Development of bio-fusion materials with skin penetrating property derived from Aurelia aurita (경피 침투율이 높은 보름달 물해파리 유래 바이오 융합 소재 개발)

  • Previously, we reported LVH peptiede derived from Aurelia aurita as cosmecetuticals with anti-winkle activity. In this study, we synthesized pal-LVH using palmitic acid to enhance skin permeability of LVH and examined the effects as cosmeceuticals of pal-LVH. To evaluate these effects, we performed cell toxicity, wound healing, and patch test for skin irritation with LVH and pal-LVH and compared these results for their effects. As a result. pal-LVH was not showed in cytotoxicity and allergenic effect like as LVH. Besides, pal-LVH had almost same excellent anti-ageing properties in high concentration and anti-winkle effect in low concentrationwas as LVH. These results suggested synthesis of palimitic acid and LVH did not affect any functions as cosmeceuticals with increasing skin permeability. Therefore, pal-LVH can be adaptable as new cosmecetuticals with anti-winkle and anti-ageing materials and applied in the development of medicine through various convergence study.

Clinical comparison studies on 20 cases of stroke patients with dysarthria by Sa-Am & General acupuncture (중풍후 언어장애 환자 20례에 대한 체침과 사암침 치료의 임상적 비교 고찰)

  • Objective : The study was designed to evaluate the possibility of treatment of stroke patients with dysarthia by the sa-am acupuncture. Methods: From March 1st 2003 to June 30th 2003, the clinical comparison studies were carried out 20 cases of stroke patients with dysarthria treated by Sa-Am acupuncture(Group I) & general acupuncture(Group II), who had been treated in Dept. of acupuncture and Moxibustion, Dae-jeon Oriental Medical Hospital, Dae-jeon University. Results : The results obtained as follows; 1. The Articuration of Group I and Group II was improved significantly. Group I is more effective than Group II significantly(p<0.05). 2. The Vowel of Group I and Group II was improved but there was no signification(p>0.05). 3. The Alternation of Group I and Group II was improved significantly. Group I is more effective than Group II significantly(p<0.05). 4. The Speed of reading the sentence of Group I and Group II was improved but only Group I was improved significantly(p<0.05). Conclusions: This result suggest that sa-am acupuncture was effective treatment of stroke patients with dysarthria. So further research is needed continuously.

  • To determine if initial infection with Mycobacterium tuberculosis changes the balance of cytokines between T cells and macrophages, we evaluated interferon (IFN)-${\gamma}$), interleukin-12 (IL)-12, and tumor necrosis factor (TNF)-${\alpha}$ productions by peripheral blood mononuclear cells (PBMC) from 15 untreated active pulmonary tuberculosis (TB) patients and 12 healthy tuberculin reactors (HTR). Freshly isolated PBMC were stimulated with Triton X-100 solubilized protein (TSP), 30-kDa or purified protein derivatives (PPD) antigen for 6, 18 and 96 hours. IL-12 p40 production by antigen-stimulated PBMC from TB patients was significantly decreased compared with that in HTR. In addition, IFN-${\gamma}$ production was significantly depressed in TB patients than that in HTR at a 96-hr stimulation. However, TNF-${\alpha}$ production was significantly higher in antigen-stimulated PBMC from TB than that of HTR. A pronounced increase in IFN-${\gamma}$ protein followed neutralization of IL-10 in early TB patients. However, neutralization of TNF-${\alpha}$ did not significantly alter IFN-${\gamma}$ induction in PBMC from TB patients. There were no significantly differences in the cytokine productions among three proteins, TSP, 30-kDa or PPD antigen. These results indicate that development of TB may be strongly associated with dysregulated productions of IL-12, IFN-${\gamma}$ and TNF-${\alpha}$, during the initial immune responses to M. tuberculosis. Further understanding of operative cytokine networks during human immune cell responses to protein antigens of M. tuberculosis may improve strategies for vaccine development.

  • 디지털화의 가속, 고속 통신 인프라의 확대 등으로 전자, 정보 통신 기기들이 단일 네트워크로 연결되어 영상 및 음향 정보를 서로 공유할 수 있으며, 생활 공간 내에서 실생활의 질 향상을 위한 지능적 정보 서비스와 자연스럽고 편한 내추럴 인터페이스 기술에 의한 지능형 반응 정보 서비스 공간 기술이 중요한 이슈로 등장하고 있다. 본 연구에서는 지능형 반응 공간의 물리적 객체로서 학교, 연구 기관 및 회사 등의 회의실을 선정한다. 그리고, 이를 대상으로 회의 참여자들이 자연스럽고 편리하게 의견 교환, 관련 자료 및 정보 처리를 할 수 있는 시스템 구축을 위한 아키텍처에 대하여 논의한다. 본 연구에서 제안하는 시스템 아키텍처는 회의와 관련된 문서나 회의 내용 등의 정보를 실감 가시화 노드로 추상화되고 메타 정보화함으로써 전체 회의 내용의 파악과 회의 정보에 대한 체계적이고 논리적인 관리를 가능하게 한다. 또한 여러 사람의 공동 작업을 필요로 하는 정보 또는 문서에 대한 동시 편집 기능과 자연스러운 동작에 의한 데이터 조작을 지원하는 실감 워크벤치 및 워크스크린 기술, 정보 핸들링의 다양성과 조작의 편리성을 위한 실감 아이콘에 의하여 자연스럽고 편리한 회의를 가능하게 한다. 그리고, 이러한 요소 기술들이 에이전트에 의해 회의 프로세스 및 요소 기술들의 시스템적 통합을 가능하게 한다.

