Multilayer Coating with Red Ginseng Dietary Fiber Improves Intestinal Adhesion and Proliferation of Probiotics in Human Intestinal Epithelial Models

  • To exert their beneficial effects, it is essential for the commensal bacteria of probiotic supplements to be sufficiently protected as they pass through the low pH environment of the stomach, and effectively colonize the intestinal epithelium downstream. Here, we investigated the effect of a multilayer coating containing red ginseng dietary fiber, on the acid tolerance, and the adhesion and proliferation capacities of three Lactobacillus strains (Limosilactobacillus reuteri KGC1901, Lacticaseibacillus casei KGC1201, Limosilactobacillus fermentum KGC1601) isolated from Panax ginseng, using HT-29 cells, mucin-coated plates, and human pluripotent stem cell-derived intestinal epithelial cells as in vitro models of human gut physiology. We observed that the multilayer-coated strains displayed improved survival rates after passage through gastric juice, as well as high adhesion and proliferation capacities within the various gut epithelial systems tested, compared to their uncoated counterparts. Our findings demonstrated that the multilayer coat effectively protected commensal microbiota and led to improved adhesion and colonization of intestinal epithelial cells, and consequently to higher probiotic efficacy.

  • A wide range of studies have steadily pointed out the relation of oxidative stress to the primary and secondary causes of human disease and aging. As such, there have been multiple misconceptions about oxidative stress. Most of reactive oxygen species (ROS) generated from chronic diseases cause oxidative damage to cell membrane lipids and proteins. ROS production is increased by abnormal stimulation inside and outside in the body, and even though ROS are generated in cells in response to abnormal metabolic processes such as disease, it does not mean that they directly contribute to the pathogenesis of a disease. Therefore, the focus of treatment should not be on ROS production itself but on the prevention and treatment of diseases linked to ROS production, including types 1 and 2 diabetes, cancer, heart disease, schizophrenia, Parkinson's disease, and Alzheimer's disease. In this regard, Korean Red Ginseng (KRG) has been traditionally utilized to help prevent and treat diseases such as diabetes, cancer, inflammation, nervous system diseases, cardiovascular disease, and hyperlipidemia. Therefore, this review was intended to summarize in vivo animal and human clinical studies on the antioxidant activities of KRG and its components, ginsenosides.

홍삼의 패키지 현황과 디자인 개선 방안 (A Study on the Korean Red Ginseng Packaging)

  • 고려인삼은 예로부터 신비의 영약으로 불려질 만큼 뛰어난 효능을 인정받아 왔다. 인삼을 수분함량 14% 이하가 되도록 하는 가공건조를 통하여 만들어진 홍삼은 부피와 무게를 감소시켜 저장과 운반이 용이해진다. 이 때 특히 사포닌을 비롯한 인삼의 유효성분들이 추가로 생성된다. 고려홍삼에 대한 선행연구들은 홍삼이 두뇌활동 촉진을 비롯한 항암작용, 인슐린억제작용, 지질대사 개선 등과 같은 성인병 치료 및 예방 효과가 있음을 입증하고 있으며, 최근에는 에이즈와 비만치료 효과에 이르기까지 다양한 효능이 밝혀지고 있다. 이처럼 고려홍삼은 약효와 품질의 우수성으로 인하여 세계인삼시장에서 인정받고 있으나, 증가된 생산과 수요, 수출 부진 등으로 인한 어려움을 겪고 있다. 실제 판매와 밀접한 관계가 있는 패키지 디자인에 대한 문제점과 개선 전략 등 디자인 측면에서의 연구는 매우 미흡한 실정이다. 따라서 변화하는 사회 환경과 국제 시장에 대응하는 적절한 패키지디자인 방안이 요구되고 있다. 본 논문에서는 홍삼제품 특성과 시장 환경의 변화, 그리고 세계인삼시장의 현황과 같은 실질적인 분석 결과를 바탕으로 국내외 홍삼과 홍삼류 제품들의 종류 및 패키지 실태를 비교, 분석하여 그 문제점과 해결방안을 모색해 본다. 이를 위한 홍삼제품은 크게 홍삼근류, 진액제품, 분말, 캡슐 및 타블렛, 드링크, 연질제품, 차 그리고 기타 캔디 등으로 나누어 살펴보았다. 앞으로 우리는 이러한 문제점과 방안을 바탕으로 적절한 포장재와 포장방법, 포장단위를 배려한 패키지디자인을 개발하는데 관심을 두어야 할 것이다.

