Xanthomonns curnpestris M12, Xunthornonas sp. M28, Acinetuhucter Iwofz GI, and Klebsiella pneumoniae L25, Pseudomonas rnaltophiliu N246 were screened to increase the ability for crude oil utilization. All of these could utilize hexadecane and octane with the exception of N246 strain for only octane biodegradation. Thus N246, M12, and M28, strains were specially examined for octane oxidation. Octane biodegradation by three strains showed the optimal conditions at $30^{\circ}C$, pH 7.0~9.0, and 0.2~0.3% octane concentration as a substrate. It was found that P. multofihila N246 and X. curnpestns M12 had plasmid and the cured plasmid from N246 strain lost octane uitilization. Therefore, it was confirmed that certain genes for octane utilization were Iocated on OCT plasmid in N246 strain. The size of OCT plasmid in N246 strain was 118 kb. The N246 strain was resistant to ampicillin.

Purpose Phenylketonuria is an inborn error of metabolism, which is inherited as an autosomal recessive trait. PKU is resulting from deficiency of phenylalanine hydroxylase. PAH gene spans about 90 kb on chromosome 12q and comprises 13 exons. In order to define the genetic basis of PKU and the frequencies and distribution of PAH mutations in the Korean population, we analyzed PAH gene in independent 80 patients with PKU. Methods All 13 exons including exon-intron boundaries and 2 kb of 5' upstream region of the PAH gene were analyzed by PCR-direct sequencing methods. Results PAH gene analysis revealed 39 different mutations including 10 novel mutations. The novel mutations consisted of 9 missense mutations (P69S, G103S, N207D, T278S, P281A, L293M, G332V, S391I and A447P) and a novel splice site variant (IVS10-3C>G). R243Q, IVS4-1G>A, and E6-96A>G were the most relevant mutations and they accounted in the whole for 38% of the mutant alleles identified in this study. We also observed that. $BH_4$ responsibility was. associated with genotype of R241C, R53H and R408Q. Conc1ustion Our present study with 80 participants extends the previous results to more comprehensive understanding of PAH allele distribution and frequency in Koreans. Although Korean mutation profile of PAH is similar to those of the nearest oriental populations (Japanese, Chinese, and Taiwanese), several different characteristic features are revealed. The characterization of the genotype-phenotype relationship was also performed. Our data would be very useful information for diagnosis, genetic counseling and planning of dietary and therapeutic strategies in Korean PAH patients.
The mitochondrial DNA (mtDNA) restriction fragment length polymorphi는 (RFLPs) of five populations were analyzed to delineate the stocks of Penaeus chinensis (Osbeck) in the Yellow Sea. Comparison of P. chinensis with P. japonicus to clarify the nucleotide divergence between two species was also carried out. Based on the fragment patterns, three composite haplotypes were analyzed in P. chinensis mtDNA as four naplotypes were in P. japonicus. Most individuals of each P. chinensis population are shared by one dominant haplotype. Another two haplotypes haying variations at the C/a I and hull sites were also distributed evenly in the Korean and Chinese populations. It is suggested that the gene exchange occurring between populations in the Yellow Sea is frequent. Average length of the mtDNA molecule was estimated to be about 16.44 kb in P. chinensis and 16.31 kb in P. japonicus, Sequence divergence (p) of mtDNA between two species estimated by using Upholt's (1977) fomula was $13.7\%$.
A selective agar medium was developed and tested for the isolation of Acidovorax avenae subsp. avenae, the causal bacterial pathogen of bacterial brown stripe, from rice seeds. The new selective agar medium, designated sorbitol pyroglutamic acid agar (SPA) medium, contained 0.5 g of $K_2$HPO$_4$, 3.0 g of Na$_2$HPO$_4$, 2.0 g of D-sorbitol, 0.2 g of L-pyroglutamic acid, 10.0 $m\ell$ of tween 80, 40.0 mg of victoria blue B, 15.0 g of agar, 150.0 mg of ampicillin and 25.0 mg of vancomycin per litter. Colonies of A. avenae subsp. avenae on SPA medium were smooth, round, convex, shiny, blue and 1.5-2.0 mm in diameter 4 days after incubation at 28$^{\circ}C$. Blue colored colony having dark blue zone was typical type of A. avenae subsp. avenae colonies on the medium. Mean recovery of 8 isolates of A. avenae subsp. avenae on the selective SPA medium was 95.8% in comparison to that on KB medium. The saprophytic bacteria were reduced to 97.9% on SPA medium compared to those on KB medium. Most of other rice seedborne bacteria as well as reported pathogenic bacteria were failed to grow on SPA medium. This medium was highly selective for recovering A. avenae subsp. avenae from rice seed samples, and it could be used to enhance the recovery of this bacterium from rice seed samples, which may be contaminated with large numbers of competing microorganisms.
