Detection of TMV, ToMV, and CMV from Tomato Seeds and Plants (토마토 종자와 식물체로부터의 TMV, ToMV 및 CMV 검출)

  • For the detection of Tobacco masaic virus (TMV), Tomato mosaic virus (ToMV), and Cucumber mosaic virus (CMV), tomato seeds of 11 table tomato and 7 cherry tomato cultivars were assayed by DAS-ELISA. Among the cultivars, TMV and ToMV were detected from 9 cultivars at the rates lower than 20% and 16%, respectively. In the assay on seed transmission rates, ToMV and CMV were detected as high as 24% and 8% , respectively, but TMV was not detected. In field survey on these viruses from tomato plants of 10 different places in Chungbuk province, ToMV and CMV were detected from most fields. TMV was detected from only 3 fields. The highest detection rates of these viruses were recorded in Cheongwon for TMV Chungju for ToMV, and the other locality of Chungju far CMV. It was difficult to find any relationship between the growth stage of tomato and infection rates. TMV usually caused mosaic on leaves while ToMV caused various symptoms including yellows, necrosis, and mottling. CMV-infected tomato plants showed symptoms of shoestring, fern leaf, and yellows.

Spherical and Aspherical RGP Lens Fitting to Epithelium and Endothelium of Rabbit's Cornea -Scanning Electron Microscopy (구면과 비구면 RGP 렌즈 장기 착용시 각막 상피, 내피에 미치는 영향-주사전자현미경적 관찰)

  • To investigate the comparative effect of spherical and aspherical RGP lens were worn during 3 weeks on rabbit's cornea. Four white rabbits were worn right eyes with spherical lens and 4 white rabbits were worn right eyes with aspherical RGP lens. Left eyes were served as control. The rabbits were sacrificed at 3 weeks after fitting and observed morphological changes by scanning electron microscopy and also investigate proliferation rate of the corneal epithelium with RGP wearing. After spherical RGP lens wearing, the epithet layer damaged compared to aspherical lens. The superficial cell layer strip off seriously, cell size significantly changed abnormal. Both spherical and aspherical RGP lens fitting group showed so many bacteria and back surface of lens was found like a fern shape. The aspherical RGP lens original material type was some formal than spherical lens. We thought that these pattern was significantly altered with spherical lens by prohibited transmitter oxygen from atmosphere therefore the epithelium shape was changed. This suggested wearing the aspherical lens might be less physiologic than shperical lens fitting.

Effects of Medium Components and Composition on Mass Propagation of Arachniodes aristata (G. Forst.) Tindale (가는쇠고사리의 대량번식에 미치는 배지구성물질과 배양토의 영향)

  • This study was conducted to investigate in vitro mass propagation methods suitable for each growth stage of A. aristata (G. Forst.) Tindale, from spore germination to sporophyte formation. Among spores germinated in $1/8-1{\times}MS$ medium and Knop medium, Knop medium yielded the highest germination percentage (87.1%). We cultured prothalli obtained from germinating spores for 8 weeks on media with different concentrations of sucrose and active carbon, as well as different concentrations and ratios of nitrogen, to select a suitable growth medium. A. aristata (G. Forst.) Tindale prothalli grew most actively in MS medium with 3% sucrose and 20 : 40 mM of $NH_4Cl$ and $KNO_3$ (total concentration of 60 mM). We investigated sporophyte formation according to soil type, finding that bedding soil mixed with perlite at a 2 : 1(v / v) ratio yielded the highest number of sporophytes per pot ($73.8/7.5{\times}7.5cm\;pot$). By contrast, when peat moss was used alone or mixed with other substrates, prothallus development and sporophyte formation were suppressed. Therefore, the most effective propagation method for A. aristata (G. Forst.) Tindale is to grow prothalli in MS medium and to induce sporophyte formation in a mixture of bedding soil and perlite (v / v = 2 : 1).

