Morphological and molecular evidence of the hybrid origin of Crepidiastrum ×muratagenii in Korea (홍도고들빼기의 형태 다양성 및 잡종 기원의 분자 증거)

  • The plant "Hong-do-go-deul-ppae-gi" has been considered as Crepidiastrum × muratagenii, a hybrid between C. denticulatum and C. lanceolatum, based on its morphological traits and geographical distribution. To reveal the hybrid origin of Hong-do-go-deul-ppae-gi, we examined additional morphological traits of this plant and its putative parents (C. denticulatum, C. lanceolatum, C. platyphyllum) and analyzed one nuclear ribosomal internal transcribed spacer (ITS) region and four chloroplast regions (trnT-L, trnL-F, rpl16 intron, and rps16 intron). As a result of examining the morphological traits, putative hybrid individuals were classified into three types based on the habit, cauline leaf, outer phyllary, and achene beak traits. A molecular analysis found that the ITS sequences of Type 1 and Type 2 individuals showed additive species-specific sites of C. denticulatum and C. lanceolatum. Plastid sequences of Type 1 and Type 2 individuals showed C. denticulatum and C. lanceolatum sequences, respectively. However, Type 3 individuals had ITS and plastid sequences corresponding to C. denticulatum. Accordingly, Type 1 and Type 2 individuals not only share morphological traits with C. denticulatum and C. lanceolatum but also show additive species-specific sites for C. denticulatum and C. lanceolatum, and not C. platyphyllum, supporting its origin as a hybrid between C. denticulatum and C. lanceolatum. Type 3 had morphological traits similar to other hybrid types but was distinguished with respect to outer phyllaries and demonstrated some resemblance to C. denticulatum. In a molecular analysis, Type 3 was found to be identical with regard to the sequence of C. denticulatum and was judged to be an ecological variation of C. denticulatum.

A Study on Recognition of Operating Condition for Hydraulic Driving Members (유압구동 부재의 작동조건 식별에 관한 연구)

  • The morphological analysis of wear debris can provide early a failure diagnosis in lubricated moving system. It can be effective to analyze operating conditions of oil-lubricated tribological system with shape characteristics of wear debris in a lubricant. But, in order to predict and recognize an operating condition of lubricated machine, it is needed to analyze and to identify shape characteristics of wear debris. Therefore, If the morphological characteristics of wear debris are recognized by computer image analysis using the neural network algorithm, it is possible to recognize operating condition of hydraulic driving members. In this study, wear debris in the lubricating oil are extracted by membrane filter (0.45 ${\mu}{\textrm}{m}$), and the quantitative values of shape parameters of wear debris are calculated by the digital image processing. This shape parameters are studied and identified by the artificial neural network algorithm. The result of study could be applied to prediction and to recognition of the operating condition of hydraulic driving members in lubricated machine systems.

  • Fractal dimension is the method to measure the roughness and the irregularity of something that cannot be defined obviously by Euclidean dimension. And the analysis method of this dimension don't need perfect, accurate boundary and color like analysis lot diameter, perimeter, aspect or reflectivity of wear particles or surface. If we arranged the morphological characteristic of various wear particle by using the characteristic of fractal dimension, it might be very efficient to the diagnosis of driving condition. In order to describe morphology of various wear particle, the wear test was carried out under friction experimental conditions. And fractal descriptors was applied to boundary and surface of wear particle with image processing system. These descriptors to analyze shape and surface wear particle are boundary fractal dimension and surface fractal dimension.

Three Dimensional Study of the Tidal Flat Geomorphology in the $K\breve{u}$m River Estuary by Multidate Image Analysis

  • The geomorophology of the tidal near $K\breve{u}$m river estuary on the west coast of Korea was studied using satellite images. There Landsat image of different tidal moment were geometrically corrected to be overlapped one another. Then topographic contours were extracted for the analysis with concept of 3-D. According to the morphological analysis, the tidal flats around $K\breve{u}$m river estuary area can be classified in three morphologically different groups: one is developped in a belt type along the shore; another is a delta type in the estuary; the other is developped between the shore line and islands. The morphological characteristics of the tidal flats in the study area seems to be mainly influenced by the tidal dynamics which are very storg on the weatern coastal zone of Korea. However the waves and human activities seem also to be important in some part of the area.

