A Comparative Study on the Efficacy of Zea mays L. Extracts as a Natural Ingredient in Cosmetics (화장품 천연원료로서 옥수수잎 추출물의 효능 비교연구)

  • In this study, Zea mays L., which were discarded after harvesting in Korea, were collected and used for experiment. The Zea mays L. were dried in the shade and ground before being extracted using two solvents. Subsequently, DPPH, ABTS, the total flavonoid content and the total polyphenol content were measured to analyze antioxidant activity. The diameter of clear zone was measured by using the paper disc diffusion method. The results showed that the ethanol extract had a slightly higher antioxidant and antibacterial activity. The total polyphenol content was measured using gallic acid as standard, and CE(ethanol extract) was found to contain $31.2{\pm}5.2mg\;GAE/g$ of polyphenols, indicating a considerably high activity. When DPPH radical scavenging ability was measured, CE was found to have $4.6{\pm}0.9%$, $6.4{\pm}0.5%$, $22.9{\pm}0.6%$ and $83.2{\pm}0.2%$ at different concentrations of $1{\mu}g/mL$, $10{\mu}g/mL$, $100{\mu}g/mL$, and $1,000{\mu}g/mL$, respectively, with a dependent tendency at the concentration of $1,000{\mu}g/mL$ and a considerably high activity at the concentration of $1,000{\mu}g/mL$. This study is expected to be used as preliminary data to develop new natural antioxidant, antibacteria raw materials, as only little research has been undertaken on Zea mays L., which are mostly discarded after harvesting, while many studies have been so far conducted on corn and corn silks.

Effect of Jasmonic Acid on the Root Growth and the Eleutheroside Accumulation in the Adventitious Root Culture of Eleutherococcus senticosus (가시오갈피나무 부정근(不定根) 배양(培養)에서 부정근(不定根)의 생장(生長)과 Eleutheroside유(類)의 생산(生産)에 미치는 Jasmonic acid처리(處理)의 영향)

  • This study was carried out to investigate the dose-dependent effect of jasmonic acid on both the adventitious root growth and the accumulation of various eleutherosides in the Eleutherococcus senticosus bioreactor culture. The highest biomass production (4.6 g DW/L) was observed in the absence of jasmonic acid and the root growth was significantly decreased by increasing the jasmonic acid concentration. However, jasmonic acid stimulated the production of both eleutheroside B, E and $E_1$. The highest level of eleutheroside B and E ($476.3{\mu}g/g\;DW$ and $676.0{\mu}g/g\;DW$) was obtained by 0.5 mg/L jasmonic acid treatment, while eleutheroside $E_1$ was accumulated at the highest level by 0.01 mg/L jasmonic acid treatment. The highest content of total eleutheroside was $2468.6{\mu}g/L$ when 0.01 mg/L jasmonic acid was applied. In addition, when the adventitious roots were cultured with 0.01 mg/L jasmonic acid, the highest levels of eleutheroside B, E and $E_1$ were observed at the 6th, 8th and 4th day of culture, respectively.

Combined Treatment of Livestock Wastewater with Sewage Using Phanerochaete chrysosporium PSBL-1 (Phanerochaete chrysosporium PSBL-1을 이용한 축산폐수와 하수의 연계처리)

  • We studied possibility of mixing treatment of livestock wastewater and sewage using Phanerochaete chrysosporium PSBL-1. Our study showed that 97.6% of SS and 95% of T-P removal efficiency was achieved when 2 mL BF02(a coagulant) and 100 mL C-210EL(a cationic polymer) were added to the mixture(2:1, v/v) of livestock wastewater and sewage. We studied treatment characteristic of Phanerochaete chrysosporium PSBL-1, after were mixed pretreated wastewater and sewage by dillution ten times about livestock wastewater. The removal efficiency of NBDCOD(non-biodegradable COD), $NH_3-N$ and T-N was increased according to increase of pH. That is, T-N concentration of effluent was satisfied 60 mg/L by drain water waterqulity standard of livestock wastewater public treatment facilities with 35 mg/L from a lapse of five days at pH 6.7, 51 mg/L from a lapse of three days at pH 8 and 33 mg/L from a lapse of one day at pH 10. Moreover $COD_{Mn}$ concentration of effluent was satisfied 40 mg/L by drain water waterqulity standard of livestock wastewater public treatment facilities after a laps of one day at all pH. Organics and nitrogen concentrations of effluent were higher case with addition of V.A.(veratryl alcohol) than case without addition of V.A.(veratryl alcohol). $COD_{Mn}$ concentration of effluent satisfied drain water qulity standard of livestock wastewater public treatment facilities from a lapse of one day, when C/N rate(3:1) of influent was not controled. T-N satisfied that from a lapse of two days, when C/N rate was controled with $4{\sim}6$.