Medium Composition Affecting In Vitro Regeneration of Matteuccia struthiopteris (청나래고사리의 기내 포자체 재생에 미치는 배지 구성물질의 영향)

  • This study was carried out to investigate the efficient in vitro mass propagation methods for juvenile sporophytes of Matteuccia struthiopteris. Chopped segments of pinnae, petiole and rhizome were cultured on 1/2MS with 0.1% activated charcoal. Among these explant sources only rhizome segments produced young sporophytes, regenerating vigorously on 1/2 MS medium. Adjusting sucrose concentration to 2% and supplement to $50mgL^{-1}$ $NaH_2PO_4$ in 1/2MS medium proved to be more efficient for plant regeneration. Various combinations of growth regulators such as kinetin, BA, NAA, and IBA were added to the growing media, and the best sporophyte regeneration was obtained by $1{\mu}M$ kinetin. The BA addition resulted in vigorous proliferation of meristematic tissues, but without differentiation to sporophytes. Three types of culture methods, solid using agar, liquid stationary, and liquid shaking culture, were employed with or without activated charcoal. The addition of 0.1% activated charcoal to modified 1/2MS media (2% sucrose, $50mgL^{-1}$ $NaH_2PO_4$, $1{\mu}M$ kinetin, pH 5.8 and 0.8% agar) yielded highest sporophyte regeneration in liquid shaking culture.

Optimal Culture Conditions for Masspropagation of Gametophytes and Sporophytes of Pyrrosia linearifolia by Tissue Culture (조직배양을 이용한 우단일엽의 대량번식을 위한 전엽체와 포자체의 적정 배양조건)

  • This study was carried out to investigate the optimal culture conditions for gametophytes growth and sporophytes regeneration of Pyrrosia linearifolia in order to provide for the masspropagation system foundation of Pyrrosia linearifolia using their life cycle. Among many different media, 2MS medium was most effective in prothallus proliferation. Prothallus growth was promoted as the total concentration of nitrogen sources increased, and the best result was observed on 120 mM nitrogen. The best concentration of sucrose was 3%. The addition of 5~20 mM IAA, NAA, BA and kinetin promoted the propagation of prothallus. But 2iP demonstrated the most inhibitory effect on prothallus proliferation. Gametophytes shaking-cultured with liquid medium showed similar growth with solid medium and normal formation of reproductive organs. Shoot regeneration was most effective on 1/8MS medium, but growth was promoted on 1/2MS medium. For promotion of shoot regeneration and growth, the suitable concentrations of sucrose and $NaH_2PO_4$ were 1% and $50{\sim}100mg{\cdot}mL^{-1}$ in 1/8MS medium, respectively.

  • Recent studies have shown that sodium/glucose cotransporter 2 (SGLT2) inhibitors might exert favourable changes on cardiac parameters as observed on cardiovascular imaging. We conducted a systematic review and meta-analysis to determine the effects of SGLT2 inhibitors on cardiac imaging parameters. Four electronic databases (PubMed, Embase, Cochrane, Scopus) were searched for studies in which the effects of SGLT2 inhibitors on cardiac imaging parameters were examined. Studies in which a population was administered SGLT2 inhibitors and analysed by echocardiography and/or cardiac magnetic resonance (CMR) imaging were included. Random-effects pair-wise meta-analysis models were utilized to summarize the studies. A total of 11 randomized controlled trials was included with a combined cohort of 910 patients. Comparing patients receiving SGLT2 inhibitors with subjects receiving placebo, the mean change in CMR-measured left ventricular mass (LVM) was -3.87 g (95% confidence interval [CI], -7.77 to 0.04), that in left ventricular end-systolic volume (LVESV) was -5.96 mL (95% CI, -10.52 to -1.41) for combined LVESV outcomes, that in left atrial volume index (LAVi) was -1.78 mL/m2 (95% CI, -3.01 to -0.55) for combined LAVi outcomes, and that in echocardiography-measured E/e' was -0.73 (95% CI, -1.43 to -0.03). Between-group differences were not observed in LVM and LVESV after indexation. The only between-group difference that persisted was for LAVi. Treatment with SGLT2 inhibitors resulted in reduction in LAVi and E/e' on imaging, indicating they might have an effect on outcomes associated with LV diastolic function.

