Spatio-temporal Fluctuation of Phytoplankton Size Fractionation in the Uljin Marine Ranching Area (UMRA), East Sea of Korea (동해 울진 바다목장해역의 크기별 식물플랑크톤 생물량의 시·공간적 분포 특성)

  • To understand size fractioned chlorophyll a and material cycles of coastal ecosystem in Uljin marine ranching area (JMRA) of East Sea, 4 times of survey were conducted from April to November 2008. Picoplankton, nanoplankton and netplankton in the surface of UMRA fluctuated with an annual mean of $0.26{\mu}g\;L^{-1}$ between the lowest value of $0.03{\mu}g\;L^{-1}$ and the highest value of $0.87{\mu}g\;L^{-1}$, annual mean $1.32{\mu}g\;L^{-1}$ between $0.11{\mu}g\;L^{-1}$ and $5.60{\mu}g\;L^{-1}$, annual mean $0.45{\mu}g\;L^{-1}$ between no detected (nd) and $4.68{\mu}g\;L^{-1}$, respectively. And the relative ratio of picoplankton, nanoplankton and netplanktons on the phytoplankton biomass was on annual average 12.9%, 65.0% and 22.1%, respectively. The 10 m layer was similar to the surface. The relative ratio of pico- and nano-plankton was higher throughout the year. That is, the material cycle of UMRA consists of a microbial food web rather than traditional food chain at a lower trophic levels. Primary production is deemed to have a higher possibility of being adjusted by top-down dynamics, such as micro-zooplankton grazing pressure rather than nutrients supply.

Recovery of Copper from Synthetic Leaching Solution of Manganese Nodule Matte by Solvent Extraction-electrowinning Process (망간단괴 매트상 모의 침출용액으로부터 용매추출-전해채취 공정에 의한 구리의 회수)

  • A scale-up test with a continuous solvent extraction and electro-winning system was carried out to separate and recover copper from a synthetic sulfuric acid solution (Cu 10.5 g/L, Co 2.0 g/L, Ni 15.0 g/L, Fe 0.2 g/L). The solution was introduced into mixer-settlers with four stages of extraction and two stages of stripping for continuous countercurrent solvent extraction to separate copper from nickel and cobalt. The loading was carried out using 40% LIX 84-I(v/v) as extractant with a phase ratio of A : O = 1 : 1. Meanwhile, the stripping was undertaken at a phase ratio of A : O = 1 : 1.5 using depleted electrolyte containing 35.0 g/L Cu and 180 g/L $H_2SO_4$ as stripping solution. The extraction and stripping efficiencies were found to be 96.7% and 91.0%, respectively. The copper composition of the stripped solution (pregnant electrolyte) was 50.0 g/L Cu with impurities of 25 ppm nickel, 5 ppm cobalt and 3 ppm iron. In the electro-winning process, copper metal of 99.833 purity was yielded with current efficiency of 98.9% and current density of $1.50A/dm^2$.

Characteristic of Alcohol Fermentation by the Culture of High Cell Density (Functional Relationship among Specific Growth Rate, Sugar Concentration, Cell Concentration and Alcohol Concentration) (고농도 균체배양에 의한 알콜발효특성 규명 (비성장속도와 당농도, 균체농도, 알콜농도의 함수관계))

  • Experiments of alcohol fermentation of the yeast,K. fragi1is CBS 1555 were performed to obtain the following results. In these experiments, the initial concentrations of sugar which was composed of inulin and fructose as weight ratio of one to one were 30, 50, 75, 100 and 150g/l and the initial densities of the microorganism were less than 0.5g/l, 10g/1 to 15g/1, and 50g/l. The functional relationship among specific growthrates, sugar concentrations, and alcohol concentrations could be expressed by Aiba-shoda equation and the specific growth rate represented the trend that decreased with increase in the initial concentration of the microorganism. Also, $\mu$max and Ks of Monod's equation could be expressed as the function of initial cell concentration like the following equations. $\mu$max=0.8-0.008X Ks=0.54X+8 In the region that sugar, alcohol and cell concentrations were 10g/1 to 120g/l, 0g/l to 60g/l and 0.5g/l to 50g/l respectively, the differences between the experimental values and the calculated ones for specific growth rate approached to 40% with respect to experimental values at the worst cases, but in most cases, those were distributed in the range of less than 20%.

