Effect of Gelatin on the Stability of Heavy Chain Monoclonal Antibody Production from Plant Suspension Cultures

  • The heavy chain monoclonal antibody (HC MAb) was produced in suspension cultures of genetically modified Nicotiana tabacum. The HC MAb secreted to the medium was unstable due to unfavorable interactions in the plant cell medium. The addition of gelatin (5g/l) stabilized the extracellular HC MAb and increased its production 10-fold. A kinetic model was developed describing the interaction between the secretedprotein and the stabilizer. The model accounted for the inactivation of the protein by simple aggregation and general instability. It was assumed that the secreted protein and the stabilizer form a stable complex. Culturing the cells semicontinuously could further increase the productivity of HC MAb.

  • The dissociation kinetics of linear polyaminopolycarboxylate complexes of lanthanide ions (L$n^{3+}: Ce^{3+},\;Eu^{3+}\;and\;Yb^{3+}$) has been studied in an aqueous solution of 0.10 M (NaCl$O_4$) at 25.0${\pm}0.1^{\circ}C$ using Cu(II) ions as a scavenger. The dissociation rates of acid-catalyzed pathway decrease in the order Ln(EPDTA$)^- > Ln(DPOT)^- > In(TMDTA)^- > Ln(MPDTA)^- > Ln(EDTA)^- > Ln(PDTA)^- > Ln(DCTA)^-$ according to the present and literature data. An increase in the N-Ln-N chelate ring from 5 to 6 and substitution of two methyl groups, one ethyl and hydroxyl group on a chelate ring carbon of these ligands leads to a decrease in kinetic stabilities of the complexes. The substitution of one methyl group and cyclohexyl ring on a ring carbon, however, results in a significant increase in the kinetic stability of the resulting $Ln^{3+}$ complexes. Individual reaction steps taking place for each system, with different copper, acetate buffer concentration and pH dependence, are also discussed.

  • 전산유동 수치모형을 이용하여 다양한 대기안정도 상태하에서 부산광역시내 승학산과 구덕산의 초고해상도 풍력자원을 평가하였다. 연구에 사용된 수치모형은 중규모와 미규모 기상현상의 재현에 널리 사용되는 전산유동 수치모형인 A2C이다. 대기안정도가 강할 때, 위치에너지의 크기가 상대적으로 강해지기 때문에 산을 넘어가는 경향이 나타난다. 반면 대기안정도가 약해지면서 산악후면의 후류 발생이 증대되며, 난류에너지가 증가한다. 그리고 연평균 풍력밀도, 난류운동에너지, 연직 바람전단력 분석을 통하여 구덕산 정상의 남쪽 부근이 다른 구역보다 가용 풍력자원이 풍부함을 확인하였다.

  • We investigate the austenite stability in nanocrystalline Fe-7%Mn-X%Mo (X = 0, 1, and 2) alloys fabricated by spark plasma sintering. Mo is known as a ferrite stabilizing element, whereas Mn is an austenite stabilizing element, and many studies have focused on the effect of Mn addition on austenite stability. Herein, the volume fraction of austenite in nanocrystalline Fe-7%Mn alloys with different Mo contents is measured using X-ray diffraction. Using a disk compressive test, austenite in Fe-Mn-Mo alloys is confirmed to transform into strain-induced martensite during plastic deformation by a disk d. The variation in austenite stability in response to the addition of Mo is quantitatively evaluated by comparing the k-parameters of the kinetic equation for the strain-induced martensite transformation.

Comparison of EMG Activity of the Posterior Oblique Sling Muscles and Pelvic Rotation During Prone Hip Extension With and Without Lower Trapezius Pre-Activation

  • Background: Prone hip extension (PHE) can be performed to measure the lumbopelvic motor patterns and motions. Imbalances in lumbopelvic muscle activity and muscle weakness can result in instability including pain in lumbopelvic region. The posterior oblique sling (POS) muscles contribute to dynamic lumbopelvic stability. In addition, POS are anatomically aligned with the trapezius muscle group according to shoulder positions. Objects: This study compared the electromyography (EMG) activity of POS and pelvic compensations during PHE with and without pre-activation of lower trapezius muscle (lowT). Methods: Sixteen healthy males were recruited. PHE was performed in randomized order: PHE with and without lowT pre-activation. Surface EMG signals were recorded for biceps femoris (BF), gluteus maximus (GM) (ipsilateral), lumbar multifidus (MF) (bilateral), and the lowT (contralateral). An electromagnetic tracking motion analysis was used to measure the angle of pelvic rotation and anterior tilting. Results: The ipsilateral GM and bilateral MF EMG amplitudes were greater during PHE with lowT pre-activation compared to PHE without lowT pre-activation (p<.05). The BF amplitude during PHE without lowT pre-activation was significantly greater than that during PHE with lowT pre-activation (p<.05). The angles of pelvic rotation and anterior tilting during PHE with lowT pre-activation were significantly smaller compared to PHE without lowT pre-activation (p<.05). Conclusion: PHE with lowT pre-activation, which is aligned with the POS, showed more increased MF and GM muscular activity with smaller lumbopelvic compensations in rotation and anterior tilting compared to PHE without lowT pre-activation.