A Survey on the Usage of Wild Grasses (산채류의 이용실태에 대한 조사)

  • This survey investigated on the degree of acknowledgment, intake frequency and using methods of wild grasses of the residents in Kyeonggi, Kangwon, Chunnam, Chungbook, Kyeongnam & Cheju area. The results were as follows: 1. An answer that impression of wild grasses is 'nature food' was the highest(42.6%). It was founded that interest of nature food has been increasing. 2. It has been shown that the most common method for elimination of astringent taste is to blanch and then wash several times with water.(62.9%) 3. The most common obtaining routes was traditional market. It has been shown that rate of gathering in the fields is higher in rural community and the group of over 60 years old than that of urban community and the other groups. 4. In rural area, people(28.7%) ate wild grasses more frequently than in urban area. Family with the old and the group of over 40 years old eat wild grass often. 5. More than 95% of answerers were familiar with the names of wild grasses, codonopsis lanceolata, chinese bellflower, braken, mugwort, wild rocambole and edible shoots of a fatsia & Korean lettuce are known to them with over 88.6%, 85.1% respectively. Frequently eaten wild grasses were braken, chinese bellflower, wild tocambole, codonopsis lanceolata, mugwort and korean lettuce orderly. But in Chunnam & Kyeongnam, they were braken, chinese bellflower, mugwort, wild rocambole and Korean lettuce orderly. 6. Wild grasses eaten with rice were total 25 species, and mugwort had the highest usage rate with 41.9%. Mugwort was used for $D'{\breve{o}}k$(rice cake)(77.8%), T'wigim $J{\breve{o}}n$(deep fat fried dish swallow fat fried dish)(50.1%) and liquor(4.6%). In the cooking of Guk(soup), Chigae(stew), Sengch'ae(raw vegetable) and kinds of wild grasses for Namul(cooked seasonal vegetable) & Bokkum(saute) are the most various with 58 species, especially braken was most commonly used for saute. Sedum & Korean lettuce were the common ingredients of Kimch and codonopsis lanceolata was used in liquer & Jangachis(pickle) with 40%, 46% each. Chinese bellflower was used with the most variable cooking method. 7. 43 species of wild grasses were stored by blanching and drying, and braken had the highest rate(32.3%) then aster scaber, flowering fern, Pimpinella brachycarpa, mugwort and ligularia orderly.

STUDIES ON THE EXTRACTION OF SEAWEED PROTEINS 4. Precipitation Conditions and Nutritional Evaluation of Isolated Seaweed proteins (해조단백질 추출에 관한 연구 4. 추출단백질의 심전조건 및 영양적 평가)

  • For the effective utilization of diverse and abundant resource of seaweeds in Korea as a food protein supplment, extraction conditions of water, salt, and alkali soluble proteins were investigated in previous work(Ryu and Lee, 1977: Lee et al., 1977: Lee et al., 1978). The present study as a part of the serial work was thus aimed to find the conditions of isolation and purification of extracted proteins, and to evaluate the nutritional quality of the isolated seaweed proteins in terms of amino acid composition, chemical score, protein score, modified essential amino acid index(MEAAI), and in vitro digestibility presented as pepsin-pancreatin digest residue index (PPDRI). As for the isolation of extracted proteins, TCA treatment was more effective for the proteins from rhodophyceae and Chlorophyceae while the precipitation at isoelectric point was more desirable for Phaeophyceae proteins. In amino acid composition, water soluble protein fraction was superior to the other fractions in Porphyra suborbiculata whereas both water and alkali soluble fractions seemed to bo more benefitial for Enteromorpha linza and Ulva pertusa; the extraction with alcohol-alkali mixed solvent for Undaria pinnatifida and Sargassum fulvellum. Glutamic acid and aspartic acid content was particularly high in all protein fractions examined. The total amino acid content of Porphyra suborbiculata and Enteromorpha linza was almost equivalent to that of dried whole egg although the essential amino acid content was lower. A comparative analysis was made on the inedexes between raw seaweed powder and isolated protein. Chemical score of Porphyra suborbiculata and Ulva pertusa was approximately 35 and 56 in cafes of raw powder and isolated protein respectively while only 10 to 16 for raw powder of Undaria pinnatifida and Sargassum fulvellum and 30 to 35 for their isolated proteins. Protein score of all isolated proteins was in the range of 63 to 73 which indicates that isolated protein would be mere valuable than the fern of raw seaweed powder. Digestibility by means of PPDRI was found to be extremely low in case of raw powder but it could be doubled in case of isolated protein yielding 67 to 70 for Porphyra suborbiculata and Ulva pertusa.