Change of Survival and Reproductive Indicator of the Manila Clam, Ruditapes philippinarum Following Chronic Exposure to Naphthalene (나프탈렌에 노출된 바지락, Ruditapes philippinarum의 생존 및 생식지표 변화)

  • Manila clam, Ruditapes philippinarum (SL; $32.74{\pm}2.18mm$, TW; $8.29{\pm}1.41g$, N; 450) were exposed to various concentrations of naphthalene for 20 weeks. Exposure concentrations of naphthalene established control, solvent control (ethanol), 30, 60, 90 and $120{\mu}g$ Nap $L^{-1}$ following results of 96h acute exposure. After exposure during 20 weeks, survival rate of the manila clam was lowed in 60, 90 and $120{\mu}g$ Nap $L^{-1}$ exposure group compared control group. Also, sex ratio of male was higher in $60{\mu}g$ Nap $L^{-1}$ exposure group (${\chi}^2=5.492$, P < 0.05) but lowed $90{\mu}g$ Nap $L^{-1}$ exposure group (${\chi}^2=4.214$, P < 0.05) and $120{\mu}g$ Nap $L^{-1}$ exposure group (${\chi}^2=30.118$, P < 0.05). Gonad development was delayed in female (> $60{\mu}g$ Nap $L^{-1}$) and male (> $30{\mu}g$ Nap $L^{-1}$). Intersex was 16.28% in female, 1.68% in male. In this result, naphthalene caused survival decrease, imbalance of sex ratio, delay of gonad development of the manila clam. Also, chronic exposure to naphthalene suggested abnormal effects in reproduction of the manila clam.

Effect of Reaction Factors on Reducing Sugar Production from Enteromorpha intestinalis Using Solid Acid Catalyst (고체 산촉매를 이용한 창자파래로부터 환원당 생산에 미치는 인자들의 영향)

  • In this study, the hydrolysis of green macro-algae Enteromorpha intestinalis using solid acid catalyst was conducted to obtain total reducing sugar. The hydrolysis was optimized with four reaction parameters of liquid-to-solid (L/S) ratio, catalyst amount, reaction temperature, and reaction time. As a optimized result, the highest TRS of 7.74 g/L was obtained under condition of 7.5 L/S ratio, $140^{\circ}C$, 15% catalyst amount and 2 hr. By the way, at this condition, only 0.13 g/L 5-HMF was detected. The solid acid-catalyzed hydrolysis of marine resources had the potential in the field of bioenergy.

Effects of Ixeris dentata Extract on Radical Oxygen Species and Bcl-2 Family in Human Breast Cancer Cells (씀바귀 추출물이 인체유방암세포의 활성 산소 및 Bcl-2 Family에 미치는 영향)

  • The aim of the study was to determine the effects of Ixeris dentata extract (IDE) on anticancer activity in human breast cancer MDA-MB-231 cells at both cellular and molecular levels. The cells were cultured in the presence of 0, 20, 30 and $40{\mu}g/mL$ Ixeris dentata extract for 24 hours, respectively. At the end of culture, cytochemical analyses for MTT activity, trypan blue dye exclusion, Annexin V-FITC Apoptosis, and radical oxygen species (ROS) were conducted. RT-PCR was also performed to determine whether or not alterations in cell viability affect the Bax/Bcl-2 ratio. MTT assay showed that relative cell viability decreased in a dose-dependent manner (p<0.05). Reduction of cell viability matched well with increased cell membrane permeability as determined by trypan blue dye exclusion test (p<0.05). The rates of intracellular ROS also increased in a similar manner to those of TB-stained cells. There was an associated shift of apoptotic cells from early to late apoptosis between the 30 and $40{\mu}g/mL$. Bax/Bcl-2 ratio significantly increased along with significant decreases in Bcl-2 expression between 30 and $40{\mu}g/mL$ groups (p<0.05). In conclusion, anticancer activity of Ixeris dentata extract is modulated by a reduction in cell viability along with increased membrane permeability, leading to ROS accumulation within cells, and subsequently cell death through an apoptotic pathway that involves Bax and Bcl-2 in human breast cancer MDA-MB-231 cells.

Optimization of Heteropolysaccharide-7 Production by Beijerinckia Indica (Beijerinckia Indica 배양을 통한 Heteropolysaccharide-7 생산 최적화)

  • Beijerinckia indica was cultured in mineral salts medium (MSM) medium with various carbon and nitrogen sources to improve the production yield of heteropolysaccharide-7 (PS-7). At high C/N ratio, the high concentration of PS-7 was produced until 40 h of the culture, whereas most of the glucose as a carbon source was used for the cell growth at low C/N ratio. However, at the high C/N ratio, PS-7 accumulation stopped at 48 h of the culture due to the increasing viscosity of the culture broth would inhibit the cell growth. Therefore, the optimized value of C/N ratio was 33.3 (20 g/L glucose, 7.5 mM $NH_{4}NO_3$) for the high production of PS-7. In the culture with various carbon sources, B. indica effectively used the hexoses or glucose-generating sugars for PS-7 formation. Especially, sucrose was the best carbon source for the high production of PS-7 (6.96 g/L) with a high viscosity (40772 cp). In the culture of B. indica with MSM medium containing 20 g/L glucose and 7.5 mM $NH_{4}NO_3$ in a 51 fermentor, the highest cell concentration was 2.5 g/L and the highest concentration of PS-7 was 7.5 g/L (35174 cp). The additional nitrogen sources of 7.5 mM $NH_{4}NO_3$, glutamine and glutamate at 12 h of the culture after exhaustion of a nitrogen source regulated the metabolism of carbon sources, therefore the nitrogen sources could control PS-7 synthesis.

