gamma}$, as adipogenic transcription factors, were suppressed compared with those of control. Conclusions : Taken together, these results provide evidence that CCE has a regulatory role in lipid metabolism that is related to differentiation into adipocytes, adipogenesis and apoptosis.

Cultivation, UHPLC Pattern Analysis, and Inhibitory Effect on Respiratory Inflammation of Lysimachia mauritiana Lam. (갯까치수염(Lysimachia mauritiana Lam.)의 재배 및 UHPLC 패턴 분석, 호흡기염증 억제 효과)

  • Objectives : Lysimachia mauritiana Lam. is known as a medicinal plant native to Korea that has antioxidant, anticancer, antibacterial, and antiviral activities. However, until now, research on the cultivation technology of L. mauritiana is insufficient, and there are no research data on the systematic cultivation method and mass production of L. mauritiana. Therefore, this study aims to establish a cultivation system of L. mauritiana. Methods : The cultivation environment of open land and facilities according to the growth of L. mauritiana was compared and tested. In addition, the equivalence of the origin collection extract and the cultivation extract was evaluated through Ultra high performance liquid chromatography (UHPLC) patterns analysis according to cultivation and comparison of the effect of inhibiting respiratory inflammation using BEAS-2B human bronchial epithelial cells. Results : The cultivation technology system was established through cultivation research of L. mauritiana raw materials. In addition, as a result of comparing and evaluating the equivalence of cultivated plants and L. mauritiana raw materials for suppressing respiratory inflammation, the same results were confirmed, and the equivalence was confirmed as a result of analyzing the UHPLC pattern with L. mauritiana raw materials. Conclusions : This study suggests that extract from cultivation research of L. mauritiana plants, which are native to Korea, can be used as a health functional food or medicine to improve respiratory health.

Thin dielectric diaphragm pressure sensor with optical readout (광학적 신호감지의 유전박막 다이아프레임을 이용하는 압력센서)

  • Optical intensity-type pressure sensor was fabricated by coupling optical fiber with a micromachined thin dielectric diaphragm, which consists of a 300 nm thick $SiO_{2}$ layer sandwiched between 150 nm thick top and bottom $Si_{3}N_{4}$ layers. At the wavelength of the sensor light source near $1.3\;{\mu}m$, the optical transmittance of the diaphragm was about 50 %, but it was decreased to a few percents by depositing $1,000\;{\AA}$ thick gold(Au) layer on the diaphragm, which is sufficient enough to be used as a light reflection layer of the sensor. From the optical output power-pressure characteristics of the sensors, it was found that the output power linearly decreased with increasing applied pressure from 0 to 77 torr regardless of the diaphragm size. The respective sensitivities were 0.52, 0.65, and 0.77 nW/torr for the diaphragm sizes of $3{\times}3$, $4{\times}4$, and $5{\times}5\;mm^{2}$, indicating that the sensitivity increases as diaphragm size decreases.

Lp error estimates and superconvergence for finite element approximations for nonlinear parabolic problems

  • In this paper we consider finite element mathods for nonlinear parabolic problems defined in ${\Omega}{\subset}R^d$ ($d{\leq}4$). A new initial approximation is taken. Optimal order error estimates in $L_p$ for $2{\leq}p{\leq}{\infty}$ are established for arbitrary order finite element. One order superconvergence in $W^{1,p}$ for $2{\leq}q{\leq}{\infty}$ are demonstrated as well.

Frequency and Geographical Distribution of B Chromosomes of Rye (Secale cereale L.) in Korea (한국 호밀(Scale cereale L.)의 B 염색체 출현빈도와 지리적 분포)

  • The frequencies and geographical distribution of B chromosomes on 15 strains of rye (Secale cereale L.) collected from various localities in Korea were investigated. All of the 15 strains of rye investigated were found to have B chromosomes, and the frequencies of B chromosomes ranged from 6% to 51% with 20.1% average. Plants with 2Bs seem to be the most stable in populations with B chromosomes. Of 1400 plants examined, one plant was observed to have a deficient-B chromosome in Buyo rye.

Water Purification and Ecological Restoration Effects of the Keumeo Stream Sustainable Structured wetland Biotop (SSB) System Established on the Floodplain of Kyungan Stream (경안천 고수부지에 조성한 금어천 생태적수질정화비오톱 시스템의 수질정화 및 생태복원 효과)

