SUGGESTIONS FOR IMPROVMENTS OF THE RADIATION PROTECTION FOR THE EMERGENCY WORKERS DURING THE FUKUSHIMA NUCLEAR POWER PLANT ACCIDENT

  • Following the emergency work in Fukushima Daiichi nuclear power plant, more attention was paid for the radiation protection of workers working under severe accident condition. The protection procedure for the emergency workers, including the on-site emergency center, the seismic isolated building and the reestablishment of the radiation protection framework were analyzed to investigate drawbacks and deficiencies which led to adverse effects on the emergency planning and on emergency workers' health and comfort. Those drawbacks were identified and studied, and then suggestions were made to enhance the emergency working condition to avoid any future problems during severe accident emergency work and management.

미국 화훼류 품종보호제도 및 식물특허 운영실태 (Introduction of Plant Variety Protection Right and Plant Patent System of Ornamental Crops in U.S.A)

  • 국내 화훼류 육성품종의 미국 출원에 대한 안내 및 미국 육성 품종의 국내 출원 시 품종보호심사에 활용하는 자료로 제공하고자 미국의 식물신품종보호제도와 식물특허에 대한 운영실태를 조사하였다. 미국은 신품종 대상에 따라 종자번식 품종과 괴경식물은 식물신품종보호법으로, 무성번식품종은 식물특허로 구별되어 보호를 받게 된다. 품종보호나 특허를 받기위한 조건은 신규성, 구별성, 균일성, 안정성이며, 과수나 묘목의 품종보호 보호기간은 25년이고 나머지 대상의 품종보호와 식물특허의 보호기간은 20년이다. 식물신품종보호와 식물특허의 심사 모두 재배심사나 현지심사를 하지 않고 출원인이 제출한 서류만으로 심사를 한다. 화훼류 출원은 영양체번식 품종의 식물특허 출원이 대부분이고 종자번식 품종의 품종보호 비율은 낮으며, 전체 식물특허 출원 중 화훼류가 87%를 차지하며, 자국과 외국간 출원 품종수에는 큰 차이가 없다.

발전기 보호용 계자상실 계전기의 적용 분석 (Application Analysis for Loss of Excitation Relay of Generator Protection)

  • Recently, the LOE(loss of excitation) incidents are occurred in domestic synchronous generator frequently, the synchronous generator protection system has been much attention for the LOE protection of the incidents that threats synchronous generators and power systems. This paper was showed the characteristics and practices of distance relay that widely used LOE protection relaying in generator. Firstly, the operating characteristics and the impedance locus for LOE of the generator protection were introduced. Even if the conventional simulation program is used for modeling, but it is difficult to implement a LOE modeling and simulation of synchronous generator. So, the LOE relay operation data collected from thermal power plant and nuclear power plant in real fields were analyzed. By reviewing the applications of GE Mho relays, the reliability of LOE for synchronous generator protection in domestic were improved.

Partial Biological and Molecular Characterization of Tomato yellow fruit ring virus Isolates from Potato

  • Eight potato-producing provinces of Iran were surveyed during the growing seasons of 2004-2006 to detect the presence of Tomato yellow fruit ring virus (TYFRV), a tentative species in the genus Tospovirus. A total of 1,957 potato leaf samples were collected from plants with tospovirus-like symptoms of chlorotic or necrotic spots, chlorosis and necrosis. The samples were tested by enzyme-linked immunosorbent assay using TYFRV-specific antibodies. Among those tested, 498 samples (25.4%) were found to be infected with the virus. The virus was detected in 72.4% of the potato fields in all provinces surveyed. Thirteen potato isolates of TYFRV were selected for further biological and molecular studies. Based on their reactions on Nicotiana tabacum plants, the isolates were separated into two groups, namely L (local infection) and N (systemic infection). The nucleotide sequences of the nucleoprotein (N) genes of the isolates were determined and compared with the homologous sequences in Genbank. No recombination evidence was found in the isolates using different recombination-detecting programs. In the phylogenetic tree, the potato isolates fell into two major groups: IRN-1 and IRN-2 corresponding to the two biologically separated groups. This study shows for the first time the biological and phylogenetic relationships of geographically distant TYFRV isolates from potatoes in the mid-Eurasian country of Iran.

