A Potent Anti-diabetic Agent from Kalopanax pictus

  • To search for the anti-diabetic principle from the stem bark of Kalopanax pictus, seven kinds of chemical constituents including hederagenin glycosides and phenolic glycosides wre isolated. The anti-diabetic evaluation of these isoltes in the streptozotocin-induced diabetic rats exhibited that kalopanaxsaponin A has a potent anti-diabetic activity in contrast to a mild activity of hederagenin. In addition, significant hypocholesterolemic and hypolipidemic activities of kalopanaxsaponin A and hederagenin were observed. The structure-activity relationship of kalopanaxsaponin A was also investigated in the present work.

Monthly Variation of the Kalopanaxsaponin Content in the Leaves of Kalopanax pictus (개두릅에서 칼로파낙스사포닌의 월별 함량 변화의 추이)

  • To find the monthly variation of kalopanaxsaponin contents in the leaves of Kalopanax pictus with thorns (KPT) and with no thorns (KPN), the leaves monthly collected from May to September were extracted with MeOH and then the kalopanaksaponin fractions were prepared. KPT collected on May showed the lowest saponin content of the KPTs whereas KPT on August exhibited the highest saponin content. From September, the saponin content in the loaves decreased. The highest saponin content was shown to be 7.3% in KPN collected on August. Evaluation of six kalopanaxsaponins A, I, J, B, H and K (KPA, KPI, KPJ, KPB, KPH, and KPK) were performed using TLC densitometer. In this measurement, considerably higher KPB and KPH, both hederagenin bisdesmosides, were found whereas very low contents In monodesrnosides KPI and KPJ were observed. In conclusion, it was shown that the leaves of KPN of August could be a biomaterial source for the kalopanaxsaponin fraction. It was also suggested that measurement of the weight of kalopanaxsaponin fraction and the content of KPB as the representative compound for kalopanaxsaponins will be used for the quantitative evaluation of the kalopanaxsaponins of K. pictus.

  • An experiment was conducted to examine the effects of Kalopanax pictus Nakai (Kalopanax) on in vitro rumen fermentation and methane (CH4) reduction. Kalopanax trunk was extracted with 70% ethanol and 70% methanol. Rumen fluid, alfalfa hay and buffer (control: C) supplemented with 0.3% Kalopanx juice (T1), 0.3% ethanol extract (T2) and 0.3% methanol extract (T3) in the total volume of culture medium were incubated at $38^{\circ}C$ for 24h and 48h. Rumen pH was lower in all Kalopanax treatments during all incubations than that in control (p<0.05). Total VFA and total gas production in T2 and T3 was significantly higher than that in C at 48h incubation (p<0.05). Ammonia-N was decreased in all treatments compared with C during the incubation periods (p<0.05). At 24h incubation, $CH_4$ contents were significantly reduced by both alcohol extracts. It is concluded that supplementing Kalopanax extracts can stimulate ruminal fermentation of rumen microorganisms and inhibit methanogenesis.

Mass Loss Rates and Nutrient Dynamics of Oak and Mixed-Hardwood Leaf Litters in a Gyebangsan (Mt,) Forest Ecosystem

  • Patterns of mass loss and nutrient release from decomposing oak (Quercus mongolica) and mixed litters (Q. mongolica, Betula schmidtii, Acer pseudo-sieboldianum, Kalopanx pictus and Tilia amurensis) in a natural hardwood forest in Gyebangsan (Mt.) were examined using litterbags placed on the forest floor for 869 days. Mass loss rates from decomposing litter were consistently higher in mixed litter (59%) than in oak litter types (52%) during the study period. Nutrient concentrations such as nitrogen (N), phosphorus (P), potassium (K), and magnesium (Mg) from decomposing litter were also higher in mixed litter than in oak litter types. Nutrient concentrations (N, P, Ca, and Mg) increased compared with initial concentration of litter, while K concentrations dropped rapidly at the first 5 months and then stabilized. The results suggest that mas loss and nutrient release obtained from decomposing litter of single species in mixed hardwood forest ecosystem should be applied with caution because of the potential differences of mass loss and nutrient release between single litter and mixed litter types.

Effect of Inorganic Components on the Seedling Growth Performance of Kalopanx pictus Nakai (음나무의 유묘생장에 영향을 미치는 엽중 무기성분 함량)

  • Terminal buds and young leaves of Kalopanax pictus are in a great demand as a edible vegetable. Its bark of stems and roots have been used as the resources of folk medicine in Korea. Recently, attempts to cultivate the plant have being tried in farms and mountain villages. This study was conducted to determine effect of inorganic elements on seedlings growth of K. pictus. Levels of inorganic elements in the leaves were variable based on macro and micro inorganic elements. Among the inorganic elements in the leaves of K. pictus seedlings, level of N was high, while other ions were low in the order of K, Ca, P, Mg, Mn, Fe and Zn. A highly significant correlations between the seedling growth and the level of N, P, K, and Na in the leaves, while a low positive significant correlations between the growth and the level of either Mg or Mn and no significant correlations between the growth and the level of Ca, Fe, Cu, and Zn. The most affecting inorganic ion on the seedling growth was N, and followed by the order of K, P, Mg, Mn and Na.
