Effect of Hfe Deficiency on Memory Capacity and Motor Coordination after Manganese Exposure by Drinking Water in Mice

  • Excess manganese (Mn) is neurotoxic. Increased manganese stores in the brain are associated with a number of behavioral problems, including motor dysfunction, memory loss and psychiatric disorders. We previously showed that the transport and neurotoxicity of manganese after intranasal instillation of the metal are altered in Hfe-deficient mice, a mouse model of the iron overload disorder hereditary hemochromatosis (HH). However, it is not fully understood whether loss of Hfe function modifies Mn neurotoxicity after ingestion. To investigate the role of Hfe in oral Mn toxicity, we exposed Hfe-knockout ($Hfe^{-/-}$) and their control wild-type ($Hfe^{+/+}$) mice to $MnCl_2$ in drinking water (5 mg/mL) for 5 weeks. Motor coordination and spatial memory capacity were determined by the rotarod test and the Barnes maze test, respectively. Brain and liver metal levels were analyzed by inductively coupled plasma mass spectrometry. Compared with the water-drinking group, mice drinking Mn significantly increased Mn concentrations in the liver and brain of both genotypes. Mn exposure decreased iron levels in the liver, but not in the brain. Neither Mn nor Hfe deficiency altered tissue concentrations of copper or zinc. The rotarod test showed that Mn exposure decreased motor skills in $Hfe^{+/+}$ mice, but not in $Hfe^{-/-}$ mice (p = 0.023). In the Barns maze test, latency to find the target hole was not altered in Mn-exposed $Hfe^{+/+}$ compared with water-drinking $Hfe^{+/+}$ mice. However, Mn-exposed $Hfe^{-/-}$ mice spent more time to find the target hole than Mn-drinking $Hfe^{+/+}$ mice (p = 0.028). These data indicate that loss of Hfe function impairs spatial memory upon Mn exposure in drinking water. Our results suggest that individuals with hemochromatosis could be more vulnerable to memory deficits induced by Mn ingestion from our environment. The pathophysiological role of HFE in manganese neurotoxicity should be carefully examined in patients with HFE-associated hemochromatosis and other iron overload disorders.

A Novel Transmission Scheme for Compressed Health Data Using ISO/IEEE11073-20601

  • In view of personal health and disease management based on cost effective healthcare services, there is a growing need for real-time monitoring services. The electrocardiogram (ECG) signal is one of the most important of health information and real-time monitoring of the ECG can provide an efficient way to cope with emergency situations, as well as assist in everyday health care. In this system, it is essential to continuously collect and transmit large amount of ECG data within a given time and provide maximum user convenience at the same time. When considering limited wireless capacity and unstable channel conditions, appropriate signal processing and transmission techniques such as compression are required. However, ISO/IEEE 11073 standards for interoperability between personal health devices cannot properly support compressed data transmission. Therefore, in the present study, the problems for handling compressed data are specified and new extended agent and manager are proposed to address the problems while maintaining compatibility with existing devices. Extended devices have two PM-stores enabling compression and a novel transmission scheme. A variety of compression techniques can be applied; in this paper, discrete cosine transformation (DCT) is used. And the priority of information after DCT compression enables new transmission techniques for performance improvement. The performance of the compressed signal and the original uncompressed signal transmitted over the noisy channel are compared in terms of percent root mean square difference (PRD) using our simulation results. Our transmission scheme shows a better performance and complies with 11073 standards.