The Endo-β-1,4-Glucanase of Bacillus amyloliquefaciens Is Required for Optimum Endophytic Colonization of Plants

  • The eglS gene in Bacillus amyloliquefaciens encodes an endo-β-1,4-glucanase that belongs to glycosyl hydrolase family 5. In this study, a disruption mutant of gene eglS was constructed to examine its role in bacterial adaptation in plants. The mutant TB2k, eglS gene inactivated bacterial strain, was remarkably impaired in extracellular cellulase activity. When inoculated on Brassica campestris, the TB2k population was reduced by more than 60% compared with the wild-type strain in the root, stem, and leaf tissues. Overexpression of eglS in the wild-type strain increased the bacteria population in the plant tissues. Further studies revealed that the transcription level of eglS was correlated with bacterial population. These data demonstrate that endo-β-1,4-glucanase of B. amyloliquefaciens is required for its optimal endophytic colonization.

The Mitogen-Activated Protein Kinase Signal Transduction Pathways in Alternaria Species

  • Mitogen-activated protein kinase (MAPK) cascades are conserved signaling modules in the eukaryotic cells. They are involved in many major cell processes in fungi such as stress responses, vegetative growth, pathogenicity, secondary metabolism and cell wall integrity. In this review, we summarized the advances of research on the MAPK signaling pathways in Alternaria species. As major phytopathogenic fungi, Alternaria species reduce crop production. In contrast to the five MAPK pathways known in yeast, only three MAPK pathways as Fus3/Kss1-type, Hog1-type, and Slt2-type have been characterized in Alternaria. The Fus3/Kss1-type MAPK pathway participates in regulation of vegetative growth, conidiation, production of some cell-wall-degrading enzymes and pathogenicity. The Hog1-type pathway is involved in osmotic and oxidative stress, fungicides susceptibility and pathogenicity. The Slt2-type MAP kinases play an important role on maintaining cell wall integrity, pathogenicity and conidiation. Although recent advances on the MAPK pathways in Alternaria spp. reveal many important features on the pathogenicity, there are many unsolved problems regarding to the unknown MAP kinase cascade components and network among other major signal transduction. Considering the economic loss induced by Alternaria spp., more researches on the MAPK pathways will need to control the Alternaria diseases.

Tomato Yellow Leaf Curl China Virus Impairs Photosynthesis in the Infected Nicotiana benthamiana with βC1 as an Aggravating Factor

  • Tomato yellow leaf curl China virus is a species of the widespread geminiviruses. The infection of Nicotiana benthamiana by Tomato yellow leaf curl China virus (TYLCCNV) causes a reduction in photosynthetic activity, which is part of the viral symptoms. ${\beta}C1$ is a viral factor encoded by the betasatellite DNA ($DNA{\beta}$) accompanying TYLCCNV. It is a major viral pathogenicity factor of TYLCCNV. To elucidate the effect of ${\beta}C1$ on plants' photosynthesis, we measured the relative chlorophyll (Chl) content and Chl fluorescence in TY-LCCNV-infected and ${\beta}C1$ transgenic N. benthamiana plants. The results showed that Chl content is reduced in TYLCCNV A-infected, TYLCCNV A plus $DNA{\beta}$ (TYLCCNV A + ${\beta}$)-infected and ${\beta}C1$ transgenic plants. Further, changes in Chl fluorescence parameters, such as electron transport rate, $F_v/F_m$, NPQ, and qP, revealed that photosynthetic efficiency is compromised in the aforementioned N. benthamiana plants. The presense of ${\beta}C1$ aggravated the decrease of Chl content and photosynthetic efficiency during viral infection. Additionally, the real-time quantitative PCR analysis of oxygen evolving complex genes in photosystem II, such as PsbO, PsbP, PsbQ, and PsbR, showed a significant reduction of the relative expression of these genes at the late stage of TYLCCNV A + ${\beta}$ infection and at the vegetative stage of ${\beta}C1$ transgenic N. benthamiana plants. In summary, this study revealed the pathogenicity of TYLCCNV in photosynthesis and disclosed the effect of ${\beta}C1$ in exacerbating the damage in photosynthesis efficiency by TYLCCNV infection.

