Kinetic Parameters 결정을 위한 Equivalent Point Method의 이용 (Application of the Equivalent Point Method for Estimation of Kinetic Parameters)

  • 본 고에서는 Equivalent time과 Equivalent temperature를 활용하여 Kinetic parameters를 결정하는 새로운 방법을 제안하였다. 본 방법의 타당성을 두 가지의 kinetic data 즉, 계산치와 실험치를 이용하여 예시하였다. 계산치는 그 Kinetics가 잘 알려진 세 가지 화학반응에 대해 임의의 등온가열조건을 적용하여 계산하였고 실험치는 2% 설탕용액을 사용하여 0.0005N 염산용액을 사용하여 가수분해가 일어나는 정도를 효소반응을 이용하여 측정하였다. 본 방법에 의해 결정된 활성화 에너지와 Frequency factor는 각각 $104.74{\pm}1.87KJ/mol$$5.26{\times}10^{14)hr^{-1}$이었으며 이들 값은 보고된 결과와 잘 일치되었다.

  • 식품의 품질변화에 미치는 온도의 영향을 분석하기 위한 kinetic parameters를 추정하는데 있어서 표준온도 상당시간이 유효함을 밝히기 위하여 비효소적 갈색반응 액체 모델 및 식품시스템을 직선적으로 온도를 증가시키는 조건$(6^{\circ}\;/h)$에서 반응시켰다. 표준온도 상당시간은 $t_E=-{\exp}[-(E_a/R)\;(1/T-1/T_{ref})]$로 정의되며, 임의의 온도 T에서 1일 동안에 일어난 품질변화는 표준온도 $T_{ref}$에서 몇일 동안에 일어난 변화에 상당하는가를 의미한다. 이와 같이 정의되는 표준온도 상당시간을 이용하여 glucose-lysine모델, 여러종류의 아미노산과 당의 액체모델 및 액체 음료 식품시스템의 실험자료로부터 비선형 최소자승법으로 kinetic parameter, n, $k_{ref}$$E_a$를 추정하였다. Glucose와 lysine의 손실속도는 1차 반응, 갈색물질 형성속도는 0차 반응이었다. 추정한 kinetic parameters를 이용하여 computer simulation한 결과 실험값과의 상관계수가 0.96 이상으로 잘 일치하여 $t_E$를 이용한 1단계 비선형 최소자승법으로 kinetic parameters를 효율적으로 정확히 추정할 수 있음을 확인하였다.

  • 주요 식중독균인 E. coli O157:H7, L monocytogenes, S. aureus, S. Typhimurium과 S. Enteritidis를 주어진 이산화염소수 농도에서 시간별로 처리한 inactivation data를 first-order kinetic model과 Weibull model을 이용하여 kinetic parameter를 비교, 분석하였다. 그 결과 Weibull model이 first-order kinetic model보다 inactivation kinetic data에 더 적합한 것을 보였다. 또한 특정 농도의 이산화염소수에서 처리한 survival plot의 tR values(특정 이산화염소수 농도에서 미생물 90%를 감소시키기 위해 필요한 시간)는 Weibull model을 적용 시, E. coli O157:H7가 5 ppm에서 2.49 min, L. monocytogenes가 5 ppm에서 1.47 min, S. aureus가 5 ppm에서 0.94 min, S. Typhimurium이 1 ppm에서 0.87 min 그리고 S. Enteritidis가 1 ppm에서 0.08 min으로 측정 되었다.

닫힌사슬과 열린사슬 자세에서 편측상지에 적용된 PNF가 양측 하지의 근 활성도에 미치는 영향 (Effect of Close kinetic chain and Open kinetic chain Position on Proprioceptive Neuromuscular Facilitation Applied to the Unilateral Upper Extremity on the Muscle Activation of Lower Extremity)

