Outbreak of Fowl Cholera in native chickens (토종닭에서 가금콜레라(Fowl Cholera) 발생)

  • Fowl cholera(FC), which has been classified as reportable animal disease in Korea, occurred in the backyard farm located in Chungju, Chungbuk province. The farm was not ordinary domestic poultry farm but backyard farm with main cultivation of fruits. The farmer reported unusual mortality of chickens which were grazed in his own fruit farm without any poultry house. There was a lots of opportunities to contact with wild animals including wild birds freely. Clinical signs observed in the chickens were torticolis, mucous excretion from mouth and greenish diarrhea. On the necropsy we found multifocal necrosis on the surface of liver, hemorrhages in the epicardium and ovary and mucopurulent exudates in the joint. The causative agents was isolated from the liver of chickens collected from the farms and identified as Pasteurella multocida using biochemical tests of bacteria. Also, the isolated bacteria were inoculated into specific pathogen free chickens to evaluate the pathogenicity. High mortality and similar pathological lesions compared to those of chickens died in the farm were observed. We report the first case of fowl cholera of the chickens in Korea and further studies are needed to evaluate the serotype, genotype and pathogenicity of isolated Pasteurella multocida.

Growth performance of crossed breed Korean domestic chickens for twelve weeks after hatching

  • The study was conducted to investigate the growth performance of two strains of Korean domestic chickens (KDC, i.e., 1A, and 2A). A total of 600, one-day-old female chicks were allocated in a completely randomized design to give 15 replicates for each strain (10 birds per pen). The chickens were fed (week 0 to 6: ad libitum; week 6 to 12: restricted feeding) commercial diets (week 0 to 5: crude protein 20.3% and metabolizable energy 3,059 kcal/kg; week 5 to 8: crude protein 18.6% and metabolizable energy 3,123 kcal/kg, and week 8 to 12: crude protein 16.7% and metabolizable energy 3,187 kcal/kg) together with fresh water. Body weight (BW) and feed intake were measured bi-weekly to calculate the average daily feed intake (ADFI), average daily gain (ADG), and subsequently the feed conversion ratio (FCR). The results indicate that the 1A crossbreed had a higher (p < 0.05) BW compared to the 2A crossbreed from week 6 to 10. A higher ADG (p < 0.05) was seen in the 1A crossbreed on week 6 and in the 2A crossbreed on week 8. Moreover, no difference (p > 0.05) was observed in the FCR between crossbreed 1A and 2A during the 12 weeks with the exception of week 8. Crossbreed 1A had (p < 0.05) an improved FCR compared to the 2A crossbreed on week 8. In conclusion, the 1A crossbreed had a higher growth performance compared to the 2A crossbreed for 12 weeks after hatching. This data can contribute by helping the KDC market.

Polygranulomatosis in the domestic chickens (닭에서 발생한 다발성 육아종증)

  • Three mature layer chickens from a farm in which chickens showed green diarrhea, cyanosis, lethargy, loss of appetite were pathologically examined. Grossly, multiple variable sized caseous nodules were detected in the liver, intestinal serosa and mesentery. In addition, parathypoid nodules in the liver and fibrous serositis on the several peritoneal organs and tissues were noticed. One of spleens had multiple infarction areas. Histologically caseous nodules consisted of central caseous core and peripheral epithelioid cells overlying the fibrous connective tissue. Multinucleated giant cells were scattered between the epithelioid cells and fibrous connective tissue. In these nodules Gram negative cocobacilus bacterial colonies were present, whereas Periodic Schiff reaction and Ziehl-Neelsen stain detected neither fungi nor acid fast bacteria. From these results multiple granulomas might be induced by Escherichia coli. In addition, severe Ascafdiodf and Salmonellosis were coinfected in these chickens.

Comparison of the performance of the laying period of crossbred Korean native chickens from 20 to 40 weeks of age

  • This study was conducted to investigate the laying performance of six strains of Korean domestic chickens (KDC 1A, 2A, 3A, 1B, 2B and 3B). A total of 288 20-week-old KDC hens were allocated in a completely randomized design to give 12 replicates per each strain (4 chickens per pen). The chickens were fed commercial diets (i.e., week 20 - 32, crude protein [CP] 18.0% and metabolizable energy [ME] 2,850 kcal/kg; week 32 - 40, CP 17.0% and ME 2,800 kcal/kg) ad libitum together with fresh water during the 20-week experiment. Body weight, age of sexual maturity, egg weight, hen-day egg production (HDP) and feed conversion ratio (FCR) were measured during the experiment period (week 20 - 40). Our results show that strain 3B had an improved (p < 0.05) HDP and FCR compared with the other strains on week 24 - 28. However, strain 3A had an improved (p < 0.05) HDP and FCR compared to the other strains on week 28 - 40. Similarly, strain 3A had a higher egg weight (p < 0.05) compared to the other strains on week 28, 32 and 40. In conclusion, the higher laying performance of strain 3A indicates its potential in the development of a KDC strain with improved commercial laying performance.

