Physiological Activities According to Cultivars and Parts of Ulsan Pear (울산지역 생산 배의 품종 및 부위별 생리활성)

  • This study was performed to confirm physiological activities according to cultivars and parts of Ulsan pear (wonhwang, pungsu, whangkeum, whasan and shingo). Total contents of phenolic compounds from peel, pulpy substance and core were 699.3-800.6, 51.5-112.5 and 254.0-401.5 mg/100 g as tannic acid equivalent, respectively. There were difference contents by cultivars, and peel and core of shingo and pulpy substance of wonhwang showed high contents, respectively. Total flavonoid contents of peel, pulpy substance and core were 125.2-164.2, 25.9-35.9 and 45.1-60.0 mg/100g, respectively and those of shingo cultivar showed comparatively high. Electron donating ability was in the order of peel (66.1-90.7%), core (48.5-82.8%) and pulpy substance (24.9-58.2%), and whasan cultivar showed comparatively low. Nitrite scavenging activity was in the order of peel (58.2-100.8%), core (59.5-86.2%), pulpy substance (39.9-82.5%). There were little difference by cultivars of core but peel and pulpy substance of shingo cultivar showed comparatively low nitrite scavenging activity. And as the concentration of each extract increased, nitrite scavenging activity increased. Xanthine oxidase inhibition rate was in the order of peel (14.1-75.4%), core (5.3-71.8%), pulpy substance (2.2-67.5%). And there little difference by cultivars and which was increased as the concentration of each extract increased.

Occurrence of Weed Flora and Changes in Weed Vegetation in Orchard Fields of Korea (우리나라 과수원에 발생하는 잡초 현황과 군락 변화)

  • This study was investigated in order to occurrence of weed flora in orchard (apple, grape, peach, pear etc.) fields and to the establishment of weed control basic data in these weeds. And this study was survey on 2015. As a result of the study on orchard weeds, there were total of 492 weeds including 63 families. Among all the weeds, 159 species were annual, 97 species were winter annual and 236 were perennial. Each fruit tree patterns of occurrence weeds, 306 species 50 families in apple orchard, 304 species 50 families in pear orchard, 286 species 50 families in grape orchard, and 288 species 48 families in peach orchard. Dominance was the highest with Digitaria ciliaris followed by Artemisia princeps, Stellaria aquatica, Commelina communis, Acalypha australis etc. in order. And exotics weeds occurred 127 species. Trifolium repens was the highest in importance analysis and the followings were in order of Conyza canadensis, Chenopodium album, Taraxacum offcinale, Rumex crispus etc. Changes in weed vegetation in orchard during 30 years, did not very many changed. In 1990, D. ciliaris, Persicaria hydropiper, Portulaca loeracea were dominant. In 2003, D. ciliaris was the most abundant species, followed by A. australis, A. princeps species. And in 2015, also D. ciliaris was the most dominant, followed by A. princeps, S. aquatica.

Growth and Survival of the Spat of Arkshell, Scapharca broughtonii in Intermediate Culture with Different Shape of Protective Net and Type of Preventive Net of Spat Loss (피조개, Scaphara broughtonii 부착치패의 중간양성시 보호망 형태와 유실방지망 종류에 따른 성장과 생존)

  • Growth and survival of the spat of arkshell, Scapharca broughtonii were investigated for the improvement of survival in intermediate culture with different shape of protective net and type of preventive net of spat loss. The growth performance of the spat of arkshell was observed for 60 days by using different forms of protective net, as the exposure form with an average shell length of $12.8{\pm}3.2$ mm(P<0.05), the fish pot form with $12.2{\pm}3.5$ mm, the cylinder form with $11.9{\pm}3.8$ mm and the last one is the rectangular form with $10.9{\pm}3.7$ mm. Their numerical value did not show any marked difference with each other. But in case of survival rate, the fish pot form showed highest survival rate which is 43.7% and the significantly lowest was showed by the exposure form, which is 5.4%(P<0.05). The cylinder form and rectangular form showed 41.2 and 31.6% respectively. And then the growth of the spat of arkshell was observed for 163 days by rearing in a sort of preventive net, the first group was in a blackout curtain with average shell length of $13.9{\pm}3.1$ mm, the next group was in a balsam pear net with $12.9{\pm}3.0$ mm, in a polyethylene net with $11.8{\pm}3.1$ mm and the control with $12.6{\pm}3.3$ mm which was not installed by preventive net of spat loss. The survival rate was 91.5% in a blackout curtain, 90.1% in a polyethylene net, 88.5% in a balsam pear net and 61.5% in a controlled group. It is seen that the highest growth and survival rate were observed in a fish pot form and cylinder form. These results were a little bit difference from those of the spat of arkshell cultured in the form of different preventive nets. There was no significant difference in survival rate in relation to the sorts of preventive net against carrying-away, but there was a difference in survival rate by more than 30% as against the non-installed controlled group. We expect that survival rate would be highly improved in intermediate culture carried out with fish pot form and cylinder form of protective net and preventive net of spat loss.