Isolation and Identification of Lactic Acid Bacteria from Commercial Kimchi (시판김치로부터 젖산균의 분리 및 동정)

  • This study was carried out to identify lactic acid bacteria isolated from commercial Kimchi. Twelve lactic acid bacteria strains were isolated from Chinese cabbage kimchi (Baechu kimchi) that was fermented for 4 days at room temperature after making kimchi, 6 strains from pickled ponytail radishes (Chongkak kimchi) that was fermented for 2 days, and 15 strains in radish cube kimchi (Kaktugi) that was fermented for 5 days, and 23 strains were isolated in pickled Wakegi (Pa kimchi) that was fermented for 4 days. Eight strains among the lactic acid bacteria of 12 strains isolated from Baechu kimchi (pH 4.0) were identified as Lactobacillus plantarum, 1 strain as Leuconostoc lactis, 2 strains as Lactobacillus casei subsp. pseudoplantarum, and 1 strain as Lactobacillus sake. Three strains among the lactic acid bacteria of 6 strains isolated from Chongkak kimchi (pH 4.5) were identified as Leuconostoc paramesenteroides, 2 strains as Leuconostoc mesenteroides subsp. mesenteroides, and 1 strain as Lactobacillus plantarum. Two strains among the 15 strains isolated in Kaktugi (pH 4.0) were identified as Leuconostoc lactis, 3 strains as Leuconostoc mesenteroides subsp dextranicum, 4 strains as Lactobacillus casei subsp. pseudoplantarum, and 4 strains as Lactobacillus coryniformis subsp. torquens. Twenty-two strains among the 23 strains isolated from Pa kimchi (pH 4.1) identified as L. plantarum and 1 strain was as Lactobacillus sake. From the results above, the dominant species of Baechu kimchi was confirmed as L. plantarum, Chongkak kimchi as L. paramesenteroides, Kaktugi as L. casei subsp. pseudoplantarum and L. coryniformis subsp. torquens, and Pa kimchi as L. plantarum.

A STUDY ON THE GROWTH OF THE MUSSEL, Mytilus edulis, IN A SALT-FIELD RESERVOIR (염전 저수지에서의 진주담치 (Mytilus edulis) 생장에 관한 연구)

  • The growth of the mussel, Mytilus edulis, in a salt-field reservoir was investigated at Dangjin, Choongnam, from June to October in 1970. l. Temperature, salinity, specific gravity, and pH of water were $22.5-27.5^{\circ}C,\;28.6-34.8\%$, 1.019-1.024, and 7.6 respectively in the salt-field reservoir from June 20 to July 19. 2. Three species of Gastropoda and seven species of Pelecypoda were collected. 3. The shell height of the mussel group in the highest abundance increased from 200mm($15\~25mm$) on June 15 to 40mm ($20\~50mm$) on September 26 in the salt-field reservoir, whereas the average shell heights of them were 19.80mm (June 15) and 35. 10mm (Sept. 26) respectively. 4. The daily growth of the mussel from July 10 to August 12 was 0.22 mm in shell height and highest during the experimental period. 5. The relationships between shell length (L) and shell height (H), and between shell length (L) and shell breadth (B) of the young mussel are represented by the following equations respectively : H=1.26776L+5.07978 B=35041L+3.05502 6. From the above results, it is suggested that the culture of the mussell, Mytilus edulis, in a salt-field reservoir is possible.

Phylogenetic Analysis of Phenanthrene-Degrading Sphingomonas

  • Soil samples were obtained from 5 sites contaminated with polycyclic aromatic hydrocarbons (PAHs). These soil samples were cultured in using phenanthrene as a sole carbon and energy source, and 36 strains of phenanthrene-degrading bacteria were isolated from 3 sites. Most of them degraded 500 ppm of phenanthrene within 8 to 10 days, and these isolates could degrade a few other PAHs other than phenanthrene. Their genotypes were determined by restriction digests of the l6S rRNA genes [amplified ribosomal DNA restriction analysis (ARDRA)]. It was found that all the phenanthrene degrading isolates were included in 4 ARDRA types, and they showed a strict site endemism. l6S rDNAs of 12 strains selected from different sites were sequenced, and they were all confirmed as Sphingomonas strains. Their l6S rDNA sequences were compared for phylogenetic analysis; their sequence showed a similar result to ARDRA typing, thus indicating that these heterotrophic soil bacteria are not regionally mixed. In addition, it was found that the microbial diversity among sampling sites could be monitored by l6S rDNA PCR-RFLP pattern alone, which is simpler and easier to perform, without l6S rDNA sequence analysis.