Serum Ferritin as a Risk Factor in Type 2 Diabetes Mellitus (2형 당뇨병 발생위험인자로서의 혈청 Ferritin의 의의)

  • Purpose : Iron accumulation interferes with hepatic insulin extraction and affects insulin synthesis and secretion. The purpose of this study is to investigate the correlation between serum ferritin and type 2 diabetes mellitus. Methods : We compared the serum ferritin level among 18 patients in an impaired glucose tolerance (IGT) group, 36 in a type 1 diabetes group, eight in a type 2 diabetes group and 29 in a healthy control group. The correlation between serum ferritin levels and sex, body mass indices(BMI), blood pressure(BP), serum fasting sugar level and serum fasting insulin level were also analyzed. Results : The mean log ferritin were $1.33{\pm}0.32$(healthy control group), $1.63{\pm}0.19$(IGT group) and $1.90{\pm}0.30$(type 2 diabetes group). In the IGT group, log ferritin was higher than in the healthy control group(P=0.001). The log ferritin of the type 2 diabetes group was higher than that of the healthy control group(P=0.001). Comparing log ferritin to other factors, log ferritin had a significant positive correlation with body mass indices(P<0.001), systolic blood pressure(P=0.001), and fasting glucose(P=0.001), fasting insulin(P=0.002). Conclusion : Compared to the normal healthy group, serum ferritin concentrations were significantly higher in the IGT group and the type 2 diabetes group. The elevation of serum ferritin concentration may be a risk factor of type 2 diabetes mellitus.

Effect of Areca Nut on Helicobacter pylori-Induced Gastric Diseases in Mice

  • Areca nut (AN) chewing is a habit in many countries in Central, Southern, and Southeast Asia. It is strongly associated with the occurrence of oral, pharyngeal, and esophageal cancer as well as systemic inflammation. However, the association between AN intake and the development of gastric lesions has not yet been identified. The aim of this study was to investigate the effect of AN on gastric diseases using a mouse model for Helicobacter pylori infection. We studied four groups of mice: those fed a normal diet (ND), those fed a diet containing 2.5% AN (AD), those fed ND and infected with H. pylori PMSS1 strain (ND/HP), and those fed AD and infected with H. pylori PMSS1 strain (AD/HP). Food intake and body weight were monitored weekly during the experiments. At 10 weeks, the mice were sacrificed, and the stomach weight, H. pylori colonization, and gastric inflammation were evaluated. The stomach weight had increased significantly in the ND/HP and AD/HP groups along with increases in H. pylori colonization; however, there was no significant difference between these two groups with respect to stomach weight and colonization. On histological grading, mononuclear cell infiltration was severer in the AD/HP group than in the ND/HP group. These data suggest that chronic gastric inflammation was aggravated by AN treatment in the mice with H. pylori-induced gastric lesions. Furthermore, as previously suggested, this animal model is useful to determine the effect of potential carcinogens on gastric lesions induced by H. pylori infection.

Study on the correlation between the soil bacterial community and growth characteristics of wild-simulated ginseng(Panax ginseng C.A. Meyer) (토양세균군집과 산양삼 생육특성 간의 상관관계 연구)

  • The studies regarding soil bacterial community and correlation analysis of wild-simulated ginseng cultivation area are insufficient. The purpose of this study was to investigate the correlation between soil bacterial community and growth characteristics of wild-simulated ginseng for selection of suitable cultivation area. The bacterial community was investigated by high throughput sequencing technique (Illumina platform). The correlation coefficient between soil bacterial community and growth characteristics were analyzed using Spearman's rank correlation. The soil bacterial community from soil samples of 8 different wild-simulated ginseng cultivated area exhibited two distinct clusters, cluster 1 and cluster 2. The relative abundance of Proteobacteria (35.4%) and Alphaproteobacteria(24.4%) was observed to be highest in all soil samples. The lower soil pH and higher abundance of Acidobacteria resulted in increased growth of wild-simulated ginseng. Additionally, abundance of Acidobacteriia (class) and Koribacteraceae (family) demonstrated significant positive correlation with fresh weight of wild-simulated ginseng. The results of this study clearly state the correlation between growth characteristic and soil bacterial community of wild-simulated ginseng cultivation area, thereby offering effective insight into selection of suitable cultivation area of wild-simulated ginseng.