Foot-and-mouth disease (FMD) is a highly contagious disease of mammals and has a great potential for causing severe economic loss in susceptible cloven-hoofed animals, such as cattle, pigs, sheep, goats and buffalo. FMDV, a member of the Aphthovirus genus in the Picornaviridae family, is a non-enveloped icosahedral virus that contains a positive sense RNA of about 8.2 kb in size. The genome carries one open reading frame consisting of 3 regions: capsid protein coding region P1, replication related protein coding region P2, and RNA-dependent RNA polymerase coding region P3. FMDV infects pharynx epithelial cell in the respiratory tract and viral replication is active in lung epithelial cell. Morbidity is extremely high. A FMD outbreak in Korea in 2002 caused severe economic loss. Although intense research is undergoing to develop appropriate drugs to treat FMDV infection, there is no specific therapeutic for controlling FMDV infection. Moreover, there is an increasing demand for the development of vaccine strategies against FMDV infection in many countries. In this report, more effective prevention strategies against FMDV infection were reviewed.
The Samkwang mine is one of gold deposits distributed mainly in the southwestern province (Chungnam) of the Korean peninsula. Golds occur in quartz veins aged of $223{\pm}4MA$ according to K-Ar age dating from muscovite in a pegmatitic quartz vein. Quantz veins intrude Precambrian biotite-granite gneiss and mica schist of unknown age. Fluid inclusions in the quartz show a range of homogenization temperatures from 159 to $274^{\circ}C$. A calculated temperature from the isotopes of the galena-sphalerite pair is $375^{\circ}C$. Two phases-fluid inclusions homogenized either by liquid or vapor phase are frequently observed in specimens over $260^{\circ}C$, which may indicate the boiling of the fluids. Pressure of formation of the quartz veins inferred by the homogenization temperatures of liquid-$CO_2$ bearing fluid is 1kb. Based on these data, it is assumed that the temperature of the formation of the Samkwang mine may lie in between $350^{\circ}-230^{\circ}C$. ${\delta}^{34}S_{{\Sigma}s}$ values of sulphide minerals show narrow range of +2.1 to +4.6, and show a trend of enrichments of $^{34}S$ in the fluid from deep to the surface. ${\delta}^{34}S_{{\Sigma}s}$ in the fluid estimated is less than 3 permil, suggesting sulphur fluid originated from the magma.
C. elegans DLK-1 has been reported to play an important role in synaptogenesis by shaping the structure of presynaptic terminal. In this study, we investigated the expression pattern and regulation of the dlk-1 gene in C. elegans. To determine the expression pattern, we made a dlk-1::gfp fusion construct, named pPDdg1, which consisted of -2.2 kb 5' upstream region, the first exon, the first intron, and a part of the second exon of the dlk-1 gene. By microinjecting this construct into the worm, we observed that the DLK-1::GFP was expressed mainly in neurons. We next examined the regulatory elements of gene expression by deletion analysis of pPDdg1. Removal of a large portion of the 5' upstream region (${\Delta}-361$ to -2246) of the gene had little effect on the expression pattern, whereas deletion of the first intron led to elimination of the DLK-1::GFP expression in most of the neurons. Our results suggest that the first intron of the C. elegans dlk-1 gene contains the regulatory element critical for gene expression.
Rat liver LDH A-cDNA has been isolated from a .lambda.gt11-rat lover cDNA library and partially characterized. The size of the isolated rat liver LDH A-cDNA if shown to be 1.6Kb and restriction enzyme sites for the rat liver LDH A-cDNA are also mapped. 682-nucleotide sequence coding for 3'-end of rat liver LDH A-cDNA has been analyzed and compared to the nucleotide sequence of the same region of rat $C_6$-glioma cell LDH A-cDNA which has been cloned from the hormonally stimulated cell cultures. The result shows that 177 nucleotide sequences coding for the C-terminal 59-amino acids are identical but 505 nucleotide sequences of 3'-nontranslated region of the two LSH A-cDNA exhibit characteristic differences in thier nucleotide sequences. Computer analysis for the folding structures for 3'-end 400 nucleotide sequences of the two LDH A-cDNA shows a possibility implying that the two LDH A-mRNAs isolated from different tissues of rats may have different half life and therefore their translational efficiency may be different. It has been previously demonstrated that isoproterenol stimulated rat $C_6$ -glioma cell cultures produce LDH A-mRNA showing 2 to 3-fold longer half life in comparison to that of noninduced LHD A-mRNA. The result therefore support for the idea that hormonally stimulated rat $C_6$-glioma cells may produce LDH A-mRNA containing different nucleotide sequences at the 3'-end nontranslated region by which the hormonally induced LDH A-mRNA could have more stable secondary mRAN structure in comparison to that of noninduced LDH A-mRNA.