  • The purpose of this study was to investigate the effect of close kinetic chain(CKC) and open kinetic chain(OKC) posion on proprioceptive neuromuscular facilitation applied to the unilateral upper extremity on the muscle activation of lower extremity. All subjects were randomly assigned to two groups: open kinetic chain group(n=5),closed kinetic chain group(n=5). All participants were PNF patterns applied on the unilateral upper extremity in all subjects were the kinetic chain(CKC) and open kinetic chain(OKC) posion on flexion/abduction/external rotation. The hold and approximation techniques for the irradiation were applied to end range. All measurements for each subject took the following tests: pre-test, post - test in 4weeks, post-testin 8weeks. EMG data was collected from the vastus medialis, tibialis anterior, biceps femoris, and gastrocnemius muscle of both lower extreamity using surface EMG system, Each EMG value in individual muscle was normalized for maximal voluntary contraction. The data were analyzed using Two-way analysis of variance(ANOVA) with repeated measures to determine the statistical significances. The results of this study are summarized as follows. First, during for close kinetic chain(CKC) and open kinetic chain(OKC) posion on PNF pattern application, all of the %MVIC values of close kinetic chain and open kinetic chain posion increased sign ificantly compared(p<0.05). Second, The close kinetic chain(CKC) and open kinetic chain(OKC) posion on PNF pattern application was significantly increased with in the intervention period(p<.05). Third, there was a no significant open kinetic chain posion on PNF pattern application of sing muscle group with in the intervention period.(p<0.05) there was a significant close kinetic chain posion on PNF pattern application of sing muscle group with with in the intervention period(p<0.05). Forth, interaction of the exercise position and muscle was also significant. Post-hoc tests revealed that the activation levels of vastus medialis muscle and tibialis anterior muscle was higher in the closed kinetic chain position(p<.05). that the activation levels of vastus medialis and gastrocnemius muscle was higher in the open kinetic chain position(p<.05). In conclusion, it was found that the application of PNF patterns to the unilateral upper extremity effect on the muscle activation of lower extremity and both open kinetic chain exercise and closed kinetic chain exercise was significantly increased muscle activity. Further studies are needed to analyzed long term effects and subjects resulting from these changed.

열린사슬운동 및 닫힌사슬운동에서 위팔굽힘 각도에 따른 앞톱니근의 근활성도 비교 (A Comparison of the Serratus Anterior Muscle Activity according to the Shoulder Flexion Angles in a Closed Kinetic Chain Exercise and an Open Kinetic Chain Exercise)

  • PURPOSE: The purpose of present study was to ascertain how the activity of the serratus anterior muscle, the upper trapezius muscle and the pectoral major muscle was affected while the upper arm was being flexed at 70, 90 and 110 degrees respectively in a closed kinetic chain exercise (wall push up plus) and an open kinetic chain exercise (static hug). METHODS: Sixteen healthy young men subjects participated in the study. Surface electromyography (EMG) data were collected from the dominant-side muscles during a closed kinetic chain exercise and an open kinetic chain exercise. The activity of each muscle was measured quantitatively, and by the use of the two-way repeated ANOVA, the data were compared with each other according to exercises and shoulder flexion angles. RESULTS: Results indicated that the closed kinetic chain exercise did not interact with the open kinetic chain exercise (p>.05). In both the closed kinetic chain exercise and the open kinetic chain exercise, the activity of the serratus anterior muscle became different significantly according to angles (p<.05). Its activity increased in order of 70, 90 and 110 degrees (p<.05). In both exercises and all angles, muscle activity was significantly higher in the serratus anterior muscle than in the upper trapezius muscle and the pectoral major muscle (p<.05). CONCLUSION: The above results show that there is a need to selectively control the exercise stress of the serratus anterior muscle in the case of the patients with the shoulder impingement syndrome characterized by the winged scapula, insufficient scapular protraction and upward rotation.

Analysis of Kinetic Data of Pectinases with Substrate Inhibition

  • Enzyme kinetics data play a vital role in the design of reactors and control of processes. In the present study, kinetic studies on pectinases were carried out. Partially purified polymethylgalacturonase (PMG) and polygalacturonase (PG) were the two pectinases studied. The plot of initial rate vs. initial substrate concentration did not follow the conventional Michaelis-Menten kinetics, but substrate inhibition was observed. For PMG, maximum rate was attained at an initial pectin concentration of 3 g/l, whereas maximum rate was attained when the initial substrate concentration of 2.5 g/l of polygalacturonic acid for PG I and PG II. The kinetic data were fitted to five different kinetic models to explain the substrate inhibition effect. Among the five models tested, the combined mechanism of protective diffusion limitation of both high and inhibitory substrate concentrations (semi-empirical model) explained the inhibition data with 96-99% confidence interval.

