  • Endometriosis is classically defined as the growth of endometrial glands and stroma at extra-uterine sites. Although it is a common gynecological problem accompanied by chronic pelvic pain, infertility, and adhesion formation, the etiology of this disease is unknown. Endometriosis pathogenesis may involve endocrine and immune dysregulation since uterine endometrial growth is regulated by sex hormone in concert with bioactive mediators produced by uterine immune and endocrine cells. IL-8/CXCL8 Upregulates 12-Lipoxygenase Expression in Vascular Smooth Muscle Cells from Spontaneously Hypertensive Rats

  • Background: We previously demonstrated remarkable differences in the expression of IL-8/CXCL8 in aortic tissues and vascular smooth muscle cells (VSMC) from spontaneously hypertensive rats (SHR) compared to VSMC from normotensive Wistar-Kyoto rats (WKY). In the present study, we investigated the direct effect of IL-8/CXCL8 on expression of 12-lipoxygenase (LO), a hypertensive modulator, in SHR VSMC. Methods: Cultured aortic VSMC from SHR and WKY were used. Expression of 12-LO mRNA was determined by real-time polymerase chain reaction. Phosphorlyation of ERK1/2 and production of 12-LO and angiotensin II subtype 1 ($AT_1$) receptor were assessed by Western blots. IL-8/CXCL8-stimulated DNA synthesis was determined by measuring incorporation of [$^3H$]-thymidine. And effect of IL-8/CXCL8 on vascular tone was determined by phenylephrine-induced contraction of thoracic aortic rings. Results: Treatment with IL-8/CXCL8 greatly increased 12-LO mRNA expression and protein production compared to treatment with angiotensin II. IL-8/CXCL8 also increased the expression of the $AT_1$ receptor. The increase in 12-LO induced by IL-8/CXCL8 was inhibited by treatment with an $AT_1$ receptor antagonist. The induction of 12-LO mRNA production and the proliferation of SHR VSMC by IL-8/CXCL8 was mediated by the ERK pathway. The proliferation of SHR VSMC and the vascular contraction in the thoracic aortic ring, both of which were induced by IL-8/CXCL8, were inhibited by baicalein, a 12-LO inhibitor. Conclusion: These results suggest that the potential role of IL-8/CXCL8 in hypertensive processes is likely mediated through the 12-LO pathway.

  • 현재 여러 기관에서 사용하고 있는 방송 시스템은 스튜디오, 방송 장비의 설치 및 확장에 대한 불편함과 비용의 부담, 거리에 따른 방송품질의 저하 등 많은 문제점을 가지고 있다. 이를 해결하기 위한 대안으로 인터넷을 이용한 방송 시스템이 등장하였다. 이는 별도의 시스템 추가 없이 인터넷이 연결되어 있는 장치를 이용하므로 비용절감과 설치의 간편함, 일정한 방송품질을 제공할 수 있다. 그러나 현재 소개되어 있는 인터넷을 이용한 방송 시스템들은 독자적 합의에 의한 호 설정으로서, 시스템 확장과 선택에 대하여 매우 낮은 퍼포먼스를 제공한다. 이를 해결하기 위해 차세대 네트워크 서비스로 각광받고 있는 SIP(Session Initiation protocol)을 이용하여 방송 시스템을 설계 및 구현 하였다.

Inhibitory Effect of Coicis Semen Extract(CSE) on Pro-inflammatory Mediatory (의이인(薏苡仁)의 염증성 사이토카인 발현 및 조절에 관한 연구)

  • Objectives This study was evaluated the effects of CSE the regulatory mechanism of NO and cytokines in the LPS-stimulated Raw 264.7 cells. Methods The Coicis Semen MeOH extract dissolved in EMEM for 1 hour prior to the addition of LPS(1${mu}g/ml$). The cell viability was measured by MTT assay, and Nitric Oxide production was monitored by measuring the nitrite content in culture medium. The levels of cytokine and PGE2 were analyzed by sandwich immunoassays. Results CSE inhibited the production of NO (0.03 and 0.1 mg/ml), $TNF-{\alpha}$ (0.03 and 0.1 mg/ml), $IL-1{\beta}$ (0.03 and 0.1 mg/ml), IL-6 (0.03, 0.1 mg/ml) and PGE2(0.03 and 0.1 mg/ml) in Raw 264.7 cells activated with LPS(lipopolysaccharide). Conclusion According to the results above, Coicis Semen can produce anti-inflammatory effect, which may play a role in adjunctive therapy in Gram-negative bacterial infections.

Anti-inflammatory effects of porcine placenta in forced swimming tested fatigue mice and RAW264.7 cells

  • Inflammation has been linked to various diseases. Especially, fatigue is a frequent symptom in several inflammatory disorders. Therefore, blocking inflammatory process is effective in fatigue. We investigated whether Denmark porcine placenta (DPP) alleviates fatigue by inhibiting inflammatory reaction using forced swimming test (FST) animal model and RAW264.7 cells. In FST-induced fatigue animal model, the mice which received the DPP for 21 days showed decreases of interleukin $(IL)-1{\beta}$ and IL-6 serum levels. Furthermore, our data revealed that lipopolysaccharide (LPS)-induced $IL-1{\beta}$, IL-6, and tumor necrosis $factor-{\alpha}$ secretion were markedly inhibited by DPP in RAW264.7 cells without inducing cytotoxicity. LPS-enhanced nitric oxide secretion and inducible nitric oxide synthase expression were inhibited by DPP. The present study also figured out that these effects of DPP were mediated by blockade of caspase-1 and nuclear $factor-{\kappa}B$ activation. Taken together, our results indicated that DPP could be alleviating fatigue as candidate of anti-inflammatory agent.

  • This paper presents design and characteristics analysis of transverse flux switched reluctance generator(TFSRG) for wind turbine. Dimension is calculated by using output equation and maximum magnetomotive force(mmf) equation per pole. Design specification within effective range of mechanical and electrical energy is suggested in order to perform the analysis. it is confirmed to torque, inductance and induced electromotive force(emf) for one phase through three dimension Finite Element Analysis. Then design specification can be verified by comparing with proposed specification.

  • This paper introduces novel a two-phase permanent magnet BLDC (PMBLDC) motor with both an asymmetric tooth and an auxiliary tooth in order to improve the dead point, efficiency and torque ripple. To calculate the merits of introducing each of the asymmetric tooth and the auxiliary tooth, characteristic analysis is performed respectively using finite element method (FEM). To maximize performance, we propose a novel model which combines the asymmetric tooth and the auxiliary tooth. To maximize the efficiency of the novel model, an optimal design is processed using the Kriging method and a genetic algorithm. Finally, an experiment is used to confirm the initial and optimal design results.

  • The Single Species, Unreacted Flow Model which is effectively applicable on the computational analysis of rocket exhaust flow is introduced in this paper. The basic concept of this model had been originated from chemically frozen analysis of hot air but it was complemented by compensating molecular weight and specific heat which was obtained CEA code analysis of exhaust plume. Comparing single species, unreacted model with the finite chemistry model, unreacted model can reduce calculation time to 1/5 while it makes similar simulation results.
