Relationship between Ovarian Development and Plasma Levels of Steroid Hormones, and Induction of Oocyte Maturation and Ovulation in the Cultured Female Korean Sea Bass, Lateolabrax japonicus (양식산 농어, Lateolabrax japonicus 암컷의 난소발달과 혈중 스테로이드 호르몬 양상 및 난모세포 성숙 및 배란유도)

  • Gonad and blood samples were taken from the cultured female Korean sea bass, Lateolabrax japonicus from October to February between 1997 and 1999. Gonadosomatic index began to increase in November and reached the highest value in December (12.8$\pm$1.5) and January (14.8$\pm$3.5), and then decreased sharply in February (2.6$\pm$1.5, p<0.05). The ovarian oocytes developed to tertiary yolk stage and reached fully-Brown stage in December and January, and then underwent atresia without maturation and ovulation in February. The plasma estradio3-17 $\beta$ level increased from November, and reached the highest value in December (1,152.3$\pm$107.2 pg/ml) and January (1,315.4$\pm$99.5 pg/ml), after then decreased in February (P<0.05). The concentration of plasma 17 $\alpha$ ,20 $\beta$-dihydroxy-4-pregnen-3-one was not significantly changed at low levels (86.6$\pm$6.5∼93.8$\pm$2.8 pg/ml) during the experimental period (P<0.05). All the fish with fully-grown oocytes in the ovary were matured and ovulated by HCG injection. The number of floating eggs were 325,000$\pm$26,000 at HCG 1,000 luhg and 195,000$\pm$35,000 at 2,000 lUikg. There was no difference in fertilization rate and hatching rate of the eggs (P<0.05). Considering these results, we could infer that the ovarian oocyte of the cultured Korean sea bass were not matured and ovulated because of the lack of gonadotropin surge. Moreover, HCG injection could induce oocyte maturation and ovulation in the cultured fish, and the effective dose was 1,000 IU/kg.

Studies on Sexual Maturation of Spotted Sea Bass, Lateolabrax maculatus (점농어, Lateolabrax maculatus의 성 성숙에 관한 연구)

  • The gametogenesis, reproductive cycle, frequency distribution of egg diameter and fecundity of spotted sea bass Lateolabrax maculatus were investigated by histological observation. The samples were collected at Goheung, Jeonam from December 1996 to November 1997. Gonadosomatic index (CSI) was appeared less than 1.0 in female and male from December to July and began to increase from August and reached the maximum values of female 7.50 and male 2.98 in October. Hepatosomatic index (HSI) was appeared annual maximum value of female 1.97 and male 1.64 in March and then decreased from April. The annual reproductive cycle of this species can be divided into four successive stages: growing stage (July to August), maturation stage (September to October), ripe and spawning stage (October to November) and degeneration and resting stage (November to July). According to the frequency distribution of egg diameter, the development mode of egg was not seperated, formed one mode for the maturation and spawning season, The individual absolute fecundity ranges from $1,726,242\~17,822,576$ eggs, and individual relative fecundity ranges from $29,458\~214,730$ eggs in total length (cm) and $938\~3,363$ eggs in body weight (g). Female and male of L. maculatus were matured when total length were over 50.1 and 45.1 cm, respectively.

Survey of Sanitary Indicative Bacteria and Pathogenic Bacteria in Fish Farms on the Southern Coast of Korea (남해안 어류양식장 해수 및 양식어류의 위생지표세균 및 병원성세균 조사)

  • The distributions of sanitary indicative bacteria and pathogenic bacteria in seawater and four species of farmed fishes, including oliver flounder (Paralichthys olivaceus), black rock fish (Sebastes schlegeli), red sea bream (Pagrus major) and sea bass (Lateolabrax japonicus), collected at fish farms located in the southern coastal area of Korea were investigated from May to October in 2004. The detection rates of fecal coliform and Entirococcus spp. of sanitary indicative bacteria in all samples were $38.9\%$ and $23.8\%$, respectively. The occurrence of fecal coliform was highest of $58.3\%$ in Busan, Geoje and Wando area, followed Yeosu $33.3\%$, Jeju $12.5\%$, Tongyeong $11.1\%$. The occurrence of Enterococcus spp. was highest In Wando area ($45.8\%$), followed by Yeosu ($33.3\%$), Tongyeong ($22.2\%$), Busan ($16.7\%$), Geoje and Jeju ($12.5\%$). The detection rate of fecal coliform was higher than that of Enterococcus spp., except in the Tongyeong area. There was no difference in the detection rate of fecal coliform from May to October, but the detection rate of Enterococcus spp. increased with seasonal warming seawater temperature. Among the pathogenic bacteria, the detection rate of Vibrio alginolyticus ($49.2\%$) in all samples was highest, followed by V. parahaemolyticus ($36.5\%$), Staphylococcus aureus ($6.3\%$), Salmonella sp. ($2.4\%$). However, V cholerae, V. vulnificus and Shigella sp. were not detected in all tested samples. The detection rates of V. parahaemolyticus and V. alginolyticus increased with seasonal warming seawater temperature from May to August.