Contrasts in the style of the gold-silver mineralization in geologic and tectonic settings in Korea, together with radiometric age data, reflect the genetically different nature of hydrothermal activities, coinciding with the emplacement age and depth of Mesozoic magmatic activities. It represents a clear distinction between the plutonic settings of the Jurassic Daebo orogeny and the subvolcanic environments of the Cretaceous Bulgugsa igneous activities. During the Daebo igneous activities (about 200-130 Ma) coincident with orogenic time, gold mineralization took place between 197 and 127 Ma. The Jurassic deposits commonly show several characteristics: prominent association with pegmatites, low Ag/Au ratios in the ore-concentrating parts, massive vein morphology and a distinctively simple mineralogy including Fe-rich sphalerite, galena, chalcopyrite, Au-rich eIectrum. pyrrhotite and/or pyrite. During the Bulgugsa igneous activities (120-60 Ma), the precious-metal deposits are generally characterized by such features as complex vein morphology, medium to high AgiAu ratios in the ore concentrates, and abundance of ore minerals including base-metal sulfides, Ag sulfides, native silver, Ag sulfosalts and Ag tellurides. Vein morphology, mineralogical, fluid inclusion and stable isotope results indicate the diverse genetic natures of hydrothermal systems. The Jurassic Au-dominant deposits were formed at the relatively high temperature (about 300 to 450$^{\circ}$C) and deep-crustal level (>3.0 kb) from the hydrothermal fluids containing more amounts of magmatic waters (3180; 5-10 %0). It can be explained by the dominant ore-depositing mechanisms as CO2 boiling and sulfidation, suggestive of hypo/mesothermal environments. In contrast, mineralization of the Cretaceous Au-Ag type (108-71 Ma) and Agdominant type (98-71 Ma) occurred at relatively low temperature (about 200 to 350$^{\circ}$C) and shallow-crustal level «1.0 kb) from the ore-fonning fluids containing more amounts of less-evolved meteoric waters (15180; -10-5%0). These characteristics of the Cretaceous precious-metal deposits can be attributed to the complexities in the ore-precipitating mechanisms (mixing, boiling, cooling), suggestive of epilmesothermal environments. Therefore, the differences of the emplacement depth between the Daebo and the Bulgugsa igneous activities directly influence the unique temporal and spatial association of the deposit type.
A kanamycin resistance($Km^r$) gene was studied for its transfer in natural freshwater environments by using the natural bacterial isolate(M1) of DK1 and the DKC601 strain, $Km^r$ plasmid of which was genetically engineered from the NI strain. The transfer frequency ofthe $Km^r$ gene and rearrangement of the $Km^r$ plasmid were compared between the gnetically engineered microorganism(GEM) and the NI parental strain by conjugation with the same recipient strain. The transfer frequency of the $Km^r$ gene was about $9.1\times 10^{-12}-1.8\times 10^{-11}$ in both the GEM and NI strains at 5 to $10^{\circ}C$, but the frequency of the NI was about 10 times higher than that of the GEM at 20 to $30^{\circ}C$. The $Km^r$ plasmid in the transconjugants obtained by conjugation of the NI with the MY1 strain as a ricipient showed alot of rearrangement, but the $Km^r$ plasmid transferred from the GEM was stable without alteration of its size. When the MT2 strain was used as a recipient, however, such a rearrangement of the $Km^r$ plamid was observed in the transconjugants obtained from the GEM as well as the NI strain. In those transconjugants obtained from different mating pairs and water environments, the plasmid were appeared to decrease in their number as the period of conjugation time was prolonged, but only the $Km^r$ plasmid transferred from the GEM kept having its size of 52kb. Therefore, the $Km^r$ gene was transferred at the same rate from the GEM and NI strains in natural freshwater environment, but the gene of the GEM strain was more stable than the NIduring conjugation and the $Km^r$ plasmid was rearranged by changing the recipient strain for conjugation in any water environments.