Effects of Replacing Mushroom By-product with Tofu By-product on the Chemical Composition, Microbes, and Rumen Fermentation Indices of Fermented Diets (두부비지의 버섯 폐배지 대체 수준이 발효사료의 영양소 함량, 미생물 성상 및 반추위 내 발효특성에 미치는 영향)

  • This study aimed to estimate the effects of replacing Mushroom By-Product (MBP) with Tofu By-Product (TBP) on the chemical composition, microbes, and rumen fermentation indices of Fermented Diets (FDs). The basal diet was formulated using MBP, TBP, rice bran, molasses, and inoculants. The MBP in the basal diet was replaced with TBP at 0, 5, and 10% on Dry Matter (DM) basis for the experimental diets. The experimental diets were fermented at $39^{\circ}C$ for 144 h. Chemical composition, pH, microbes, and rumen fermentation indices of the FDs were analyzed. With increasing TBP replacement, crude protein content of FDs increased (L, P < 0.001), whereas crude ash content decreased (L, P = 0.002). Lactic acid bacteria and Bacillus subtilis contents in the TBP-replaced FDs were higher than those in the control (P < 0.05), whereas pH level and mold count were lower (P < 0.05). With increasing TBP replacement, in vitro rumen digestibility of DM (L, P = 0.053) and neutral detergent fiber (L, P = 0.024) increased, wheres rumen pH changed (P = 0.026) quadratically. Rumen total volatile fatty acid (L, P = 0.001) and iso-butyrate contents (Q, P = 0.003) increased with increasing TBP replacement. In conclusion, this study indicates that the replacement of MBP with TBP could improve the quality of FD.

Autophagy Inhibition with Monensin Enhances Cell Cycle Arrest and Apoptosis Induced by mTOR or Epidermal Growth Factor Receptor Inhibitors in Lung Cancer Cells

  • Background: In cancer cells, autophagy is generally induced as a pro-survival mechanism in response to treatment-associated genotoxic and metabolic stress. Thus, concurrent autophagy inhibition can be expected to have a synergistic effect with chemotherapy on cancer cell death. Monensin, a polyether antibiotic, is known as an autophagy inhibitor, which interferes with the fusion of autophagosome and lysosome. There have been a few reports of its effect in combination with anticancer drugs. We performed this study to investigate whether erlotinib, an epidermal growth factor receptor inhibitor, or rapamycin, an mammalian target of rapamycin (mTOR) inhibitor, is effective in combination therapy with monensin in non-small cell lung cancer cells. Methods: NCI-H1299 cells were treated with rapamycin or erlotinib, with or without monensin pretreatment, and then subjected to growth inhibition assay, apoptosis analysis by flow cytometry, and cell cycle analysis on the basis of the DNA contents histogram. Finally, a Western blot analysis was done to examine the changes of proteins related to apoptosis and cell cycle control. Results: Monensin synergistically increases growth inhibition and apoptosis induced by rapamycin or erlotinib. The number of cells in the sub-$G_1$ phase increases noticeably after the combination treatment. Increase of proapoptotic proteins, including bax, cleaved caspase 3, and cleaved poly(ADP-ribose) polymerase, and decrease of anti-apoptotic proteins, bcl-2 and bcl-xL, are augmented by the combination treatment with monensin. The promoters of cell cycle progression, notch3 and skp2, decrease and p21, a cyclin-dependent kinase inhibitor, accumulates within the cell during this process. Conclusion: Our findings suggest that concurrent autophagy inhibition could have a role in lung cancer treatment.

Identification and gene expression profiling of chicken Pumilio family, Pum1 and Pum2

  • Members of the Pumilio are the RNA binding proteins acting as translational repressors and required for germ cell development and asymmetric division. We identified chicken Pum1 and Pum2 that are similar to mouse and human in highly conserved C-terminal RNA-binding domain and eight tandem repeats. The comparative sequence analysis of Pum1 and Pum2 from fly, chicken, mouse and human shows high degree of evolutionary conservation in the homology of the peptide sequence and the structure of PUM-HD (Pumilio homology domain) with similar spacing between adjacent Pum repeats. Also, structures of chicken Pum1 and Pum2 genes are almost identical to those of mouse and human. We revealed that the expression levels of Pum1 and Pum2 were the highest in hatched female gonad among various embryonic tissues, and Pum2 expressed highly in 12-day and hatched gonad by real-time RT-PCR. These results suggest that Pum1 and Pum2 might have an effect on the development of chicken gonad.