A Novel Redundant Data Storage Algorithm Based on Minimum Spanning Tree and Quasi-randomized Matrix

  • For intermittently connected wireless sensor networks deployed in hash environments, sensor nodes may fail due to internal or external reasons at any time. In the process of data collection and recovery, we need to speed up as much as possible so that all the sensory data can be restored by accessing as few survivors as possible. In this paper a novel redundant data storage algorithm based on minimum spanning tree and quasi-randomized matrix-QRNCDS is proposed. QRNCDS disseminates k source data packets to n sensor nodes in the network (n>k) according to the minimum spanning tree traversal mechanism. Every node stores only one encoded data packet in its storage which is the XOR result of the received source data packets in accordance with the quasi-randomized matrix theory. The algorithm adopts the minimum spanning tree traversal rule to reduce the complexity of the traversal message of the source packets. In order to solve the problem that some source packets cannot be restored if the random matrix is not full column rank, the semi-randomized network coding method is used in QRNCDS. Each source node only needs to store its own source data packet, and the storage nodes choose to receive or not. In the decoding phase, Gaussian Elimination and Belief Propagation are combined to improve the probability and efficiency of data decoding. As a result, part of the source data can be recovered in the case of semi-random matrix without full column rank. The simulation results show that QRNCDS has lower energy consumption, higher data collection efficiency, higher decoding efficiency, smaller data storage redundancy and larger network fault tolerance.

Real-time Imaging of Inositol 1,4,5-trisphosphate Movement in Mouse Salivary Gland Cells

  • Inositol 1,4,5-trisphosphate ($IP_3$) plays an important role in the release of $Ca^{2+}$ from intracellular stores into the cytoplasm in a variety of cell types. $IP_3$ translocation dynamics have been studied in response to many types of cell signals. However, the dynamics of cytosolic $IP_3$ in salivary acinar cells are unclear. A green fluorescent protein (GFP)-tagged pleckstrin homology domain (PHD) was constructed and introduced into a phospholipase C ${\delta}1$ (PLC ${\delta}1$) transgenic mouse, and then the salivary acinar cells were isolated. GFP-PHD was heterogeneously localized at the plasma membrane and intracellular organelles in submandibular gland and parotid gland cells. Application of trypsin, a G protein-coupled receptor activator, to the two types of cells caused an increase in GFP fluorescence in the cell cytoplasm. The observed time course of trypsin-evoked $IP_3$ movement in acinar cells was independent of cell polarity, and the fluorescent label showed an immediate increase throughout the cells. These results suggest that GFP-PHD in many tissues of transgenic mice, including non-cultured primary cells, can be used as a model for examination of $IP_3$ intracellular dynamics.

Encryption Scheme for MPEG-4 Media Transmission Exploiting Frame Dropping

  • Depending on network conditions, a communication network could be overloaded when media are transmitted. Research has been carried out to lessen network overloading, such as by filtering, load distribution, frame dropping, and other methods. Among these methods, one of the most effective is frame dropping, which reduces specified video frames for bandwidth diminution. In frame dropping, B-frames are dropped and then I- and P-frames are dropped, based on the dependency among the frames. This paper proposes a scheme for protecting copyrights by encryption, when frame dropping is applied to reduce the bandwidth of media based on the MPEG-4 file format. We designed two kinds of frame dropping: the first stores and then sends the dropped files and the other drops frames in real time when transmitting. We designed three kinds of encryption methods using the DES algorithm to encrypt MPEG-4 data: macro block encryption in I-VOP, macro block and motion vector encryption in P-VOP, and macro block and motion vector encryption in I-, P-VOP. Based on these three methods, we implemented a digital rights management solution for MPEG-4 data streaming. We compared the results of dropping, encryption, decryption, and the quality of the video sequences to select an optimal method, and found that there was no noticeable difference between the video sequences recovered after frame dropping and the ones recovered without frame dropping. The best performance in the encryption and decryption of frames was obtained when we applied the macro block and motion vector encryption in I-, P-VOP.

