Detection for demineralization of dental hard tissues using index-sequenced radiographs (방사선사진의 인덱스-순서 분석을 통한 치아경조직의 탈염화 검출)

  • Purpose : The purpose of this study was to develop the radiographic technique for detecting the demineralization which is known as indication of dental caries. Materials and Methods: This technique was based on the comparing of multiple radiographs which was irradiated by multiple X-ray spectra. For the meaningful comparing, the multiple radiographs were reconstructed to the dosimetrically consistent images using a standard material. The difference of resulting images of same target with multiple spectra represents the difference of response of material as regards the spectra. Results : We have found about 10% of demineralization of dental hard tissues particularly in the proximal region through the analyzing of differences. Conclusion: Most intriguing thing in this investigation was that the method to analyze difference shows us to an anatomic structure of dental hard tissues even if absolute values of optical density were excluded during the procedures.   • Health industry, a knowledge based high value-added industry, is being considered as a strategic area for the 21C and many advanced countries are making every endeavors for the promotion of health industry along with information technology, new materials, and mechatronics. Korean health industry, however, has been excluded from the governmental supports as well as bound by strict regulation so far, and there is a significant gap in technology compared with advanced countries. In 21C, technology is the main factor of national competitiveness and that is why the role of R&D institutes are so important in the high level of competition to cope with the technology protection policies of advanced countries. In this article, with Directory of Korean R&D Institutes published by Korea Industry Technology Association, I reviewed the trends of R&D institute of health industry. Main findings of the research can be summarized as follows. The portion of health industry R&D institute is 3.6% of total R&D institute but the amount of R&D investment is over than 5% This means health industry are knowledge based and R&D intensified industry, meanwhile the variations of same industry R&D institutes of health industry is huge in R&D investments and other activities. Regional distributions of health industry institutes show some kind of different patterns in each industry areas. Medical devices and Medical informatics's preference of metropolitan region are distinguished with other industry areas. Many of the institutes are located in same site of it's company rather than operating separate building for R&D specific uses. It is better for transforming ideas to products and close cooperation of research body with product lines, but it is a handicap for networking and communicating with other research institutions too. It takes 18.4yrs for bearing R&D institute on the average. For a long times 'copy products' or 'me too products' policies were easy way to maintain business entities. But recently, it is recognized that research activities are essential component of sustaining it's own business firms. This means technology itself is leading power of corporation itself in the high level of competition.

Study on the Coke Oven Emissions in Cokes Using and Manufacturing Workplaces (코크스 제조 및 사용 공정에서의 코크스오븐 배출물질 연구)

  • This study was performed to evaluate the coke oven emissions (COE) and polynuclear aromatic hydrocarbon levels in coke manu-facturing industry, secondary lead smelting industry and glass bottle manufacturing industry. 1. There were no significant difference between the means of personal samples and area samples by the types of industry(p>0.05). The levels of airborne total particulates of the secondary lead smelting industry was the highest($2.30mg/m^3$), and those of the coke manu-facturing industry and glass bottle manu facturing industry were $1.95mg/m^3$ and $1.37mg/m^3$. The concentration of COE was the highest in the glass bottle manufacturing industry($0.79mg/m^3$), and in order of $0.19mg/m^3$ in the coke manufacturing industry and $0.06mg/m^3$ in the secondary lead smelting industry. COE/total particulates(%) was highest in the glass bottle manufacturing industry(58.1%) and in order of 10.3% in the coke manufacturing industry and 3.1% in secondary lead smelting industry. There were significant differences in the total particle concentration and COE by the types of industry(p<0.05). 2. The levels of airborne total particulates was the highest at the smelting process of secondary lead smelting industry($2.30{\pm}0.72mg/m^3$), and the lowest at the smelting process of glass bottle manufacturing industry ($0.99{\pm}1.22mg/m^3$) Concentration of COE was the highest at the casting process of glass bottle manufacturing industry ($1.09{\pm}1.15mg/m^3$), the lowest at the smelting process of secondary lead smelting industry ($0.06{\pm}0.03mg/m^3$). The COE/total particulates(%) was the highest at the casting process of glass bottle manufacturing industry($65.9{\pm}20.5%$), and the lowest at the smelting process of secondary lead smelting indusry($3.1{\pm}2.7%$). 3. There were positive correlations between level of The airborne total particulates and concentration of COE in coke manufacturing industry and glass bottle manufacturing industry (p<0.05), but negative correlation in secondary lead smelting industry. 4. The numbers of case and rates that over the Threshold Limit Values(TLVs) were 24 (77.4%)cases in glass bottle manufacture, 14(23.7%) cases in the coke manufacturing industry and no one case in secondary lead smelting industry. Total numbers of case and rates that over TLVs were 38( 35.5%) cases. 5. The limit of detection(LOD) for PAH was $10{\mu}g/ml$ in standard sample. All PAH levels of the cokes manufacturing industry and the secondary lead smelting industry and the glass bottle manufacturing industry were trace or not to detect.