Enhancement of Herboxidiene Production in Streptomyces chromofuscus ATCC 49982

  • Structurally, herboxidiene contains the tetrahydropyran acetic acid moiety and a side chain including a conjugated diene, and has been isolated from Streptomyces chromofuscus ATCC 49982. Its production was significantly elevated nearly 13.5-fold (0.74 g/l) in a medium supplemented with glycerol (medium No. 6A6), and was more efficacious (1.08 g/l; 19.8-fold) in fed-batch fermentation at 36 h in medium No. 6A6, from Streptomyces chromofuscus. For further enhancement, regulatory genes metK1-sp and afsR-sp from Streptomyces peucetius were overexpressed using an expression vector, pIBR25, and similarly ACCase from Streptomyces coelicolor and two genes, metK1-sp and afsR-sp, were also overexpressed using an integration vector, pSET152, under the control of the strong $ermE^*$ promoter in Streptomyces chromofuscus. Only the recombinant strains S. chromofuscus SIBR, S. chromofuscus GIBR, and S. chromofuscus AFS produced more herboxidiene than the parental strain in optimized medium No. 6A6 with an increment of 1.32-fold (0.976 g/l), 3.85-fold (2.849 g/l), and 1.7-fold(1.258 g/l) respectively.

Antioxidative and Antimicrobial Activities of Lilium Species Extracts Prepared from Different Aerial Parts (부위별 식용백합 추출물의 항산화 및 향균효과)

  • In this study, Lilium sp. were separated into bulbs, leaves, and flowers. Then, total polyphenol contents, electron donating ability (EDA), superoxide dismutase (SOD)-like activity, hydroxyl radical scavenging activity, and antimicrobial activity were measured from the extracts of each of the three aforementioned parts. The examination of physiologically active substances in the three parts revealed that Lilium davidii leaves had high total polyphenol contents, SOD-like activity, hydroxyl radical scavenging activity, and EDA, while the flowers of L. lancifolium showed high SOD-like activity, hydroxyl radical scavenging activity, and EDA, as well as a high level of total polyphenols in the bulb. Measurements of the antimicrobial activities of the extracts against Gram positive bacteria revealed that the leaves and flowers of L. davidii and L. lancifolium caused Bacillus subtilis and Salmonella enteritidis to form clear zones greater than 10 mm. Furthermore, the flowers of L. lancifolium showed particularly high antimicrobial activity against B. subtilis, and the flowers of L. davidii had high activity against S. enteritidis. For the Gram negative bacteria, the leaves and flowers of L. davidii and L. lancifolium caused Listeria monocytogenes and Escherichia coli to form clear zones greater than 10 mm, and finally, the flowers of L. davidii and L. lancifolium showed high antibacterial activity, with inhibition exceeding 12 mm.

Analysis of Hematologic Characteristics of Endangered Korean Native Cattle according to the Age (성장단계별 멸종위기 희소한우의 혈액학적 특성분석)

  • The importance of genetic resource preservation has been highlighted in the literature as a means of maintaining genetic diversity. Investigations for hematologic values and the differential count of white blood cell count (WBC) for Korean indigenous cattle (KIC) and endangered indigenous cattle (EIC) are rarely performed. Therefore, the objective of this study was to investigate the hematologic values of total 40 EIC (White, Black, Mini cattle) and 35 KIC as control by analysis of hematologic characteristics. As a result, the mean values of RBC and platelet of EIC were significantly decreased by age (p<0.05). The mean values of RBC, HCT, MCV and MCHC between EIC and KIC of the same age (2~3 years) showed the statistical significance (p<0.05). Also, in the WBC of EIC, the mean values were decreased according to the age from $13.9{\times}10^3/{\mu}L{\sim}12.7{\times}10^3/{\mu}L$ under 1 year to $9.1{\times}10^3/{\mu}L{\sim}11.5{\times}10^3/{\mu}L$ over 2 years respectively. In the differential count of WBC of EIC (White, Black, Mini cattle), it showed generally the rates of 40.2%, 52.2%, 49.0% lymphocyte and 27.2%, 33.9%, 32.0% segmented neutrophil from 2~3 years respectively. Result of this study will be used for establishing reference range for blood analysis in EIC such as white, black and mini cattle. This study reported hematological values which could serve as baseline information for comparison in conditions of nutrient deficiency, physiological and health status of endangered Korean native cattle. In addition, this study provides a valuable resource for further investigations of the preservation of rare genetic stocks underlying traits of interest in cattle.

Rhombifiline and 5,6-Dehydrolupanine from Angyrus foetida L.

  • Rhomobifoline and 5,6-dehydrolupanine were isolated for the frist time from the leaves and stems of A. foetida L. indigenous to Saudil Arabia. In addition, five other alkaloids, previously identified in A, foetidia L., namely N-methylcytisine, sparteine, anagyrine, lupanine and cytisine, were isolated. The isolated alkaloids were characterized by UV, $^1-NMR,{\;}^{13}H-NMR,{\;}^{13}C-NMR{\;}and{\;}Mass{\;}spectral{\;}data.{\;}^{13}C-NMR$ data of rhombifoline and 5,6-dehydroupanine are reported for the first time.