Optimization of Cell Concentration and Dilution Rate in Cell Recycled Ethanol Fermentation (세포재순환 에탄올 발효에서 세포농도와 희석률의 최적화)

  • The rheological characteristics of the ethanol fermentation broth were pseudoplastic when the yeast concentration was above 150g/L. From the viewpoint of rheological properties, the cell concentration below 150g/L was recommended for ethanol fermentation. Since the cell floc was formed at the cell concentration of 100 g/L, yeast cells were not much plugged in the pores of the membrane. The cell concentration above 100g/L was desirable when considering the permeability of the membrane. Since ethanol productivity was the highest when the cell concentration was 130 g/L in cell recycled ethanol fermentation. The optimal dilution rate was determined at 1.3 h-1 at constant cell mass of 130g/L. At this dilution rate, the ethanol productivity and glucose conversion ratio ware 80 g/L$\cdot$h and 0.94, respectively.

Characteristics of Disinfection By-Products Formation in Korea (국내 정수장의 소독부산물 생성 특성)

  • The characteristics of disinfection by-products (DBPs) formation at 28 water treatment plants in Korea were investigated. Investigated species of DBPs were trihalomethanes (THMs), haloacetic acids (HAAs) and chloral hydrate (CH). The maximum concentration of THMs was $84.1{\mu}g/L$, minimum and the averages were $6.9{\mu}g/L$ and $27.8{\mu}g/L$, respectively; the maximum concentration of $HAA_5$ was $90.8{\mu}g/L$, minimum and the averages were $3.8{\mu}g/L$ and $26.7{\mu}g/L$, respectively; while the maximum concentration of CH was $29.5{\mu}g/L$, minimum and the averages were $0.5{\mu}g/L$ and $7.4{\mu}g/L$, respectively. On the other hand, DBPs levels during summer months, when the water temperature was near $25^{\circ}C$, were nearly twice as great as DBPs levels during the winter season. The ratio of $THMs/HAA_5$ was 1.07, and $HAA_5$ and THMs were the dominant species of DBPS in the Kum-Sumjin river and Nakdong river, respectivley.

Contribution of Marine Microbes to Particulate Organic Matter in the Korea Strait

  • To assess the relative contribution of bacterial and phytoplankton biomasses to particulate organic matter (POM) in the water column, microbial abundance and biomass were from two transects in the western channel of the Korea Strait in 1996. Bacterial abundance had a mean value of $5.9{\times}10^5$ cells/ml and chlorophyll-a averaged 0.14 ${\mu}g/l$. Bacterial abundance in the Korea Strait showed a positive relationship with chlorophyll-a concentration, while the distribution of POM did not covary with chlorophyll-a. Particulate organic carbon (POC) and nitrogen (PON) concentrations were greater in August than in October. Bacterial carbon (POC) and nitrogen (PON) concentrations were greater in August than in October. Bacterial carbon and nitrogen biomasses were 7.29 ${\mu}gC/l$ and 1.24 ${\mu}gN/l$, respectively, during the study periods. Bacterial biomass was larger in October than in August due to the autumn phytoplankton bloom. Phytoplankton biomass based on chlorophyll-a was 7.67 ${\mu}gC/l$ for carbon and 1.10${\mu}gN/l$l for nitrogen. The ratio of bacterial carbon (BC) to phytoplankton carbon (Cp) averaged 0.95 in the Korea Strait in 1996. Bacteria may play a more significant role in the dynamics of POM than phytoplankton do in August, with BC/Cp ratio of 1.26. The ratio of BC to Cp increased with a decrease in chlorophyll-a concentration. Averaged over all the samples in both cruises, the contribution of microbial biomass to POC and PON was about 43% and 51%, respectively. Bacterial assemblage constituted a significant fraction of POC (21%) and PON (27%). Phytoplankton accounted for 22% of POC and 24% of PON. Microbial biomass played a more important role in the dynamics of POC and PON in October than in August due to a significant increase in microbial biomass in the southern transect (transect-B) in October by the autumn phytoplankton bloom. This study showed that marine microbes may constitute a significant part in the reservoir of POM in the Korea Strait.