Pathogenicity of Salmonella typhimurium and S enteritidis isolated from domestic animals in Gyeongbuk province (경북지역 가축에서 분리된 Salmonella typhimurium과 S enteritidis의 병원성 시험)

  • The result of studying the pathogenicity of Salmonella typhimurium and S enteritidis isolated from domestic animals in Gyeongbuk province were summarized as follows. In Congo-red binding test, S typhimurium had much more rough types than S enteritidis. In colicin production test, 4 strains of S typhimurium were positive but all of S enteritidis were negative. In hemolysin production test, all of S typhimurium and S enteritidis were negative. In Guinea pig serum resistant test, all of S typhimurium and S enteritidis were positive. As a result of pathogenicity test to mice, 54.4% of mice were died. Therefore, S typhimurium and S enteritidis were considered as highly pathogenic. S typhimurium DT104 and S enteritidis PT4 were more pathogenic to mice than other phage types of same serovar. S typhimurium and S enteritidis were considered not so pathogenic for 6-day-old chickens. The recovery rates of Salmonella stains from mice and chickens inoculated were 96.8%, and 54%, respectively. In chickens, proportional to the time from 2 weeks after challenge inoculation, the recovery rates were noticeably decreased.

Serological survey of Japanese encephalitis virus in domestic animals (축종별 일본뇌염바이러스에 대한 항체보유율 조사)

  • A total of 1,024 sera were collected from cattle(227), pigs(465), chickens(257) and dogs(75) raised or slaughtered in Daejeon metropolitan city from April to September 2002. Japanses encephalitis virus(JEV) antibodies in sera were detected by the haemagglutination inhibition test. The prevalence rates of JEV antibodies were 99.1 %, 54.0 %, 63.0 % and 98.7 % in cattle, pigs, chickens and dogs, respectively. In case of cattle and dogs, the monthly antibody-positive rates were as high as 85.7∼100.0 % and there were no differences among six months. In case of pigs, the monthly antibody-positive rate showed the lowest in April(6.4 %) and the highest in July(100.0 %) and it remained above 50 % during the summer-time. In case of chickens, the monthly antibody-positive rate was 100.0 % in July & August, 80.5 % in June, 40.0 % in May, 7.5 % in September and 5.0 % in April in order and there were distinct differences in seasons.

Morphology and Molecular Phylogeny of Raillietina spp. (Cestoda: Cyclophyllidea: Davaineidae) from Domestic Chickens in Thailand

  • Raillietina species are prevalent in domestic chickens (Gallus gallus domesticus) in Phayao province, northern Thailand. Their infection may cause disease and death, which affects the public health and economic situation in chicken farms. The identification of Raillietina has been based on morphology and molecular analysis. In this study, morphological observations using light (LM) and scanning electron microscopies (SEM) coupled with molecular analysis of the internal transcribed spacer 2 (ITS2) region and the nicotinamide adenine dinucleotide dehydrogenase subunit 1 (ND1) gene were employed for precise identification and phylogenetic relationship studies of Raillietina spp. Four Raillietina species, including R. echinobothrida, R. tetragona, R. cesticillus, and Raillietina sp., were recovered in domestic chickens from 4 districts in Phayao province, Thailand. LM and SEM observations revealed differences in the morphology of the scolex, position of the genital pore, number of eggs per egg capsule, and rostellar opening surface structures in all 4 species. Phylogenetic relationships were found among the phylogenetic trees obtained by the maximum likelihood and distance-based neighbor-joining methods. ITS2 and ND1 sequence data recorded from Raillietina sp. appeared to be monophyletic. The query sequences of R. echinobothrida, R. tetragona, R. cesticillus, and Raillietina sp. were separated according to the different morphological characters. This study confirmed that morphological studies combined with molecular analyses can differentiate related species within the genus Raillietina in Thailand.