Antimicrobial Susceptibility of Escherichia coli Isolated from Fish Farms on the Southern Coast of Korea (남해안 어류양식장에서 분리된 Escherichia coli에 대한 항균제 감수성)

  • Three-hundred and sixteen Escherichia coli strains from seawater, and a variety of farmed fishes, including oliver flounder (Paralichthys olivaceus), black rock fish (Sebastes schlegeli), red sea bream (Pagrus major) and sea bass (Lateolabrax japonicus) between May to October in 2004, were tested by agar dilution method to determine their susceptibility patterns to 17 antimicrobial agents. Overall, 92.1% of Escherichia coli isolates from samples showed antimicrobial resistance to at least one antimicrobial agent and the multiple resistance was seen in 173 isolates (54.7%). The resistance of E. coli isolates to tetracycline (74.1%) was highest, followed by cephalothin (69.9%), doxycycline (66.5%), streptomycin (47.2%), ampicillin (46.2%), cefazolin (31.6%), enrofloxacin (31.0%). norfloxacin (28.2%). The most frequent resistance pattern was TE-D-CF-CIP-ENO-NOR-AM-S-C-SXT-AmC-CZ (14.7%), followed by CF (6.2%), TE (5.1%), TE-CF (4.5%) in 177 isolates from fishes and TE-D-CF (7.2%) followed by TE-D-CF-S (5.8%), CF and TE-D-S (3.6%) in 139 isolates from seawater.

Application of Single Cell Gel Electrophoresis for Detection of DNA Single Strand Breaks in DNA of Fish Blood Cell (어류혈구세포에 있어서 Single Cell Gel Electrophoresis를 응용한 DNA Single Strand Breack의 측정)

  • Single-cell gel electrophoresis (comet assay) was used to detect DNA single strand break in blood cells from several marine fish species. Three fish species were collected from Georgia coastal area. Mummichog, Fundulus heteroclitus showed higher DNA damage than sea bass, Lateolabrax japonicus and trout, Oncorhynchus masou masou under the same experimental conditions. Mummichogs had more alkaline-labile sites on their DNA than other fish species. The comet assay with mummichog blood cells at pH 12.5 showed a dose-response curve with the increasing concentrations of hydrogen peroxide. While the isolated leucocytes showed no increase of DNA damage after in vitro exposure to 2-methyl-1,4-naphthoquinone (MNQ), erythrocytes showed dose-dependent DNA damage. These results indicate that the comet assay can be applied successfully as a bioassay using erythrocyte for environmental monitoring.

Backscattering strength and vertical distribution of dominant fishes in inland waters by hydroacoustics (음향기술을 이용한 내수면 주요어종의 음향산란특성과 연직분포)

  • This study is aimed to understand the vertical distribution of fish in the daytime and nighttime using an acoustic survey in Yondam reservoir of Jeollabuk-do, and an acoustical backscattering strength of dominant species, bass (Micropterus salmoides) and a bluegill (Lepomis macrochirus), which are classified as the ecosystem disturbing species. The results showed that the fish school was distributed in the shallow areas of less than 10 m depth during the period of Aug. and Oct. where it had a strong thermocline, otherwise, it was distributed over 10-times density under stable water temperatures as $13^{\circ}C$ in Nov. There was no vertical patchiness difference between daytime and nighttime unlike the typical marine ecosystem. The dominant species were crucian carp, bluegill, bass. The hydroacoustic method can provide the spatial distribution and effective removal ways of the ecosystem disturbing species in inland fisheries.

Antimicrobial Susceptibility Pattern of Enterococcus faecalis and E. faecium from Fish Farms in the Southern Coast of Korea (남해안 어류양식장에서 분리 된 Enterococcus Faecalis와 E. faecium의 항균제 감수성 비교)

  • The antimicrobial resistance of 160 strains of Enterococcus faecalis and 173 strains of E. faecium to 12 antimicrobial agents was investigated. The test strains were isolated from 126 wild seawater and farmed fish, including olive flounder (Paralichthys olivaceus), black rock fish (Sebastes schlegeli), red sea bream (Pagrus major), and sea bass (Lateolabrax japonicus), in 2005 and 2006. Overall, 91.9% of the E. faecalis isolates and 88.4% of the E. faecium isolates showed antimicrobial resistance to at least one antimicrobial agent. The pattern of antimicrobial resistance of the isolates differed little according to the species of fish. The percentage of E. faecalis and E. faecium with specific antimicrobial resistance differed according to the sample source. For the isolates from farmed fish samples, 66.7% of E. faecalis were tetracycline resistant and 54.5% of E. faecium were erythromycin resistant. By contrast, in the wild fish seawater samples, 92.0% of E. faecalis were rifampin resistant and 88.5% of E. faecium were tetracycline resistant.

