A STUDY ON THE COLOR STABILITY OF PORCELAIN REPAIR RESINS (도재 수리용 레진의 색안정성에 관한 연구)

  • Statement of problem : Porcelain restorations are widely used in restorative dentistry with a high degree of general success. However, porcelain is brittle and may fracture intraorally. Sometimes a repair may be attempted using composite resin and bonding system available. Color stability of porcelain repair resins affects esthetics and prognosis of prosthesis, so that provides important criteria for the choice of porcelain repair resins. Purpose : The purpose of this study was to investigate the color stability of composite resins used widely as porcelain repair system. Materials and method : Composite resins used for this study were Carisma(Kulzer, Germany), Tetric ceram(Ivoclar, Liechtenstein), Filtek Z250(3M, USA), and Arabesk Top(VOCO, Germany). Thirty disc-shaped specimens($10{\times}2mm$) were fabricated from each composite resin. Color changes of specimens after treatment using thermocycling system(KD-TCS30, Kwang-duk F.A. Korea) and after immersion in methylene blue(0.05% by Wt) were measured with colorimeter(Model Tc-6Fx, Tokyo Denshoku Co.) and obtained data were analyzed. Results and Conclusion: The results obtained from this study were as fo11ows. 1. The color change of Filtek Z250 was lesser than those of Tetric ceram and Arabesk Top after thermocycling (12000 times). 2. All of the composite resins in this study showed increase of color change according to increase of thermocycling frequency. 3. After treatment with methylene blue, the color change of Arabesk Top was greater than those of Carisma and Tetric ceram, and Tetric ceram was more stable than others.

Extraction and fractionation of proteins haying both chitinase and ${\beta}-1,3-glucanase$ canase activities from rice leaves ($Chitinase/{\beta}-1,3-glucanase$ 활성 동시보유 벼잎단백질 분획의 성질)

  • Five electrophoretic bands of crude enzyme extracted from rice leaves were found to possess both chitinase and ${\beta}-1,3-glucanase$ activities. These $chitinase/{\beta}-1,3-glucanase$ were resolved into acidic and basic fractions of protein by DEAE-cellulose and chitin affinity column chromatography. The optimal pH and temperature for ${\beta}-1,3-glucanase$ activity of two fractions were in the same extent as pH 5 and $60^{\circ}C$, whereas those for chitinase activity differed from one another; pH 3 and $60^{\circ}C$ for the acidic and pH 4 and $50^{\circ}C$ for the basic fraction, respectively. In addition, lysozyme activity was found in both fractions.

The Study of Indoor Air Quality at Schools in Chung-Nam Area (충남 지역 학교의 실내공기질에 관한 연구)

  • The concentrations of HCHO(formaldehyde), $PM_{10}$(particulate matter), $CO_2$(carbon dioxide) and TBC(total bacteria counter) distribution in schools(Chung-Nam Area) were examined, and the results were compared with the recommended criterion of the administration law of indoor air. The subjects were an elementary school, a middle school and a high school in Chung-Nam area, and the concentration of TBC was examined by Single Stage Air Cascade Sampler, which applied the inertia collision catching method of 28.29L/min(flux) during 5 months from March, 2007 to July, 2007. The instrument(LD-3B, SIBATA Company)was used to examine $PM_{10}$, by a light scattering method and a light transmission method. The instrument(Airboxx(KD Engineering) was used to examine $CO_2$. The instrument(Z300XP(Environmental sensor)was used to examine HCHO. The result indicated that the $PM_{10}$ average concentrations of the surveyed classrooms were $49{\mu}g/m^3$ in Spring and $59{\mu}g/m^3$ in Summer. The $CO_2$ average concentration of the surveyed schools were 576 ppm in the classroom and 527 ppm in the stateroom. The average concentration of TBC were $729CFU/m^3$ in an elementary school, $401CFU/m^3$ in a middle school, $381CFU/m^3$ in a high school. The HCHO average concentration of the surveyed schools were 0.03 ppm in the classroom, 0.02 ppm in the stateroom.

Chemical Changes of Fruit-Vegetable Juice during Mixed Culture Fermentation of Lactic Acid Bacteria Isolated from Kimchi and Yeast (김치 젖산균과 효모의 혼합배양 방법에 의한 과채류즙 발효과정중의 주요 성분변화)

  • Lactic acid bacteria KL 1, KD 6, KL 4 strains isolated from kimchi, or obtained Lactobacillus acidophilus, Lactobacillus plantarum, Leuconostoc mesenteroides with and without yeast(Saccharomyces cerevisiae) were inoculated in fruit vegetable juice for mixed culture fermentation 3 days at 3$0^{\circ}C$, and then their chemical changes were studied during fermentation. The amount of organic acid produced by the mixed culture fermentation of KL 1 and yeast was 0.82%(3 days) or 0.58%(1 day) and with the final pH of 3.3(3 days) or 4.2(1 day). These mixed culture systems of isolated strains or other bacterial strains had almost similar results of growth rate and acid production. The contents of vitamin C and carotene were retained and stabilized as 70~80% level of their initial values after 24 hrs fermentation. And also ethanol was produced as of the range in 9.6mg%(W/V) by the mixed culture fermentation of KL 1 and yeast, however, the content of ethanol in single culture fermentation by KL 1 strain was much lower than that of mixed culture. The major components of organic acids in fermented juice by mixed culture were considered as malic(26.0%), lactic(49.9%), succinic and citric acid, whereas these of unfermented juice were malic(53.2%), citric and other acids. On other hand, reducing sugar was decreased from 18.3mg/ml in fresh juice to about 12mg/ml in juice by mixed culture fermentation. Concentrations of fructose, glucose and sucrose were also greatly reduced in fermented juice.