The Characteristics of Anatomical Structure and Fruit Quality According to Fruit Developmental Stage of Pyrus pyrifolia Nakai cv. Manpungbae ('만풍배'의 생육기별 해부학적 구조와 과실품질)

  • This study was carried out to understand the physiological characteristics of 'Manpungbae' (Pyrus pyrifolia Nakai) pears through the seasonal changes of pericarp structure and anatomical differences between bagging and non-bagging treatment, and also fruit quality and peel coloration characteristics at the harvest time. The pericarp at full bloom was consists of outer epidermis, hypodermis, parenchyma cell, and inner epidermis from the exterior. The cell layers from the outer epidermis to vascular bundle increased rapidly 7-10 layers to 18-26 layers from full bloom (FB) to 77 days after full bloom (DAFB) and did not change significantly until maturity. Thus, the cell division period of 'Manpungbae' pear was until 77 DAFB and during this period, the thickness from hypodermis to vascular bundle increased from $73.1{\mu}m$ to $195{\mu}m$ in this period. Stone cells were formed from seven to 21 DAFB and stone cell clusters were formed around 49 DAFB. The cork cell layer was formed between 49 and 77 DAFB. 'Manpungbae' fruit pericarp was consists of 4.5 layers of the cork cell layers and seven layers of hypodermis which has the tannin at harvest time (161 DAFB). Comparison of the fruit enlargement and fruit structure development by bagging or non-bagging showed that 'Manpungbae' fruits without bagging had more than three cork cell layer than those with bagging at maturity. The size of stone cell clusters were varied in two treatments. Fruit weight was higher in the non-bagging treatment but there was no difference in soluble solid contents (SSC) between two treatments. The weight of the 'Manpunbae' fruit was distributed from 301 g to more 900 g and the average fruit weight was 677.2 g at harvest time, and fruits in the range of 551-800 g accounted for 71.6% of total production. The SSC, acidity and SSC/acidity ratio was $10.2-12.1^{\circ}Brix$, 0.10-1.24% and 9.76-14.31 respectively, and the SSC was higher in bigger fruit which had a very higher positive correlation with a fruit weight. However, the fruit firmness tended to be lower with fruit size which had a very higher negative correlation with the fruit weight and SSC. The cork cell layer numbers between yellowish brown and green pericarp were not different significantly, in 3.8 and 3.5 respectively.

  • Brassinosteroid (BR), a plant steroid hormone, plays key roles in numerous growth and developmental processes as well as tolerance to both abiotic and biotic stress. To understand the biological networks involved in BR-mediated signaling pathways and stress tolerance, we performed comparative genome-wide transcriptome analysis of a constitutively activated BR bes1-D mutant with an Agilent Arabidopsis $4{\times}44K$ oligo chip. As a result, we newly identified 1,091 (562 up-regulated and 529 down-regulated) significant differentially expressed genes (DEGs). The combination of GO enrichment and protein network analysis revealed that stress-related processes, such as metabolism, development, abiotic/biotic stress, immunity, and defense, were critically linked to BR signaling pathways. Among the identified gene sets, we confirmed more than a 6-fold up-regulation of NB-ARC and FLS2 in bes1-D plants. However, some genes, including TIR1, TSA1 and OCP3, were down-regulated. Consistently, BR-activated plants showed higher tolerance to drought stress and pathogen infection compared to wild-type controls. In this study, we newly developed a useful, comprehensive method for large-scale identification of critical network and gene sets with global transcriptome analysis using a microarray. This study also showed that gain of function in the bes1-D gene can regulate the adaptive response of plants to various stressful conditions.

Comparison of Changes in Cell Wall Characteristics during Storage in 'Niitaka' and 'Pingguoli' Pear Fruits (동양배 '신고'와 '사과배' 과실의 저장 중 세포벽 특성의 변화 비교)