Anti-oxidation and Anti-aging Effect of Mixed Extract from Korean Medicinal Herbs (한약재 복합 추출물이 항산화 및 항노화에 미치는 영향)

  • Objectives : The present study was designed to investigate effects of mixed extract from korean medicinal herbs (MIX) on oxidation/reduction reaction-related and aging-related enzyme in vitro. Methods : We performed MTT assay, collagenase inhibition assay, elastase inhibition assay, tyrosinase inhibition assay, DPPH free radical scavenging assay, SOD-like activity and xanthine oxidase inhibition assay. Results : Recently, many studies have reported that elastin is also involved in inhibiting or repairing wrinkle formation, although collagen is a major factor in the skin wrinkle formation. The MIX showed 97% inhibition of collagenase activity, and 64% inhibition of elastase activity at 1 mg/ml concentration of MIX, next only to positive control, which indicate good efficacy for anti-wrinkle ingredient. Also it's treatment showed 34% inhibition of tyrosinase activity, to relate whitening effect, at the same dose of MIX. Antioxidant activities were determined by DPPH radical scavenging, xanthine oxidase (XO) inhibiting activity and SOD-like activity. Also these scavenging, XO-inhibiting and SOD-like activities were measured in 91%, 80%, and 63% inhibition, respectively, at a treated dose of 1 mg/ml, compare to control. Conclusions : These results suggest that possibility of mixed korean medicinal herbs as a functional ingredient for anti-wrinkle and whitening, anti-oxidation and anti-aging cosmetic formula.

Anti-diabetic Effect of Opuntia humifusa Stem Extract (손바닥선인장(Opuntia humifusa) 줄기 추출물의 항당뇨 효과)

  • Opuntia humifusa known as the Eastern prickly pear have been used as a treatment of burns, diarrhea, asthma, rheumatism, gonorrhea, and diabetes in alternative medicine. O. humifusa is widely cultivated in the middle and southern provinces of Korea and distributed in North America. The aim of this study is to investigate anti-diabetic effect of O. humifusa stem (OHS) water or 80% MeOH extract using 3T3-L1 adipocytes and db/db mice animal models. OHS 80% MeOH extract at a dose of $250{\mu}g/ml$ significantly increased the glucose uptake and lipid accumulation compared with the control in 3T3-L1 adipocytes. Blood glucose, plasma total cholesterol and triglyceride levels were significantly reduced by oral treatment of OHS 80% MeOH extract (200 mg/kg BW) for 6 weeks in db/db mice. Also, the oral treatment of OHS 80% MeOH extract slightly changed the plasma insulin and insulin resistance levels in db/db mice, but were no significance in comparison to control. Glucose transporter(GLUT)4 expressions of adipose tissue and muscle were significantly increased more than that in the control. Therefore, these results suggest that OHS 80% MeOH extract inhibits the blood glucose level through regulation of lipid profile, insulin resistance, and GLUT4 expression in db/db mice and its diabetic effect is effective more than water extract.