An Optimal Driving Support Strategy(ODSS) for Autonomous Vehicles based on an Genetic Algorithm

  • A current autonomous vehicle determines its driving strategy by considering only external factors (Pedestrians, road conditions, etc.) without considering the interior condition of the vehicle. To solve the problem, this paper proposes "An Optimal Driving Support Strategy(ODSS) based on an Genetic Algorithm for Autonomous Vehicles" which determines the optimal strategy of an autonomous vehicle by analyzing not only the external factors, but also the internal factors of the vehicle(consumable conditions, RPM levels etc.). The proposed ODSS consists of 4 modules. The first module is a Data Communication Module (DCM) which converts CAN, FlexRay, and HSCAN messages of vehicles into WAVE messages and sends the converted messages to the Cloud and receives the analyzed result from the Cloud using V2X. The second module is a Data Management Module (DMM) that classifies the converted WAVE messages and stores the classified messages in a road state table, a sensor message table, and a vehicle state table. The third module is a Data Analysis Module (DAM) which learns a genetic algorithm using sensor data from vehicles stored in the cloud and determines the optimal driving strategy of an autonomous vehicle. The fourth module is a Data Visualization Module (DVM) which displays the optimal driving strategy and the current driving conditions on a vehicle monitor. This paper compared the DCM with existing vehicle gateways and the DAM with the MLP and RF neural network models to validate the ODSS. In the experiment, the DCM improved a loss rate approximately by 5%, compared with existing vehicle gateways. In addition, because the DAM improved computation time by 40% and 20% separately, compared with the MLP and RF, it determined RPM, speed, steering angle and lane changes faster than them.

Effects of Histamine on Cultured Interstitial Cells of Cajal in Murine Small Intestine

  • Interstitial cells of Cajal (ICCs) are the pacemaker cells in the gastrointestinal tract, and histamine is known to regulate neuronal activity, control vascular tone, alter endothelial permeability, and modulate gastric acid secretion. However, the action mechanisms of histamine in mouse small intestinal ICCs have not been previously investigated, and thus, in the present study, we investigated the effects of histamine on mouse small intestinal ICCs, and sought to identify the receptors involved. Enzymatic digestions were used to dissociate ICCs from small intestines, and the whole-cell patch-clamp configuration was used to record potentials (in current clamp mode) from cultured ICCs. Histamine was found to depolarize resting membrane potentials concentration dependently, and whereas 2-PEA (a selective H1 receptor agonist) induced membrane depolarizations, Dimaprit (a selective H2-agonist), R-alpha-methylhistamine (R-alpha-MeHa; a selective H3-agonist), and 4-methylhistamine (4-MH; a selective H4-agonist) did not. Pretreatment with $Ca^{2+}$-free solution or thapsigargin (a $Ca^{2+}$-ATPase inhibitor in endoplasmic reticulum) abolished the generation of pacemaker potentials and suppressed histamine-induced membrane depolarization. Furthermore, treatments with U-73122 (a phospholipase C inhibitor) or 5-fluoro-2-indolyl des-chlorohalopemide (FIPI; a phospholipase D inhibitor) blocked histamine-induced membrane depolarizations in ICCs. On the other hand, KT5720 (a protein kinase A inhibitor) did not block histamine-induced membrane depolarization. These results suggest that histamine modulates pacemaker potentials through H1 receptor-mediated pathways via external $Ca^{2+}$ influx and $Ca^{2+}$ release from internal stores in a PLC and PLD dependent manner.

PB상품에 대한 소비자의 지각특성이 관계지속의도에 미치는 영향: 브랜드신뢰 및 브랜드자산을 매개로 한 정책적 접근 (The Effect of Consumer's Perceptual Characteristics for PB Products on Relational Continuance Intention: Mediated by Brand Trust and Brand Equity)