Evaluation for Soil Moisture Stabilization and Plant Growth Response in Horizontal Biofiltration System Depending on Wind Speed and Initial Soil Moisture (풍속과 초기 토양수분에 따른 평면형 바이오필터 내 토양수분 안정화 및 식물 생육반응 평가)

  • The final aim of this study is to develop a biofiltration system integrated with plant vegetation for improving indoor air quality effectively depending on indoor space and characteristics. However, to approach this final goal, several requirements such as constant pressure drops (PDs) and soil moisture contents (SMCs), which influence the capacity design for a proper ventilation rate of biofiltration system, should be satisfied. Thus, this fundamental experiment was carried out to adjust a proper wind speed and to ensure a stabilization of initial SMCs within biofilter for uniform distribution of SMCs and PDs, and for normal plant growth, especially avoiding root stress by wind. Therefore, we designed horizontal biofliter models and manufactured them, and then calculated the ventilation rate, air residence time, and air-liquid ration based on the biofilter depending on three levels of wind speed (1, 2, and $3cm{\cdot}s^{-1}$). The relative humidity (RH) and PD of the humidified air coming out through the soil within the biofilter, and SMC of the soil and plant growth parameters of lettuce and duffy fern grown within biofilter were measured depending on the three levels of wind speed. As a result of wind speed test, $3{\cdot}sec^{-1}$ was suitable to keep up a proper RH, SMC, and plant growth. Thus, the next experiment was set up to be two levels of initial SMCs (low and high initial SMC, 18.5 and 28.7%) within each biofilter operated and a non-biofiltered control (initial SMC, 29.7%) on the same wind speed ($3cm{\cdot}sec^{-1}$), and measured on the RH and PD of the air coming out through the soil within the biofilter, and SMC of the soil and plant growth parameters of Humata tyermani grown within biofilter. This result was similar to the first results on RHs, SMCs, and PDs keeping up with constant levels, and three SMCs did not show any significant difference on plant growth parameters. However, two biofiltered SMCs enhanced dry weights of the plants slightly than non-biofiltered SMC. Thus, the stability of this biofiler system keeping up major physical factors (SMC and PD) deserved to be adopted for designing an advanced integrated biofilter model in the near future.

Relationship of Depression, Irrational Faith and the School Adaptation of Elementary School Students (초등학생의 우울성향, 비합리적 신념, 학교적응의 관계)