Three new records of mushroom species from Hangyeong-Andeok Gotjawal, South Korea

  • This study introduces one specimen each of the genera Gymnopilus, Marasmius, and Mycena as an unrecorded species found in the Hwasun Gotjawal and Jeju Gotjawal Provincial Park of the Hangyeong-Andeok terrain, South Korea. Morphological analyses of macroscopic and microscopic features, and molecular analysis of the nuclear ribosomal internal transcribed spacer region were conducted to identify the specimens. Our results indicated that the newly identified specimens were Gymnopilus dilepis, Marasmius tenuissimus, and Mycena seminau. Here, we describe these newly identified mushroom species and provide baseline data on their diversity in Gotjawal, South Korea.

Identification of Some Phellinus spp.

  • Four strains of Phellinus spp. was identified based on internal transcribed spacer(ITS) region of rDNA sequence analysis and morphological characteristics. Basidiocarps of all strains were effused-reflexed and hymenial surface was poroid. Hyphal system was dimitic and basidiospore was globose to ellipsoid. The amplification of ITS regions produced a DNA fragment of 500 to 780 by in all strains examined. The determined sequences were analyzed for the reconstruction of phylogenetic tree. From these results, Phellinus sp. KM-1, KM-2, and KM-4 was identified as P. hartigii, P. baumii, and P. linteus, respectively.

  • In the course of screening for microbial metabolites employing human cancer cell line, we identified a mycelial extract of Streptomyces sp. 1365, which are effective on growth inhibition and morphological change of MCF-7, human breasr cancer cell line. By repeased column chromatography and recrystallization process, yellow needle crystals were obtained as an active compound and identified as resistomycin by spectral analysis.

Morphological Analysis of Wear Particles using Fractal Parameters

  • The fractal dimension is the characteristics that can quantitatively define the irregularity in natural. It is useful in describing the morphology or various rubbed surface for hydraulic piston motor instead of the stylus profiling method. But fractal parameters had not constructed on the morphological characteristic or rubbed surface because of the insufficient knowledge about a conception of fractal dimension. In this study, for the purpose or applying fractal I parameters practically, we have suggested way to establish the morphological characteristic of rubbed surface with fractal parameters, and we carried out an experiment on the lubricant friction and wear by using Ball-ON-Disk type tester. Materials were the brass and the bronze which are used to slipper-pad in the hydraulic piston motor. We searched for fractal parameters or surface structure with the digital image processing, Surface fractal dimension can be determined by sum of intensity difference of surface pixel. Using the image processing and fractal parameters for rubbed surface in the friction and wear test, morphology of rubbed sur race can be effectively obtained by fractal dimensions.

Morphological and Genetic Variation of Two Populations of Platichthys stellatus (Pleuronectidae, PISCES) from the East Sea (동해 강도다리(Platichthys stellatus) 2개체군의 형태 및 분자변이)

  • Morphological and genetic variation of two populations of Platichthys stellatus were investigated based on 30 individuals each, collected from Uljin (seedling release area) and Pohang (control) in Korea. Morphological analyses demonstrated that the two populations of P. stellatus were well distinguishable in body color of the blind side and fin shape. Mitochondrial DNA control region analysis indicated no significant differences between the two populations ($F_{ST}=-0.00849$, P>0.05). We also analyzed microsatellite DNA loci of the two populations using six markers. Observed heterozygosity ($H_O$) and expected heterozygosity ($H_E$) were 0.550 and 0.592, respectively, in P. stellatus from Uljin, but 0.700 and 0.737 in P. stellatus from Pohang. An index of differentiation in genetic structure revealed significant differences between the two populations ($F_{ST}=0.0208$, P<0.05). Our results suggest that the Uljin population may be comprised of released P. stellatus, whereas the Pohang population may be wild P. stellatus, highlighting the necessity of continuous monitoring of the two populations.
