  • A new bacterial infection occurred among the cultured Korean catfish, Silurus asotus, in Chunbuk prefecture, Korea, 1993. This disease produced about 30% mortality in the fish for 4 months. The diseased fish was swimming listless at the water surface with head up and tail down, sometimes spinnig in circles. The most outstanding clinical sign was ulceration on the skull and at the base of the pectoral fins. The causative organism was isolated from the brain, kidney, spleen and liver of diseased fish, and identified as Edwardsiella icaluri by the biochemical and biophysical characteristics. After intraperitoneal innoculaton of the isolate, the pathogenicity was proved positive for Korean catfish, S. asotus, and channel catfish, Ictalurus punctatus, but negative for carp, Cyprinus carpio.

Ultrastructural Changes of Oocyte in Korean Catfish, Silurus asotus (발달 단계에 따른 한국산 메기(Silurus asotus) 난모세포의 미세구조적 변화)

  • This study was carried out to investigate the histomorphological changes and the electrophoretic patterns of egg components, obtained from 100 of 1-year-old female catfish(Silurus asotus). Especially, the light microscopic and ultrastructural changes of ooplasm and follicular membranes of oocytes, were observed by light and transmission electron microscope. All data were collected from October in 1992 to May in 1993. The size of the nucleoli and number of the yolk granules increased as the oocyte grew. Yolk granules were deposited in the oocyte as fluid. Due to the presence of large early and late maturing oocytes, their ovaries were large, transparent, granular, and greenish in color. As the percentages of fish in LMO and RO stage increased from March to April, mean of GSI values(19.95%) increased. Follicle cells such as granulosa cell and thecal cell change a squamous into cuboid shape in LPO and EMO stage. Processes, microvilli, from the granulosa cells and from the oocyte grow and make contact with each other in the pore canals of the zona radiata during vitellogenesis, but are withdrawn as the zona radiate becomes more compact and devoid of pore canals during oocyte maturation. The electrophoretic pattern of major band in mature stage was much thicker(21k, 24k, 32k, 45k, 67∼110k, 170k dalton) than that in previtellogenic phase.

  • Temperature-related cleavage rates and mitotic intervals (${\tau}_o$) in the far eastern catfish Silurus asotus were studied by averaging the duration of the second and third embryonic divisions to establish effective procedures for chromosome manipulations. At higher temperatures eggs developed faster and underwent more asynchronous development. Mitotic intervals for far eastern catfish were $21.5{\pm}1.4$ min at $22^{\circ}C$, $18.5{\pm}1.2$ min at $24^{\circ}C$, and $14.0{\pm}2.1$ min at $26^{\circ}C$. There was a strong negative correlation between ${\tau}_o$ and water temperature (Y=-1.85X+21.9, $R^2=0.9868$, where Y is ${\tau}_o$ and X is temperature).

  • Histological study of normal and albinic catfish Silurus asotus produced in aquaculture has been carried out on organs such as the eye, the barbel, the dorsal fin, and the skin (including dorsal, ventral and lateral regions). Although individuals have no differences in morphological appearance, their colors showed clear difference as follows: the normal catfishes were black overall, but the albinic ones have a yellowish-white color over the whole body, red eyes and white barbels. All the organs investigated were fundamentally very similar in structure. Regarding the distribution and density of the melanin pigments, however, the albinic catfish have no melanin in the skin and eye, particularly in the basement membrane of the epidermis and the pigment epithelial layer in the retina, while the normal catfish have lots of melanin. This phenomenon may coincide with the so-called $i^1/i^1$ type of oculocutaneous albinism.

Renal effect of experimental feeding of melamine and cyanuric acid in different concentrations on Japanese catfish (Silurus asotus) (멜라민과 cyanuric acid의 농도별 혼합투여에 따른 메기(Silurus asotus) 신장에서의 조직병리학적 소견)

  • The recent outbreak of renal failure in infants in China and in animals in USA and Europe has been determined to be caused by melamine adulterated in the food. In the course of the investigation, cyanuric acid was identified in addition to melamine in the offending food. Fish feeds were also recently found to be contaminated with melamine. The purpose of this study was to characterize the histopathological effect and toxicity potential of different concentrations of melamine and cyanuric acid in the kidney of Japanese catfish (Silurus asotus). The fish were administered melamine and cyanuric acid in combination at the concentrations of 12.5, 25, 50, 100 and 200 mg/kg/day for 3 days by single oral administration dissolved in carboxymethyl cellulose. The results showed that renal crystals were observed in renal tubules and collecting ducts at the concentration over 25 mg/kg dose group and the number of crystals in kidney were in proportion to the concentrations of melamine and cyanuric acid.