  • A Sustainable Structured wetland Biotop (SSB) system was constructed on the floodplain of Kyungan stream in December, 2006. It purifies polluted water of Keumeo stream which flows into the stream. Water were sampled once a month at inlet and outlet from December, 2007 to December, 2008. $BOD_5$, SS, T-N and T-P were analyzed. Plant and fish species of the system were monitored twice during the period. Average influent and effluent BOD5 concentration was 6.2 and 2.2 mg/L, respectively and BOD5 removal was 50.8%. SS concentration of influent and effluent was averaged 10.1mg/L and 1.5mg/L, respectively and SS abatement amounted to 77.0%. Average influent and effluent T-N concentration was 4.9mg/L and 2.9 mg/L, respectively and T-N retention was 50.8%. T-P concentration of influent and effluent was averaged 0.386mg/L and 0.107mg/L, respectively and T-P removal amounted to 77.0%. Twenty two plant species were naturally introduced into the system, however, they didn't make up a significant portion of the plant populations compared with the planted species. Dominant plant species were in the following order; Phragmites communis > Typha latifolia > Iris pseudoacorus > Persicaria thunbergii. Five families and 15 species of fish were observed in the system including Chinese minnow (Moroco oxycephalus) which inhabits in clean water. Six more fish species were monitored in the system compared with ones living in Kyungan stream. Amphibia and reptiles accounted for 11 species of 4 orders and 7 families including Korean Salamander (Hynobius leechi) which also lives in cleanwater.

A Study on the Behaviour of Organics and Nitrogen Using Upflow Anaerobic Reactor When Acid Fermenter is Added (상향류식 혐기성반응조와 산발효조의 병합처리시 유기물질과 질소거동에 관한 연구)

  • This study was aimed to behaviour of organics and nitrogen on the upflow anaerobic reactor when a acid fermenter is added. Up flow anaerobic reactor (UAR) reaction will result which operates, COD removal efficiencies of reactor with nitrate loading rate 0.11, 0.66g/L/d were over 77%, but one with 1.0g/L/d was 73.5%. Especially, on NLR 0.11g/L/d, COD removal was 77% and nitrate removal efficiency was 93% simultaneously. The other side upflow anaerobic reactor and acid fermenter (UAR+AF) reaction will result witch operates, COD removal efficiencies of reactor with nitrate loading rate 0.11, 0.66g/L/d were over 85%, but one with 1.0g/L/d was 80%. Especially, on NLR 0.11g/L/d, COD removal was 85% and nitrate removal efficiency was 98% simultaneously. Also, without in reaction condition increase of influent nitrate concentration resulted in the linear decrease of nitrate removal efficiency and nitrate removal efficiency at influent nitrate-nitrogen 800mg/L was 50%. Alkalinity was increased theoretically by denitrification at low nitrate-N concentration, however, it was not increased theoretically at high nitrate-No 40% nitrate-N of UAR was denitrified until 70% height of reactor and 90% nitrate-N of UAR+AF was denitrified until 30% height of reactor Upflow anaerobic reactor was to occur accumulate acid, which TVA/Alkalinity is 0.3$\sim$0.47.   • Thermoplastic elastomers are being used increasingly throughout industry owing to their superior properties, such as superior elasticity, formability, and recoverability. Currently, research related to thermoplastic elastomers is focused on the development of composite elastomers by combining with various materials and the development of equipment. On the other hand, in the field of small and medium sized companies, it is necessary to study not only the application of these new materials, but also the process conditions that enable the extrusion of thermoplastic elastomers in inexpensive uniaxial screwing equipment. If extrusion is performed in a single screw extruder, it is important to maintain a uniform thickness through process control of the extruder. This study examined the effects of the processing temperature, which is an extrusion process variable, on the formability of a tube in the thermoplastic elastomer TPE-800L uniaxial extrusion process. The nozzle zone temperature is the most important factor in the extrusion of thermoplastic elastomer TPE-800L; the most excellent moldability was confirmed at $165-170^{\circ}C$.

Rapid Detection of Lactobacillus Genus Inhabiting in Vagina of 20's Healthy Women Using Multiplex PCR (Multiplex PCR 분석을 통한 건강한 20대 여성의 질 내에 서식하는 Lactobacillus 속 유산균의 신속한 검출)

  • The purpose of this study is to simultaneously detect and identify specific genus of Lactobacillus distributed in vagina of healthy women in their twenties by using multiplex primer. Vaginal fluid samples were taken from 166 women who were healthy and did not have vaginosis symptoms caused by bacterial infection. Six species of lactic acid bacteria belong to Lactobacillus genus that are known to inhabit in healthy vagina were selected to make a species-specific multiplex primer. Multiplex primer I was specified to selectively detect L. iners, L. crispatus, L. gasseri and multiplex primer II was specified to selectively detect L. acidophilus, L. jensenii, L. fermentum. L. crispatus (77%) was most frequently detected, and L. acidophilus (57%) and L. jensenii (57%) were relatively higher than others. Although the proportion of L. iners (59%) was a little higher than those of L. acidophilus and L. jensenii, this should be further validated in healthy women's vagina to be sure since they often appear in women having bacterial vaginosis and/or during the antibiotic therapy. Conclusively, the multiplex PCR technique using the species-specific primer could a useful tool to predict variation of vaginal health condition and process of recovery from vaginal disease.

This device is composed of a mechanical glove put on the user's hand and a muscle sensor to measure the activity of his or her muscle.