동결건조 분말된장의 흡습 거동에 대한 속도론적 연구 (Kinetic Study for Hygroscopic Behavior of Freeze Dried Soy Paste Powder)

  • 서로 다른 외관색택을 갖는 (적갈색 및 담백색) 2종의 된장을 동결건조하고 분말화 하였다. 이들의 흡습거동을 여러 수분찰성도$(0.11{\sim}0.88)$ 및 서로 다른 세온도(25, 40 및 $50^{\circ}C$)에서 조사하였으며, 품질 특성 및 저장안정성의 관점에서 동력학적 및 열역학적 해석을 하였다. 시료 분말된장의 등온수분흡착은 BET분류에 따른 typell에 속하였고, Henderson의 경험식으로 잘 설명할 수 있었다. Caurie식으로부터 계산한 저장 안전수분량은 온도에 의존하였으며 $5.5{\sim}3.98%$(건량기준)범위 이었다. 수분흡습과정은 평형수분함량과 임의의 수분함량과의 차이에 대한 1차 속도식에 따랐으며, 초기에 높은 흡습성을 나타내는 특징을 보였다. 분산성 및 색차의 변화는 저장수분활성도에 민감한 의존성을 보였으며, 최대의 저장안정성은 저장안전 수분함량수준에서 나타났다. 또한, 수분함량에 대한 열역학적 변수의 변화도 저장안전 수분함량수준에서 유의성이 있었으며, 분말된장이 저장안정성과 높은 상관성을 갖는 것으로 나타났다.

  • As for the purpose, we first introduce an random mutation into wild-type gene to expand a mutation space, and then further recombine the mutant genes by staggered extension process PCR. As a result, we obtained the best clones 6-52 that showed a high activity and stability, from a round of error prone and staggered extension process PCR. The purified enzyme showed a similar pH stability to the wild-type enzyme and reveal a slightly high optimum pH at 12. In the optimum temperature, an identical dependency was also showed and a quite high stability in the thermal stability was obtained. Along with this, the enzyme was also stable at a reaction that supplement with a 15 % of ethanol as an additive. The addition of other solvents and surfactants did not improve the reaction and thus resulted in a similar profile to those of wild-type enzyme. The specific activity on the target compound rac-ketoprofen ethyl ester was calculated to be about 85, 000 unit, and the kinetic constants Km and Vmax were determined to be 0.2 mM and 90 mM/mg-protein/min respectively. The deduced amino acid alignment with the wild type enzyme revealed five mutations at L120P, I208V, T249A, D287H and T357A. Based on these observations, the site directed mutagenesis to delineate the mutagenic effect is under progress.

저질이 천연수중 구리와 카드뮴의 화학종 분포에 미치는 영향 (Influence of Sediment on the Chemical Speciation of Copper and Cadmium in an Aquatic System)

  • In order to predict the distribution of chemical species of copper and cadmium in water, conditional stability constant and complexation capacity between copper or cadmium and natural aquatic sediment have been determined in a shallow lake in Haman, Kyungnam. Kinetic parameters were calculated by Langmuir isotherm equation. Conditional stability constant was log $K_{cuSed}=4.78 and log K_{cdSed}=4.45$. Complexation capacity was $1.70{\times}10^{-4}$moles/g for copper and $5.54{\times}10^{-5}$moles/g for cadmium. Accuracy of experimental values of conditional stability constant was checked by comparing the calculated concentration of the metals with the measured one. Relatively good agreement between these values was obtained. Relative errors were 8.9% for copper and 6.5% for cadmium. Data of the measured conditional stability constant were put into data base of MINEQL computer program, and concentration of various chemical species of copper and cadmium in a model aquatic system was calculated. Aquatic sediment was associated with copper at the concentration of $10^{-5M}(0.059g/\ell)$10-5M(0.059g/l) and with cadmium at the concentration of $10^{-6M}(0.018g/\ell)$, and it significantly influenced on the distribution of chemical species of the metals. This result showed that prediction of chemical species of the heavy metals in an aquatic system should be taken into account the influence of the sediment.

  • Purpose To investigate the physico-chemical properties of CB-ph [2-benzoyloxycinnamaldehyde], an anticancer drug obtained from Cinnamomum cassia using methylenechloride. and its stability in various aqueous solutions. Results CB-ph was rarely soluble in water but soluble in methanol and very soluble in ether. Kinetic salt effect on degradation of CB-ph in buffer solutions at pH 4.0 and 6$0^{\circ}C$ showed a linear relationship having a positive slope that means reactions between hydronium ions and protonated substrates.   • In this paper, an optimal motion control scheme is proposed for robot manipulators. A simple explicit solution to the Hamilton-Jacobi equation is presented. The optimization of motion control is based on the mininization of the torque term affecting the kinetic energy and the augmented error which has the first-order stable dynamics for the position and velocity tracking error. In the presence of parametric uncertainty, an adaptive control scheme using the optimal principle is proposed. The global stability of the closed-loop system is guaranteed by the Lyapunov stability approach, The effectiveness and feasibility of the proposed control schemes are shown by simulation results.
