Effects of Feeding Herb Resources Powder on Meat Quality and Sensory Properties in Korean Native Black Goat (한방제재 급여가 재래 흑염소육의 육질 및 관능적 특성에 미치는 영향)

  • This study was carried out to investigate the effect of different herb resource powders on meat quality and sensory properties in Korean native black goat for 500 d. The experimental treatment was arranged with 24 heads (3 treatment/8 heads) by control group, T1 and T2 group respectively (Control, not added herb powders; T1, 1% added herb powders; T2, 2% added herb powders). Total weight gain for the Korean native black goat was the highest in T2 group feeding for 500 days, and daily gain tended to be similar to the total weight gain. The total feed intake were the highest as 519.20 kg in T2 group, although feed conversion showed 18.35 in the T2 group, which means it had the best feed efficiency compared to the other treatment groups. The carcass rate was higher in the T1 group (51.10%) than in the other groups (p<0.05). The cooking loss and drip loss of Korean native black goat was the highest as 34.72% and 3.83% in the control group (p<0.05). However, total cholesterol amounts in the treatment group were not significantly different from, although tended to be higher than, the control group (p>0.05). Also, the overall sensory evaluation of the treatment group revealed low scores, meaning more meat flavor than those of the control in tenderness, flavor, texture, and black goat off-flavor and overall evaluation (p<0.05). Total synthesis evaluation was higher for the treatment group (3.71, 3.90 point) than that of the control group (4.82 point) (p<0.05). Comparative Quality Traits, Flavor Compounds, and Metabolite Profile of Korean Native Black Goat Meat

  • Although goat meat has several health benefits than other red meats but comsumers reluctant it due to its unpleasant flavor. This study aimed to investigate the odorant of goat meat as well as compare the quality traits of meat regarding sex status. The loin meats [non-castrated male (NCM), castrated male (CM), and female (FM)] were collected and stored at 4℃ in a laboratory refrigerator and analyzed on the 1st, 5th, and 8th consecutive days. The moisture content was the lowest and the protein content was highest in FM (p<0.05). Fat and ash content in NCM and FM were similar while lowest in CM. The CIE L* was significantly higher in NCM, but there were no significant differences of CIE a* and CIE b* within groups at initial day. The color intensity increased on the 5th storage day and decreased again after the 8th storage day, except in NCM. NCM displayed the highest thiobarbituric acid reactive substances value (p<0.05), whereas CM displayed a higher pH value than other groups throughout the storage period. Indole levels were the highest in NCM (0.031 mg/kg); however, skatole levels were not significant differences across all treatments (p>0.05). No significant difference was observed in the fatty acid composition between NCM and CM (p>0.05), whereas a significant difference was observed in FM (p<0.05). Most of the water-soluble metabolites showed significant differences between sexes. Overall, sex status effects on quality properties of meat and castration can improve the overall sensory acceptance by reducing goaty flavor of Korean native black goat meat.

Application of artificial insemination and pregnancy diagnosis kit for Korea native black goats (한국 재래흑염소의 계통별 인공수정과 임신진단 키트의 활용)

  • This study investigated the application of artificial insemination and pregnancy diagnosis kit for Korean native black goats. Semen was collected by electrical ejaculation, followed by semen analysis and artificial insemination in three goat strains (Dangjin, Jangsu and Tongyoung). Pregnancy was confirmed using a cow pregnancy test kit (IDEXX Rapid Visual Pregnancy Test kit) and ultrasound diagnosis. Analysis revealed that semen collected from male Korean native black goats by electrical ejaculation was about 1~1.5 ml in volume, $18{\sim}25{\times}10^8/ml$ concentration, and having > 97% motility. Furthermore, confirmation of pregnancy by pregnancy test kit and ultrasound diagnosis after artificial insemination were similar. In addition, the efficiency of pregnancy was 20~40% for all three strains: Tongyoung was the highest with 44%, followed by Dangjin (%), and Jangsu (20%). This study determines the artificial fertilization efficiency and the feasibility of using a cow pregnancy test kit for early pregnancy diagnosis in Korean native black goats. Although further research is required for validation, the results of the current study contribute to the breeding and improvement of Korean native black goat in research institutions as well as in general farms.