Distribution of R Factors in Salmonella and Escherichia Coli Isolated from Korean Domestic Animals (우리나라 가축(家畜)에서 분리(分離)한 Salmonella 및 대장균(大腸菌)의 내성인자(耐性因子) R의 분포(分布))

  • Recent reports confirm that R factors is widespread in Korea among Enterobacteriaceae isolated from humans. However, no reports have been made concerning the incidence of transferable drug resistance in domestic animals in this country. A total of 211 isolates of Escherichia coli, including 94 strains from dogs, 76 strains from pigs, 30 strains from chickens, and 21 strains from cow milk, were examined for drug resistance and distribution of R factors. And, respective two strains of Salmonella E group and Salmonella cholerasuis which were isolated from dogs and pigs, respectively were also examined for the same purposes. Of 211 strains of E. coli isolated, 66.8% were found to be resistant to 8 antibacterial agents such as streptomycin(SM), tetracycline(TC), chloramphenicol(CP), ampicillin sodium(AP), nalidixic acid(NA), gentamicin(GM), and polymyxin B(PX). Among the isolates, 86.2% of the strains from dogs, 70% of the strains from chickens, 43.4% of the strains from pigs, and 28.6% of the strains from milk, respectively, were found to be resistant to the drugs. The following percentage of resistance of E. coli to each individual drugs was encountered: of 94 strains from dogs, AP, 64.9%; SM, 20.2%; NA, 12.8%, CP and PX, 8.5% each; GM, 2.1% each; GM, 2.1%. Among 76 strains from pigs, 42.2% and 2.6% each were resistant to TC, AP and PX, respectively. Among 30 strains from chickens, 43.3% were resistant to SM, TC, AP, respectively, and no strains were resistant to the other drugs. No strains of the isolated from milk were resistant to the drugs, except that 28.6% were resistant to SM and AP, respectively. Of the strains from dogs, multiply resistant strains(56.8%) were more than singly resistant one(43.2%) and sixteen different drug resistant patterns were observed. The most frequently encountered patterns were AP TC AP and SM CP AP NA. Of the isolates from other sources, the most frequently encountered resistant patterns were as follows: TC among the strains from pigs; SM TC AP from chickens; SM AP from milk. Of the resistant strains from dogs, 32% carried R factors and the most common resistance patterns of R factors were AP TC AP and SM TC CP, whereas 35.2% of the resistant strains from pigs carried R factors of which the most common encountered pattern was TC. Of the resistant strains from chickens, 46.9% carried R factors of which the most common patterns were SM TC TC AP and AP, whereas 50% of the resistant strains from milk carried R factors of which the most common pattern was SM. Of 4 strains of Salmonella isolated, no strains were resistant to the drugs, except that only one strain of Salmonella E group isolated from a dog was resistant to AP. The strain did not harbor R factor.

  • It has long been believed for the presence of infectious coryza affecting serious economic loss in domestic poultry industry. However, the etiologic agent has not been isolated until quite recently. From 1979, several strains of Haemophilus-like organism were isolated from chickens with symptoms similar to infectious coryza, and their colonial morphology, growth requirement, biochemical properties and pathogenicity were assessed. In addition, serological properties of the isolates by cross hemagglutination inhibition test was also investigated. The results indicated that all the isolates were identified as Haemophilus gallinarum which had similar characteristics to the reference strains.

Application of genomic big data to analyze the genetic diversity and population structure of Korean domestic chickens

  • Genetic diversity analysis is crucial for maintaining and managing genetic resources. Several studies have examined the genetic diversity of Korean domestic chicken (KDC) populations using microsatellite markers, but it is difficult to capture the characteristics of the whole genome in this manner. Hence, this study analyzed the genetic diversity of several KDC populations using high-density single nucleotide polymorphism (SNP) genotype data. We examined 935 birds from 21 KDC populations, including indigenous and adapted Korean native chicken (KNC), Hyunin and Jeju KDC, and Hanhyup commercial KDC populations. A total of 212,420 SNPs of 21 KDC populations were used for calculating genetic distances and fixation index, and for ADMIXTURE analysis. As a result of the analysis, the indigenous KNC groups were genetically closer and more fixed than the other groups. Furthermore, Hyunin and Jeju KDC were similar to the indigenous KNC. In comparison, adapted KNC and Hanhyup KDC populations derived from the same original species were genetically close to each other, but had different genetic structures from the others. In conclusion, this study suggests that continuous evaluation and management are required to prevent a loss of genetic diversity in each group. Basic genetic information is provided that can be used to improve breeds quickly by utilizing the various characteristics of native chickens.