기호성 개선 인삼농축액 제조를 위한 추출 조건 설정 (Characterization and optimization for beverage manufacture using Korean red ginseng extract)

  • 백삼과 홍삼의 추출수율은 각각 33.56%와 48.79%로 홍삼의 추출수율이 높았고, 산성다당체의 경우 1~4차 추출단계까지는 홍삼군이 백삼군보다 높았으나, 5차 추출단계에서 백삼군이 6차 추출단계에서는 홍삼군이 높게 나타났다. 백삼과 홍삼의 추출단계별 농축액의 조사포닌 함량은 백삼(56.44 mg/g)에 비해 홍삼(53.15 mg/g)이 약 5.8 % 증가하는 것으로 나타났다. 이들을 혼합한 선택적 홍삼농축액 모델 4종의 기호도 평가 결과 1, 2, 6차 추출물이 혼합된 RGB-4가 가장 높은 기호도를 나타내었고 시판 홍삼 농축액 3종에 대해 비교한 결과에서도 RGB-4가 쓴맛, 떫은맛, 인삼향 강도가 약해지고 종합적 기호도에서 높은 평가를 받았다. 본 실험에서는 추출방법을 달리 함으로써 추출수율 및 조사포닌 함량을 감안하여 인삼 쓴맛과 향의 강도를 낮출 수 있다는 것을 확인하였다. 이 추출방법을 이용하여 쓴맛을 저감시키고 인삼향미의 강도를 저감시킨 제품을 개발하여 좀 더 편하고 자연스럽게 홍삼제품을 접할 수 있도록 더 많은 연구가 진행 되어야 할 것이다.

인삼의 구증구폭(九蒸九曝)에 의한 갈변물질, 조지방, 조단백 및 향기성분의 변화 (Conversion of Brown Materials, Crude Lipids, Crude Proteins and Aromatic Compounds of Changed Ginseng by 9 Repetitive Steaming and Drying Process)

  • Korean ginseng (Panax ginseng C. A. Meyer) has been used as an important medicinal plant in the Orient for a long time. It has been claimed that ginseng has many beneficial bioactive effects on human health, such as antitumor, antistress, antiaging and enhancing immune functions. Red ginseng possibly have new ingredients converted during steaming and dry process from fresh ginseng. Kujeungkupo method which means 9 repetitive steaming and drying process was used for the processes of green tea, Polygonatum odoratum, and Rehmanniae radix preparata. In this study, ingredient conversion of ginseng by 9 repetitive steaming and drying process were investigated measuring conversion efficiency of brown materials, crude lipids, crude proteins and aromatic compounds. Brown materials, as an antioxidant, in red ginseng were produced through non-enzymatic reaction by heat. Repetitive steaming and drying treatments on ginseng root contiunously increased the content of brown materials and the chromaticity. Crude lipids were degraded by heat and converted into volatile aromatic ingredients. Crude lipids were degraded and decreased by 0.52% after the 5th and 7th. Crude proteins were also decomposed and converted to amino acid. Crude proteins after the 9th treatment were decreased by more than 85% as increased times of treatments. A bicyclogermacrene as aromatic material was decreased as increased treatment times, while but a aromatic caramel was increased.

Effects of Korean Red Ginseng Extract on Cisplatin-Induced Nausea and Vomiting

  • Ginseng, the root of Panax ginseng CA Meyer, is well known as a tonic medicine for restoring and enhancing human health. In traditional medicine, ginseng is utilized for the alleviation of emesis, which includes nausea and vomiting. However, it has not yet been demonstrated whether ginseng exhibits in vivo anti-nausea and anti-vomiting properties. In this study, we examined the anti-emetic effect of Korean red ginseng total extract (KRGE) on cisplatin-induced nausea and vomiting using ferrets. Intraperitoneal administration (i.p.) of cisplatin (7.5 mg/kg) induced both nausea and vomiting with one-hour latency. The episodes of nausea and vomiting reached a peak after 1.5 h and persisted for 3 h. Treatment with KRGE via oral route significantly reduced the cisplatin-induced nausea and vomiting in a dose-dependent manner. The anti-emetic effect was 12.7 $\pm$ 8.6, 31.8 $\pm$ 6.9, and 67.6 $\pm$ 4.0$\%$ with doses of 0.3, 1.0, and 3.0 g/kg of KRGE, respectively. Pretreatment with KRGE via oral route 1 and 2 h before cisplatin administration also significantly attenuated the cisplatin-induced nausea and vomiting. However this did not occur with a pretreatment 4 h before cisplatin administration. These results are supportive of KRGE being utilized as an anti-emetic agent against nausea and vomiting caused by chemotherapy (i.e. cisplatin).