Biological Control of Soilborne Diseases on Tomato, Potato and Black Pepper by Selected PGPR in the Greenhouse and Field in Vietnam

  • Bacterial wilt, Fusarium wilt and Foot rot caused by Ralstonia solanacearum, Fusarium oxysporum, and Phytophthora capsici respectively, continue to be severe problems to tomato, potato and black pepper growers in Vietnam. Three bio-products, Bacillus vallismortis EXTN-1 (EXTN-1), Bacillus sp. and Paenibacillus sp. (ESSC) and Bacillus substilis (MFMF) were examined in greenhouse bioassay for the ability to reduce bacterial wilt, fusarium wilt and foot rot disease severity. While these bio-products significantly reduced disease severities, EXTN-1 was the most effective, providing a mean level of disease reduction 80.0 to 90.0% against bacterial wilt, fusarium wilt and foot rot diseases under greenhouse conditions. ESSC and MFMF also significantly reduced fusarium wilt, bacterial wilt and foot rot severity under greenhouse conditions. Bio-product, EXTN-1 with the greatest efficacy under greenhouse condition was tested for the ability to reduce bacterial wilt, fusarium wilt and foot rot under field condition at Song Phuong and Thuong Tin locations in Ha Tay province, Vietnam. Under field condition, EXTN-1 provided a mean level of disease reduction more than 45.0% against all three diseases compared to water treated control. Besides, EXTN-1 treatment increased the yield in tomato fruits 17.3% than water treated control plants.

Nigrospora Species Associated with Various Hosts from Shandong Peninsula, China

  • Nigrospora is a monophyletic genus belonging to Apiosporaceae. Species in this genus are phytopathogenic, endophytic, and saprobic on different hosts. In this study, leaf specimens with disease symptoms were collected from host plants from the Shandong Peninsula, China. The fungal taxa associated with these leaf spots were studied using morphology and phylogeny based on ITS, TEF1, and TUB2 gene regions. In this article, we report on the genus Nigrospora with N. gorlenkoana, N. oryzae, N. osmanthi, N. rubi, and N. sphaerica identified with 13 novel host associations including crops with economic importance such as bamboo and Chinese rose.

Biological and Molecular Characterization of a Korean Isolate of Orthotospovirus chrysanthinecrocaulis (Formerly Chrysanthemum Stem Necrosis Virus) Isolated from Chrysanthemum morifolium

  • Biological and molecular characterization of a Korean isolate of Orthotospovirus chrysanthinecrocaulis (formerly known as chrysanthemum stem necrosis virus, CSNV) isolated from Chrysanthemum morifolium was determined using host range and sequence analysis in this study. Twenty-three species of indicator plants inoculated mechanically CSNV-Kr was investigated for determination of host range. CSNV-Kr induced various local and systemic symptoms in the inoculated plant species. CSNV-Kr could not infect three plant species and induced symptomless in systemic leaves in Nicotiana tabacum cultivars, though the plant samples reacted positively with the antiserum to CSNV by double-antibody sandwich-enzyme-linked immunosorbent assay. The complete genome sequence of CSNV-Kr was determined. The L RNA of CSNV-Kr consists of 8,959 nucleotides (nt) and encodes a putative RNA-dependent RNA polymerase. The M RNA of CSNV-Kr consists of 4,835 nt and encodes the movement protein (NSm) and the glycoprotein precursor (Gn/Gc protein). The S RNA of CNSV-Kr consists of 2,836 nt and encodes NSs protein and N protein. The Gn/Gc and N sequence of CSNV-Kr were compared with those of previously published CSNV isolates originating from different countries at nucleotide and amino acid levels. The Gn/GC sequence of CSNV-Kr shared 98.8-99.5% identity with CSNV isolated from other countries and the N sequence of CSNV-Kr shared 98.8-99.6% identity. No particular region of variability could be found in either grouping of viruses. All of the CSNV isolates did not show any relationship according to geographical origins and isolation hosts, suggesting no distinct segregation of the CSNV isolates.