  • The purpose of this study was to investigate the differences in depression, irrational faith and the school adaptation of elementary school students by their sex and region, to provide basic materials to teach children with the likelihood to be depressed by analyzing the correlation of depression, irrational faith and school adaptation, and help them to have healthy emotion and behaviors. To achieve these goals, following agenda were set. First, what are the differences in the depression, the irrational faith and the school adaptation of elementary school students by their sex and region? Second, what is the relationship between the depression characteristics and the irrational faith of elementary school students? Third, what is the relationship between the irrational faith and the school adaptation of elementary school students? Fourth, what is the relationship between the depression characteristics and school adaptation of elementary school students? : To resolve these agenda, the subjects were selected from Grade 6 students from the elementary schools locatec in Suwon and Hwaseong. The jubjects were selected from the two schools in Suwon and three schools in Hwaseong which were randomly chosen. The subjects were 670 in total. 70 insincere answers were excluded from the analysis. Therefore the subjects used in the analysis were 600 in total(150 male and female students in city respectively and 150 male and female students in rural area respectively). The tools used for this study were Children's Depression Inventory (CDI), Irrational Faith Test (over-self-reliance, dichotomy, determinism, over-concern, evasive faith, over dependent faith), School Adaptation Test (Attitudes to teachers and school environments, hobbies and specialties, academic achievements, peer relationship). Materials were processed with the SPSS program for ANOVA and correlation analysis. All the statistical values were verified at .05 level. The followings are the results of the analysis of the collected data. First, in the analysis of depression, there was significant difference between male and female students (F=4.75, p<.05). Female students (X =56.93) showed a little higher value than male students (X =53.83). There was significant difference between regions (F=13.02, p<.001), too. Rural area students (X =57.93) showed higher value than city area students (X =52.82). When analyzing irrational faith, there was significant difference between male and female students (F=S.60, p<.05). Female students (X =81.32) showed a little higher value than male students (X =77.72). There was no significant difference according to regions. When analyzing the school adaptation, there was no significant difference according to either sex or region. Second, depression and irrational faith showed highly positive correlations in all areas. In particular, over concern and evasive faith showed the highest correlation with depression inclination (r=.68, p<.001). When looking into the results by sex and by region, female students (r=.70, p<.001) and the rural area students (r=.69, p<.001) showed higher correlation between depression and irrational faith than male students and city area students (r=.63, p<.001). Additionally, in all areas including by sex and by region, a positive correlation was shown. In all variables such as male students (r=.63, p<.001), female students (r=.72, p<.001), city area students (r=.66, p<.001), and rural area students (r=.69, p<.001), over concern and evasive faith showed the highest correlation with depression inclinations. Third, irrational faith (overall) and its sub factors such as over-self-reliance, dichotomy, determinism, over-concern and evasive faith had significant negative correlations with school adaptation (overall) and its sub factors in the scope of $r=-.27{\sim}-.52$. Determinism out of the sub factors of the irrational faith had the highest negative correlation with school adaptation (overall) and sub factors at the scope of $r=-.37{\sim}-.51$. However, over-self-reliance showed the negative correlation with school adaptation (overall) and its sub factors only partially. When looking into the results by gender, both genders showed significant negative correlation between irrational faith (overall) and its sub factors with school adaptation. Male students showed highly negative correlations with school adaptation (overall) and its sub factors in determinism ($r=-.35{\sim}-52$) and over-concern and evasive faith($r=-.31{\sim}-.51$), and fern ale students showed in over-self-reliance ($r=-.27{\sim}-.45$). However over-self-reliance showed negative correlation with school adaptation and its sub factors only partially. When looking into by region, both city and rural areas showed significant negative correlation between irrational faith (overall) and it sub factors, and school adaptation. Rural areas showed higher negative correlation in irrational faith (overall) ($r=-.39{\sim}-.53$) and over-self-reliance ($r=-.32{\sim}-.44$) and dichotomy ($r=-.28{\sim}-.39$) than city areas. However over-self-reliance showed negative correlation with school adaptation and its sub factors only partially. Fourth, depression and school adaptation showed the negative correlation in all areas. In particular, academic achievements and peer relationship showed the highest negative correlation with depression (r=-.53 p<.001). When looking into the results by sex and by region, female students (r=-.62, p<.001) and rural area students (r=-.61, p<.001) showed higher negative correlation with depression and school adaptation than female students (r=-.56, p<.001) and city area students (r=-.57, p<.001). Although there was negative correlation in all areas by sex and by region, male students (r=-.52, p<.001) and rural area students (r=-.56, p<.001) showed the highest negative correlation in peer relationship and female students (r=-.57, p<.001) and city area students (r=-.56, p<.001) showed the highest negative correlation in academic records. Based on the results of the study, it is proposed to provide elementary school students sho are likely to be depressed very easily with careful counselling and teaching based on the attention and love in school fields so that they can adapt themselves to home, school and society with positive and reasonable thinking.