Early Osteological Development and Squamation in the Spotted Sea Bass Lateolabrax maculates (Pisces: Lateolabracidae) (한국산 점농어(Lateolabrax maculatus) 자치어의 골격발달과 비늘형성)

  • Early osteological development of the skull, vertebral column, and fins, and squamation in the spotted sea bass, Lateolabrax maculates, were studied under extensive larval rearing conditions. The first ossification during cephalic skeleton development began with the premaxillary, dentary, and parasphenoid at 6.4 mm Total length (Notochord length 6.1 mm) and was completed by 25.2 mm TL (Standard length 20.3 mm). Ossification of the cartilaginous caudal complex started simultaneously in the urostyle and two preural centra at 12.2 mm TL (10.4 mm) and it was completely ossified by 32.0 mm TL (26.4 mm). The principal caudal fin rays, with a count of 9/8, began to ossify at 11.6 mm TL (10.2 mm) and the procurrent caudal fin rays, with counts of 4 (upper) and 3 (lower), started to ossify by 12.6 mm TL (10.9 mm). Ossification of these parts was completed by 21.4 mm TL (17.5 mm). Ossification of the vertebral column was first observed in the first to fourth centra at 8.3 mm TL (7.5 mm) and was fully completed by 21.7-35.0 mm TL (17.8-29.3 mm). The pectoral girdle started to ossify by 5.6 mm TL (5.4 mm) and was completed by 26.8 mm TL (21.8 mm). Eight pectoral fin rays were ossified at 11.6 mm TL (10.2 mm) and 16-18 rays were fully ossified by 13.8 mm TL (12.0 mm). Also, the dorsal, anal, and pelvic fin rays started to ossify at 12.2 mm TL (10.4 mm) and were completed by 12.8 mm TL (11.2 mm), 23.8 mm TL (19.4 mm), and 13.8 mm TL (12.0 mm), respectively. Ossification of the anal and dorsal pterygiophores initially occurred by 12.6 mm TL (10.9 mm) and 14.3 mm TL (12.2 mm), and was completed by 21.4 mm TL (17.5 mm) and 19.3 mm TL (15.9 mm), respectively. Squamation started at three centers of differentiation: the middle region of the trunk, the anterior of the caudal peduncle, and on the caudal peduncle at 23.8 mm TL (19.4 mm). The body was covered with scales, except the snout, at 57.2-60.2 mm TL (SL 47.1-49.2 mm).

  • The author carried out an experiment to find out the response of sea-bass, Epinephelus septemfasciatus (Thunberg) to the color lights. The experimental tank (360L$\times$50W$\times$55H cm) was set up in a dark room. Six longitudinal sections with 60 cm intervals are marked in the tank to observe the location of the fish. Water depth in the tank was kept 50 cm level. Light bulbs of 20 W at the both ends of the tank projected the light horizontally into the tank. Two different colored filters were selected from four colors of red, blue, yellow, and white, and they were placed in front of the light bulbs to make different colors of light. Light intensity were controlled by use of auxiliary filters intercepted between the bulb and the filter. The fishes were acclimatized in the dark for 50 minutes before they were employed in the experiment. Upon turning on the light, the number of fish in each section was counted 40 times in 30 second intervals, and the mean of the number of fish in each section was given as the gathering rate of the fish. The colors favourited by the fish was found in the order of blue, red, white and yellow in day time, and red, blue, yellow and white at night time. The gathering rate of fish on illumination period was not constant and fluctuated with irregularity. The difference of the gathering rate on two different colors of light was small and the difference was larger in night time then in day time.

The Patterns of Oxygen Consumption In Six Species of Marine Fish (해산어류 6종의 산소소비 경향에 관한 연구)

  • Oxygen consumption of marine fishes according to different water temperatures, fish population densities and body weights was measured in the respiratory chamber for the following six species: the olive flounder Paralichthys olivaceus, the tiger puffer Takifugu rubripes, the rockfish Sebastes schlegeli, the sea bass Lateolabrax Japonicus, the red seabream Pagrus major and the black seabream Acanthopagrus schlegeli. Also the lethal concentration of dissolved oxygen in them was determined. Oxygen consumption in each fish species increased as the water temperature increased. The relationship between the oxygen consumption rate $(Oc,\;ml/kg{\cdot}\;hr)$ and the water temperature (T,$^{\circ}C$) for each species appeared as the following equations demonstrate; olive flounder: Oc=34.0515T-339.5987 $(r^2=0.9730)$, tiger puffer: Oc=34.4941T-479.8732 $(r^2=0.9483),$ rockfish: Oc=44.7970T-634.2627 $(r^2=0.9718),$ sea bass: Oc=26.1488T-318.0633 $(r^2=0.9316),$ red seabream: Oc=61.1020T-722.8926 $(r^2= 0.9805),$ black seabream: Oc=75.1460T-947.9370 $(r^2=0.9392).$ The of gen consumption of fish with different population densities decreased as the number of fish increased. As the body weight of the olive flounder increased, the mass-specific oxygen consumption decreased. The relationship between oxygen consumption and body weight (W; g) was expressed as Oc=2532.0268W-0.6565 $(r^2=0.9229)$. The levels of lethal dissolved oxygen in the olive flounder, rockfish, tiger puffer and red seabream were 0.66, 0.79, 0.75 and 1.36 m1/1, respectively.