  • Kinematic and kinetic analysis has been performed for Soft Golf swings utilizing realistic three dimensional computer simulations based on three dimensional motion tracking data. Soft Golf is a newly developed recreational sport in South Korea aimed to become a safe and easy-to-learn sport for all ages. The advantage of Soft Golf stems from lighter weight of the club and much larger area of the sweet spot. This paper tries to look into kinematic and kinetic aspects of soft golf swings compared to regular golf swing and find the advantages of lighter Soft Golf clubs. For this purpose, swing motions of older aged participants were captured and kinematic analysis was performed for various kinematic parameters such as club head velocity, joint angular velocity, and joint range of motions as a pilot study. Kinetic analysis was performed by applying kinematic data to computer simulation models constructed from anthropometric database and the measurements from the participants. The simulations were solved using multi-body dynamics solver. Firstly, the kinematic parameters such as joint angles were obtained by solving inverse dynamics problem based on motion tracking data. Secondly, the kinetic parameters such as joint torques were obtained by solving control dynamics problem of making joint torque to follow pre-defined joint angle data. The results showed that mechanical loadings to major joints were reduced with lighter Soft Golf club.

Protein Adsorption on Ion Exchange Resin: Estimation of Equilibrium Isotherm Parameters from Batch Kinetic Data

  • The simple Langmuir isotherm is frequently employed to describe the equilibrium behavior of protein adsorption on a wide variety of adsorbents. The two adjustable parameters of the Langmuir isotherm - the saturation capacity, or $q_m$, and the dissociation constant, $K_d$ - are usually estimated by fitting the isotherm equation to the equilibrium data acquired from batch equilibration experiments. In this study, we have evaluated the possibility of estimating $q_m$ and $K_d$ for the adsorption of bovine serum albumin to a cation exchanger using batch kinetic data. A rate model predicated on the kinetic form of the Langmuir isotherm, with three adjustable parameters ($q_m,\;K_d$, and a rate constant), was fitted to a single kinetic profile. The value of $q_m$ determined as the result of this approach was quantitatively consistent with the $q_m$ value derived from the traditional batch equilibrium data. However, the $K_d$ value could not be retrieved from the kinetic profile, as the model fit proved insensitive to this parameter. Sensitivity analysis provided significant insight into the identifiability of the three model parameters.

  • A kinetic model of the cyclodextrin formation in a heterogeneous enzyme reaction system using swollen extrusion starch as substrate was derived emphasing the structural features of extrusion starch. The degree of gelatinization, the ratio of accessible and inaccessible portion of extrusion starch, adsorption of CGTase on swollen starch, the structural transformation during reaction, and product inhibition caused by produced CDs were considered in deriving kinetic model. Various kinetic constants were also evaluated. The derived kinetic equation was numerically simulated, which result showed that the derived kinetic equations can be used to predict the experimental data reasonably well under the various experimental conditions. Kinetic model can be utilized for the optimization of enzyme reactor and the process development for CD production from swollen extrusion starch.

생전분을 당공여체로 한 Stevioside의 당전이 반응의 동력학적 해석 (Kinetic Analysis of Transglycosylation Reaction of Stevioside Using Raw Starch as a Glycosyl Donor)

  • 분쇄마찰매치 함유 불균일상 효소반응계에서 생전분을 당공여체로 한 stevioside의 당전이 반응의 kinetics에 관한 연구를 수행하였다. 생전분으로부터 CD를 생서하는 과정과 생성된 CD와 stevioside가 random sequenrial bireactant ,echanism으로 반응하여 당전이 equation을 유도하였다. 또한 유도된 반응식의 각종 kinetic constants을 평가하였다. 유도된 반응식을 Runge-Kutta integration법으로 계산하였으며, 계산 결과를 실험치와 비교하여 유도식의 효용성을 검토하였다. 유도된 kinetic equations는 당공여체인 생전분의 농도, stevioside의 농도, 그리고 중간산물인 CD의 농도 변화를 비교적 정획히 표시할 수 있었으며, 고효율 당전이 효소반응기 개발에 활용될 것이다.