  • The characteristics of fruit ripening and cell wall modifications of two oriental pear fruits were compared during storage. The loss of fresh weight was lower in 'Pingguoli' than 'Niitaka', probably due to the difference in wax accumulation and skin structure. 'Pingguoli' produced much higher amount of ethylene and showed climacteric ripening pattern, but this response was not found in 'Niitaka'. A significant difference in soluble pectin contents was found, thus, the amount of water soluble pectins were much higher in 'Niitaka' at harvest but this difference was not significant at later stage of storage. A severe loss of water soluble pectins in 'Niitaka' was confirmed whereas a similar tendency was not found in CDTA soluble ones. Even the amount of $Na_2CO_3$ soluble pectins was relatively low, 'Pingguoli' contained approximately 2 times higher than 'Niitaka' and both cultivars showed an increasing tendency in $Na_2CO_3$ soluble polymers at later stage of storage. No significant difference in alkali soluble polysaccharides (hemicelluloses fraction) was found between both cultivars. There was a significant change in gel filtration profiles regardless of cultivars, especially in water soluble pectins resulting from severe depolymerization probably due to degradation of higher molecular weight pectins and loss of their side chains. In gel filtration profiles of $Na_2CO_3$ soluble pectins, neutral sugars were evenly distributed regardless of molecular size of pectic polymers but the decrease of high molecular weight fraction was found. In comparison of alkali soluble polymers, a similar trend was found, that is, there was no difference in gel filtration profiles but 'Niitaka' seemed to have longer side chains in alkali soluble polysaccharides in both 4% and 24% of KOH soluble fractions.

Atomic Absorption Spectrophotometric Analysis of Copper In the Soil s of Orchards (원자흡광법(原字吸光法)에 의(依)한 과수원(果樹園) 토양중(土壤中)의 동함량(銅含量) 분석(分析)에 관(關)한 연구(硏究))

  • This study was conducted to detect copper which is considered in the soils of orchards, since copper fungicide has been applied to fruit trees. Soil samples taken from the fields of the chief producing districts of apple (Chungju, Yesan, Daegu), pear (Yangju, Bucheon, Seonghwan) and citrus (Seogypo in Jeju island) were analysed by an atomic absorption spectrophotometer. The results obtained were summarized as follows ; 1. In orchards of apple, the amount of copper of soils from Yesan, Chungju and Daegu were ranged 2.6-171.3ppm, 2.2-136.1ppm and 14.3-134.6ppm, respectively. Very little copper was detected from the soils in the field which has been cultivated for less than 20 years. About 100ppm and 130-170ppm of copper were detected in the field which has been cultivated for 30 years and for 50-60 years, respectively. Most of the copper was detected in the surface layer of soils (0-10cm), while very low content of copper was detected in the deeper layer of soils (10-20cm). 2. In orchards of pear, 20-30ppm of copper was detected from the surface of soils in the field which has been cultivated for more than 30 years and the highest level of copper, 36.8ppm, was detected from Yangju area. The amount of copper of soils from Yangju, Seonghwan and Bucheon were ranged 3.6-36.8ppm, 9.7-19.4ppm and 3.6-24.7ppm, respectively. 3. In orchards of citrus of Jeju island, only trace amount and 23-38ppm of copper were detected in the fields cultivated for 15 years and 20-30 years, respectively. The highest level of copper, 57ppm, was detected from the surface layer of soils in the field which has been cultivated for 35 years, but in most of the soil samples tested, only the natural background level of copper, about 20ppm, was detected. 4. The levels of copper residue in all the soil samples tested were lower than the tolerance level (125ppm of copper which is extracted in 0.1N-HCl solution), except those of copperr residue, 130-170ppm, that were detected from the orchards of apple which have been under cultivation for 50-60 years. Hence no problem for the farming could be speculated with the present concentration of copper analysed.

A Study on the cooking in 'The Joobang' ("주방(酒方)"의 조리가공에 관한 분석적 고찰)

  • 'The Joobang' is a cooking book written purely in Korean which came from a Mr. Lee. It is unknown when and by whom this book was written, but it is inferred from red pepper mentioned in this book and from Korean-writting type that the book was written around in the mid-nineteenth century. The contents of 'the Joobang' consist of thirty seven items in all : sixteen items of rice alcoholics, five side dishes, eleven Tucks(rice cakes) and Guaja(cookies) and five processed materials. Cereals for making rice alcoholics were regular rice and sticky rice. In the first brew, rice alcoholics is made all from regular rice, and in the second brew, fifty six percent of regular rice and forty four percent of sticky rice was made use of. For five items of alcoholics forming 31.3% of alcoholics, flour was added, in which case the proportion of leaven to cereals was lower. There were four types of processing cereals in brewing rice alcoholics Jee-ae-bop(steamed rice, 60.9%), thick gruel with cereals ground and completely cooked(8.7%), thick gruel with cereals ground and half cooked (26.1%) and GooMung Tuck(doughnut-shaped rice cake, 4.3%). The comparison of the materials for brewing rice alcoholics and of the types of processing cereals presented in the seven cooking books purely in Korean(Umsik Deemeebang, JoobangMoon, Umsikbo, Sool-mandununbop, Kyuhap chongseo. The Kims' Joobangmoon and The Lee's Umsikbup) showed us the following facts ; Yihwajoo (rice alcoholics brewed around the blooming time of pear trees) and Kwahajoo(rice alcoholics which passed the summed time) were most often introduced and commonest, and the materials for brewing and the types of processing little changed over times. The materials for side dishes were all animal food, and vegetable food was little used for side dishes. Red pepper was used for seasoning. It is a problem of Korean traditional cooking that there were no units of measuring for side dishes. The main materials for Tuck(rice cakes) and Guaja(cookies) were sticky rice, regular rice and honey. There was a variety of terms for the process, from which we can see that the procedures of making Tuck(rice cakes) and Guaja(cookies) were very complicate. Processed materials were chiefly made of sticky rice and the material of firewood(oak trees) and the strength of fire(fire burning vigorously and fire turning to ashes) were described.