Characteristics of ustilago maydis virus of SH14 killer strain isolated in Korea

  • SH-14, a novel killer strain of Ustilago maydis was isolated in Korea. It has been reported in other papers that the toxin specificity and double-stranded RNA pattern of SH-14 strain were different from other laboratory strains. In this paper, we analyzed the biochemical characteristics of U. maydis SH-14 virus. Three distinctive peaks were isolated from CsCl density gradient, designated as top (T), intermediate (I) and bottom (B) components. We found that the densities of each components, 1.285, 1.408 g/cm$\^$3/, respectively, are very similar to those of other strains. As previously reported by the analysis of dsRNA in each component, the dsRNA segments are separately encapsidated. Capsid protein of SH-14 virus consists of two proteins about 70 Kd shown by SDS-PAGE analysis. Electron microscopic examination of the virus particles revealed that UmV particles are very similar in size and morphology to all isolates as well as all lab-strains. In order to test immunological cross reactivity of UmV, werstern bolt analysis was carriedout with antiserum against A8 virus. All capsid protein had positive reaction against A8 antibody which indicated that UmV are immunologically cross-reactive with all isolates from Korea. The results presented in this paper may show that UmV isolated from SH-14 strain has very similar biochemical characteristics to those of other UmV. However, the difference in the toxin specificity and the molecular weight of toxin protein from the SH-14 strain has us to conclude that U. maydis SH-14 strain is a new killer type.

Purification of Mitochondrial Matrix Aldehyde Dehydrogenase from Pig Brain

  • The activity of aldehyde dehydrogenase (ALDH) in the cerebrum, cerebellum, striatum, and medulla oblongata was examined and mitochondrial matrix ALDH was purified prior to immunohistochemical study on the localization of ALDH isozymes in pig brain. Relatively high enzyme activity was found in the striatum and medulla oblongata when using indole-3-acetaldehyde as substrate, and in the striatum when using 3,4-dihydroxyphenylacetaldehyde (DOPAL). The main part of mitochondrial ALDH activities with both acetaldehyde and DOPAL existed in the matrix fraction. The ratio of activity of the matrix to the membrane fraction in the cerebrum was higher than in the cerebellum, suggesting that the distribution pattern of ALDH isozymes was different according to the brain regions. The 276-fold purified mitochondrial matrix ALDH from pig brain was identified to be homologous tetramers with 53 KD subunits. The enzyme showed maximal activity at pH 9.0 and was stable in the temperature range from $25^{\circ}C$ to $37^{\circ}C$. The mitochondrial matrix ALDH activity was considerably inhibited by acetaldehyde in vitro. The $K_m$ values of the enzyme for acetaldehyde and propionaldehyde were 5.8 mM and 4.9 mM, respectively, whereas $K_m$ values for indole-3-acetaldehyde and DOPAL were 44 ${\mu}M$ and 1.6 ${\mu}M$, respectively. The $V_{max}/K_{m}$ ratio was the highest with DOPAL as compared with other substrates. These results suggested that mitochondrial matrix ALDH in the present work might be a low Km isozyme involved in biogenic aldehyde oxidation in pig brain.

  • The purpose of the present study was to determine the in vivo effect of oxytocin antagonist-I(Al) on uterine oxytocin receptor number (Rn) and/or binding affinity (Kd) in the estrous rat. Anesthetized rats were given a bolus infusion of control or 5${\mu}\textrm{g}$ of AI and sacri-ficed 0.5 and 4 hours later. The uterine tissue was removed, trimmed and frozen. Membrane oxytocin receptors were isolated after homogenization of uterine tissue and differential ultracentrifugation. The oxytocin receptor assay was performed by saturation with cold oxytocin competion with a high specific activity oxytocin antagonist. Rn and Kds were determined by nonlinear curve fitting methods. No differences(p>0.05) between the AI and control treated animals in either oxytocin receptor number or binding affinity was detected in this study. These data suggest that the major mode of action of AI is via competitive inhibition at the uterine oxytocin receptor and not by altering receptor number or binding affinity.

Behaviors of the Fungicide Procymidone in Soils (살균제 Procymidone의 토양 중 동태)

  • This study was focused on adsorption, leaching, photolysis, and hydrolysis of the fungicide procymidone in soils. Adsorption type of procymidone on three different soil were well fitted to Linear and Freundlich isotherm. Distribution coefficients (Kd) were ranged from 2.75 to 12.18 and Freundlich isotherm Kf value $1.99{\sim}9.98$, 1/n value $0.74{\sim}0.89$. Desorption rates were $20.1{\sim}34.0%$ (Namgye), $26.3{\sim}44.6%$ (Jigog) and $31.6{\sim}50.9%$ (Baegsan series) and desorption hysteresis were $0.65{\sim}0.79,\;0.55{\sim}0.73\;and\;0.49{\sim}0.68$. Procymidone seemed to be stable to photolysis in acidic and neutral solutions but hydrolyzed rapidly in alkaline solution. Considering leaching properties procymidone mobility low in soils.

  • For developing high performance fuel cell, peripheral devices and key components have to be studied in priority. In this study, centrifugal pump was studied for heat recovery. For PEM fuel cell system, a four-impeller centrifugal pump was designed, tested and compared with result of commercial product (IWAKI). In addition, effects of number of impeller were analyzed by CFD. The experiment and analysis were progressed in the same conditions. The results showed the quantitative difference under 30% between the numerical and the experimental pressure difference and mass flow rate.