  • The Aeromonas strains were isolated from diseased cultured catfsh (Silurus asotus) in the ponds near by Gochang of Chunbuk. The present isolates were identified as A. hydrophila based on their biological and biochemica1 characteristics. The isolates of A. hydrophila were named GC-1, GC-2, GC-3, GC-4 and GC-5, Five strains were grew optimally at temperatures $35^{\circ}C,$ pH 7.5 and in 0.5 to l% NaCl. This bacterium(GC-1) was injected into health catfish in order to prove the causative agent of ascites and haemorrhagic ulcers. The symptoms in cat fish infected by this challenge method were observed to be very similar to the symptoms of a natural infection, but controls did not show any abnormal symptoms during the experimental period. At 24 post-injection, the red spot developed around the injection site and the haemorrhagic ulcers was extended near the gill and ventral fin. Five strains were tested for drug sensitivity by plate method. All strains were sensitivity to gentamicin and resistant to penicillin, streptomycin and tetracycline.

  • The far eastern catfish (Silurus asotus) growth hormone (GH) gene was cloned and characterized. The complete nucleotide sequences of genomic GH gene sequences as well as a catfish GH cDNA were obtained by RT-PCR and gene filter screening. The GH cDNA and genomic gene span 1.0 and 1.8 kb from the start codon to the polyadenylation signal, respectively. Both on cDNA and gDNA GH genes, the sequence polymorphism was detected including various silence mutations. The genomic GH gene comprised of only four exons and three introns, which was novel type of fish GH gene structure. The evolutionary relation of the catfish GH gene was inferred based on the comparative phylogenic analysis using the gene structures and sequences.

Effects of Starvation on Growth and Physiological Response in Cultured Catfish, Silurus asotus (기아에 따른 양식 메기, Silurus asotus의 성장 및 생리적 반응)

  • An experiment was conducted for 80 days to determine the effect of feeding and starvation on the survival, growth, gonad-somatic index (GSI), hepato-somatic index (HSI), viscera index (LSI), the morphometric changes and hematological parameter in a cultured catfish, Silurus asotus. The survival and growth of the starved group was lower than those of the fed group during the experiment. Starvation resulted in retardation of growth, which provides examples of the reduction of final body weight, growth rate of body weight, specific growth rate and condition factor, whereas the fed group continued to grow and remained in good condition. Blood analysis (cortisol, glucose, aspartate aminotransferase, alanine aminotransferase, red blood cell and hemoglobin) showed significant differences between the fed and starved groups at the end of the experiment. These results suggest that all nutritional parameters used for starvation and feeding in this study appear to be a useful index of nutritional status in the cultured catfish.

Immunosuppressive Properties of Catfish Bile from Silurus asotus: Inhibition of T Cell Activation in Mouse Splenocytes

  • Concentrated catfish Silurus asotus bile (SAB) containing high amounts of ursodeoxycholic acid (UDCA) and taurocholic acid may have immunosuppressive properties. To investigate the putative immunosuppressive properties of SAB, the anti-proliferation and suppression of early T cell activation markers, and the inhibition of cytokines induced by T cells in response to anti-CD3 mAb activation in mouse splenocytes were examined. The suppression of these activation repertoires are the main properties of calcineurin inhibitors. It was found that SAB effectively suppressed the activation of T cells, and cytokines from T cell activation, at levels similar to cyclosporine A, a calcineurin inhibitor. Although the mechanism in which suppression occurs is not clear, we speculate that SAB from Silurus asotus, which has been known to switch their intake habits to zoophagy during an early adult stage, may explain the suppressive effect of SAB as a result of high amounts of functional UDCA in bile. Our results suggest that the treatment or intake of SAB, either in therapy or as a food supplement, may act as an adjuvant therapy for the prevention of transplant rejection, although further investigation is required before this treatment can be applied clinically.

Effects of pH on Fertilization and the Hatching Rates of Far Eastern Catfish Silurus asotus

  • In this experiment, we examined the survival, fertilization, hatching times, and hatching rates of Far Eastern catfish Silurus asotus at pH ranging from 2 to 13 under laboratory conditions. Eggs could be fertilized at pH 3-12. In a hatching experiment, mortality was first observed at pH 13, when all fertilized eggs died within 8 min, followed by pH 2 (30 min), pH 12 (60 min), pH 3 (4 h), and pH 11 (5 h). Hatching only occurred at pH 4-10, with the highest hatching rate at pH 7 (52%) and the lowest at pH 10 (24%). Hatching rates in acid solutions were higher than in alkaline solutions, although the difference was not significant. Hatching was first observed at pH 10, beginning 27 h after fertilization and ending at the 31 h. A clear difference was observed between hatching times, ranging from 31 to 64 h and increasing in order with descending pH.