Inhibitory Effect of Fermented Red Ginseng against Passive Cutaneous Anaphylaxis Reaction and Scratching behaviors in Mice

  • To evaluate the antiatopic effect of Korea red ginseng (RG, steamed root of Panax ginseng CA Meyer, Family Araliaceae) fermented by Bifidobacterium longum H-1 (FRG), its inhibitory effect on passive cutaneous anaphylaxis (PCA) reaction and itching in mice was measured. FRG and its ingredient saponin fraction (FSF) potently inhibited PCA reaction and scratching behaviors. FRG at a dose of 200 mg/kg and FSF at a dose of 50 mg/kg significantly inhibited the scratching frequency by 45% and 47%, respectively. FRG and FSF also inhibited the degranulation and protein expression of tumor-necrosis $factor-{\alpha}$ and interleukin-4 of RBL-2H3 cells induced by IgE-complex. However, polysaccharide fraction of FRG (FPF) weakly inhibited it, compared with FSF. The inhibitory effect of FRG against PCA reaction and scratching behaviors more potently inhibited than that of RG. Based on these findings, FRG can improve allergic skin disorders atopic dermatitis by the regulation of $TNF-{\alpha}$, and IL-4 produced by mast cells and basophils and its degranulation.

수삼(水蔘), 홍삼(紅蔘) 및 백삼(白蔘)의 지방질성분(脂肪質成分)에 관한 연구(硏究) (Studies on the Lipid Components of Fresh Ginseng, Red Ginseng and White Ginseng)

  • Lipid and fatty acid compositions of free lipids and bound lipids from fresh ginseng, red ginseng and white ginseng were studied by means of silicic acid column chromatography, thin-layer chromatography and gas-liquid chromatography. Free lipid and bound lipid contents in those three samples were 1.21 to 1.45% and 0.32 to 0.45%. Neutral lipid fractions in free lipids from the samples were 76.6 to 79.7%, while glycolipid and phospholipid fractions were 11.6 to 14.7% and 8.5 to 8.7%, respectively. The major lipids were triglycerides, sterol esters and hydrocarbons, diglycerides and free sterols in neutral lipids, sterol glucoside, monogalactosyl diglyceride, esterified steryl glycoside, digalactosyl diglyceride in glycolipids and phosphatidyl ethanolamine, phosphatidyl glycerol, phosphatidyl choline and phosphatidyl inositol in phospholipids. Fourteen kinds of even numbered and four kinds of odd numbered fatty acids were identified in the four lipid fractions (TL, NL, GL and PL) by GLC, and the main fatty acids were linoleic acid, palmitic acid, oleic acid and linolenic acid.

인삼(Panax ginseng C.A. Meyer) 신품종 고풍의 육성경과 및 생육특성 (Breeding Process and Characteristics of Gopoong, a New Variety of Panax ginseng C.A. Meyer.)

  • KT&G 중앙연구원 원료연구소에서 육성한 신품종 고풍에 대해서 육성경과, 생산력 검정 및 지역 적응시험을 수행하였다. 1968년에 선발하여 20년 이상 육성된 고풍은 줄기, 잎자루, 꽃대 및 잎맥부위까지도 진한 자색을 띤다. 열매송이 모양은 역삼각형이며 진한 붉은 색으로 재래혼계종인 자경종이나 기개발된 천풍 및 연풍과 지상부 표현형에서의 차이를 나타내었 다. 1981-1984년에 실시한 생산력 검정시험이나 1985-1990년에 실시한 지역적응력시험에서 대비구인 재래혼계종인 자경종과의 지상부 생육 제반 특성에서는 차이가 없었으나 유전적 특성의 발현 재현성이 있었다. 지하부 형질에서는 고풍의 생존율이 대비구인 자경종보다 2.6% 높았고, 고풍의 칸당 수량은 2.3 kg으로 자경종 2.2 kg보다 0.1 kg이 높았으며, 고급홍삼인 천지삼율은 고풍은 16.6%로 대비구인 자경종 9.4%보다 7.2%가 더 높았다. 따라서 고풍은 고급 홍삼제조용 원료삼 품종으로 가능성이 매우 높은 것으로 판단되었다.

Based on these findings, RG can improve allergic skin disorders atopic dermatitis and contact dermatitis by the regulation of $TNF-{\alpha}$, and IL-4 produced by mast cells and basophils and their membrane stabilization.
