Ginsenoside Rg5 overcomes chemotherapeutic multidrug resistance mediated by ABCB1 transporter: in vitro and in vivo study

  • Background: Multidrug resistance (MDR) to chemotherapy drugs remains a major challenge in clinical cancer treatment. Here we investigated whether and how ginsenoside Rg5 overcomes the MDR mediated by ABCB1 transporter in vitro and in vivo. Methods: Cytotoxicity and colon formation as well as the intracellular accumulation of ABCB1 substrates were carried out in MDR cancer cells A2780/T and A549/T for evaluating the reversal effects of Rg5. The expressions of ABCB1 and Nrf2/AKT pathway were determined by Western blotting. An A549/T cell xenograft model was established to investigate the MDR reversal activity of Rg5 in vivo. Results: Rg5 significantly reversed ABCB1-mediated MDR by increasing the intracellular accumulation of ABCB1 substrates without altering protein expression of ABCB1. Moreover, Rg5 activated ABCB1 ATPase and reduced verapamil-stimulated ATPase activity, suggesting a high affinity of Rg5 to ABCB1 binding site which was further demonstrated by molecular docking analysis. In addition, co-treatment of Rg5 and docetaxel (TXT) suppressed the expression of Nrf2 and phosphorylation of AKT, indicating that sensitizing effect of Rg5 associated with AKT/Nrf2 pathway. In nude mice bearing A549/T tumor, Rg5 and TXT treatment significantly suppressed the growth of drug-resistant tumors without increase in toxicity when compared to TXT given alone at same dose. Conclusion: Therefore, combination therapy of Rg5 and chemotherapy drugs is a strategy for the adjuvant chemotherapy, which encourages further pharmacokinetic and clinical studies.

Correlation Analysis between Key Word Search Frequencies Related to Food Safety Issue and Foodborne Illness Outbreaks (식중독 사고 발생과 식품 안전 관련 검색어 빈도와의 상관성 분석 연구)

  • Through the increasing use of internet and smart device, consumers can search the information what they want to find. The information has been accumulated and become into a big data. Analyzing the big data regarding key words associated with foods and foodborne pathogens could be a method for predicting foodborne illness outbreaks, especially in school food services. Therefore, the objective of this study was to elucidate the correlations between key words associated with foods and food safety issues. Frequencies of the key words for foodborne pathogens and food safety issues were searched using an internet portal site from January 1, 2012 to December 31, 2014. In addition, foodborne outbreak data were collected from Ministry of Food and Drug Safety for the same period of time. There was correlation between the time having maximum key word frequencies of foods and foodborne pathogens, and the time for foodborne illness outbreak occurred. In addition, the search frequencies for foods and foodborne pathogens were generally increased right after foodborne outbreaks occurred. However, in some cases foodborne outbreaks occurred after the search frequencies for certain seasonal foods increased These results could be useful in food safety management for reducing foodborne illness and in food safety communication.

  • Parathyroid hormone is the most important endocrine regulator of calcium concentration. Its N-terminal fragment (1-34) has sufficient activity for biological function. Recently, site-directed mutagenesis studies demonstrated that substitutions at several positions within shorter analogues (1-14) can enhance the bioactivity to greater than that of PTH (1-34). However, designing the optimal sequence combination is not simple due to complex combinatorial problems. In this study, support vector machines were introduced to predict the biological activity of modified PTH (1-14) analogues using mono-substituted experimental data and to analyze the key physicochemical properties at each position that correlated with bioactivity. This systematic approach can reduce the time and effort needed to obtain desirable molecules by bench experiments and provide useful information in the design of simpler activating molecules.

  • This study addresses preliminary design and construction specification for micropiles. Especially several design methods for micropiles in sands, clays or mixed soil layers are described. The bearing capacity of micropiles are mainly depended upon the shaft resistance. Therefore, the pressure of grouting is one of the most important design parameters for the bearing capacity evaluation of micropile. There is no theoretical way to evaluate the shaft resistance of micropile up to now because grouting method is another key parameter for micropile design approach. Because of above reasons, the present design approaches of the micropile are based on the collected field data The bearing capacities of designed micropiles should be verified by static load tests before and after construction at the planned site.

  • The stocks of listed S&M sized firms in the KoreaStock Exchange(KSE) have been neglected to investors for long time in site of their good performences. The word "Neglected" means that the stocks of listed S&M sized firms in the KSE have fail to acquried liquidity. In the result, Listed S&M sized firms in the KSE have not financed equity by issuing stocks timely in the primary market. This problem has resulted in poor investment to their listed S&M sized firms in the KSE. The possible key to sovlve this problem is in the Institutional Investors, especially to the Pension fund. Korean Institutional Investors have lost their basic roles, that is, final supports to prevent the markets not to demolish. The Acts prventing Pension to invest to the stocks is to change to allowing to invest them in soon. this opportunity is good chance to solve the problems of poor liquidity of stocks of listed S&M sized firms in the KSE.

Synthesis and Binding Affinity of Homologated Adenosine Analogues as A3 Adenosine Receptor Ligands

  • Homologated analogues 3a and 3b of potent and selective A3 adenosine receptor ligands, IB-MECA and dimethyl-IB-MECA were synthesized from commercially available 1-O-acetyl-2,3,5-tri-O-benzoyl-${\beta}$-D-ribofuranose (4) via $Co_2(CO)_8$-catalyzed siloxymethylation as a key step. Unfortunately, homologated analogues 3a and 3b did not show significant binding affinities at three subtypes of adenosine receptors, indicating that free rotation, resulting from homologation, induced unfavorable interactions in the binding site of the receptor maybe due to the presence of many conformations.

  • A new transplant production system that produces high quality plug seedlings of specific crop has been studied. It is a plant factory designed to produce massive amount of virus free seedlings. The design concept for building this plant factory is to realize maximum energy efficiency and minimum initial investment and running cost. The basic production strategy is the sitespecific management. In this case, the management of the growth of individual plantlet is considered. This requires highly automated and information intensive production system in a closed aseptic environment the sterilized specific crops. One of the key components of this sophisticated system is the irrigation system. The conditions that this irrigation system has to satisfy are: 1. to perform the site specific crop management in irrigation and 2. to meet the no waste standard. The objective of this study is to develop an irrigation scheduling that can implement the no waste standard.

A Study on the Decomposition of Amygdalin Using an In Vitro Assay (Amygdalin의 in Vitro 분해에 관한 연구)

  • Amygdalin is a cyanogenic glycoside which is commonly found in almonds, bamboo shoots, and apri-cot kernels, and peach kernels. Amygdalin was first hydrolysed into prunasin, then degraded into cyanohydrin by sequential two-stage mechanism. The objective of this study was to examine the amygdalin decomposition and cyanide formation at various in vitro conditions, including acid, enzyme and anaerobic microbes (AM) in human feces (HF). In acid hydrolysis mimicking gastric environment, amygdalin was degraded to cyanide up to 0.2% in specific pH. In contrast, enzyme assay showed higher cyanide generation either by ${\beta}$-glucosidase, or by incubation with microbe. In conclusion, we are convinced of cyanide generation are occurred mainly by microbiological activities of the gut flora up to 41.53%. After ingestion with some staff, the degree and site of degradation in an organism is a key parst of regulatory decision making of that staff.