  • 본 연구는 대형할인점의 PB(Private Brand)상품에 대한 소비자의 지각특성과 관계지속의도 간의 영향관계를 규명하는 것으로서 설문조사에 의한 실증적 연구를 토대로 하였다. 실증적 연구를 위해 선행연구를 바탕으로 하여 PB상품에 대해 소비자가 지각하는 특성으로서의 지각품질, 점포이미지, 브랜드이미지, 지각가치 등의 요인을 도출하고 이들 선행요인이 PB상품의 브랜드신뢰와 브랜드자산을 매개로 하여 관계지속의도에 어떠한 영향을 미치는가를 구조적으로 규명하고자 하였다. 실증적 연구를 위한 설문조사는 2012년 1월부터 2012년 5월 말까지 경기 및 부산지역의 소비자를 대상으로 실시하였다. 설문은 전체 300부를 배부하여 253부(84.3%)를 회수하였으며, 그 중 응답이 누락되었거나 불성실한 설문을 제외한 245부(81.6%)를 유효 표본으로 최종분석에 활용하였다. 연구결과 PB상품에 대한 선택속성으로서의 지각품질, 브랜드이미지 및 지각가치는 브랜드신뢰 및 브랜드자산에 유의적인 정(+)의 영향을 미치는 것으로 나타났으며, 나아가 이들 브랜드신뢰와 브랜드자산은 관계지속의도에 유의적인 정(+)의 영향을 미치는 것으로 나타났다. 그러나 PB상품을 판매하는 할인점의 점포이미지는 브랜드신뢰에는 정(+)의 영향을 미치지만 브랜드자산에는 유의적인 영향을 미치지 않는 것으로 분석되었다. 즉, 전반적으로 PB상품에 대해 소비자가 지각하는 품질, 점포이미지, 브랜드이미지 및 지각가치와 관계지속의도간의 관계에서 브랜드신뢰 및 브랜드자산 요인이 이들 관계를 매개하고 있음을 구조적으로 확인할 수 있었다. 이러한 연구결과를 바탕으로 볼 때 PB상품에 대한 소비자의 관계지속의도를 극대화하고 재구매를 지속적으로 유도하기 위해서는 PB상품의 품질향상, 브랜드이미지 개선 및 브랜드가치를 향상시키는 기본적 노력 위에 PB상품의 브랜드에 대한 신뢰와 자산가치를 극대화시키는 전략적 접근이 요구된다고 할 수 있다.

대형마트의 점포이미지가 쇼핑가치 및 쇼핑만족에 미치는 영향: 지각된 혼잡의 역할 (The Influence of Store Images of Discount Stores on Shopping Values and Shopping Satisfaction: The Roles of Perceived Retail Crowding)

  • 본 연구는 3대 대형마트(이마트, 홈플러스 및 롯데마트)의 점포이미지 구성요인이 쇼핑가치 및 쇼핑만족에 미치는 영향을 분석하였다. 점포이미지 구성요인으로 점포분위기, 판매원서비스, 부대시설, 제품구색 및 점포입지를 포함하였고, 쇼핑가치로는 실용적 쇼핑가치 및 쾌락적 쇼핑가치 등을 포함하였다. 자료는 지방에 위치한 세 점포를 이용하는 쇼핑객 114명을 대상으로 'mall-intercept' 방법에 의해 수집한 다음, SmartPLS 2.0에 의해 분석하였다. 분석결과, 판매원서비스 및 점포입지가 실용적 쇼핑가치에 영향을 주고, 점포분위기, 판매원서비스 및 점포입지가 쾌락적 쇼핑가치에 영향을 주는 것으로 나타났다. 쇼핑가치들은 모두 쇼핑만족에 영향을 주는 것으로 나타났다. 이들 두 쇼핑가치 가운데 쾌락적 쇼핑가치가 쇼핑만족에 더 많은 영향을 주는 것으로 나타났다. 본 연구는 지각된 혼잡이 쇼핑가치와 쇼핑만족 간의 관계에 대해 조절역할을 하는지 분석하였다. 분석결과, 쇼핑가치와 쇼핑만족 간의 관계에서 지각된 혼잡이 조절역할을 하지 않는 것으로 나타났다. 이는 쇼핑가치와 쇼핑만족 간의 관계에서 지각된 혼잡의 정도에 따라 이들 관계의 크기가 크게 변하지 않음을 의미한다. 지각된 혼잡의 조절역할 외에, 지각된 혼잡이 쇼핑만족에 직접영향을 줄 것으로 가정하고 이를 WarpPLS 3.0에 의해 분석한 결과, 두 변수 간에는 비선형 관계(non-linear)가 있음이 밝혀졌다. 즉 점포혼잡을 크게 지각할수록 쇼핑만족은 더 크게 증가하는 것으로 나타났다. 본 연구가 기존의 연구와 다른 차별점은 이들 간의 관계에서 지각된 혼잡의 역할을 중점적으로 분석하였다는 점이다. 본 연구는 점포이미지가 쇼핑만족에 미치는 직접효과도 분석하였다. 즉 소비자가 지각하는 점포에 대한 이미지가 쇼핑가치를 매개하지 않고 쇼핑만족에 직접 영향을 주는가를 분석하였다. 분석결과, 점포분위기와 판매원서비스가 쇼핑만족에 직접 영향을 주는 것으로 나타났다. 이러한 연구결과를 토대로 실무적 시사점, 연구의 한계 및 향후의 연구방향 등에 대해 기술하였다.