Proximal caries detection using digital subtraction radiography in the artificial caries activity model (인공 치아우식 발생 모델에서 디지털 방사선 공제술을 이용한 인접면 치아우식증의 진단)

  • Purpose: The purpose of the experiment was to evaluating the diagnostic ability of dental caries detection using digital subtraction in the artificial caries activity model. Materials and Methods: Digital radiographies of five teeth with 8 proximal surfaces were obtained by CCD sensor (Kodak RVG 6100 using a size #2). The digital radiographic images and subtraction images from artificial proximal caries were examined and interpreted. In this study, we proposed novel caries detection method which could diagnose the dental proximal caries from single digital radiographic image. Results: In artificial caries activity model, the range of lesional depth was $572-1,374{\mu}m$ and the range of lesional area was $36.95-138.52mm^2$. The lesional depth and the area were significantly increased with demineralization time (p<0.001). Furthermore, the proximal caries detection using digital subtraction radiography showed high detection rate compared to the proximal caries examination using simple digital radiograph. Conclusion: The results demonstrated that the digital subtraction radiography from single radiographic image of artificial caries was highly efficient in the detection of dental caries compared to the data from simple digital radiograph.

Imperatorin Suppresses Degranulation and Eicosanoid Generation in Activated Bone Marrow-Derived Mast Cells

  • Imperatorin has been known to exert many biological functions including anti-inflammatory activity. In this study, we investigated the inhibitory effects of imperatorin on the production of inflammatory mediators in mouse bone marrow-derived mast cells (BMMC). Imperatorin inhibited degranulation and the generation of eicosanoids (leukotriene $C_4$ ($LTC_4$) and prostaglandin $D_2$ ($PGD_2$) in IgE/antigen (Ag)-stimulated BMMC. To elucidate the molecular mechanism involved in this process, we investigated the effect of imperatorin on intracellular signaling in BMMC. Biochemical analyses of the IgE/Ag-mediated signaling pathway demonstrated that imperatorin dramatically attenuated degranulation and the production of 5-lipoxygenase-dependent $LTC_4$ and cyclooxygenase-2-dependent $PGD_2$ through the inhibition of intracellular calcium influx/phospholipase $C{\gamma}1$, cytosolic phospholipase $A_2$/mitogen-activated protein kinases and/or nuclear factor-${\kappa}B$ pathways in BMMC. These results suggest that the effects of imperatorin on inhibition of degranulation and eicosanoid generation through the suppression of multiple steps of IgE/Ag-mediated signaling pathways would be beneficial for the prevention of allergic inflammation.

A Study on the Depigmenting Effect of Carthamus tinctorius Seed, Cyperus rotundus and Schizonepeta tenuifolia Extracts (홍화자, 향부자, 형개 추출물의 미백효과에 관한 연구)

  • The objective of the present study was to evaluate the skin depigmentation effect of the extracts of three herbs, Carthamus tinctorius seed, Cyperus rotundus and Schizonepeta tenuifolia. Their effects on tyrosinase and melanin synthesis inhibitory action were assessed. We found that the C. tinctorius seed ethanol extracts reduced the tyrosinase activity and melanin formation of B16F10 melanoma cells. The C. tinctorius seed suppressed the expression in microphthalmia associated transcription factor (MITF), tyrosinase, tyrosinase related protein 1 (TRP-1), and tyrosinase related protein 2 (TRP-2) in B16F10 melanoma cells. These results show that C. tinctorius seed inhibited melanogenesis on the B16F10 melanoma cell. The underlying mechanism of C. tinctorius seed whitening activity may be the inhibition of tyrisinase, MITF, tyrosinase, TRP-1, and TRP-2 expression. The results suggested that C. tinctorius seed has considerable potential as a natural functional ingredient with a depigmentation effect.