Effects of Sous-vide Cooking Temperature on Triceps Brachii of Black Goats

  • The aim of this study was to determine the effects of sous-vide cooking temperature on the triceps brachii of black goats. Triceps brachii of black goats (12 months) were sous-vide cooked at 55℃, 60℃, and 65℃. The samples were examined for color, scanning electron microscope photographs, sarcomere length, fiber cross-sectional area, cooking yield, shear force, sensory evaluation, and aromatic profile. The results showed that CIE a*, CIE b*, and chroma increased with increasing sous-vide cooking temperature. However, the cooking yield significantly decreased with increasing sous-vide cooking temperature, and the shear forces of the 60℃ and 65℃ samples showed no significant differences. For sensory evaluation, the 60℃ sample showed the highest scores for flavor, texture, and off-flavor. Furthermore, the 60℃ sample showed the significantly lowest value of octadienone (aroma characteristics of metallic) intensity (p<0.05). Therefore, sous-vide cooking of triceps brachii of black goats at 60℃ is effective in reducing off-flavor and improving tenderness.

Development of a Microsatellite Marker Set for the Individual Identification and Parentage Verification of Korean Native Black Goats (재래흑염소 개체식별과 친자확인을 위한 Microsatellite Marker Set 개발)

  • The Korean native black goat (Capra hircus coreanae) is the goat species to be officially registered in Korea under the Food and Agriculture Organization. The object of this study is to establish a set of microsatellite (MS) markers for the individual identification and parentage verification of goats. In this study, we analyzed alleles of MS markers in crosses between Korean native black goats and crossbred goats (n=304 animals), and, based on the diversity of alleles for each marker, we selected 11 MS markers for individual identification and parentage verification. Using these 11 MS markers, the probabilities of different individuals with the same genotype being found within random and half-sib mating populations were 5.58×10-10 and 1.15×10-7, respectively. The parentage verification accuracy was 0.999996 when information about the parents was available and 0.999833 with no information. Thus, even given the total rearing population of 576,150 animals in South Korea, we concluded that these markers could be used for the individual identification and parentage verification of goats. Moreover, by analyzing the genetic relationships between the four lines of Korean native black goats and the crossbred goats, we verified the genetic characteristics of Korean native black goats, confirming their conservation value as a unique genetic resource.

Influence of blood serum, follicular fluid and gonadotropin on in vitro maturation for goat oocytes (혈청과 난포액 및 성선자극호르몬 첨가가 염소 난자의 체외성숙에 미치는 영향)

  • This study investigated the effects of goat blood serum (gBS), goat follicular fluid (gFF) and gonadotropin (FSH) on the in vitro maturation, fertilization and development of Korean native black goat oocytes. Our results indicate that the gBS combined with FSH treated group showed significantly higher maturation rate than the other groups. Furthermore, blastocyst formation rate was significantly increased in all treated groups, and gBS and gFF combined with FSH treated groups were higher than other groups. However, gene expression levels of BMP15 and GDF9 in COC, both oocyte maturation related genes, remained unaffected after 24 h maturation. The results of the present study indicate that supplementation of the maturation medium with gBS, gFF and FSH is efficacious in improving the in vitro maturation, fertilization and development of Korea native black goat oocytes.

Effects of Dietary Energy Level on Growth and Meat Quality of Korean Black Goats (사료의 에너지 수준이 육성기 흑염소의 발육과 육질에 미치는 영향)

  • This study was conducted to investigate the optimal energy level at growing Korean black goat from April 20 to November 6, 2006. Forty male Korean black goat were divided into four treatments and were fed with concentrate feed containing ME 2.4, 2.6, 2.8 and 3.0 Mcal/kg with CP 15% and rice straw. Average daily gains of were 59.8, 65.4, 73.2 and 77.2 g/day in groups fed with ME 2.4, 2.6, 2.8 and 3.0 Mcal/kg diet, respectively. There was a tendency to increase ADG as the ME level of concentrate feed grew. Dressing percentage were 49.73, 48.64, 50.19 and 51.36% in each group, and were increased as the ME level grew. Meat percentage ranged between 53.1 and 55.3%, and were almost constant. Ranges of crude fat contents and water holding capacity were and 57.9~59.0%, which were increased as the ME level grew. On the other hand, shear force were between 2.8 and 3.1 kg/cm2, which were decreased as the ME level grew. These results suggest that the optimal energy level at growing goat's concentrate is ME 3.0 Mcal/kg.