우리나라 주요 냉장판매식품의 보관온도 실태 조사 (Survey on Storage Temperature of Domestic Major Chilled Foods in Refrigerator)

  • 본 연구는 주요 냉장식품(두부, 묵, 어육가공품(어묵), 생면, 김초밥, 샐러드)의 온도 관리실태를 조사하여 문제점을 파악하고 국내 유통업소의 냉장 냉동진열 판매 제품의 관리온도 개선 및 관리방안을 제시하고자 하였다. 주요 냉장고 내 위치별 온도는 보편적으로 아래($7.5^{\circ}C$)가 높고 윗부분($6.4^{\circ}C$)이 낮았으며, 앞쪽($8.9^{\circ}C$)이 높고 뒤쪽($7.5^{\circ}C$)이 낮은 것으로 조사되었다. 모든 냉장식품의 평균 냉장고 보관 온도는 $7^{\circ}C$(최대 $22.9^{\circ}C$, 최저 $-2^{\circ}C$)였으며, 대형할인매장이 $6.5^{\circ}C$,백화점이 $7.5^{\circ}C$, 편의점이 $7.6^{\circ}C$ 였다. 모든 냉장식품의 평균 표면온도는 $8.2^{\circ}C$(최대 $19^{\circ}C$, 최저 $-0.4^{\circ}C$)였으며, 대형할인매장이 $8.1^{\circ}C$,백화점이 $8.5^{\circ}C$, 편의점이 $7.6^{\circ}C$으로 조사되었다. 식품표면과 중심온도는 식품별 차이가 있으며 최대 $2.3^{\circ}C$ 차이로 나타났다. 두부(표면온도 $6.1{\sim}10.6^{\circ}C$, 중심온도 $4{\sim}11.1^{\circ}C$), 어묵($6{\sim}12.3^{\circ}C$, $6.5{\sim}13.6^{\circ}C$), 김밥($2.3{\sim}18.2^{\circ}C$, $2{\sim}17.3^{\circ}C$), 샐러드류($4.6{\sim}12.2^{\circ}C$, $4.1{\sim}10.4^{\circ}C$), 생면류($3.4{\sim}12.6^{\circ}C$, $3.8{\sim}10.3^{\circ}C$), 묵류($7.1{\sim}19^{\circ}C$, $8.3{\sim}17.1^{\circ}C$)의 다섯 가지 식품별로 차이를 보였다. 냉장고 표시온도와 식품표면 온도 항목간의 유의적 차이가 있는 것으로 나타났다(P<0.01). 냉장고 표시 온도는 김밥 1곳(100%)에서 규정온도인 $10^{\circ}C$를 초과했지만, 식품표면 온도에서는 두부 9곳(17%), 어묵 6곳(11.3%), 묵 10곳(18.9%), 생면 11곳(20.8%), 김밥 13곳(24.5%), 샐러드 4곳(7.5%)이 초과한 것으로 나타났다. 결론적으로 우리나라 냉장보관 식품 중 표면온도와 $10^{\circ}C$를 초과하는 경우가 많아 냉장보관식품의 온도관리가 철저히 이루어져야 할 것으로 판단된다.