A Study on Traditional Korean Medical Countermeasures of Influenza A by Case Histories (의안(醫案)을 통한 신종 인플루엔자의 한의학적 대처방안)

  • Background: The recent outbreak of the novel strain of influenza A (H1N1) virus has raised a global concern of the future risk of pandemic. Traditional Korean Medicine(TKM) has been combatting against contagious diseases and developed its own particular and efficient way in treating those diseases. Objectives: Provide a distinctive and effective TKM method in understanding the principles of treatment, prevention, and contraindications against influenza A through case histories. Method: We revised case histories of eminent doctors of Ming and Qing dynasties according to their clinical manifestations similar to those of influenza A. We also verified prescriptions of the "Dong-uibogam(東醫寶鑑)" through examining clinical practices of China today. Results: 1. The subtypes of Warm disease; Wind Warmth and Pestilence has the most similar clinical manifestation in comparison to Influenza A. Specifying these terms is important in recognizing and classifying various diseases under the name of Warm disease. 2. TKM considered not only external factors but also individual factors like general condition, diet, emotion, constitutional types, etc in the treatment of febrile diseases. 3. TKM developed a new way in the treatment of contagious diseases. considering the characteristics of each pathogen. TKM described the Warm pathogen being light as a feather, which enters through the nose, principally affecting the Lung. Therefore, they emphasized treatment with Pungent Sweet Cooling Moistening[辛甘凉潤] method. 4. As the conservation of fluid and humor is the primordial concern in the treatment of Warm disease, they restricted release of the exterior with pungent-warm and purgation method. The purgation methods was used exclusively to decrease fever and preserve fluid and humor. 5. The only differentiating characteristics of Influenza A with seasonal influenza are vomiting and diarrhea. Case research revealed the possibility of these manifestations to be a mechanism of restoration. 6. TKM provides alimento prevention method like Mint Pear Porridge, mung bean, and etc also combination of herbal medicine. Also emphasized in the conservation of essence for the prevention of contagious diseases. Conclusions: TKM developed its unique way in understanding the pathogenesis, diagnosis, treatment, and prevention of contagious diseases and formed its independent scheme of Warm Disease. This knowledge in febrile contagious disease is relevant today in providing diverse treatment and prevention for influenza A.

Relations of Glaucoma and Dietary Pattern Factor (녹내장과 식사패턴요인의 관련성)

  • Purpose: This study was performed to identify the relations between glaucoma and dietary patterns in Korean adults, aged 30 years or older. Methods: 6,453 subjects (male 2,759, female 3,694 subjects) who aged 30 years or older from the Korea National Health and Nutrition Examination Survey were included. Glaucoma was assessed based on the subjects were diagnosed by a doctor. At first, waist circumference and body mass index (BMI) were compared to investigate the association between glaucoma and obesity index. Secondly, dietary patterns using factor analysis for external blue colored fish(mackerel), seaweed(seaweed, laver), bean(bean, soybean milk), vegetables(sprout, spin, carrot, cabbage, cucumber, pepper, tomato, pumpkin) and fruits(tangerine, apple, pear, strawberry, grape, peach) were obtained. Lastly, regression analysis was used for glaucoma and emerged dietary factors. Results: In glaucoma and normal group, waist circumference and BMI were $82.25{\pm}9.96cm$ and $23.27{\pm}2.84kg/m^2$, $75.79{\pm}13.79cm$ and $22.39{\pm}4.06kg/m^2$, respectively. There was statistically significant difference (p<0.05). Three dietary patterns were emerged from the factor analysis, and factor 1 was included sprout, spin, carrot, cabbage, seaweed, cucumber, mackerel, laver. The factor 1 of dietary pattern was inversely related to the prevalence of glaucoma (OR: 0.697, 95%CI: 0.496-0.980, p=0.038), and model adjusting for age and sex was showed the same trend (OR: 0.698, 95%CI: 0.497-0.982, p=0.039). Conclusions: This study was analysed first with glaucoma and dietary pattern in Korean, and vegetables, laver and mackerel significant associated to reduce the risk of glaucoma.