Comparative Study on the Production of Interferons from Porcine Blood Leukocytes (돼지의 백혈구 인터폐론 생산에 관한 비교연구)

  • Attempts were to produce porcine leukocyte interferon(PorLeIF) and porcine immune interferon (PorIIF) in the culture of porcine leukocytes. The interferons produced were tested for antiviral activity against vesicular stomatitis virus on poreine-derived PK(15) cells, human-derived FL cells, and Korean native black goat-derived BGK cells. The results were summarized as follws: 1. In the isolation of porcine leukocytes, the mean isolation rate by the buffy coat separation method (28.7%) was higher than that by the hydroxyethyl starch-RBC sedimentation method (9.2%). 2. When NDV(BI)-induced PorLeIFs were assyed on PK(15) cells and FL cells, the mean titers were 129 IU/ml and 72 IU/ml respectively, being 55.8% of the activity in homologous species system expressed in heterologous system. 3. The activities of PHA P-induced PorIIFs were 197 IU/ml on PK (15) cells and no activity on human FL cells. The mean antiviral activity of PorIIF was 1.5 times that of PorLeIF in PK (15) cells. 4. The cytopathic effect of vesicular stomatitis virus was observed in BGK cells derived from Korean native black goat kidney permitting interferon assay on the cells. While the cross-species antiviral activity of reference human ${\alpha},\;{\beta}-interferon$ was observed on the cells, PorLeIF and PorIIF did not show any activity.

Composition in Amino Acid and Changes in Protein Mineral Contents during Storage of Black Goat Extracts (흑염소 증탕액의 아미노산 조성 및 저장 중 단백질과 무기물의 변화)

  • 본 연구는 체중 18~19kg(16~17개월령)의 측염소 3 두(♀)를 반도체로 한 후 흑염소육에 한약재를 첨가한 것을 가약 흑염소 소주로 하고, 흑염소육만 증탕한 것을 순 흑염소 소주로 하였고, 한약재만 증탕한 것을 한약재 증탕액으로 하여 4$^{\circ}C$에 30일간 저장하면서 VBN, 아미노산 무기질 조성 변화를 분석하여 다음과 같은 결과를 얻었다. VBN함량은 모든 처리구에서 저장기간이 경과하면서 증가하는 경향이었고, 가약 흑염소 소주보다 순 흑염소 소주의 VBN이높게 나타났으며(P<0.05) 처리구 모두 실험기간 동안 가식권 범위내에 있었다. 무기물 함량은 가약 흑염소소주가 순 흑염소 소주보다 높은 함량을 나타내었고, 한약재 증탕액에서 높은 함량을 나타내었던 무기물이 가약 흑염소 소주에서 높은 함량을 나타내었다. 특히 가약 흑염소 소주는 순 흑염소 소주도다 Ca, K, Mg가 2배이상 많은 함량을 나타내었다. 저장기간이 경과함에 따라 Ca, Fe는 감소하는 경향이었다. 순 흑염소 소주는 가약 흑염소소주보다 총 아미노산 함량이 높았으며, 가약 흑염소 소주는 glycine, glutamic acid, alanine, aspartic acid 순이었고, 순 흑염소 소주는 glycine, glutamic acid, alanine, aspartic acid, arginine 순이었으며, 한약재 증탕액은 gluta-mic acid와 aspartic acid 함량이 가장 높은 경향이었다.

Differential characterization of myogenic satellite cells with linolenic and retinoic acid in the presence of thiazolidinediones from prepubertal Korean black goats

  • Objective: Myogenic satellite cells were isolated from semitendinosus muscle of prepubertal Korean black goat to observe the differential effect of linolenic and retinoic acid in thepresence of thiazolidinediones (TZD) and also to observe the production insulin sensitive preadipocyte. Methods: Cells were characterized for their stemness with cluster of differentiation 34 (CD34), CD13, CD106, CD44, Vimentin surface markers using flow cytometry. Cells characterized themselves as possessing significant (p<0.05) levels of CD13, CD34, CD106, Vimentin revealing their stemness potential. Goat myogenic satellite cells also exhibited CD44, indicating that they possessed a % of stemness factors of adipose lineage apart from their inherent stemness of paxillin factors 3/7. Results: Cells during proliferation stayed absolutely and firmly within the myogenic fate without any external cues and continued to show a significant (p<0.05) fusion index % to express myogenic differentiation, myosin heavy chain, and smooth muscle actin in 2% horse serum. However, confluent myogenic satellite cells were the ones easily turning into adipogenic lineage. Intriguingly, upregulation in adipose specific genetic markers such as peroxisome proliferation-activated receptor ${\gamma}$, adiponectin, lipoprotein lipase, and CCAAT/enhancer binding protein ${\alpha}$ were observed and confirmed in all given treatments. However, the amount of adipogenesis was found to be statistically significant (p<0.01) with linolenic acid as compared to retinoic acid in combination with TZD's. Conclusion: Retinoic acid was found to produce smaller preadipocytes which have been assumed to have insulin sensitization and hence retinoic acid could be used as a potential agent to sensitize tissues to insulin in combination with TZD's to treat diabetic conditions in humans and animals in future.
