3D-QSAR and docking studies of selective COX-2 inhibitors

  • The three-dimensional quantitative structure-activity relationship (3D-QSAR) approach using comparative molecular field analysis (CoMFA) and comparative molecular similarity analysis (CoMSIA) was applied to 62 derivatives known as COX-2 selective inhibitors. Partial least square (PLS) analyses produced good predicted models with q2 value of 0.803 (s=0.285, F=215.401, r2=0.951) and 0.769 (s=0.192, F=245.364, r2=0.980) for CoMFA and CoMSIA, respectively.   • This study was carried out to simulate the change of various properties of recycled water when zero-discharge system was applied to a KOCC recycling process. contaminants such as chemical oxygen demand, anionic trash, and calcium hardness were gradually increased in the process water as the recycling was repeated . Especially, the increase of anionic trash and COD were closely related to the starch derived from corrugating adhesive in KOCC. Four kinds of water were compared in the preparation of handsheet to evaluate the performance of retention program. Waters used in this work were laboratory tab water, process water from Dong-II mill and the same one treated by UASB process, and closed white water prepared by KRICT. The result revealed that one the major factors fro reducing a retention power was the anioni trash accumulated in the recycled water.

  • 산림자원의 부족현상이 갈수록 심화됨에 따라, 재활용폐지를 원료로 활용하는 비율이 증가되고 있으며, 재활용 폐지의 품질도 지속적으로 악화되어 제지 환경이 점점 어려워지고 있다. 이에 최근에 개발되어 지대지, 라이너 판지등에 적용하여 강도상승 목적으로 사용중인 호프만계 폴라비닐아민에 대한 성능평가 결과를 고찰하고자한다. 호프만계 폴리비닐아민의 가장 큰 장점은 투입량을 증량하여도 종이의 Formation에 악영향을 마치지 않고 효과적으로 내절도, 파열강도등 강도적 적성이 향상되므로 응용 범위가 점차 확대되고 있다. 이와 더불어 제지환경이 점점 어려워짐에 따라, 보류 및 탈수 향상등 제지 공정개선제로 많이 사용되는 파우더 및 에멀젼의 성능 고도화가 절실히 요구되고 있으며, OCI-SNF의 개선 사례를 살펴보고자한다.

  • Recently lots of study on EL have been performed by other researcher. Organic multilayer system of TPD/$Alq3$ and Rhodamine 101 perchrolate/Alq3/ was constructed on ITO and finally Al as cathodic electrode. The thickness of emitting layer was $150{\AA}$ and device was fabricated by changing amount of dopant. AFM image for each surface morphology and EL spectra using fluoromax-2 was investigated. Electrical and emission properties of EL device was dependent on deposition method and condition.

  • A white-yellowish 5-7 cm sized solid mass was observed in the mammary gland of a 7 years old poodle bitch. Mass was well-defined and metastasis was not observed. In microscopic findings, mass contained hyperplasia of glandular epithelial cells, proliferation of mucin-secreting myoepithelial cells and their stroma and formation of cartilage and bone. Bone marrow with fat and hematopoietic cells was observed. Based on the microscopic findings, the mass was diagnosed as benign mixed tumor of the mammary gland in dog.

  • Pitch synthesis reaction was studied based on the effect for chemical composition of feedstock. Feedstock was selected as pyrolyzed fuel oil (PFO) and coal-tar (CT), which are by-products in petroleum and steel industry. Pitch was prepared at $420^{\circ}C$ for 180 minutes on atmospheric pressure by thermal treatment. Thermal stability and softening point (SP) of the prepared pitches were investigated and their molecular weight distribution was analyzed by MALDI-TOF. PFO has various aliphatic compounds and coal-tar has high aromaticity with 3 wt% of primary quinoline insolubles. The thermal property of PFO was enhanced with polymerization reaction during the thermal treatment with increased molecular weight range. But CT was inferior to PFO because of side reaction by hetero elements. CTP was appeared molecular weight by 0~200 m/z.

  • SOD2-transgenic $T_3$ petunia line (A2-36-2-1-1-35) was treated with different levels of methyl viologen (MV) to determine its resistance to oxidative stress. Four (4) levels of MV (0, 100, 200, and $400\;{\mu}M$) were applied. The SOD2-transgenic $T_3$ petunia line exhibited a very significant oxidative stress resistance at the highest MV concentration ($400\;{\mu}M$) treatment compared to non-transgenic plant. RNA and protein expression of SOD2 transgene and higher parenchyma cell density in the transgenic petunias exhibiting resistance to oxidative stress proves its contribution to the expression of its resistance to oxidative stress.

  • Seventy three hydrolysable tannins and related compounds were isolated from seven Euphorbia plants. Among them, 28 compounds including nine gallotannins, eleven ellagitannins and eight related compounds were selected according to the structural similarity. Cytotoxicity of them on the human tumor cell lines including A-549, SK-OV-3, SK-MEL-2, XF-498 and HCT-15 were evaluated by the SRB method in vitro. 3,4,6-Tri-O-galloyl-D-glucose was shown to exhibit most potent cytotoxic effect($4.4{\mu}g/ml).

  • 88 selective COX-2 inhibitors belonging to three chemical classes (triaryl rings, diaryl cycloalkanopyrazoles, and diphenyl hydrazides) were studied using comparative molecular field analysis (CoMFA) and comparative molecular similarity indices analysis (CoMSIA). Partial least squares analysis produced statistically significant models with q values of 0.84 and 0.79 for CoMFA and CoMSIA, respectively. The key spatial properties were detected by careful analysis of the isocontour maps. The binding energies calculated from flexible docking correlated with inhibitory activities by the least-squares fit method. (omitted)

  • The safety pharmacological core battery studies of AS2-006A. a newly developed wound healing drug, were investigated according to the ICH S7A guidelines in compliance with Good Laboratory Practice(GLP) Regulations, The doses given were 0, 100. 300 and 1000 mg/kg and drugs were administered subcutaneously. The animals used for this study were mice, rats and guinea pigs. AS2-006A showed no effects on the central nervous system such as motor activity. behaviotal changes. coordination, sensory/motor reflex responses and body temperature. no effects on blood pressure(BP). heart rate(HR), and ECG profiles and respiratory system. it was concluded that AS2-006A possess no general pharmacological effects at all doses tested.
